Small cell lung cancer (SCLC) is characterized by rapid growth and high metastatic capacity. It has strong epidemiologic and biologic links to tobacco carcinogens. Although the majority of SCLCs exhibit neuroendocrine features, an important subset of tumors lacks these properties. Genomic profiling of SCLC reveals genetic instability, almost universal inactivation of the tumor suppressor genes TP53 and RB1, and a high mutation burden. Because of early metastasis, only a small fraction of patients are amenable to curative-intent lung resection, and these individuals require adjuvant platinum-etoposide chemotherapy. Therefore, the vast majority of patients are currently being treated with chemoradiation with or …

Background: Tracheal reconstruction presents a challenge because of the difficulty in maintaining the rigidity of the trachea to ensure an open lumen and in achieving an intact luminal lining that secretes mucus to protect against infection.
Methods: On the basis of the finding that tracheal cartilage has immune privilege, researchers recently started subjecting tracheal allografts to "partial decellularization" (in which only the epithelium and its antigenicity are removed), rather than complete decellularization, to maintain the tracheal cartilage as an ideal scaffold for tracheal tissue engineering and reconstruction. In the present study, we combined a bioengineering approach and a cryopreservation technique …

Second-term general chemistry is a key gateway course at our institution for a variety of STEM (Science, Technology, Engineering, and Mathematics) majors. Herein, we examine data on DFW% (i.e., % of D and F grades and withdrawals) in our general chemistry courses over roughly a quarter-century period, focusing on sections taught by the authors. The data are discussed in light of changes in curriculum, pedagogy, and assessments, which included introduction of recitation sections, classroom response systems (clickers), and online homework, restructuring of course content into a modular format, followed by a redesign of the course modules along an anchoring concepts …

Chromatin remodeling proteins contribute to DNA replication, transcription, repair, and recombination. The chromodomain helicase DNA-binding (CHD) family of remodelers plays crucial roles in embryonic development, hematopoiesis, and neurogenesis. As the founding member, CHD1 is capable of assembling nucleosomes, remodeling chromatin structure, and regulating gene transcription. Dysregulation of CHD1 at genetic, epigenetic, and post-translational levels is common in malignancies and other human diseases. Through interacting with different genetic alterations, CHD1 possesses the capabilities to exert oncogenic or tumor-suppressive functions in context-dependent manners. In this Review, we summarize the biochemical properties and dysregulation of CHD1 in cancer cells, and then discuss CHD1's …

Pancreatic ductal adenocarcinoma (PDAC) is considered non-immunogenic, with trials showing its recalcitrance to PD1 and CTLA4 immune checkpoint therapies (ICTs). Here, we sought to systematically characterize the mechanisms underlying de novo ICT resistance and to identify effective therapeutic options for PDAC. We report that agonist 41BB and antagonist LAG3 ICT alone and in combination, increased survival and antitumor immunity, characterized by modulating T cell subsets with antitumor activity, increased T cell clonality and diversification, decreased immunosuppressive myeloid cells and increased antigen presentation/decreased immunosuppressive capability of myeloid cells. Translational analyses confirmed the expression of 41BB and LAG3 in human PDAC. Since …

Introduction: Despite significant clinical advancement with the use of immune checkpoint blockade (ICB) in non-small cell lung cancer (NSCLC) there are still a major subset of patients that develop adaptive/acquired resistance. Understanding resistance mechanisms to ICB is critical to developing new therapeutic strategies and improving patient survival. The dynamic nature of the tumor microenvironment and the mutational load driving tumor immunogenicity limit the efficacy to ICB. Recent studies indicate that myeloid cells are drivers of ICB resistance. In this study we sought to understand which immune cells were contributing to resistance and if we could modify them in a way …

Data Integration (DI) is a technical framework that facilitates the integration of different data sources into a unified system. Often, organizational data is spread across various files, databases, and personal computers, resulting in data silos and difficulties in accessing vital organizational knowledge. The solution rests in the development of Information Systems capable of handling diverse data from multiple heterogeneous sources. This study presents a systematic literature review on data integration approaches, specifically focusing on the aerospace industry. We conducted a literature search on IEEE Xplore database with keywords ‘data integration aerospace’ for articles published between January 2000 and December 2022 …

