Furthermore, there is a need for research on STS in women due to women’s unique interactions with trauma and the potential for them to develop trauma disorders. At the time of this study, there …

Mammalian Dihydropyrimidine Dehydrogenase: Employing Transient State Kinetic Methods To Determine Catalytic And Inhibitory Mechanisms, Dariush Forouzesh Jan 2023

Mammalian Dihydropyrimidine Dehydrogenase: Employing Transient State Kinetic Methods To Determine Catalytic And Inhibitory Mechanisms, Dariush Forouzesh

Dissertations

Dihydropyrimidine dehydrogenase (DPD) catalyzes the two-electron reduction of pyrimidine bases uracil and thymine as the first step in pyrimidine catabolism. The enzyme achieves this simple chemistry using a complex cofactor set including two flavins and four Fe4S4 centers. The flavins, FAD and FMN, interact with respective NADPH and pyrimidine substrates and the iron-sulfur centers form an electron transfer wire that links the two active sites that are separated by 56 Å. DPD accepts the common antineoplastic agent 5-fluorouracil (5FU) as a substrate and so undermines the establishment of efficacious toxicity. Though studied for multiple decades, a precise description of the …

Does Euphrates Softshell Turtle Nest In Unfavourable Substratum? Description Of Nests From Euphrates River, Türkiye, Yusuf Bayrakci

Turkish Journal of Zoology

Nest site selection differs among species based on a characteristic crucial to the embryo's survival. The identified nest-site characteristics of the Euphrates softshell turtle highlight that these turtles, like their other soft-shelled relatives, highly prefer the sandy substrate, even pure sand, to nest. However, alteration and loss of natural habitats for many decades limited the suitable oviposition sites along the Euphrates River. This study presents the uncharacteristic nest-site choice and the nesting time of Euphrates softshell turtle for the first time. I discovered four nests, as well as six unhatched eggs in a nest on 20 August 2017. This discovery …

Multi-Omic Analysis Of Vegetative Propagation Induced Plant Rejuvenation In Grapevines: Implications For The Generation Of Locally Adapted Cultivars Using Epigenetics, Tajbir Raihan

Theses and Dissertations--Plant and Soil Sciences

Epigenetic memory of the growing environment is a process capable of increasing plants’ resilience to abiotic stimuli. The cultivated grapevine is a long-living perennial species with a large corpus of research suggesting that plant adaptation to the environment has implications for the selection of genetic material in a changing climate. Moreover, vine age has been traditionally correlated with enhanced fruit quality. Recent studies have shown that environment/age-acquired traits in grapevine (e.g., adaptation to the growing environment and fruit/wine quality, respectively) are associated with epigenetic mechanisms. These studies also indicate that of propagation type affects the maintenance/erasure of the acquired traits …

Species-Specific Microsymbiont Discrimination Mediated By A Medicago Receptor Kinase, Xiaocheng Yu Jan 2023

Species-Specific Microsymbiont Discrimination Mediated By A Medicago Receptor Kinase, Xiaocheng Yu

Theses and Dissertations--Plant and Soil Sciences

Legumes form a mutualistic partnership with nitrogen-fixing bacteria to meet their nitrogen requirements. This collaboration leads to the development of root nodules, offering an ideal environment for the bacteria to convert atmospheric nitrogen into ammonia. A salient aspect of this symbiotic relationship is host range specificity, such that nodulation capacity and nitrogen fixation efficiency differ significantly between different host-bacterial combinations. Understanding the genetic basis of this specificity could pave the way for strategies to overcome the host range barrier. Sinorhizobium meliloti and Sinorhizobium medicae are two closely related species that display differential symbiotic behaviors with legume plants of the Medicago …

Steady-State Regulation Of Secretory Cargo Export And Er Homeostasis By Inositol Trisphosphate Receptors And Penta-Ef-Hand Proteins, Aaron J. Held

Graduate Student Theses, Dissertations, & Professional Papers

Constant protein degradation and turnover necessitates constitutive secretion that delivers the correct mix of nascent proteins to their appropriate subcellular destinations. Cells thus exhibit steady-state secretion and the additional ability to adjust secretory flux, though we lack a clear understanding of this critical process. During secretion, the COPII coat is responsible for providing a balance of actively and passively selected ER cargos to enter the secretory pathway. Furthermore, Ca2+ -binding proteins have been implicated in regulating this process in response to Ca2+ signals. In Chapter 1, we review the secretory pathway and vesicular trafficking, with a focus on …

Prediction Of Kinase-Substrate Associations Using The Functional Landscape Of Kinases And Phosphorylation Sites, Serhan Yilmaz, Filipa Blasco Tavares Pereira Lopes, Mark R. Chance, Mehmet Koyutürk

Faculty Scholarship

Protein phosphorylation is a key post-translational modification that plays a central role in many cellular processes. With recent advances in biotechnology, thousands of phosphorylated sites can be identified and quantified in a given sample, enabling proteome-wide screening of cellular signaling. However, for most (> 90%) of the phosphorylation sites that are identified in these experiments, the kinase(s) that target these sites are unknown. To broadly utilize available structural, functional, evolutionary, and contextual information in predicting kinase-substrate associations (KSAs), we develop a network-based machine learning framework. Our framework integrates a multitude of data sources to characterize the landscape of functional relationships …

Our esteemed longtime contributor George McKenna considers the question in the following article, “Getting There,” to which nine Human Life Foundation Great Defenders of Life respond with comments on his proposal and ideas …

Unleashing Carbon Removal Technologies, Sierra Apillanes, Alexa Penalosa Jan 2023

Unleashing Carbon Removal Technologies, Sierra Apillanes, Alexa Penalosa

Colorado Environmental Law Journal

Mounting climate change concerns are driving unprecedented interest in carbon dioxide removal technologies. Unlike carbon capture strategies, which trap carbon dioxide at specific emission points such as power plant smokestacks, carbon removal technologies directly remove carbon dioxide from the ambient atmosphere. Fossil fuel industry stakeholders have championed carbon capture for years as a means of reducing carbon emissions while continuing the nation's heavy reliance on carbon-intensive energy sources. This focus on promoting carbon capture has delayed the development of policies specifically aimed at promoting carbon removal. As a result, humankind has yet to effectively leverage carbon removal technologies in the …
