For giraffes, few standardized methods exist for quantifying chronic stress. Allostatic load index is quantified from a panel of multi-system stress biomarkers to estimate cumulative stress. Our objective was to determine whether a panel of biomarkers selected for their role in allostatic load would reflect the number of documented stress events experienced by giraffes. Cortisol, DHEA-S, cholesterol, non-esterified fatty acids (NEFA), and fructosamine were determined in serum samples from zoo-housed giraffes (n = 18). These were correlated with the overall number and frequency of ZIMS-documented stress events experienced prior to blood collection. We also compared giraffes grouped by high vs. …

Postpartum mood disorders (PMD) are currently among the leading causes of maternal postpartum morbidity and mortality. PMD include the conditions of postpartum blues (PB), postpartum depression (PPD), and postpartum psychosis. The pathogenesis of PMDs are ambiguous, and there are no reliable prenatal predictive markers despite current research efforts. Even though reliable indicators have not been found, leading ideas suggest an etiology of hormonal fluctuations. Mass production of plastic within the past decade has led to over 100 billion tons of plastics being added to the world’s oceans through rivers and effluent disposal and decomposition. For marine environments, the sudden and constant growth of microplastics (plastics 1 µm to 5 mm in diameter), is of particular concern to top-predatory fish such as steelhead trout (Oncorhynchus mykiss), who passively or actively uptake microplastics while consuming prey. Background Endocardial cells are a major progenitor population that gives rise to heart valves through endocardial cushion formation by endocardial to mesenchymal transformation and the subsequent endocardial cushion remodeling. Genetic variants that affect these developmental processes can lead to congenital heart valve defects. Crk and Crkl are ubiquitously expressed genes encoding cytoplasmic adaptors essential for cell signaling. This study aims to explore the specific role of Crk and Crkl in the endocardial lineage during heart valve development. The BCL-2 inhibitor Venetoclax is a promising agent for the treatment of acute myeloid leukemia (AML). However, many patients are refractory to Venetoclax, and resistance develops quickly. ATP-binding cassette (ABC) transporters mediate chemotherapy resistance but their role in modulating the activity of targeted small-molecule inhibitors is unclear. Using CRISPR/Cas9 screening, we find that loss of ABCC1 strongly increases the sensitivity of AML cells to Venetoclax. Genetic and pharmacologic ABCC1 inactivation potentiates the anti-leukemic effects of BCL-2 inhibitors and efficiently re-sensitizes Venetoclax-resistant leukemia cells. Conversely, ABCC1 overexpression induces resistance to BCL-2 inhibitors by reducing intracellular drug levels, and high ABCC1 levels …

