Bezlotoxumab Binding Specificity To Toxin B Of Clostridium Difficile, Kun Karnchanapandh
Bezlotoxumab Binding Specificity To Toxin B Of Clostridium Difficile, Kun Karnchanapandh
Chulalongkorn University Theses and Dissertations (Chula ETD)
Clostridioides difficile infection (CDI) is one of the significant global health issues. Toxin B is the main target protein preventing CDI recurrence. Clinical research has shown that bezlotoxumab (Bez), a monoclonal antibody designed to prevent CDI recurrences, is less effective against the B2 strain compared to the B1 strain, resulting in a high recurrence rate. Bez functions by binding to the epitope 1 and 2 regions in the combined repetitive oligopeptide (CROP) domain. Some binding residues are distinctively different between B1 and B2 strains. In this work, our goal was to understand and compare the interaction of toxins B1 and …

Chulalongkorn University Theses and Dissertations (Chula ETD)
Durian fruit possesses high nutritional value due to its enriched bioactive compounds such as phenolics, carotenoids, and vitamins. While various transcription factors (TFs) regulate the phenylpropanoid biosynthetic pathway, MYB TFs play a pivotal role in controlling key genes in this pathway. In this study, we identified candidate v-Myb myeloblastosis viral oncogene homolog, MYB TFs from our transcriptome database of the Monthong cultivar including five developmental/postharvest ripening stages. MYBs that were upregulated during the ripening stage were considered as candidate transcriptional activators, based on the positive correlation observed between flavonoid biosynthetic genes and flavonoid contents in ripe durian pulps. Conversely, MYBs …

3d Printing Of Hydroxyapatite/Polyhydroxyalkanoate-Based Composites, Warrayut Kanabenja
Chulalongkorn University Theses and Dissertations (Chula ETD)
This thesis focuses on developing bio-based 3D printing feedstocks for biomedical applications, such as bone scaffolds and soft tissue. However, their utility is limited by a narrow processing window which challenges manipulating filament size. These issues result in discontinuous flow and incomplete deposition of melted PHB. In part one, PHB and PHBV were blended with PLA and PBAT to enhance their processability and printability. The polymer blends exhibited lower crystallization temperature and degree of crystallinity. Assessing their 3D printability, neat PHB and PHBV filaments experienced, poor flow during extrudate deposition, and significant warping, leading to 3D printing failures. The blends …

Chulalongkorn University Theses and Dissertations (Chula ETD)
Anemonefish (Amphiprion and Premnas spp.) are renowned for their territorial nature, typically associated with sea anemones but capable of adapting to various substrates, including artificial ones. Despite their widespread popularity in aquariums, detailed documentation of their juvenile territorial behaviors remains limited. This study investigates the territorial behaviors—specifically side-by-side swimming, face-to-face swimming, attack biting, biting, and lunging—of immature pink skunk clownfish (Amphiprion perideraion) in different artificial substrates. Conducted at Si Racha Fishery Research Station, the experiment utilized six pairs of A. perideraion designated as residents. These fish were subjected to three substrate conditions: fake sea anemone, ceramic tiles, and an empty …

Introduction
Summers are usually accompanied with wildfire season, with fires burning rampant across the world. These fires can be devastating to those who live in affected areas, resulting in many short and long-term consequences. However, they also are a source of most-needed resources in the afflicted area. Natural disturbances are a key component of any ecosystem and to improve biodiversity (Krawchuk et al., 2020). Disturbances also promote community stability and landscape heterogeneity (Bai et al., 2022). Forest fires can affect interactions between plant and animal communities and its members, as well as natural processes such as nutrient cycling and soil …

Acute Copper (Cu) Toxicity In Dragonfly Naiads (Insecta: Odonata), Jeff M. Simpson
Department of Entomology: Distance Master of Science Projects
The objective of this study was to examine the effects copper (Cu) on mortality within a pair of test groups of naiads in the order Odonata, and to replicate previous observations regarding their potential uses as indicators for water quality. Few past studies have examined toxicity within relation to the odonates. It has been suggested that this order may contain valuable model organisms (Tollett et al., 2008). Very little focus has been given to odonates as bioindicators, with the few studies done being performed on species that would typically inhabit lentic environments (Trevino, 1999). Similarly, the test subjects for this …

The Design Of Primary Holding Magnet For The Lanl Neutron Electric Dipole Moment Experiment, Piya Amara Palamure
The Design Of Primary Holding Magnet For The Lanl Neutron Electric Dipole Moment Experiment, Piya Amara Palamure
Theses and Dissertations--Physics and Astronomy
The measurement of the permanent electric dipole moment of the neutron (nEDM) plays a significant role in searching for sources of beyond standard model CP violating physics. The goal of the Los Alamos National Laboratory (LANL) nEDM experiment is to push the upper limit of the nEDM to < 3 × 10−27 e·cm (68 % CL). A highly uniform magnetic field is key to achieving this sensitivity for the nEDM measurement by reducing the systematic uncertainties associated with the magnetic field non-uniformity. The B0 coil was designed to achieve a field uniformity of < 0.3 nT·m−1 at a nominal holding field of 1 µT. This document will outline a novel technique employed in the construction of the B0 coil using printed circuit boards (PCBs) and will present preliminary field maps obtained with the B0 coil housed in a magnetically shielded room (MSR) at LANL.
As Ultra Cold Neutrons (UCNs) move from the source to the measurement cells, the UCNs experience a large magnetic field gradient in the region between the layers of the MSR. This large gradient would otherwise cause depolarization of the UCNs. To mitigate this, a double cos θ coil will serve as the …

