In Silico Prediction Of Acute Chemical Toxicity Of Biocides In Marine Crustaceans Using Machine Learning, Rama Krishnan, Ian S. Howard, Sean Comber, Awadhesh N. Jha
School of Engineering, Computing and Mathematics
Biocides are a heterogeneous group of chemical substances intended to control the growth or kill undesired organisms. Due to their extensive use, they enter marine ecosystems via non-point sources and may pose a threat to ecologically important non-target organisms. Consequently, industries and regulatory agencies have recognized the ecotoxicological hazard potential of biocides. However, the prediction of biocide chemical toxicity on marine crustaceans has not been previously evaluated. This study aims to provide in silico models capable of classifying structurally diverse biocidal chemicals into different toxicity categories and predict acute chemical toxicity (LC50) in marine crustaceans using a set of calculated …

It’S About Time: Tennessee’S Modernization Of Juvenile Sentencing And Where To Go From Here, Blake Lee, J.D.
Lincoln Memorial University Law Review Archive
Tennessee has long been considered the harshest in the nation regarding its treatment of minors convicted of first-degree murder. After the United States Supreme Court determined that automatic life sentences for juveniles violated the Eighth Amendment in Miller v. Alabama, many states began the process of reforming their sentencing statutes. Tennessee did not follow this trend, as the law on the books did not prescribe a “life sentence” to youthful homicide offenders. Rather, Tennessee’s sentencing statute automatically sentenced juveniles convicted of first-degree murder to a lengthy “term of years,” and as a result, did not fall within the reach …

Chromophore Carbonyl Twisting In Fluorescent Biosensors Encodes Direct Readout Of Protein Conformations With Multicolor Switching, Malin J Allert, Shivesh Kumar, You Wang, Lorena S Beese, Homme W Hellinga
2020-Current year OA Pubs
Fluorescent labeling of proteins is a powerful tool for probing structure-function relationships with many biosensing applications. Structure-based rules for systematically designing fluorescent biosensors require understanding ligand-mediated fluorescent response mechanisms which can be challenging to establish. We installed thiol-reactive derivatives of the naphthalene-based fluorophore Prodan into bacterial periplasmic glucose-binding proteins. Glucose binding elicited paired color exchanges in the excited and ground states of these conjugates. X-ray structures and mutagenesis studies established that glucose-mediated color switching arises from steric interactions that couple protein conformational changes to twisting of the Prodan carbonyl relative to its naphthalene plane. Mutations of residues contacting the carbonyl …

Karbala International Journal of Modern Science
According to World Health Organization data, Coronavirus (COVID-19) has infected about 660, 378, 145 patients around the world. It is nonetheless difficult for physicians to detect COVID-19 infections out of CT or X-ray radiographs. Thus, several computer-aided diagnosis (CAD) systems based on deep learning and radiographs were developed to detect COVID-19 infections. However, the majority of approaches considered small datasets, which is ineligible to provide diverse COVID-19 radiographs. This work utilizes a massive number of X-ray radiographs, and compared standard CNN, DenseNet-121, and GoogLeNet for isolating COVID-19 infections out from normal and other pneumonia radiographs. The dataset in this work …

Prosecutorial Actus Reus: Appellate Review Of Prosecutorial Misconduct And The Diminishment Of Responsibility, Elizabeth Griffiths, Heather L. Scheuerman, Sandy Xie
International Journal on Responsibility
The socio-historical concept of criminal responsibility links the action (actus reus) and mental state (mens rea), or intention, of the actor (i.e., the defendant) to determine legal and moral liability for his or her behavior and to apportion punishment. When the actor responsible for immoral conduct is the prosecutor in pursuit of a conviction, the courts respond very differently. More specifically, because prosecutors are presumed to be moral and ethical system actors, assumptions about their good character likely influence the ways in which they are held to account. This study explores the content and arguments made …

Exosomes derived from endothelial cells and Schwann cells have been employed as novel treatments of neurological diseases, including peripheral neuropathy. Exosomal cargo plays a critical role in mediating recipient cell function. In this study, we thus performed a comprehensive proteomic analysis of exosomes derived from healthy mouse dermal microvascular endothelial cells (EC-Exo) and healthy mouse Schwann cells (SC-Exo). We detected 1,817and 1,579 proteins in EC-Exo and SC-Exo, respectively. Among them, 1506 proteins were present in both EC-Exo and SC-Exo, while 311 and 73 proteins were detected only in EC-Exo and SC-Exo, respectively. Bioinformatic analysis revealed that EC-Exo enriched proteins were …

Spinal and bulbar muscular atrophy (SBMA) is a neuromuscular disease with substantial mitochondrial and metabolic dysfunctions. SBMA is caused by polyglutamine (polyQ) expansion in the androgen receptor (AR). Activating or increasing the NAD+-dependent deacetylase, SIRT3, reduced oxidative stress and death of cells modeling SBMA. However, increasing diminished SIRT3 in AR100Q mice failed to reduce acetylation of the SIRT3 target/antioxidant, SOD2, and had no effect on increased total acetylated peptides in quadriceps. Yet, overexpressing SIRT3 resulted in a trend of motor recovery, and corrected TCA cycle activity by decreasing acetylation of SIRT3 target proteins. We sought to boost blunted …

Human actions, both legal and illegal, affect wildlife in many ways. Inaccurate diagnosis of cause of death undermines law enforcement, management, threat assessment, and mitigation. We found 410 dead birds collected along 196 km of power lines in four western USA states during 2019–2022. We necropsied these carcasses to test conventional wisdom suggesting that electrocution is the leading cause of death of birds at electrical infrastructure. Of 175 birds with a known cause of death, 66% died from gunshot. Both raptors and corvids were more likely to die from gunshot than from other causes, along both transmission and distribution lines. …

A Thesis On The Human Genetic Basis Of Severe Pneumococcal Infection In Papua New Guinea, Emma Catherine Walker
Arts & Sciences Graduate Student Theses and Dissertations
Pneumonia remains a major cause of childhood mortality, especially in under-resourced communities. The profound morbidity of pediatric pneumonia in Papua New Guinea (PNG) compels innovative approaches to ameliorate disease. While the high incidence of pneumonia has been ascribed to environmental factors, we here identify a heritable allele increasing risk of severe pneumonia. In a pilot study, whole exome sequencing reveals a single nucleotide variant (SNV) in coenzyme Q6 (COQ6), a mitochondrial enzyme essential for ubiquinone biosynthesis. Significant association of SNV homozygosity with pneumonia replicates in an independently recruited cohort (p=0.036). Mice homozygous for the murine variant exhibit increased mortality after …
Investigating The Effects Of Opioid Exposure And Withdrawal On Endogenous Opioid Peptide Systems, Neuroimmune Interactions, And Striatal Neuron Activity, Marwa Mikati
Arts & Sciences Graduate Student Theses and Dissertations
Endogenous opioid peptides and their receptors perform critical roles in reward, aversion, and drug exposure processes. Both the peptides and the receptors perform distinct roles in modulating behavior depending on the brain region and stimulus in question. It has long been a challenge to study opioid peptides specifically because they are structurally similar to each other and occur at very low concentrations. Therefore, understanding their roles in stress, drug exposure, and drug withdrawal has presented difficulty for researchers over the years. To address this difficulty, I helped develop a method for the detection of enkephalins, a class of opioid peptides, …
