My dissertation argues we would benefit from focusing on the voice when analyzing gothic and horror texts. That is, I contend, there remains significant fertile ground for us to till in these texts if we shift our focus to the voice and its various iterations across these texts’ long history. To demonstrate this point, I delineate three variations of this theme: the doubled voice in the possession narrative, the split voice in the ventriloquist-dummy dynamic, and the inherent uncertainty of the voice without discernable origin. Representations of Military Women in Contemporary War Stories seeks to understand how war stories influence our perception of who belongs in military service. With the canon of western war writing dominated by the memoirs and stories of white men, what happens when service women enter into and author war stories, and how does their appearance destabilize questions of who is fit for military service? War literature provides an important lens through which to observe how military service is scripted by culturally and socially constructed expectations of one’s gender, race, and occupation. Caeneus, as written in Ovid’s Metamorphoses, is a notable blend of ancient and Hellenistic versions of the myth. Ovid’s Caeneus can be understood as a transgender man, since he was assigned female at birth, but asks for his gender to be changed by the god Neptune, and goes on to live the rest of his life with the body, appearance, and social roles of a man. Ovid incorporates Caeneus’ trans identity with his use of grammatical gender endings for Caeneus, using masculine gender for Caeneus except when discussing his pre-transition childhood, or when the centaurs address him mockingly while fighting. …

Cutaneous touch is facilitated by discrete cellular complexes composed of non-neuronal cells associated with mechanoreceptor neuron endings. The non-neuronal cells of these cutaneous end organ complexes (CEOCs) are believed to contribute to touch, but their role in touch sensation remains unclear. To better understand the function of CEOC cells, we sought to characterize the transcriptional profile of CEOC-rich tissue and identify genes expressed in CEOC cells. Bill skin of the tactile foraging Pekin duck (Anas platyrhynchos) is dense with CEOCs, specifically the avian analogs of mammalian Pacinian and Meissner corpuscles, while corpuscles in duck foot skin are scarce. My dissertation investigates how animals behaviorally respond to environmental change, especially when there is limited information about that change. Uncertainty about the environment comes in various forms, including food resources which vary unpredictably and novel cues which present an unknown level of potential risk or benefit. Various behavioral strategies help animals cope with such uncertainty. Tactics to manage unpredictable variation in food include gathering information to reduce uncertainty (sampling) and strategically adjusting preference or aversion to variation in resources (variance-sensitivity). Divided between Norway and Russia, the Barents Sea shelf (BSS) is an ~1.4 million km2 Arctic province, containing significant hydrocarbon accumulations. However, much of the area is frontier, and geologic data are often restricted or unavailable. One strategy to mitigate lack of geologic data is the use of analogues from well-known, mature basins. Even though there have been attempts to use analogues to study the geology of the BSS, such use is limited. Moreover, no analogue, to my knowledge, has been capable of addressing the regional tectonostratigraphic development of the shelf as a whole.

Perceptual Dialectology Of Tulsan Speakers Of English, Andrew Carter

The present study analyzes new interviews from fifteen Tulsan speakers of English and maps produced by draw-a-map tasks. The interviews and subsequent map tasks were conducted over the course of the winter of 2021 for the purpose of this thesis and have been analyzed for folk linguistic information about the perceptions of Tulsa and the rest of the state. The study finds that Tulsan speakers hold similar dialectal perceptions about both their own city and other locations as their fellow Oklahomans, and also provides evidence of a deteriorating faith among Tulsans in the existence of the northern-southern dialectal divide compared …

