The recent identification of skull bone marrow as a reactive hematopoietic niche that can contribute to and direct leukocyte trafficking into the meninges and brain has transformed our view of this bone structure from a solid, protective casing to a living, dynamic tissue poised to modulate brain homeostasis and neuroinflammation. This emerging concept may be highly relevant to injuries that directly impact the skull such as in traumatic brain injury (TBI). Regular aerobic activity is associated with a reduced risk of chronic pain in humans and rodents. Our previous studies in rodents have shown that prior voluntary wheel running can normalize redox signaling at the site of peripheral nerve injury, attenuating subsequent neuropathic pain. However, the full extent of neuroprotection offered by voluntary wheel running after peripheral nerve injury is unknown. Here, we show that six weeks of voluntary wheel running prior to chronic constriction injury (CCI) reduced the terminal complement membrane attack complex (MAC) at the sciatic nerve injury site. Dent disease type 1 (Dent 1) is a rare X-linked genetic condition which impacts kidney function and is caused by pathogenic variants in CLCN5. Affected males typically develop low molecular weight proteinuria, hypercalciuria, nephrocalcinosis, nephrolithiasis, and other symptoms. Kidney failure often occurs between the third to fifth decade of life. Here, we report an 11-year-old boy with Dent 1 and a severe kidney disease phenotype. The patient presented with flank pain, nocturnal enuresis, foamy urine, and increased urinary frequency. He was found to have nephrotic-range proteinuria, without hypoalbuminemia, and a significantly decreased estimated glomerular filtration rate at presentation. Further, he …

Pancreatic cancer serves as the third leading cause of cancer-associated morbidity and mortality in the United States, with a 5-year survival rate of only 12% with an expected increase in incidence and mortality in the coming years. Pancreatic ductal adenocarcinomas constitute most pancreatic malignancies. Certain genetic syndromes, including Lynch syndrome, hereditary breast and ovarian cancer syndrome, hereditary pancreatitis, familial adenomatous polyposis, Peutz-Jeghers syndrome, familial pancreatic cancer mutation, and ataxia telangiectasia, confer a significantly higher risk. Screening for pancreatic malignancies currently targets patients with germline mutations or those with significant family history. Screening the general population is not currently viable owing …

Introduction: Longitudinal effect of diet-induced obesity on bone is uncertain. Prior work showed both no effect and a decrement in bone density or quality when obesity begins prior to skeletal maturity. We aimed to quantify long-term effects of obesity on bone and bone marrow adipose tissue (BMAT) in adulthood.
Methods: Skeletally mature, female C57BL/6 mice (n = 70) aged 12 weeks were randomly allocated to low-fat diet (LFD; 10% kcal fat; n = 30) or high-fat diet (HFD; 60% kcal fat; n = 30), with analyses at 12, 15, 18, and 24 weeks (n = 10/group). Tibial microarchitecture was analyzed …

Introduction/aims: Prior studies have emphasized the role of inflammation in the response to injury and muscle regeneration, but little emphasis has been placed on characterizing the relationship between innate inflammation, pain, and functional impairment. The aim of our study was to determine the contribution of innate immunity to prolonged pain following muscle contusion.
Methods: We developed a closed-impact mouse model of muscle contusion and a macrophage-targeted near-infrared fluorescent nanoemulsion. Closed-impact contusions were delivered to the lower left limb. Introduction: Nadofaragene firadenovec (Ad-IFNα/Syn3) is now approved for BCG-unresponsive bladder cancer (BLCA). IFNα is a pleiotropic cytokine that causes direct tumor cell killing via TRAIL-mediated apoptosis, angiogenesis inhibition, and activation of the innate and adaptive immune system. We established an immunocompetent murine BLCA model to study the effects of murine adenoviral IFNα (muAd-Ifnα) gene therapy on cancer cells and the tumor microenvironment using a novel murine equivalent of Nadofaragene firadenovec (muAd-Ifnα).

VMP-like sequence (vls) antigenic variation systems are present in every Lyme disease Borrelia strain with complete genome sequences. The linear plasmid-encoded vls system consists of a single expression site (vlsE) and contiguous array(s) of silent cassettes that have ~90% identity with the central cassette region of the cognate vlsE gene; antigenic variation occurs through random, segmental, and unidirectional recombination of vls silent cassette sequences into the vlsE expression site. Automated annotation programs do not accurately recognize vls silent cassette sequences, so these regions are not correctly annotated in most genomic sequences. In this study, the vls …

PURPOSE: To provide an automated system for synthesizing fluorescein angiography (FA) images from color fundus photographs for averting risks associated with fluorescein dye and extend its future application to spaceflight associated neuro-ocular syndrome (SANS) detection in spaceflight where resources are limited.
DESIGN: Development and validation of a novel conditional generative adversarial network (GAN) trained on limited amount of FA and color fundus images with diabetic retinopathy and control cases.

