Spatial Distribution And Insect Associations Of Frangula Alnus In The Allegheny National Forest, Pennsylvania: Implications For Management, Jennifer S. Greenleaf
Graduate Theses, Dissertations, and Problem Reports (ETD)
Glossy buckthorn (Frangula alnus) (Rosales: Rhamnaceae) is an invasive European shrub that threatens to displace native vegetation in roadsides and disturbed forests in North America due to its lack of natural enemies, long leaf-out periods, and its ability to survive in a multitude of soil conditions. While well understood in its native range, information regarding F. alnus in North America is lacking. Currently, the management of this invasive shrub relies on an understanding of where it can be found and thus where treatments should be applied. This research investigated the spatial distribution patterns of F. alnus and profiled …

Role Of Cytochrome P450-Epoxygenases And Adenosine A2a Receptor In The Modulation Of Vascular Response, Stephanie Onyeka Agba
Graduate Theses, Dissertations, and Problem Reports (ETD)
Coronary Artery Disease (CAD) is one of the leading causes of mortality worldwide, and endothelial dysfunction is a major precursor for cardiovascular diseases (CVDs). Arachidonic acids in the vascular endothelial membrane are metabolized by the cytochrome P450-epoxygenases (CYP-2Js and 2Cs) to produce epoxyeicosatrienoic acid (EETs). These anti-inflammatory metabolites or oxylipins have been shown to be cardio-protective against ischemic/reperfusion injury and mediate vasodilation through hyperpolarization of smooth muscle cells. However, the enzyme called soluble epoxide hydrolase (sEH), mainly hydrolyzes EETs to its inactive form (DHETs) and an increase in sEH upregulates Angiotensin-II type1-receptor and facilitates endothelial dysfunction.
we previously showed vascular …
Health-Related Quality Of Life And Disease Severity Among Patients With Idiopathic Pulmonary Fibrosis, Rowida Mohamed
Health-Related Quality Of Life And Disease Severity Among Patients With Idiopathic Pulmonary Fibrosis, Rowida Mohamed
Graduate Theses, Dissertations, and Problem Reports (ETD)
Background: Predicting the survival of patients with Idiopathic pulmonary fibrosis (IPF) remains challenging due to the remarkable heterogeneity of the disease prognosis. Therefore, developing reliable prognostic tools is of utmost importance in IPF management. Nomograms currently represent the most accurate and discriminatory tools for predicting outcomes, with superior performance compared to the commonly used integer score approach. Therefore, developing a valid local nomogram to predict mortality among patients with IPF is warranted. Additionally, IPF is characterized by a high symptom burden significantly impacting Health-related quality of life (HRQoL). Accurately measuring HRQoL in IPF is crucial as it guides treatment decisions …

Microfluidics be used for a multitude of applications, including bioanalysis, cellular assays, organ-on-a-chip, and separations. Microfluidics have great potential to overcome many challenges in conventional chemical and biological applications. However, complex fabrication procedures in silicon, glass, thermoplastics, and (polydimethylsiloxane) PDMS devices make rapidly optimizing and prototyping devices challenging. Microfluidic fabrication techniques have been a long-standing challenge in successfully translating these …
Localized Delivery Of Gm-Csf In The Regulation Of Breast Tumor Oxygen And Anti-Tumor Immunity Through Macrophages, Nicole E. Mihalik
Localized Delivery Of Gm-Csf In The Regulation Of Breast Tumor Oxygen And Anti-Tumor Immunity Through Macrophages, Nicole E. Mihalik
Graduate Theses, Dissertations, and Problem Reports (ETD)
GM-CSF has been employed as an adjuvant to cancer immunotherapies in solid tumors due to its ability to drive immune stimulatory macrophages and other myeloid cells that promote anti-tumor responses. However, GM-CSF does not always exert immune stimulatory effects on myeloid cells, and sometimes even drives immune suppressive phenotypes. GM-CSF effects on myeloid cells and the tumor microenvironment (TME) appear to vary across dosages and even cancer types. However, an optimal dosing regimen for GM-CSF has yet to be determined to-date, and this is further complicated by the fact that GM-CSF is often administered via oncolytic viruses or tumor vaccines …