Globally, pests (invertebrates, vertebrates, pathogens, weeds) can cause estimated annual losses of between 20% and 40%, but higher losses are disproportionately experienced by many low-income countries, as agriculture is the mainstay of the majority of the people and of national economies. Pests pose a major barrier to these countries’ ability to meet the aims of the UN Sustainable Development Goals (SDGs), particularly SDG2, “End hunger, achieve food security and improved nutrition and promote sustainable agriculture.” However, solutions, in the form of pest risk alert systems, coupled with major advances in technology, are now providing opportunities to overcome this barrier in …

Smart control tactics, wider stability region, rapid reaction time, and high-speed performance are essential requirements for any controller to provide a smooth, vibrationless, and efficient performance of an in-house fabricated active magnetic bearing (AMB) system. In this manuscript, three pre-eminent population-based metaheuristic optimization techniques: Genetic algorithm (GA), Particle swarm optimization (PSO), and Cuckoo search algorithm (CSA) are implemented one by one, to calculate optimized gain parameters of PID controller for the proposed closed-loop active magnetic bearing (AMB) system. Performance indices or, objective functions on which these optimization techniques are executed are integral absolute error (IAE), integral square error (ISE), integral …

Empirical interdisciplinary research has explored the role of spatial ability in STEM learning and achievement. While most of this research indicates that fostering spatial thinking in educational contexts has the potential to positively impact students’ enrollment and performance in STEM subjects, there is less agreement on the best approach to do so. This article provides an overview of various types of effective spatial interventions and practices in formal or informal educational contexts, including targeted training of STEM-relevant spatial skills, spatialized curricula embedded in schools, integrated STEM practices addressing students’ use of spatial skills, and spatial activities in informal STEM education. …

We compared perceived duration, ratings of enjoyment and RPE between virtual vs. standard cycling. Participants completed a VO2 peak test and four counterbalanced trials (two 20-min, two 40-min) one at each duration on the interactive Expresso bike (EXP) and a workload matched trial on a Velotron (VEL). Acute RPE were assessed every 5-min. Perceived duration (min) and exercise enjoyment were assessed 15-min post. No significant differences were found for perceived duration between trials (SDE 23.9 ± 8.4; SDV 27.1 ± 9.6; LDE 40.0 ± 12.1; LDV 38.9 ± 10.8). For long duration expresso (LDE) RPE-overall were significantly lower at 25, …
Proton Beam Therapy And Photon-Based Magnetic Resonance Image-Guided Radiation Therapy: The Next Frontiers Of Radiation Therapy For Hepatocellular Carcinoma, Michael Chuong
All Publications
External beam radiation therapy (EBRT) has increasingly been utilized in the treatment of hepatocellular carcinoma (HCC) due to technological advances with positive clinical outcomes. Innovations in EBRT include improved image guidance, motion management, treatment planning, and highly conformal techniques such as intensity-modulated radiation therapy (IMRT) and stereotactic body radiation therapy (SBRT). Moreover, proton beam therapy (PBT) and magnetic resonance image-guided radiation therapy (MRgRT) have expanded the capabilities of EBRT. PBToffers the advantage of minimizing low- and moderate-dose radiation to the surrounding normal tissue, thereby preserving uninvolved liver and allowing for dose escalation. MRgRT provides the advantage of improved soft tissue …

Decades have now passed since Colin Pittendrigh first proposed a model of a circadian clock composed of two coupled oscillators, individually responsive to the rising and setting sun, as a flexible solution to the challenge of behavioral and physiological adaptation to the changing seasons. The elegance and predictive power of this postulation has stimulated laboratories around the world in searches to identify and localize such hypothesized evening and morning oscillators, or sets of oscillators, in insects, rodents, and humans, with experimental designs and approaches keeping pace over the years with technological advances in biology and neuroscience. Here, we recount the …

Introduction: Neuropeptide signaling modulates the function of central clock neurons in the suprachiasmatic nucleus (SCN) during development and adulthood. Arginine vasopressin (AVP) and vasoactive intestinal peptide (VIP) are expressed early in SCN development, but the precise timing of transcriptional onset has been difficult to establish due to age-related changes in the rhythmic expression of each peptide.
Methods: To provide insight into spatial patterning of peptide transcription during SCN development, we used a transgenic approach to define the onset of Avp and Vip transcription. Avp-Cre or Vip-Cre males were crossed to Ai9+/+ females, producing offspring in which the fluorescent protein …

