Much of our current understanding of how small-molecule ligands interact with proteins stems from X-ray crystal structures determined at cryogenic (cryo) temperature. For proteins alone, room-temperature (RT) crystallography can reveal previously hidden, biologically relevant alternate conformations. However, less is understood about how RT crystallography may impact the conformational landscapes of protein-ligand complexes. Previously, we showed that small-molecule fragments cluster in putative allosteric sites using a cryo crystallographic screen of the therapeutic target PTP1B (Keedy et al., 2018). The progressive evolution of post-traumatic vertebral necrosis and consequent loss of structural integrity of the vertebral body along with neurological risk, makes it one of the most feared and unpredictable pathologies in spine traumatology. Problem

Milton Keynes International Seventh-day Adventist Church (MKI) is not able to evangelize its multiethnic/multicultural neighbors. This is because individual members of the church consider evangelism to be an annual event where speakers are invited from abroad to hold two- or three-week campaigns. Despite the high cost involved in this method, few people are baptized and even then, it is difficult to retain the new members. The culprit is the church's inability to connect with its multiethnic and multicultural community because of not using the evangelistic method of Jesus Christ effectively.

Although the Adventist Church upholds the importance of a Christlike and stable marriage, the Italian Union historically never developed a systematic and comprehensive approach to both premarital counseling and enrichment programs for married couples, whose preparation is aleatory and depends on both/either the pastor’s approach and/or the couple’s request. The results could be a weakening of both marital stability and marital satisfaction. We assume that the root problem is a naïve understanding of marriage which causes couples to put more effort into preparing for the wedding day rather than for the marital journey.

The environmental legal framework in Cuba is based on constitutional article 75 and Law 81, of July 11, 1997, on the environment, a framework law on the matter, with a series of complementary provisions with the rank of Decree Law or Ministerial Resolution. Under a Florida law enacted in 2021, any child over the age of six years old can be arrested and subjected to juvenile delinquency proceedings. Florida, as well as the United States in general, is an outlier when it comes to statutory minimum ages for juvenile delinquency. The most common and recommended minimum age internationally is fourteen years old, and many studies show that arresting, charging, and adjudicating children below the age of fourteen is counterproductive, as it leads to increased recidivism, potentially violates due process, and leaves lasting negative effects on children. Air pollution impacts economic activity, and economic activity creates air pollution. Understanding this endogenous nexus is critical as any policy designed to affect one will inevitably affect the other. My research explores both sides of this nexus. Specifically, I identify the economic effects from the implementation of an international climate agreement (the Kyoto Protocol); I determine the impact of air pollution on physical productivity and determine which pollutants are responsible for this effect; and I link urban form to vehicular emissions via the structure of road networks.

One of the goals of endodontic therapy is to debride infected pulps of teeth and return the tooth to a state of health in preparation of restorative care and return to function. Chemically cleansing of the canal system augments mechanical debridement by flushing out debris, dissolving tissue, and disinfecting the root canal system. The purpose of this research is to compare the efficacy of five irrigants: 0.008% HOCl acid (Endocyn®), 0.03% HOCl (Vashe®), 6% and 8.25% sodium hypochlorite (NaOCl), and Ozone against a typical bacterial species found in infected pulps, Enterococcus faecalis (E. faecalis).

