Graft versus host disease (GVHD) represents the major complication after allogeneic hematopoietic stem cell transplantation (Allo-SCT). GVHD-prone patients rely on GVHD prophylaxis (e.g. methotrexate) and generalized anti-GVHD medical regimen (glucocorticoids). New anti-GVHD therapy strategies are being constantly explored, however there is an urgent need to improve current treatment, since GVHD-related mortality reaches 22% within 5 years in patients with chronic GVHD. This review is an attempt to describe a very well-known receptor in lipoprotein studies - the low-density lipoprotein receptor related protein 1 (LRP1) - in a new light, as a potential therapeutic target for GVHD prevention and treatment. Our …

Exercise And Ischemia-Activated Pathways In Limb Muscle Angiogenesis And Vascular Regeneration, Vihang A Narkar

Faculty, Staff and Student Publications

Exercise has a profound effect on cardiovascular disease, particularly through vascular remodeling and regeneration. Peripheral artery disease (PAD) is one such cardiovascular condition that benefits from regular exercise or rehabilitative physical therapy in terms of slowing the progression of disease and delaying amputations. Various rodent pre-clinical studies using models of PAD and exercise have shed light on molecular pathways of vascular regeneration. Here, I review key exercise-activated signaling pathways (nuclear receptors, kinases, and hypoxia inducible factors) in the skeletal muscle that drive paracrine regenerative angiogenesis. The rationale for highlighting the skeletal muscle is that it is the largest organ recruited …

Tuning The Thermodynamic, Optical, And Rheological Properties Of Thermoresponsive Polymer Solutions Via Silica Nanoparticle Shape And Concentration, Christopher A P Neal, Valeria León, Michelle C Quan, Nondumiso O Chibambo, Michelle A Calabrese Jan 2023

Tuning The Thermodynamic, Optical, And Rheological Properties Of Thermoresponsive Polymer Solutions Via Silica Nanoparticle Shape And Concentration, Christopher A P Neal, Valeria León, Michelle C Quan, Nondumiso O Chibambo, Michelle A Calabrese

Faculty, Staff and Student Publications

HYPOTHESIS: The shape and quantity of hydrophilic silica nanoparticles (NPs) can be used to tune the microstructure, rheology, and stability of phase-separating polymer solutions. In thermoresponsive polymer systems, silica nanospheres are well-studied whereas anisotropic NPs have little literature precedent. Here, we hypothesize that NP shape and concentration lower the onset of rheological and turbidimetric transitions of aqueous poly(N-isopropyl acrylamide) (PNIPAM) solutions.

EXPERIMENTS: Differential scanning calorimetry (DSC), Fourier-transform infrared spectroscopy (FTIR), turbidimetry, and oscillatory rheology are utilized to examine interactions between NPs, PNIPAM, and water and to track changes in phase separation and rheological properties due to NP concentration and shape. …

Faculty, Staff and Student Publications

We develop a methodology to create data-driven predictive digital twins for optimal risk-aware clinical decision-making. We illustrate the methodology as an enabler for an anticipatory personalized treatment that accounts for uncertainties in the underlying tumor biology in high-grade gliomas, where heterogeneity in the response to standard-of-care (SOC) radiotherapy contributes to sub-optimal patient outcomes. The digital twin is initialized through prior distributions derived from population-level clinical data in the literature for a mechanistic model's parameters. Then the digital twin is personalized using Bayesian model calibration for assimilating patient-specific magnetic resonance imaging data. The calibrated digital twin is used to propose optimal …

However, people tend to focus more on individuals who share social values while making friends. On the other hand, in a multicultural society, people work, leave, or learn together to share such values. The diversity of the origin of people living in the United States (US), especially in major cities, varies significantly regarding race, ethnicity, and religion, resulting in super-diverse urban setups. This study examined the multicultural friendship between international Arab students studying in the US and their American counterparts. The study was guided by …

Ccl2 Suppression In Lewis Lung Carcinoma Model Enhances Host Survival And Immune Response In The Tumor Microenvironment, Nivedita Biswas Jan 2023

Ccl2 Suppression In Lewis Lung Carcinoma Model Enhances Host Survival And Immune Response In The Tumor Microenvironment, Nivedita Biswas

Theses and Dissertations

CCL2, a chemokine also referred to as monocyte chemoattractant protein 1 (MCP-1) and small inducible cytokine A, is chemotactic for monocytes. CCL2-induced recruitment of CCR2+Ly6C(hi) monocytes increases vascular permeability CCR2+ endothelium aiding the escape and migration of tumor cells. CCL2 is produced by cancers of varied immunogenicity and augments tumor proliferation and metastasis. Elevated CCL2 expression by tumor cells is linked to poor prognosis. We have previously shown that two bacterially derived superantigens, Staphylococcal enterotoxin G and I (SEG/SEI) stimulate large numbers of T cells in an antigen-independent fashion and promote enhanced survival in the poorly immunogenic B16-F10 melanoma model. Conversely, …