This research work explores the construction of the masculinity of the rebellious gaucho in several literary works written by authors from the River Plate, and how it serves as a vehicle for the transmission of ideas –often of political content– of their creators. The basic concept underlying this study is that some writers of works set in the rural world show a serious social commitment, and often combine their literary activity with politics. This work examines the development and collapse of the slave trade in Kentucky. It pushes readers to think about the slave trade in an environmental context. The development of the slave trade in the state depended on the violent acquisition of Indigenous lands. The networks of credit and capital that settler colonists established—wealth concentrated in land, stock, crops, and people—fueled the development of a slave system west of the southern mountains. That system increasingly relied on the forced movement of enslaved people in and through Kentucky. Burkholderia cepacia complex is a group of closely related environmental bacteria that can exacerbate disease in immunocompromised individuals. B. cepacia bacteria are equipped with an arsenal of mechanisms to cooperate and compete for nutrients in various polymicrobial environments. Contact-dependent growth inhibition (CDI) systems are antagonistic mechanisms widespread among proteobacteria. CDI systems consist of Two Partner Secretion pathway proteins that function to deliver the toxic C-terminal portion of the large effector exoprotein (termed ‘BcpA’ in Burkholderia-type CDI systems) to the cytoplasm of a compatible recipient bacterial cell upon direct cell to cell contact. Chronic, low-grade systemic inflammation rises in obesity and promotes type 2 diabetes (T2D). Circulating immune cells are key indicators of obesity and T2D pathogenesis. T cells outnumber monocytes, in blood, suggesting that T cells might fuel peripheral inflammation in obesity/T2D. Our lab’s work supports this idea by identification of a Th17 cytokine profile in T2D from T-cell stimulated peripheral blood mononuclear cells. Work described herein further supported this work by demonstrating that T cells dominate peripheral inflammation over monocytes across the spectrum of obesity and glycemic control. Helicobacter pylori is a recognized carcinogen and gastric colonization by strains that harbor the cag type IV secretion system (1) is the strongest known risk factor for stomach malignancy. Gastric adenocarcinoma is the fourth leading cause of cancer-related deaths world-wide ( > 700,000 deaths annually), with H. pylori directly contributing to the development of more than one million new cases of cancer per year accounting for 5.5% of all malignancies. Professionals of the child welfare system in Kentucky have continuously worked to retain and recruit new foster parents for the foster care system. Foster parents are uniquely placed in a surrogate caring position for children removed from their homes for reasons of abuse or neglect. Studying cancer evolution is an important task which contributes to better understanding of cancer biology and facilitates identification of new therapeutic targets. Among statistical intervals, confidence intervals and prediction intervals are well-known and commonly used. In many applications, the problem becomes finding an interval that covers at least a certain proportion $P$ of the population for a characteristic of interest with a specified confidence level $(1-\alpha)$. And such interval is named a $P$-content, $(1-\alpha)$-confidence Tolerance Interval (TI). The topic of the dissertation is the utility of tolerance intervals for various regression models. The exponentially-modified Gaussian (EMG) distribution is well-suited for analyzing data with positive skewness due to its characteristic positive skew from the exponential component. Despite its popularity in various fields, the EMG distribution has only been analyzed for univariate data without any regression settings. Adverse childhood experiences (ACEs) pose a major public health issue as they are associated with poor mental and physical health outcomes. In the United States (US), more than 60% of adults report at least one ACE, and around 15% experienced four or more. Repeated exposure to ACEs stressors can convert ACEs to toxic stress by altering the brain's architecture and function. Evidence suggests that exposure to four or more ACEs, which are indicators of exposure to toxic stressors, is associated with nicotine dependence (ND) and depressive symptoms. Ubiquitin (UB) and ubiquitin-like protein (UBL) pathways have emerged as important targets for oncology drug discovery based on the success of proteasome inhibitors (bortezomib or carfilzomib), E3 inhibitors, and the NEDD8 E1 inhibitor pevonedistat (MLN42924). Chemical inhibitors have also proven to be useful probes for identifying and dissecting multifactor UB and UBL regulatory networks. Toward this end, we have pursued approaches to target NEDD8 ligation to Cullins, through developing small molecule inhibitors of DCN1 (defective in Cullin Neddylation 1). Individuals residing in Appalachian regions have significant health disparities, including higher cancer incidence and mortality rates. Previous studies have addressed the impact of socioeconomic status and environmental risk factors on Appalachia cancer disparities, while few studies have evaluated genetic risk factors.
Germline whole exome sequencing samples from 7,078 individuals with cancer (759 Appalachians) were evaluated. Demographics and relatedness were assessed using KING. Ethnicity was verified by principal component analysis using TRACE, which included 6,034 individuals (85%) of European genetic ancestry. After QC filtering, 5,980 individuals were analyzed. Recent lung cancer research has led to advancements in molecular immunology, resulting in development of small molecule inhibitors, or immune checkpoint inhibitors, that propagate an anti-tumor T cell response. Over the last century, advancements in medicine have been made to improve cancer patients' outcomes. The discovery of new druggable targets is essential to achieving this goal. Defective in Cullin Neddylation 1 is a proven oncogenetic driver that may lead to better clinical outcomes when targeted. In efforts to further improve a series of novel small molecular inhibitors of the DCN1 and UBE2M protein-protein interaction, the Guy lab has designed and synthesized 26 new compounds. These compounds were designed to specifically target the UBE2M N-Acetyl sub-pocket on DCN1 that was not occupied by compounds previously described in the literature.

Gene therapy is currently being studied as a treatment for a variety of indications, including cancer, infectious disease, and cardiovascular diseases, among others. While many of the early treatments in the field involved the use of viral delivery methods, various safety, ethical, and financial concerns limit the potential uses of this methodology. As such, more recent research has focused on developing non-viral delivery platforms to alleviate some of the issues inherent in viral delivery. Methyltransferases (MTs) are ubiquitous enzymes commonly involved in biosynthesis and regulation. The fundamental goal of this thesis was to explore the permissive nature of methyltransferases and leverage this unique biocatalytic feature to develop a new platform for chemoselective intramolecular cyclization reactions. To realize this ambitious goal, this thesis project set out to probe the substrate specificity of multiple natural product tailoring enzymes (MTs and glycosyltransferases), identify model substrates common to multiple such tailoring enzymes and subsequently establish multi-enzyme tandem reactions that would set the stage for a subsequent chemoselective intramolecular cyclization reaction. Sepsis, or the organ damage that ensues after the body fails to properly contain a local infection, is the leading cause of in-patient hospitalization in the United States. Advances in critical care medicine over the last 20 years have enabled most sepsis patients to survive the life-threatening dysregulated immune response. However, a majority of survivors report chronic weakness and fatigue years after sepsis, and the cause of this weakness remains largely unknown. Pediatric brain cancer is a rare and potentially deadly diagnosis that affects children in Kentucky at a higher incidence than those nationally. Limited exposures in children early in life to warrant the development and progression of brain cancer point to the role parental exposures experienced in the period prior to conception and throughout pregnancy may play in its development. Many of these exposures may be experienced through occupation.

Air overpressure is a shock wave that occurs when explosives detonate and travel through the air. This can cause damage and annoyance in mining blasts, making it a significant concern for the surroundings of the operation.