Advancing Assessments Of Climate Change Vulnerability Of West Virginia Watersheds, Joseph T. Molina
Graduate Theses, Dissertations, and Problem Reports (ETD)
As climate change is becoming increasingly recognized as a threat to aquatic ecosystems, climate adaptation planning, in both the long- and short-term, is becoming more common in natural resource management. There is a need within conservation agencies to develop methodologies and assessments that support adaptation planning to efficiently disperse conservation dollars and effectively strengthen ecological and community resilience as climate changes continue. My thesis aims to provide West Virginia natural resources managers with climate vulnerability assessments that can be used to determine where and how conservation efforts should be administered. Additionally, I demonstrate a methodology that can be built upon …

Diversification Of Ergot Alkaloid Biosynthesis In Natural And Engineered Fungi, Kyle Austin Davis
Graduate Theses, Dissertations, and Problem Reports (ETD)
Ergot alkaloids are a complex family of tryptophan-derived mycotoxins produced by a diverse range of fungi that occupy a wide variety of ecological niches including soil saprotrophs, plant endophytes, pathogens of plants or insects, and opportunistic pathogens of humans and other mammals. Ergot alkaloids are a similarly diverse family of chemicals that elicit a variety of pharmacological activities in animals due to their resemblance to neurotransmitters and high binding affinity for neurological receptors, including those that bind adrenaline, dopamine, and 5-hydroxytryptamine receptors. These structural similarities allow us to create medicines aimed at treating a range of neurological diseases and disorders …

Graduate Theses, Dissertations, and Problem Reports (ETD)
Chronic lower back pain (CLBP) is one of the leading causes of disability worldwide, with an estimated direct cost of over 600 billion dollars in the US alone. CLBP profoundly impacts the patient, their support team, and the healthcare system. Widespread changes to the patient's well-being, including mental health, are seen in CLBP and contribute to a downward spiral that often leads to disability. Chronic pain is poorly understood and is difficult to treat because of its subjective nature. Recent improvements in pain assessment in the clinic and in biomedical research have had a profoundly impacted the field. More than …
Elucidating Mechanisms Of Proteasome Modulation By Small Molecules To Treat Neurodegenerative Diseases And Cancer, Janelle Joo Yee Chuah
Elucidating Mechanisms Of Proteasome Modulation By Small Molecules To Treat Neurodegenerative Diseases And Cancer, Janelle Joo Yee Chuah
Graduate Theses, Dissertations, and Problem Reports (ETD)
Maintaining protein homeostasis, or proteostasis, is crucial for cellular function, and protein degradation through the proteasome plays a pivotal role in this process. Dysregulation of the proteasome can lead to proteostasis collapse and has been linked to the development of neurodegenerative diseases and cancers. Neurodegenerative diseases often exhibit proteasome impairment, whereas proteasome upregulation is frequently observed in cancer. Although the use of proteasome inhibitors as a pharmaceutical approach has shown success in certain cancers, it has yet to demonstrate efficacy in neurodegenerative diseases due to the absence of drug-like small molecules capable of effectively reversing proteasome impairment and/or activating the …
Forage Grass Growth Habit And Allelopathy In Birdsfoot Trefoil Germination And Establishment, Nathan El Blake
Forage Grass Growth Habit And Allelopathy In Birdsfoot Trefoil Germination And Establishment, Nathan El Blake
Graduate Theses, Dissertations, and Problem Reports (ETD)
Birdsfoot trefoil (Lotus corniculatus L.) (BFT) is a high quality, tanniniferous forage legume that has been shown to mitigate gastrointestinal nematode (GIN) infection in sheep. Reliable establishment of birdsfoot trefoil in existing pastures is difficult due to BFT’s poor seedling vigor, susceptibility to crown and root rot, and, perhaps chiefly, competition from other pasture plants. Most existing pastures in the Northeastern United States are primarily composed of cool-season grasses, many of which can be highly competitive. A common form of interplant competition is allelopathy, which occurs when secondary metabolites produced by a plant inhibit a necessary growth process of …

