BACKGROUND: Hybrids are expected to show greater phenotypic variation than their parental species, yet how hybrid phenotype expression varies with genetic distances in closely-related parental species remains surprisingly understudied. Here, we investigate pelage and morphometric trait variation in anthropogenic hybrids between four species of Brazilian Callithrix marmosets, a relatively recent primate radiation. Marmoset species are distinguishable by pelage phenotype and morphological specializations for eating tree exudates. In this work, we (1) describe qualitative phenotypic pelage differences between parental species and hybrids; (2) test whether significant quantitative differences exist between parental and hybrid morphometric phenotypes; and (3) determine which hybrid morphometic …

Ibogaine and its main metabolite noribogaine provide important molecular prototypes for markedly different treatment of substance use disorders and co-morbid mental health illnesses. However, these compounds present a cardiac safety risk and a highly complex molecular mechanism. We introduce a class of iboga alkaloids - termed oxa-iboga - defined as benzofuran-containing iboga analogs and created via structural editing of the iboga skeleton. The oxa-iboga compounds lack the proarrhythmic adverse effects of ibogaine and noribogaine in primary human cardiomyocytes and show superior efficacy in animal models of opioid use disorder in male rats. They act as potent kappa opioid receptor agonists …

Calcium imaging has become a popular way to probe astrocyte activity, but few techniques holistically capture discrete calcium signals occurring across the astrocyte domain. Here, we introduce STARDUST, a pipeline for the spatio-temporal analysis of regional dynamics and unbiased sorting of transients from fluorescence recordings of astrocytes. We describe steps for installing software, detecting active pixel patches, obtaining region of activity (ROA) maps, and extracting time series from ROAs. We then detail procedures for extracting signal features using custom-made code.

Over the past three years, new SARS-CoV-2 variants have continuously emerged, evolving to a point where an immune response against the original vaccine no longer provided optimal protection against these new strains. During this time, high-throughput neutralization assays based on pseudoviruses have become a valuable tool for assessing the efficacy of new vaccines, screening updated vaccine candidates against emerging variants, and testing the efficacy of new therapeutics such as monoclonal antibodies. Lentiviral vectors derived from HIV-1 are popular for developing pseudo and chimeric viruses due to their ease of use, stability, and long-term transgene expression. However, the HIV-based platform has …

Cancer cachexia mouse models are needed to recapitulate the clinical features of patients with cachexia. Here, we present a protocol for the establishment and evaluation of cancer cachexia mouse models. We delineate the steps in preparing tumor cells for inoculation and surgical procedures. After the establishment of these mouse models, we describe essential techniques to assess cancer cachexia, including grip strength evaluation, tissue collection, and the calculation of cross-sectional areas of muscle tissue. For complete details on the use and execution of this protocol, please refer to Liu et al.,

Spinal cord injury (SCI) triggers microglial/monocytes activation with distinct pro-inflammatory or inflammation-resolving phenotypes, which potentiate tissue damage or facilitate functional repair, respectively. The major integrin Mac-1 (CD11b/CD18), a heterodimer consisting of CD11b and CD18 chains, is expressed in multiple immune cells of the myeloid lineage. Here, we examined the effects of CD11b gene ablation in neuroinflammation and functional outcomes after SCI. qPCR analysis of C57BL/6 female mice showed upregulation of

Cancer patients often experience changes in mental health, prompting an exploration into whether nerves infiltrating tumors contribute to these alterations by impacting brain functions. Using a mouse model for head and neck cancer and neuronal tracing, we show that tumor-infiltrating nerves connect to distinct brain areas. The activation of this neuronal circuitry altered behaviors (decreased nest-building, increased latency to eat a cookie, and reduced wheel running). Tumor-infiltrating nociceptor neurons exhibited heightened calcium activity and brain regions receiving these neural projections showed elevated Fos as well as increased calcium responses compared to non-tumor-bearing counterparts. The genetic elimination of nociceptor neurons decreased …

Variable-rate delivery of intravenous drugs is difficult to achieve with a tethered infusion system in a freely moving animal. Here, we present a protocol for continuous intravenous delivery of oxytocin in pregnant rats and mice. We describe steps for using an implantable, preprogrammed, microprocessor-controlled infusion pump connected to the jugular vein to induce labor. This protocol can be adapted to a variety of experimental paradigms in non-pregnant animals where precise intravenous pharmacological manipulation is desired. For complete details on the use and execution of this protocol, please refer to Giri et al.
Long-Term Clinical Outcome Of Corneal Endothelial Degeneration Managed With Keratoleptynsis In A Chihuahua Dog, Manbok Jeong
Long-Term Clinical Outcome Of Corneal Endothelial Degeneration Managed With Keratoleptynsis In A Chihuahua Dog, Manbok Jeong
The Thai Journal of Veterinary Medicine
A 9-year-old spayed female Chihuahua presented with a 2-month history of progressive corneal edema in the left eye. Slit lamp biomicroscopy showed moderate corneal edema, instability of the axial corneal epithelial surface, and an anterior cortical opacity of the lens. Based on these results, a diagnosis of canine endothelial degeneration was made with the incidental finding of an incipient cataract in the left eye. Keratoleptynsis successfully managed the corneal edema for 28 months after surgery without corneal ulceration. This case study underscores the need for an early surgical approach using keratoleptynsis for dogs with a progressive nature of canine endothelial …

