We created a generalizable pipeline for antibiotic-resistance-gene-free plasmid (ARGFP)-based cloning using a dual auxotrophic- and essential-gene-based selection strategy. We use auxotrophic selection to construct plasmids in engineered E. coli DH10B cloning strains and both auxotrophic- and essential-gene-based selection to (1) select for recombinant strains and (2) maintain a plasmid in E. coli Nissle 1917, a common chassis for engineered probiotic applications, and E. coli MG1655, the laboratory "wild-type" E. coli strain. We show that our approach has comparable efficiency to that of antibiotic-resistance-gene-based cloning. Subdural empyema is a collection of pus in the subdural space between the dura mater and the arachnoid. It carries very high morbidity and mortality as it can spread anywhere in the brain; however, the risk can be mitigated with appropriate surgical and medical intervention. Being protected by the skull, cranial infections are usually preceded by a significant risk factor, either an external invader such as skull fractures secondary to trauma, penetrating injury, prior surgery, or, more commonly, in more than 50% of cases, due to spread of an internal infection such as ear or sinus infections. Thrombocytopenia with concomitant anemia is a serious condition with a high mortality risk. Destruction of platelets, i.e., thrombocytopenia, can be secondary to either auto-antibodies (immune-mediated) or mechanical destruction (non-immune-mediated). The Coombs test is a widespread tool to differentiate between the two categories, resulting in different specific treatment approaches for each diagnosis. A peripheral blood smear can also help make the diagnosis; for instance, in cases of mechanical destruction such as thrombotic thrombocytopenic purpura (TTP), the red blood cell (RBC) shape looks fragmented, forming schistocytes. Abstract Background Many people with dementia are reliant on family caregivers to provide daily care to maintain quality of life and dignity. As a result, caregivers can experience increased burden, poorer health outcomes and increased stigma. During the last decade jacket-frames have emerged as the main kind of substructure for bottom-mounted offshore wind farms in intermediate water depths. With the offshore wind industry moving towards deeper waters, the predominance of jacket-frames is expected to increase in future years. Multipurpose platforms combining wind and wave energy are proposed as an innovative solution to enhance the sustainability of offshore wind energy. In this research, a multipurpose platform is investigated with a novel feature in its oscillating water column (OWC) wave energy converter—a variable geometry skirt. Reducing the time required to predict design loads and responses would result in significant efficiency improvements to the floating offshore wind turbine (FOWT) design process. Present methods of predicting design values require at least six, 1-hour sea condition simulations. It would therefore be highly beneficial to identify what conditions lead to the extremes of a range of responses of interest to reduce the length of the time series required. In the last few years, there has been a lot of interest in the effect of nutrition on cognitive function, as it affects insulin signals inside the body, whose role is not limited to controlling blood glucose levels but also extends to the learning process and cognitive function. Nutrition-induced insulin resistance is one of the present diseases, which impairs brain insulin signaling and leads to various forms of dementia. This review discusses the different roles of insulin inside the body, the negative effect of insulin resistance on brain function, and the use of some drugs that may improve cognitive dysfunction.

Conflict, and violence related events have been found to have significant effects on the cognitive thinking and mental well-being of individuals. Although there is ample evidence suggesting negative association of conflict with schooling outcomes, there is non-existent research on how violence can impact degree choices made by students at the university level. By using university level admissions data between 2014 and 2016 from Pakistan, this paper examines the differential in preference for degree choices of students who live in conflict-affected areas compared to students who live in conflict-free areas. Discourse analysis and the theories of war and peace journalism are used to investigate the newspaper coverage of the Pulwama attack of 2019, a recent development in the Kashmir conflict between India and Pakistan that brought the nuclear-armed Asian neighbours on the brink of a war. To identify the journalistic discourses of the Pulwama attack, 686 headlines of news articles published in six popular newspapers on the attack and its aftermath during February 15-25, 2019, are analysed. While Northern Ireland is still recovering from the violent legacy of the 30-year Troubles during a global COVID-19 pandemic, people still experience violence daily. Economic and political inequalities heighten intergroup tensions and insecurity contributing to the promotion of destructive stories. Brexit has escalated the culture wars between the Protestant Unionist Loyalist and Catholic Nationalist Republican communities as Loyalist youth and paramilitary groups protest on Belfast’s streets. Through 120 semi-structured interviews, we explore the experiences and understandings of Civil Society Organization (CSO) peacebuilding and reconciliation workers that liaison with ex-combatants and youth in Derry and the Border area of Northern Ireland. …

The subfield of food and eating practices has registered a significant volume of theoretical and empirical studies. However, there is very limited research targeting non-state armed groups. This article contributes to understanding the nuanced role of food and eating practices (or commensality) in conflict, and its significance in the construction and sustenance of sense of community in non-state armed groups that use particularly elaborate means of indoctrination to build a following. This article explores the failure of top-level interventions in the resolution of local conflicts and the role of local agency in peacebuilding. The article relied primarily on qualitative techniques and instruments to collect data to examine why top-level interventions by successive governments have failed to resolve the Bawku chieftaincy conflict. The article finds that top-level interventions have failed to resolve the conflict because they are introduced with little to no participation by local communities. These interventions are, therefore, unable to garner grassroot support and commitment. In contrast, the BIEPC led peace process has been more successful in the management of …

Academic research has highlighted the gendered impacts and amplifications of gender disparities of COVID-19. Traditionally, Pakistan is a patriarchal society, where it is a parenthood norm to socialize specific gender social roles.

Objectives

The current research asserts that these normative gender roles may influence individuals throughout their life course, even during the COVID-19 pandemic. Climate change in the Arctic is more rapid than anywhere on the globe and changes in the marine environment can impact the distribution and abundance of Arctic and sub-Arctic species. Understanding how a species responds to climate change can aid conservation planning and recovery. Spectacled eiders (Somateria fischeri), sea ducks listed as "threatened" under the Endangered Species Act, winter at the Bering Sea and nest along the coastal areas of Alaska and Arctic Russia. In cold climates during winter, surface-based temperature inversions reduce vertical dispersion within the atmospheric boundary layer. Reduced vertical dispersion coupled with stagnant horizontal winds causes pollution emitted near the ground level to accumulate to unhealthy amounts. To study pollution trapping, we measured vertical differences in fine particulate matter and ozone across a shallow 20-meter vertical scale during surface-based inversions in Fairbanks, Alaska and showed that pollution trapping occurs on this extremely shallow scale. In winter 2022, trace gases were measured by long-path differential absorption spectroscopy to probe vertical dispersion on a taller scale, up to ~200 m above the surface. …

The collapse of the Soviet Union in 1991 opened Siberia as a new "travelers' frontier." Travel writers seeking destinations unspoiled by the encroachment of global capitalism and the resultant "placeless" landscapes made Siberia the subject of numerous travel books. This thesis critically examines the geopolitical discourses embedded in those books. Through a close reading and discourse analysis of three representative samples spanning the post-Cold War period, it highlights the key tropes, images and discourses that Anglophone travel writers have deployed in their constructions of Russia as a geopolitical Other. Analyzing and comprehending co-production of knowledge (CPK) in the context of working with Arctic Indigenous communities on climate change research is the main goal for this interdisciplinary doctoral dissertation. CPK is shared decision-making on every step of the research process with research partners from communities, agencies, or organizations. CPK with Arctic Indigenous communities requires dedicated consideration of equity, ethics, cultural worldviews, and colonization.