This study aimed to assess the effectiveness of different types of respiratory protection, including National Institute for Occupational Safety and Health (NIOSH) Approved respirators and surgical masks in healthcare settings, as well as barrier face coverings (BFCs) used by the public as source control. The assessment was based on three components: total outward leakage (TOL), particle size distribution, and exhalation airflow velocity. To achieve this, the study utilized headform testing systems to measure TOL and airflow velocity. Specifically, a hard surface headform was used for NIOSH Approved respirators and surgical masks, while two soft surface headforms of different sizes were …
Sense And Sensitivity: Spatial Structure Of Conspecific Signals During Social Interaction, Keshav Ramachandra
Sense And Sensitivity: Spatial Structure Of Conspecific Signals During Social Interaction, Keshav Ramachandra
Graduate Theses, Dissertations, and Problem Reports (ETD)
Organisms rely on sensory systems to gather information about their environment. Localizing the source of a signal is key in guiding the behavior of the animal successfully. Localization mechanisms must cope with the challenges of representing the spatial information of weak, noisy signals. In this dissertation, I investigate the spatial dynamics of natural stimuli and explore how the electrosensory system of weakly electric fish encodes these realistic spatial signals. To do so In Chapter 2, I develop a model that examines the strength of the signal as it reaches the sensory array and simulates the responses of the receptors. The …

Spatial Variability In Above Ground Carbon Within An Appalachian Forest, Megan A. Ponczek
Spatial Variability In Above Ground Carbon Within An Appalachian Forest, Megan A. Ponczek
Graduate Theses, Dissertations, and Problem Reports (ETD)
Forests have long been recognized for their provision of air and water quality ecosystem services to society; but more recently, they have become valuable to the carbon credit markets used as a tool for mitigating climate change. Quantifying above ground carbon storage in forest ecosystems is essential for these carbon credit markets and can also provide insight into factors that control the spatial distribution of carbon in forests. The goal of this study is to assess the degree to which three factors: topography, tree species, and legacies of logging, impact the spatial variability in above ground carbon within a 400x500m …

The Birds And The Bees And The Willows: Understanding The Reproductive Biology Of Salix Nigra Marsh., Julianne Gmys Grady
Graduate Theses, Dissertations, and Problem Reports (ETD)
Salix nigra is a common tree-form willow found throughout the eastern United States and Canada in riparian and wetland habitats. Like other members of the Salicaceae, it is dioecious with separate male and female individuals. While Salix has been thought to be strictly insect pollinated and Populus wind pollinated, several species of Salix have had wind pollination present in varying amounts by species. S. nigra’s pollination syndrome has not been studied extensively, though it is assumed to be insect pollinated. Unlike other willows with wind pollination, S. nigra is basal on the Salix phylogenetic tree. This study was conducted …
Energy Conversion In Plasmas Out Of Local Thermodynamic Equilibrium: A Kinetic Theory Perspective, Mahmud Hasan Barbhuiya
Energy Conversion In Plasmas Out Of Local Thermodynamic Equilibrium: A Kinetic Theory Perspective, Mahmud Hasan Barbhuiya
Graduate Theses, Dissertations, and Problem Reports (ETD)
The study of energy conversion in collisionless plasmas that are not in local thermodynamic equilibrium (LTE) is at the leading edge of plasma physics research. Plasma constituents in such systems can exhibit highly structured phase space densities that deviate significantly from that of a Maxwellian. A standard approach has emerged in recent years for investigating energy conversion between bulk flow and thermal energy in collisionless plasmas using the non-LTE generalization of the first law of thermodynamics. The primary focus is placed on pressure-strain interaction (PS) term, with a particular emphasis on its non-LTE piece called Pi − D. Recent studies …
Intersectional Microaggressions And Disordered Eating In Asian American Women, Spring Szoka
Intersectional Microaggressions And Disordered Eating In Asian American Women, Spring Szoka
Graduate Theses, Dissertations, and Problem Reports (ETD)
Disordered eating in college-age women is well-documented. Less is known about minoritized populations and disordered eating for college-age women, especially Asian American women. As Asian Americans are often subjected to the “model minority” myth, less research has been dedicated to understanding disordered eating for this population. Small, everyday slights motivated by the model minority myth and other racial stereotypes are known as microaggressions. Asian American women hold an intersectional identity that places them at risk for experiencing microaggressions from their identity as both a woman and as an Asian American. The purpose of this study was to examine relationships between …

Exploring Prenatal Care In A Rural Appalachian State: A Project Watch Study Of Barriers And Facilitators In All Births From May 2018 To March 2022, Madelin Gardner
Graduate Theses, Dissertations, and Problem Reports (ETD)
Introduction: PNC is essential in protecting the health of birthing person and infant. Teenage and advanced maternal age (AMA) birthing persons are known risk factors for poor birth outcomes. However, less is known about whether these age groups are associated with inadequate PNC. Births to teenagers continue to be of concern in rural areas however, little is known about the association between inadequate PNC and poor infant outcomes in teenage populations. Previous studies have determined that greater risk of inadequate PNC has been linked to more rural areas compared to more urban areas. WV is the third most rural state …

