Flecainide acetate is a Class 1c anti-arrhythmic with a potent sodium voltage gated channel blockade which is utilized for the second-line treatment of tachyarrhythmias in children and adults. Given its narrow therapeutic index, the individualization of drug therapy is of utmost importance for clinicians. Despite efforts to improve anti-arrhythmic drug therapy, there remain knowledge gaps regarding the impact of variation in the genes relevant to flecainide's disposition and response. This variability is compounded in developing children whose drug disposition and response pathways may remain immature. The average flood event in the United States costs Americans $4.7 billion. Flood events have become more frequent, widespread, and of longer duration due to climate change. The United States attempts to combat the emerging threat of flooding through federal and private insurance programs. Still, as flooding risk rises, property owners are burdened by the responsibility to assess and prepare for these risks independently since standard homeowners’ and renters’ insurance does not cover flood damage under any circumstances in the United States. Multiple paternity is an ecological phenomenon that plays an important role in reproductive dynamics and population genetics; the presence and rate of multiple paternity can shed light on reproductive behavior and the potential genetic effects of human management. Previous methods have estimated multiple paternity using genetic markers such as microsatellites, and require DNA from both parents and their offspring. However, evolving genomic technology promotes use of alternative genomic methods and acquiring paternal DNA from free-ranging animals is often difficult or impossible. We propose kinship values derived from single-nucleotide polymorphisms (SNPs) as a tool to predict multiple paternity. Calculating kinship values …

Epoxyeicosatrienoic acids (EETs) are cytochrome P450-epoxygenase-derived metabolites of arachidonic acid that act as endogenous signaling molecules in multiple biological systems, including their controversial effects on pain, including reports of the central analgesic effect and its action in inducing pain. The aim of this study was to verify the peripheral antinociceptive effect of EETs and the effect of the selective EET receptor antagonist, 14,15-EEZE, on this antinociception. The nociceptive threshold was determined by paw pressure withdrawal, and hyperalgesia was induced by intraplantar injection of PGE2 to evaluate the effect of EETs administration. Decoding cellular state transitions is crucial for understanding complex biological processes in development and disease. While recent advancements in single-cell RNA sequencing (scRNA-seq) offer insights into cellular trajectories, existing tools primarily study expressional rather than regulatory state shifts. We present CellTran, a statistical approach utilizing paired-gene expression correlations to detect transition cells from scRNA-seq data without explicitly resolving gene regulatory networks. Applying our approach to various contexts, including tissue regeneration, embryonic development, preinvasive lesions, and humoral responses post-vaccination, reveals transition cells and their distinct gene expression profiles. Short-Term Territoriality: The Influence Of Time And Gender On Territorial Behavior, Audra Kooi, Celia Vander Woude, Elijah Vander Woude

Territoriality, the behavior of claiming and defending space or objects, has been widely studied in both animal and human contexts, but most research focuses on long-term patterns. Forty-seven undergraduate students were instructed to sit and watch videos of varying lengths. After leaving the room, participants were instructed to sit again, and their seat choices were recorded to assess territorial behavior, measured by displacement. Results showed a strong preference for returning to the same seat, which suggests territorial behavior is not exclusively established in the long term but can emerge within a few minutes.

Cerebellar nuclei (CN) neurons serve as the primary output of the cerebellum and originate from the cerebellar primordium at early stages of cerebellar development. These neurons are diverse, integrating information from the cerebellar cortex and relaying it to various brain regions. Employing various methodologies, we have characterized a specific subset of CN neurons that do not originate from the rhombic lip or ventricular zone of the cerebellar primordium. Embryos were collected at early stages of development and processed for immunohistochemistry (IHC), western blotting, in situ hybridization (ISH), embryonic culture, DiI labeling, and flow cytometry analysis (FCM). Introduction: Pneumonia and anemia are prevalent medical conditions with significant implications on patient health, resulting in numerous hospitalizations and deaths. Various studies have been done on the mortality rates of pneumonia and anemia individually. However, fewer describe the mortality of patients diagnosed with pneumonia superimposed on anemia.
Methods: This retrospective study used data from electronic medical records obtained from rural Midwestern Missouri, including 9,879 patients who were admitted with either anemia or pneumonia. The primary outcome was in-hospital mortality.
Results: The study found that patients with pneumonia had a higher mortality rate of 25.8% when diagnosed with comorbid anemia compared …