Immunotherapies have emerged as promising strategies for cancer treatment. However, existing immunotherapies have poor activity in high-grade serous ovarian cancer (HGSC) due to the immunosuppressive tumor microenvironment and the associated low tumoral CD8+ T cell (CTL) infiltration. Through multiple lines of evidence, including integrative analyses of human HGSC tumors, we have identified miR-146a as a master regulator of CTL infiltration in HGSC. Tumoral miR-146a expression is positively correlated with anti-cancer immune signatures in human HGSC tumors, and delivery of miR-146a to tumors resulted in significant reduction in tumor growth in both ID8-p53−/− and IG10 murine HGSC models. Emerging evidence implicates microbiome involvement in the development of pancreatic cancer (PaCa). Here, we investigate whether increases in circulating microbial-related metabolites associate with PaCa risk by applying metabolomics profiling to 172 sera collected within 5 years prior to PaCa diagnosis and 863 matched non-subject sera from participants in the Prostate, Lung, Colorectal, and Ovarian (PLCO) cohort. We develop a three-marker microbial-related metabolite panel to assess 5-year risk of PaCa. The addition of five non-microbial metabolites further improves 5-year risk prediction of PaCa. We introduce a pioneering approach that integrates pathology imaging with transcriptomics and proteomics to identify predictive histology features associated with critical clinical outcomes in cancer. We utilize 2,755 H&E-stained histopathological slides from 657 patients across 6 cancer types from CPTAC. Our models effectively recapitulate distinctions readily made by human pathologists: tumor vs. normal (AUROC = 0.995) and tissue-of-origin (AUROC = 0.979). We further investigate predictive power on tasks not normally performed from H&E alone, including TP53 prediction and pathologic stage. The study of IRS-1 has increased over the years due to the prevalence of cardiometabolic diseases. An increase in IRS-1 serine phosphorylation is often associated with the development of insulin resistance due to the downregulation bestowed upon it by different kinases such as p70 S6K-1. Recently, p300 has been shown to acetylate IRS-1 thus leading to its downregulation. However, studies have not looked into the relationship that exists between serine phosphorylation and lysine acetylation of IRS-1. Research has demonstrated that mental health illnesses have increased, particularly among college students. Although there is a general understanding of the implications of this trend, there remains a lack of evidence on the impact of mental health diagnoses on residential undergraduate minority ethnic students. This qualitative phenomenological study aims to gain insight into the lived experiences of 10 residential undergraduate minority students aged 18 to 36 diagnosed with one or more mental health disorders. During the COVID-19 lockdowns, experts predicted an increase of children’s mental health issues. However, not all children showed symptoms after lockdowns lifted. The study’s purpose was to examine parental stress and resilience during COVID-19 lockdowns and their relationship to children’s behaviors, specifically anxiety and aggression after lockdown. Participants included 61 parents of children currently aged 7-11 who did not have mental health disorders before COVID lockdowns. Most parents were white, non-Hispanic within middle to high income ranges. The quantitative study used a correlational design. The purpose of this transcendental, phenomenological study was to describe the lived experiences of educators with classroom-based crises at military-connected schools and the crisis prevention and intervention training they have received. Knowles’ theory on adult education guided this study as it provides a useful framework for evaluating the provision and implementation of effective staff training. This qualitative study followed a phenomenological research design and involved 10 special education and general education teachers from military-connected schools. Shotgun proteomics is essential for protein identification and quantification in biomedical research, but protein isoform characterization is challenging due to the extensive number of peptides shared across proteins, hindering our understanding of protein isoform regulation and their roles in normal and disease biology. We systematically assess the challenge and opportunities of shotgun proteomics-based protein isoform characterization using in silico and experimental data, and then present SEPepQuant, a graph theory-based approach to maximize isoform characterization. Background: An accurate measurement of the glomerular filtration rate (GFR) is essential for detecting renal insufficiency in living kidney donors. Iohexol is a "near-ideal" exogenous filtration marker for GFR measurements that has attracted increasing interest in clinical practice because it is non-toxic, non-radioactive, readily available, and easy to measure. In this study, we aimed to set up a laboratory test to conveniently assess the plasma clearance of iohexol in living kidney donors.

Societal stigma, criminal sanctions, and the mental illness or disorder of pedophilia are explored in this paper because of the considerable research gaps that have accumulated over the past decade. Most of the data acquired regarding individuals with pedophilia have been predominantly from male offenders, with most of the information coming from reports to local law enforcement and the FBI. The outcome of multiple studies shows that neuroimaging, associated brainwaves, and related sexual attraction are significantly different for individuals with pedophiliac tendencies than for others. This transcendental phenomenological study aimed to describe the lived experiences of addiction counselors who appeared resilient to burnout (BO) and compassion fatigue (CF). The research question to frame this study was: What are the lived experiences of addiction counselors who appear resilient to severe levels of burnout and compassion fatigue, leading to longevity in the addiction counseling field? This included resiliency factors to resolve BO and CF, coping skills to resolve symptoms of BO and CF, and internal and external factors that contributed to BO and CF. The purpose of this quantitative, quasi-experimental nonequivalent group research was to analyze hypothesized differences in gifted students’ perceptual reasoning based on the type of instructional strategy they receive. Specifically, this study examined possible statistically significant differences among gifted students’ reasoning after receiving at least three years of full-time differentiated instruction, cluster grouping, or general education-focused teaching. Comparing gifted students’ perceptual reasoning and analyzing the impact of gifted instructional strategies and general systems will influence improvements in effective strategies for teaching gifted children. Copious research demonstrates an epidemic of sexual trauma (ST) among females resulting in excruciating shame and fear of exposing oneself to others. Shame is an under-researched emerging mental health concern. This study addressed a gap in the literature regarding interventions for shame symptoms experienced by female survivors of ST. Specifically, it was an intervention study that applied a spiritually integrated form of the Creative Arts Personal Growth Group (CAPG) with Centering Prayer (CAPG-CP).