Chemotherapy-Induced Pdl-1 Expression In Cancer-Associated Fibroblasts Promotes Chemoresistance In Nsclc, Gayathri Heenatigala Palliyage
Graduate Theses, Dissertations, and Problem Reports (ETD)
A cure for cancer is out of reach for most patients due to chemoresistance. Cancer-associated fibroblasts (CAFs), one of the most abundant types of stromal cells in the tumor microenvironment, are known to provide functional and structural support for tumor progression. These cells also play a role in cancer chemoresistance, but a detailed understanding of the process, particularly in chemoresistant lung cancer, is lacking. In this study, we investigated programmed death-ligand 1 (PDL-1) as a potential biomarker for CAF-induced chemoresistance and evaluated its role and underlying mechanisms of chemoresistance in non-small cell lung cancer (NSCLC). We demonstrated that chemotherapy-stimulated CAFs …
Targeting Hiv-1 Reservoirs In T Cell Subsets, Min Li, Marietta M Budai, Min Chen, Jin Wang
Targeting Hiv-1 Reservoirs In T Cell Subsets, Min Li, Marietta M Budai, Min Chen, Jin Wang
Faculty, Staff and Students Publications
The HIV-1 reservoirs harbor the latent proviruses that are integrated into the host genome. It is a challenging task to eradicate the proviruses in order to achieve an HIV cure. We have described a strategy for the clearance of HIV-1 infection through selective elimination of host cells harboring replication-competent HIV (SECH), by inhibition of autophagy and promotion of apoptosis during viral re-activation. HIV-1 can infect various CD4+ T cell subsets, but it is not known whether the SECH approach is equally effective in targeting HIV-1 reservoirs in these different subsets in vivo. In a humanized mouse model, we found …
Learning Representations For Novelty And Anomaly Detection, Ranya Almohsen
Learning Representations For Novelty And Anomaly Detection, Ranya Almohsen
Graduate Theses, Dissertations, and Problem Reports (ETD)
The problem of novelty or anomaly detection refers to the ability to automatically
identify data samples that differ from a notion of normality. Techniques
that address this problem are necessary in many applications, like in medical
diagnosis, autonomous driving, fraud detection, or cyber-attack detection, just to
mention a few. The problem is inherently challenging because of the openness of
the space of distributions that characterize novelty or outlier data points. This is
often matched with the inability to adequately represent such distributions due
to the lack of representative data.
In this dissertation we address the challenge above by making several …

Response Of Running Buffalo Clover (Trifolium Stoloniferum Muhl Ex. A. Eaton) To Herbaceous Competition And Transplanting In Monongahela National Forest, Ruben E. Sabella
Graduate Theses, Dissertations, and Problem Reports (ETD)
Running buffalo clover (RBC) is a rare perennial plant that grows throughout the American Midwest and the Appalachian Mountains. It requires disturbed forests to establish and proliferate. It has been suggested that, in the past, these conditions were created by buffalo; now logging operations maintain RBC populations. However, forest managers have been looking for ways to create suitable habitat for RBC that do not involve harvesting practices. This could help create new populations in areas that cannot be logged. Once established, competing vegetation might influence RBC abundance and flowering. This study seeks to quantify this influence by measuring the vegetation …