Analyzing The Impact Of Zip Subfamily Ii Mutations On The Zinc And Iron Transport Of Zupt From Escherichia Coli, Bradley Vandecar
Wayne State University Theses
The disfunction or misregulation of the ZIP family of metal ion transporters has been linked to a plethora of diseases and cancers in humans. Zn2+ is the primary substrate for ZIPs and is transported through the M1 metal binding site, though this activity is stimulated through the binding of Fe2+ in the M2 metal binding site. The members of ZIP Subfamily II are predicted to have an inactive M2 site due to the lack of Fe2+ transport and from a previously generated homology model. This work utilizes homology modelling, whole cell metal ion uptake assays, and growth studies in minimal …

SARS-CoV-2 Papain-like protease is an essential viral protein for viral replication. In our lab, we hypothesize that successful inhibition of this protease will stop viral replication and allow for clearance of the infection by the host. The work described here aimed to design and execute an in silico drug discovery workflow for successfully selecting PLpro inhibitors from the Mcule purchasable compound database. This work reports such a workflow and nine potential compounds for in vitro testing. This work also describes structural activity relationship analysis of a proprietary set of PLpro inhibitors synthesized by medicinal chemistry collaborators and makes recommendations for …
Identifying Host-Intrinsic Resistance Mechanisms To Therapeutic Anti-Tumor Antibody, James Edward Glassbrook
Identifying Host-Intrinsic Resistance Mechanisms To Therapeutic Anti-Tumor Antibody, James Edward Glassbrook
Wayne State University Dissertations
The use of monocloncal antibodies (mAbs) directed at tumor-associated antigens (TAA), often termed “targeted immunotherapy” has revolutionized cancer treatment, markedly improving patient outcomes. As a prime example, the mAb trastuzumab targets human epidermal growth factor 2 (HER2). Like many targeted immunotherapies, only a subset of patients with HER2+ cancer exhibit long-term benefit from trastuzumab. In addition to tumor-intrinsic factors, such as HER2 heterogeneity, we hypothesize that host genetics may influence subsequent immune activation after treatment. To model this interaction, we utilized the genetically heterogeneous Diversity Outbred (DO) mouse model in an F1 cross with BALB/c (producing DOCF1 mice). This cohort …

Background: Family caregivers are at a high risk for low quality of life due to caregiving-related stress. Caregivers' stress is commonly assessed using self-reported measures, which reflect relatively subjective and long-term stress related to caregiving, but objective biological markers of stress are rarely used for caregivers. The purposes of this study were (1) to determine whether caregiver characteristics were associated with stress assessed using a stress biomarker (serum cortisol) and a self-reported caregiving distress measure (Caregiver Burden Inventory) and (2) to determine the predictability of both stress measures for quality of life in caregivers of patients with heart failure. Methods: …
Pregnancy Risk And Coerced Interventions After Dobbs, Elizabeth Kukura
Pregnancy Risk And Coerced Interventions After Dobbs, Elizabeth Kukura
SMU Law Review
Only nine months after the Supreme Court eliminated the federal constitutional right to abortion in Dobbs v. Jackson Women’s Health Organization, fourteen states had banned abortion entirely, and experts estimate the ultimate number of states imposing complete or near-complete restrictions on abortion care will likely rise to twenty-four. Millions of people with the capacity for pregnancy now (or will soon) live in places where getting pregnant means there is no choice other than to carry the pregnancy to term and give birth. One underappreciated, though critically important, impact of Dobbs is the extent to which newly enacted abortion restrictions will …

Lower-limb wearable resistance (WR) facilitates targeted resistance-based training during sports-specific movement tasks. The purpose of this study was to determine the effect of two different WR placements (thigh and shank) on joint kinematics during the acceleration phase of sprint running. Eighteen participants completed maximal effort sprints while unloaded and with 2% body mass thigh- or shank-placed WR. The main findings were as follows: 1) the increase to 10 m sprint time was small with thigh WR (effect size [ES] = 0.24), and with shank WR, the increase was also small but significant (ES = 0.33); 2) significant differences in peak …

Standard off-host tick census techniques are generally inefficient for larval stages. Also, such techniques expose the investigator to environmental hazards including tick bites. Mechanical devices are an option which were tested under field conditions for proof of concept and in side-by-side comparison to the standard techniques. The tick-vac collected more ticks than the legging technique, but with lower frequency of positive detections. It may have utility for sampling small areas, such as weedy corrals suspected of harboring ticks. Different configurations of tick-bots, robotic vehicles with flags, depended on intensive operator control to avoid obstacles, limiting operation to line of sight. …

Despite over a decade of both quantitative and qualitative studies, food insecurity among United States college/university students remains a pervasive problem within higher education. The purpose of this perspective piece was to highlight research gaps in the area of college food insecurity and provide rationale for the research community to focus on these gaps going forward. A group of food insecurity researchers from a variety of higher education institutions across the United States identified five thematic areas of research gaps: screening and estimates of food insecurity; longitudinal changes in food insecurity; impact of food insecurity on broader health and academic …