Trichomonas vaginalis is responsible for 156 million new cases per year worldwide. When present asymptomatically, the parasite can lead to serious complications, such as development of cervical and prostate cancer. As infection increases the acquisition and transmission of HIV, the control of trichomoniasis represents an important niche for the discovery and development of new antiparasitic molecules. This urogenital parasite synthesizes several molecules that allow the establishment and pathogenesis of infection. Among them, peptidases occupy key roles as virulence factors, and the inhibition of these enzymes has become an important mechanism for modulating pathogenesis. Based on these premises, our group recently …

Cronobacter sakazakii is an opportunistic pathogen linked to outbreaks in powdered infant formula (PIF), primarily causing meningitis and necrotizing enterocolitis. Whole-genome sequencing (WGS) was used to characterize 18 C. sakazakii strains isolated from PIF (powdered infant formula) manufacturing plants (2011–2015). Sequence Type (ST) 1 was identified as the dominant sequence type, and all isolates carried virulence genes for chemotaxis, flagellar motion, and heat shock proteins. Multiple antibiotic resistance genes were detected, with all isolates exhibiting resistance to Cephalosporins and Tetracycline. A significant correlation existed between genotypic and phenotypic antibiotic resistance. The plasmid Col(pHAD28) was identified in the isolates recovered from …

The Major Questions Doctrine And The Threat To Regulating Emerging Technologies, Johnson, Walter G., Tournas, Lucille M.
Santa Clara High Technology Law Journal
Emerging technologies offer the potential to improve health and quality of life but also pose notable risks to safety, wellbeing, and equity. Law and technology scholarship posits that robust policy and regulatory strategies in the public interest are required to manage these complex benefits, risks, and uncertainties. At the same time, the Supreme Court in its recent jurisprudence appears eager to revitalize nondelegation legal norms, especially through the major questions doctrine—a shifting administrative law doctrine that increasingly appears to act as a clear statement rule when interpreting statutory grants of authority to regulatory agencies. This article argues the major questions …

The Impact Of Poorly Facilitated Anti-Racist Conversations, Brandon Kirkwood
Antioch University Dissertations & Theses
It is well established that the graduate school experience is significantly different for students of color; on top of a rigorous training program, they face additional distinct challenges including racism, discrimination, and feelings of isolation. Insidious Trauma theory provides the framework for this quantitative exploration assessing the impact of poorly facilitated anti-racist conversations on the physical and emotional wellbeing of clinical psychology graduate students of color. George Floyd’s murder, among many other incidents of racial injustice, was a powerful catalyst that propelled academic institutions into action. Anti-racist dialogue became the intentional focus of graduate classes almost overnight. Well-meaning but insufficiently-prepared …

Meeting The Client Halfway: A Relational Revision To Account For Intra-Actions In Psychotherapeutic Space, Lara Pirro Jancetic
Antioch University Dissertations & Theses
This post-qualitative study branching from new-materialist, post-humanist epistemology and ontology examines relationality through the lens of three different theories: agential realism, John Shotter’s communication theory, and relational mindfulness. These theories each describe the dynamics involved in relationships, how they each make sense of these dynamics, and what they entail for human relations. Using a post-qualitative diffractive methodology, this study examines these theories and how they intersect to bring about new insights in our understanding of relationships and how this information can support therapy practice. The results are a series of nine principles of orientation, which were applied to marriage and …

Connecting Antibiotic Resistance To The Environment (Care): Introducing A Novel Framework Integrating Chemical Cross-Resistance And Place-Based Engagement To The Blue Marsh Watershed In Reading, Pennsylvania, Jill Felker
Antioch University Dissertations & Theses
Antibiotic resistance is a serious health threat around the world. Millions of individuals are infected with antibiotic-resistant bacteria yearly, and thousands die from previously curable illnesses. Although antibiotic resistance occurs naturally, misuse of antibiotics accelerates the loss of their effectiveness. Public health campaigns focusing on antibiotic awareness have not effectively communicated and educated the public on this health crisis. New efforts to combat antibiotic resistance are urgently needed. This dissertation focuses on the ecological and public health components of antibiotic resistance research that must be addressed to decelerate antibiotic resistance. A new interdisciplinary theoretical framework was developed to Connect Antibiotic …