Burkholderia Collagen-Like Protein 8, Bucl8, Is An Outer-Membrane Component Of A Novel Efflux Pump And Immunogenic Vaccine Candidate, Megan Elizabeth Grund
Burkholderia Collagen-Like Protein 8, Bucl8, Is An Outer-Membrane Component Of A Novel Efflux Pump And Immunogenic Vaccine Candidate, Megan Elizabeth Grund
Graduate Theses, Dissertations, and Problem Reports (ETD)
Burkholderia spp. are Gram‐negative intrinsically resistant bacteria that are responsible for environmental and nosocomial infections of clinical and biodefense concern. Our laboratory previously identified in silico Burkholderia collagen-like protein 8 (Bucl8) in the BSL-3 pathogens Burkholderia pseudomallei and Burkholderia mallei (Figure 1). The protein sequence of Bucl8 contains tandem outer-membrane efflux protein domains (OEP) typical of efflux systems and a unique collagen-like domain (CL) consisting of a repeating sequence of Glycine-Alanine-Serine (GAS). Because efflux pumps are often a mechanism of pathogenicity and antibiotic resistance, we investigated Bucl8 further. Here, we determined that the locus of bucl8 contains genes …
Evaluating The Degradation Relationship Between Latent Print Impressions And Touch Dna Under Varying Environmental Conditions, Katrina M. Rupert B.S.
Evaluating The Degradation Relationship Between Latent Print Impressions And Touch Dna Under Varying Environmental Conditions, Katrina M. Rupert B.S.
Graduate Theses, Dissertations, and Problem Reports (ETD)
Latent prints have been processed as a source of touch DNA evidence in the hopes of attaining partial or even full DNA profiles. However, the relationship between the latent quality and the DNA quality has not yet been fully explored, resulting in gaps in understanding how the two are connected. A common source of touch DNA is from latent print evidence, which is typically processed and photographed at a scene, and subsequently processed for DNA in the laboratory. However, there is skepticism in the forensic community around touch DNA as laboratory procedures are often manipulated beyond standard operating procedures to …

Stability Properties Of A System Of Receptors In The Presence Of Attractive Domains, Basim Mohammed Al-Maliki
Stability Properties Of A System Of Receptors In The Presence Of Attractive Domains, Basim Mohammed Al-Maliki
Graduate Theses, Dissertations, and Problem Reports (ETD)
I use mathematical modeling to understand aspects of molecular processes in living cells. Specifically, we look at the first few steps in cell signaling. Signal initiation requires that receptors located on the cell membrane bind together to form dimers. Microscopic imaging of receptors on the cell membrane revealed that receptors tend to accumulate in small areas (clusters). Studies also show that the membrane has a complex landscape consisting of domains and barriers of various origin.
Here we study the way receptor dimerization is influenced by domains on the cell membrane. Our underlying hypothesis is that the physical and chemical properties …
Enabling Technologies For Chemical Synthesis: I. Selective Microwave Heating; Ii. Synthesis And Regioselective Cyclotrimerizations Of Tethered 1,6-Diynes, Amir Tavakoli
Graduate Theses, Dissertations, and Problem Reports (ETD)
Reaction discoveries, method developments, and technology advancements lie at the heart of synthetic organic chemistry. These innovations are essential for creating and manipulating complex molecules, which are the building blocks of many important chemical compounds, including pharmaceuticals, materials, and agrochemicals. Here, we first describe new methods to prepare neopentylene-tethered (NPT) 1,6-diynes which are valuable substrates for reaction discovery and target-oriented synthesis, especially in benzannulation strategies toward illudalane natural products. NPT 1,6-diynes have been employed as coupling partners in cyclotrimerization reactions for the synthesis of highly substituted benzene rings which present a persistent challenge in chemical synthesis and are underrepresented scaffolds …
Enantioselective Dearomatization Facilitated By Non-Covalent Interactions Of N-Heterocyclic Carbene Catalysts And Pyridinium Salts, Jack A. Patterson
Enantioselective Dearomatization Facilitated By Non-Covalent Interactions Of N-Heterocyclic Carbene Catalysts And Pyridinium Salts, Jack A. Patterson
Graduate Theses, Dissertations, and Problem Reports (ETD)
Dihydropyridines are a practical organic scaffold commonly used for their pharmaceutical properties. Nucleophilic dearomatization of pyridines has proven to be a useful method in obtaining dihydropyridines. By using N-heterocyclic carbene (NHC) organocatalysts, dihydropyridines can be synthesized with high regioselectivity of the 1,4 regioisomer over the 1,2- regioisomer but often with low enantioselectivity. By incorporating non-covalent interaction contact points, we hypothesize that enantioselectivity can be enhanced. By synthesizing numerous NHCs, each proposed to exhibit a different type of non-covalent interaction (i.e., hydrogen bonding or ion pairing), an understanding of the secondary interactions within this reaction system can be studied and …

This research investigates the historical and current focus of the field of marketing on the disability community and considers whether marketers can positively impact attitudes toward the group through their firm media content. My first essay uses the Rocky Road to Policy Pavers model to discuss the importance of this large and underserved consumer group, and highlights several opportunities for the marketing community to reduce existing barriers to disabled consumers in the marketplace. One way that marketers can reduce stigma and improve attitudes toward the disability community is by strategically portraying individuals with disabilities in their firm media content. My …