Acetyl CoA acetyltransferase 1 (ACAT1), a mitochondrial enzyme, is mainly involved in the formation and decomposition of ketones, isoleucine, and fatty acids. Previous clinical studies showed that mutations in the ACAT1 gene lead to ketoacidosis, Notably the role of ACAT1 in human cancer' pathogenesis varies depending on cancer type, and its specific role in gastric cancer remains largely unknown. In the current study, we found that the expression of ACAT1 in primary late-stage gastric cancer tumor tissues was significantly lower than in early-stage tumors. The sandy beach surf zone (SBSZ) represents an ecologically, economically, and culturally important marine habitat around the world. Despite its significance, the SBSZ has been historically underrepresented in marine and coastal ecology research. This in turn has caused many inshore marine species that prefer this habitat to be largely ignored. This article focuses on the ethnic history and religious traditions of the Khanty (formerly known as Ostiaks), a Finno-Ugric people dwelling in the north of Western Siberia and its responses to the global influences. With 40 other widely dispersed small peoples of the Russian North and Siberia, they are classified as aboriginal small-numbered people of the North and Siberia. The olfactory system of extant amphibians changes as the animal transitions from a fully aquatic to a terrestrial lifestyle at metamorphosis. Cellular morphology of the nose and expression patterns of olfactory genes in the nasal cavity have been examined for a variety of frogs and salamanders, but among plethodontid salamanders, molecular data are available only for Plethodon shermani. Using standard histology and micro-CT reconstruction, I investigated the structure of the olfactory organs of larvae, juveniles, and adults of six plethodontid species, with terrestrial, streamside, semiaquatic, and aquatic adults. This study was motivated by the need to develop a noninvasive and highly sensitive monitoring tool for determining local occupancy of an endangered aquatic species To make inference into the occupancy of fishes within small stream sites, we developed a method to determine whether environmental DNA (eDNA) concentrations from a target species were elevated relative to a tracer, formulated to mimic eDNA and introduced at a single location. This pilot project seeks to nurture space for Indigenous perspectives while equipping early childhood educators with tools for integrating diverse epistemology into outdoor learning practices to foster collaborative educational approaches. To align with this effort of shared knowledge, the project intends to offer a culturally sustaining educational tool that can help disrupt dominant approaches by evaluating alternative teaching strategies that center partnerships with local Indigenous communities to move toward equity-centered education. The bryozoan Watersipora subtorquata and its cryptic congener Watersipora ‘new species’ are now globally invasive species. Previous studies have found this species complex to have a high tolerance to copper-based antifouling paints, which represent a large portion of the mitigation effort toward preventing hull fouling by invasive invertebrates. Tolerance alone, however, is unlikely to explain the full extent of Watersipora’s massive and rapid worldwide invasion success. The symbiotic relationship between the chlamydomonad green alga Oophila amblystomatis and embryos of certain amphibian species is often presumed to be mutualistic. However, the existence of a mutualism has only been experimentally tested and established in two closely related ambystomatid salamanders. These experiments showed a positive correlation between intracapsular algal density and embryonic growth, survival, hatching synchrony, and hatchling body size. Oophila has been documented within egg capsules of a growing number of amphibian species, including several frogs in the family Ranidae. In a literary analysis of selected fiction by Philip K Dick through an ecocritical framework, the focus of this study reveals the consequences of ecological destruction on futuristic societies reflecting the Anthropocene. The hypothesis of adaptive breeding habitat selection proposes congruence between the evolved selection of habitats and fitness components such as reproductive success. However, mismatches between habitat selection and observed fitness outcomes are not unusual and can arise for multiple reasons. Variation in individual qualities of animals has also been found to affect reproduction and survival. I examined associations between barn owl nest box selection, reproductive output, and plumage characteristics previously shown to correspond to individual quality. I found that barn owls selected tall nest boxes made of wood, mounted high off the ground, and with increased proportions of grassland around …

Prescribed grazing management can have myriad effects on wildlife, with low to moderate grazing being associated with an increased abundance of some grassland birds. Specifically, Grasshopper Sparrow (Ammodramus savannarum) and Savannah Sparrow (Passerculus sandwichensis) abundance have been shown to be positively associated with both grazing and a higher percentage of native plant cover. However, measures of abundance provide an incomplete assessment of habitat quality for birds. Physiological and morphological measurements can help indicate environmental stress in individual birds, which reveals more information about habitat quality. BACKGROUND: Oxytocin (OXT) has been used as a post-treatment for post-traumatic stress disorder (PTSD). Previous research suggests that pre-treatment of intranasal OXT attenuates signs of fear in rats; however, the impact of intraperitoneal (i.p) OXT pre-treatment on signs of fear in rats is unknown. The Klamath River tends to experience impaired upstream water quality, which improves downstream. Although this consensus is accepted, an updated spatiotemporal analysis of certain water quality parameters has not been conducted since 2017. With four of the Klamath River’s dams slated for removal by the end of 2024, it is critical to have an updated assessment of current water quality conditions from which to compare conditions following dam removal. In this study, total nitrogen (TN), total phosphorus (TP), and microcystin data from 2010-2021 were analyzed to determine concentration trends from Link Dam to the Klamath River Estuary.

Currently, more than 700 valid species of Streptomyces have been identified worldwide. Streptomyces produce the majority of antibiotics, antivirals and antifungals, as well as important agricultural bioactive molecules due to their unique genetic capabilities.