Rare earth elements are critical materials for many technologies driving the energy industry forward. However, there is increasingly low security and lack of sustainability of current supplies. New sources and processing methods are needed and are being intensely investigated among U.S. energy leaders such as rare earth extraction from lignite coal in North Dakota. A critical need is to confirm the technological and economic viability of these approaches, aspects which are inherently interconnected, would benefit from a dynamic approach. The current approach is Techno-Economic Assessments (TEAs). TEAs evaluate the economics of the process and commercialization of the technology for viability …

Using Rna-Based Therapies To Target The Kidney In Cardiovascular Disease, Trecia C Palmer, Robert W Hunter Jan 2023

Using Rna-Based Therapies To Target The Kidney In Cardiovascular Disease, Trecia C Palmer, Robert W Hunter

Faculty, Staff and Student Publications

RNA-based therapies are currently used for immunisation against infections and to treat metabolic diseases. They can modulate gene expression in immune cells and hepatocytes, but their use in other cell types has been limited by an inability to selectively target specific tissues. Potential solutions to this targeting problem involve packaging therapeutic RNA molecules into delivery vehicles that are preferentially delivered to cells of interest. In this review, we consider why the kidney is a desirable target for RNA-based therapies in cardiovascular disease and discuss how such therapy could be delivered. Because the kidney plays a central role in maintaining cardiovascular …

Gibbs Process Distinguishes Survival And Reveals Contact-Inhibition Genes In Glioblastoma Multiforme, Afrooz Jahedi, Gayatri Kumar, Lavanya Kannan, Tarjani Agarwal, Jason Huse, Krishna Bhat, Kasthuri Kannan

Faculty, Staff and Student Publications

Tumor growth is a spatiotemporal birth-and-death process with loss of heterotypic contact-inhibition of locomotion (CIL) of tumor cells promoting invasion and metastasis. Therefore, representing tumor cells as two-dimensional points, we can expect the tumor tissues in histology slides to reflect realizations of spatial birth-and-death process which can be mathematically modeled to reveal molecular mechanisms of CIL, provided the mathematics models the inhibitory interactions. Gibbs process as an inhibitory point process is a natural choice since it is an equilibrium process of the spatial birth-and-death process. That is if the tumor cells maintain homotypic contact inhibition, the spatial distributions of tumor …

Evaluating The Impact Of Electricity And Electron Movement On The Electric Grid From Distributed Generation, Cooper R. Wade Jan 2023

Evaluating The Impact Of Electricity And Electron Movement On The Electric Grid From Distributed Generation, Cooper R. Wade

Theses and Dissertations

The United States’ electric grid is currently undergoing a monumental energy transition involving the incorporation of new fuel sources, generation types, and operational modifications. Since 1920, the U.S. energy collective has been overseen by the Federal Energy Regulatory Commission (FERC), where during this tenure they have pursued the direct regulation of Distributed Energy Resources (DERs). This reasoning has been utilized over 300 times in federal cases and proceedings as the adoption rate of DERs has increased, which is based upon the language allowing the federal regulation of electricity based upon the flow of electrons. This is based solely upon the …

BACKGROUND: Percutaneous thermal ablation is a curative-intent locoregional therapy (LRT) for selected patients with unresectable colorectal liver metastasis (CLM). Several factors have been identified that contribute to local tumour control after ablation. However, factors contributing to disease progression outside the ablation zone after ablation are poorly understood.

METHODS: In this retrospective study, using next-generation sequencing, we identified genetic biomarkers associated with different patterns of progression following thermal ablation of CLM.

RESULTS: A total of 191 ablation naïve patients between January 2011 and March 2020 were included in the analysis, and 101 had genomic profiling available. Alterations in the TGFβ pathway …

Osteoblast-Specific Overexpression Of Nucleolar Protein No66/Riox1 In Mouse Embryos Leads To Osteoporosis In Adult Mice, Qin Chen, Krishna M Sinha, Benoit De Crombrugghe, Ralf Krahe Jan 2023

Osteoblast-Specific Overexpression Of Nucleolar Protein No66/Riox1 In Mouse Embryos Leads To Osteoporosis In Adult Mice, Qin Chen, Krishna M Sinha, Benoit De Crombrugghe, Ralf Krahe

Faculty, Staff and Student Publications

In previous study, we showed that nucleolar protein 66 (NO66) is a chromatin modifier and negatively regulates Osterix activity as well as mesenchymal progenitor differentiation. Genetic ablation of the NO66 (RIOX1) gene in cells of the Prx1-expressing mesenchymal lineage leads to acceleration of osteochondrogenic differentiation and a larger skeleton in adult mice, whereas mesenchyme-specific overexpression of NO66 inhibits osteochondrogenesis resulting in dwarfism and osteopenia. However, the impact of NO66 overexpression in cells of the osteoblast lineage in vivo remains largely undefined. Here, we generated osteoblast-specific transgenic mice overexpressing a FLAG-tagged NO66 transgene driven by the …