The scaffolding function of receptor interacting protein kinase 1 (RIPK1) confers intrinsic and extrinsic resistance to immune checkpoint blockades (ICBs) and emerges as a promising target for improving cancer immunotherapies. To address the challenge posed by a poorly defined binding pocket within the intermediate domain of RIPK1, here we harness proteolysis targeting chimera (PROTAC) technology to develop a RIPK1 degrader, LD4172. LD4172 exhibits potent and selective RIPK1 degradation both in vitro and in vivo. Degradation of RIPK1 by LD4172 triggers immunogenic cell death, enhances tumor-infiltrating lymphocyte responses, and sensitizes tumors to anti-PD1 therapy in female C57BL/6J mice. We present a patient with human immunodeficiency virus who developed multiple painful lesions that previously in the past had speciated as Cryptococcus neoformans cutaneously, and in the lung. Despite induction therapy for presumed re-infection, the patient did not improve so a biopsy was performed and this was speciated as Mycobacterium avium complex, with final diagnosis being disseminated Mycobacterium avium complex. This case highlights the importance of considering a broad differential diagnosis for any new lesions regardless of prior culture data.

Cancers frequently co-opt lineage-specific transcription factors (TF) utilized in normal development to sustain proliferation. However, the effects of these TFs on tumor development depend considerably on where in the genome they bind. A new article by Taylor and colleagues expands on previously developed diamidine compounds that obstruct the DNA binding sites of the pioneer TF PU.1 (SPI1) in acute myeloid leukemia. Immobilization and sequencing of genomic DNA targeted by these compounds revealed that these inhibitors alter the genomic binding patterns of PU.1. The authors report that their strategy constrains the genomic binding preferences of PU.1, leading to redistribution of PU.1 …

The paper aims to identify the streams and influences of protest drama in the rich theatrical productions during Martial Law 1972-1986. The focus is on the different perspectives and commitment that shaped the struggles in form and content in the plays staged by selected groups in a number of provinces in the country. The primary staging ground for discussion is the politics of the spectacle which will be traced through the different dynamics of creating a play from the staged text through the process of producing a play.

Lopez’s position as a secular critic by analyzing his milieu during the Commonwealth period, criticism of his collection Literature and Society, and the content of essays from the same collection in light of the debate between “art for art’s sake” and proletarian literature. Espiritu writes a book which may be described as a “collective intellectual biography” of five figures in Philippine letters whose lives and texts were influenced by their expatriation in America: Carlos P. Romulo, Carlos Bulosan, Jose Garcia Villa, N.V.M. Gonzalez, and Bienvenido Santos. With a biographer’s touch, Espiritu identifies the themes that are more or less common and played out differently among them: performativity, ambivalence, nationalism in their self-representation, cultural hybridity, and patronage relations—leading to what may be called a genuine “transnational” perspective for Asian American intellectual history. Through a consideration of narrativity, the paper explores different structures of recognition in the corrido and 1941 filmic versions of the Ibong Adarna. The paper compares the chapbook’s context and narrative techniques with that of the film’s capacity to address an audience and creating subject-forms. It explores the applicability of James Siegel’s concept of the lingua franca’s communicative power to the Philippine situation, and suggests that different textual technologies contain different though overlapping grammars that affect their ideological functions.

What do we make of the abiding significance of state-sanctioned human captivity and imprisonment on a massive, unprecedented scale as a primary American modality of civilization? Democracy? Self-fidelity is a human ethical task. But how can we be faithful to a self that is both individual and social? This paper takes up this question by comparing the basic ethical frameworks of Nikolai Berdyaev and Watsuji Tetsurō. In the first section of this paper, Berdyaev’s notion of personality and his three forms of ethics (ethics of law, ethics of redemption, and ethics of creativeness) are discussed. In telling the story of PETA, or the Philippine Educational Theater Association, this article reflects on the distinct characteristics that PETA plays have acquired in the more than 40 years it has served as the people’s theater—from the martial law years to the present, for the continuous expression of Filipino culture and heritage. The article shows the variety of PETA plays and the people behind their collaborative production, and ends with the present challenges of PETA as it reaches out to a younger generation of audience.

This article will discuss the production design approaches and techniques used in plays by the Philippine Educational Theater Association (PETA) from 1973-1986, using the principles of “aesthetics of poverty.” The “aesthetics of poverty” came about in consideration of the poverty situation of the Philippines and is really an attitude more than anything else, appropriate for the social conditions of Philippine society. This was also the period when there was very limited budget for productions, the result of which was creating with one’s imagination and clear imaging for meanings and symbolic representation.