Ergot alkaloids are a complex family of tryptophan-derived mycotoxins produced by a diverse range of fungi that occupy a wide variety of ecological niches including soil saprotrophs, plant endophytes, pathogens of plants or insects, and opportunistic pathogens of humans and other mammals. This phenomenological dissertation explores the lived experiences of secondary social studies educators situated in the Appalachian region. Hermeneutic phenomenology was used as a philosophical and methodological approach to gather insights into this phenomenon. Interviews were conducted with three educators to capture their experiences from their childhoods, to their teaching careers, and into their current personal lives. Salix nigra is a common tree-form willow found throughout the eastern United States and Canada in riparian and wetland habitats. Like other members of the Salicaceae, it is dioecious with separate male and female individuals. While Salix has been thought to be strictly insect pollinated and Populus wind pollinated, several species of Salix have had wind pollination present in varying amounts by species. S. nigra’s pollination syndrome has not been studied extensively, though it is assumed to be insect pollinated. In examining the marital communication of recent empty nesters, the aim of this study was to test the theory of resilience and relational load (TRRL) (Afifi et al., 2016). The postparental period that follows directly after children leave their parents’ places of residence may present unique challenges for married individuals to navigate together. Bioenergy can help mitigate climate change by providing a carbon-neutral fuel source. However, multiple challenges exist to achieving carbon neutrality including converting lignocellulosic materials to fuel and enhancing soil C sequestration during the growth of the feedstocks. To address these challenges, there have been recent efforts to genetically modify feedstocks to produce more energy dense oils that increase fuel conversion efficiency and to cultivate deep-rooted perennial feedstocks that can enhance soil C storage. Our understanding of how plant-microbe interactions regulate carbon (C) and nitrogen (N) retention in soil organic matter (SOM) remains uncertain. Conflicting evidence suggests that microbial decomposition can lead to the loss of unprotected particulate SOM, but also that microbial decomposition produces simpler compounds that preferentially form more persistent aggregate- or mineral-protected SOM pools. Further, uncertainty remains in how plant-microbe interactions alter SOM retention in these different pools through litter chemistry controls on microbial decomposition traits and rhizosphere priming. A novel, base-mediated cyclization was developed to form N-hydroxy- and N-alkoxyindoles from 2-(2-nitrophenyl)butenoates. Potassium tert-butoxide or sodium tert-pentoxide, with or without the in-situ addition of an electrophile, formed a multitude of N-hydroxy- and N-alkoxyindoles. The formation of these butanoate precursors was improved via a Barluenga coupling reaction. Novel 4,5,6,7-tetramethoxy-1,10-phenanthroline was utilized as an ancillary ligand within the Watanabe-Cenini-Söderberg reductive cyclization reaction conditions. These new reaction conditions produced high yields for sterically hindered 2-nitrostyrenes and the first synthesis of dilemmaone A using this reductive cyclization. Land managers need tools to mitigate wildfire proliferation in the western United States. Prescribed burning may be an effective tool to reduce wildfire severity and promote healthy landscapes. In 2020, a large-scale wildfire provided a rare opportunity to compare early postfire data between areas with and without a history of prescribed burning. Field data were collected approximately one year after a wildfire in fifty 20-meter plots from sites treated with prescribed fire in either 1999, 2007, or 2011 and fifty from sites without a history of prescribed fire. In order to study these irregularities, a cell culture perfusion system is necessary. Bubbles can proliferate in closed-loop systems and are detrimental to cells. To mitigate loss of habitat, protected lands are dedicated to preserving ecosystems, but such areas often allow outdoor recreational activities. In this thesis, I present multidisciplinary research applicable to deceased unhoused individuals in urban San José, California. I present the problem of homelessness through a “hybrid collectif,” a conceptual framework that encompasses the factors that drive homelessness in life and death. I use taphonomic methodology to examine the problem of homelessness in this urban region by experimentation with porcine carcasses in simulated death scenarios. Western snowy plovers (Charadrius nivosus nivosus) breed and winter along sandy beaches and salt pannes of the Pacific coast of the U.S. and Mexico. Since 2015, the percentage of successful nests in Monterey Bay has declined due to predation. This study examines the site fidelity and dispersal of western snowy plovers after repeated nest loss using nesting data from 2013-2020 from a banded population in Monterey, California. Dispersal distance within a breeding season reflected plovers serially polygamous breeding strategy, with more site fidelity within a breeding season than between two breeding seasons. Canary rockfish are a profitable fishery resource that has failed and successfully recovered in the 21st century. This study aimed to evaluate their trophic ecology through stomach content and stable isotope analysis and relate these to their mercury concentrations, biological traits, and environmental conditions. Canary rockfish consume mostly krill and teleosts with their geographic location affecting the proportion of prey items, suggesting regional environmental effects: chlorophyll-a, relief, port, and depth impact dietary choices. Mean ??13C values (-17.18 ± 0.54) significantly increased in individuals residing in deeper depths, higher latitudes, higher productivity, and higher temperatures, and in sexually mature individuals. It is unknown if a relationship exists between prebiotic intake and depression status. Using 2015-2018 National Health and Nutrition Examination Survey cycles data, adult American estimated mean daily prebiotic intake across demographic groups, gender, and depression status were reported. Additionally, the 10 highest prebiotic content Food and Nutrient Database for Dietary Studies foods were identified. Adults (≥ 18 years) with adequate sleep and at least one day of dietary recall data were included. Participants with excessive alcohol intake, BMI ≤ 18, BMI ≥ 30, or current depression medication use were excluded. The confounding variables were inadequate sleep, unemployment, and age. …