The Thai Journal of Veterinary Medicine
Osteoporosis is a systemic bone disorder characterized by decreased bone density, which can weaken bone strength and increase the risk of fractures. Osteogenesis imperfecta (OI), one of the main causes of osteoporosis in children, results in fragile bones and multiple fractures. The present study documented the case of a dog with an undetermined osteoporotic disease managed with intramedullary rod fixation. A 3-month-old intact male Alaskan Malamute dog, weighing 11.6 kg, was presented with lameness in the left pelvic limb. Radiography revealed left pelvic and femoral fractures, and the cortex of the long bones was thin. On CT scanning, a significant …

Synchronous Tri-Cavitary Biphasic Malignant Mesothelioma In A Dog Presenting With Dyspnea, Nawarus Prapaiwan, Tawewan Issarankura Na Ayudhaya, Poonavit Pichayapaiboon, Onrampha Tanglakmankhong
The Thai Journal of Veterinary Medicine
Malignant mesothelioma is an uncommon tumor that has a challenging diagnosis and a tendency for late diagnosis. A 7-year-old intact male dog presented with respiratory distress. Thoracic radiography revealed pleural effusion and bone reactions at multiple ribs. A computerized tomography scan with rib biopsy was suggested as a diagnosis plan. Unfortunately, the dog died. Gross pathological findings demonstrated multifocal to coalescing, pinkish-white firm masses scattered over the parietal pleural surface, extended to the serosa of the diaphragm and the pericardium, making them adhere to each other and causing the coalescence of a white irregular mass at the diaphragm. Multiple white …

Long-Term Tapering Regimen Use Of Oclacitinib For The Control Of Pruritus In An Atopic Dog, Piyarat Chansiripornchai
The Thai Journal of Veterinary Medicine
Abstract
Canine atopic dermatitis (CAD) is a genetically predisposed inflammatory skin disease with a global distribution, yet it is incurable. A key clinical sign of CAD is pruritus. Oclacitinib has proven effective in managing pruritus in atopic dogs. This case report details the long-term administration of oclacitinib every other day for controlling pruritus in an atopic dog. An 8-year-old atopic Bangkaew dog presented with pruritus, alopecia, and recurrent Malassezia pachydermatis infection. To control the pruritus, the dog received oclacitinib at a dose of 0.4 mg/kg orally every 24 hours for 30 weeks. The pruritus visual analog scale (PVAS) decreased from …

Fatal Disseminated Toxoplasmosis In Captive Red Kangaroo (Macropus Rufus) And Parma Wallaby (Macropus Parma): Pathological And Immunohistochemical Investigations, Sawang Kesdangsakonwut
The Thai Journal of Veterinary Medicine
Abstract
Toxoplasmosis is a protozoan zoonosis caused by Toxoplasma gondii. Marsupials are considered a susceptible intermediate host. T. gondii is mainly transmitted to intermediate hosts via the fecal-oral route. A captive red kangaroo was diagnosed with disseminated toxoplasmosis based on clinicopathological and immunohistochemical (IHC) investigations. Two years later, a five-year-old Parma wallaby was kept in the same area as the red kangaroo and showed clinical signs of depression, withdrawal from the troop, and weakness. The wallaby died within two days after showing the clinical signs. Pathological findings in this wallaby were compatible with disseminated toxoplasmosis. Tachyzoites and tissue cysts …

Interzone/cavitation are key steps in early stage joint formation that have not been successfully developed in vitro. Further, current models of endochondral ossification, an important step in early bone formation, lack key morphology morphological structures such as microcavities found during development in vivo. This is possibly due to the lack of appropriate strategies for incorporating chemical and mechanical stimuli that are thought to be involved in joint development. We designed a bioreactor system and investigated the synergic effect of chemical stimuli (chondrogenesis-inducing [CIM] and hypertrophy-inducing medium [HIM]) and mechanical stimuli (flexion) on the growth of human mesenchymal stem cells (hMSCs) …

Designing A Novel Approach To Cadaveric Dissection And Evaluation Of The Lacrimal Gland Anatomy, Maresa Joanna Mcdowell
Doctoral Dissertations and Projects
During our study, the orbital dissections of 44 cadavers were analyzed, and the lacrimal gland's condition after dissection was evaluated. Medical students previously dissected twenty-two of the cadavers using the dissection steps outlined in Grant’s Dissector. From these, 21 student groups performed orbital dissection using the two approaches outlined in Grant’s Dissector. Post-evaluation of Grant’s anterior approach (GAA) showed that 50% of the lacrimal glands had signs of damage to the orbital lobe, and 88.89% of the palpebral lobe were damaged. When Grant’s supraorbital approach (GSA) was used, 70% of the palpebral lobes showed signs of damage compared to the …
