Characterization Of Cell Type-Specific Molecular Heterogeneity In Cancer Using Multi-Omic Approaches, Min Kyung Lee Jan 2023

Characterization Of Cell Type-Specific Molecular Heterogeneity In Cancer Using Multi-Omic Approaches, Min Kyung Lee

Dartmouth College Ph.D Dissertations

Tumors are composed of heterogeneous cell types each with its own unique molecular profiles. Recent advances in single cell genomics technologies have begun to increase our understanding of the molecular heterogeneity that exists in tumors with particular focus on gene expression and chromatin accessibility profiles. However, due to limitations in methods for certain sample types and high cost for single cell genomics, bulk tumor molecular profiling has been and remains widely used. In addition, other facets of single cell epigenomic profiling, particularly methylation and hydroxymethylation, remains underexplored. Thus, investigations to understand the cell type specific epigenetic heterogeneity and the cooperation …

Zinc Oxide Based Nanomaterials And Device Designs - For Bioenergy Harvesting, Sensing And Water Purification, Congran Jin

Dartmouth College Ph.D Dissertations

Biomechanical energy harvested from the human body can power implantable electronics such as cardiac pacemakers, replacing conventional batteries and offering a lifelong energy solution. The electrical signals converted from biomechanical motions contain vital physiological information, making them suitable as a sensor to monitor one's health condition. Zinc Oxide (ZnO) is a versatile material with various intriguing properties, including piezoelectricity, photo-catalytic and piezo-catalytic properties. This thesis aims to exploit and enhance these properties of ZnO by rationally designing its nano- and micron-structures and employing them, along with other functional materials, in the development of novel biomechanical energy harvesting, water purification, and …

100 Years Of International Ip - Reflections On Past, Present And Future, Frederick M. Abbott Jan 2023

100 Years Of International Ip - Reflections On Past, Present And Future, Frederick M. Abbott

Scholarly Publications

We have been asked to reflect on the past 100 years of international intellectual property law and to try to project forward about what changes might be necessary or desirable in the future. Only a science fiction writer would purport to have some idea about what things might look like a hundred years in the future, including from the standpoint of international intellectual property, so my remarks on that will be somewhat more proximate to the present.

The ‘Homework Gap’ And Academic Achievement In High School Science: An Ecological Perspective For Policymakers And Practitioners, Tianca Crocker, David Kleitsch

National Youth Advocacy & Resilience Journal

Information and communication technologies (ICTs) are increasingly required within K-12 educational settings, yet the impact of youth’s access to and use of digital resources outside of the classroom on academic achievement is only beginning to be explored in the literature. This study used data from the Fragile Families & Child Wellbeing Study to assess the relationship between grades in high school science and digital equity across the learner’s ecological environment. Digital inequities experienced by school-aged youth are referred to as the homework gap. The COVID-19 pandemic placed new urgency on resolving the homework gap as millions of students shifted to …

Microdebris Abundance, Distribution, And Ingestion By Sargassum-Associated Juvenile Fishes In The Gulf Of Mexico, Olivia L. Lestrade, Frank Hernandez Jan 2023

Microdebris Abundance, Distribution, And Ingestion By Sargassum-Associated Juvenile Fishes In The Gulf Of Mexico, Olivia L. Lestrade, Frank Hernandez

Gulf and Caribbean Research

Holopelagic Sargassum is a critical nursery habitat for the early life stages of many marine fishes, including several federally managed species in the United States and Caribbean. Sargassum is often aggregated along surface convergence features where microdebris (synthetic, semi—synthetic, and naturally—derived particles size) have also been found in relatively high concentrations. In this study, we collected microdebris from Sargassum and adjacent open water habitats (in 2018), and juvenile fishes from Sargassum (in 2017 and 2018) in the northern Gulf of Mexico to quantify habitat—specific microdebris concentrations and the degree to which Sargassum—associated juvenile fishes ingest microdebris. Microdebris concentrations within …

A Miocene Nannofossil Biostratigraphic Case Study: Alaminos Canyon Block 627 And Mississippi Canyon Block 555, And Sedimentation Rates In The Gulf Of Mexico, Bethany L. Cobb Faulk, Murlene W. Clark Jan 2023

A Miocene Nannofossil Biostratigraphic Case Study: Alaminos Canyon Block 627 And Mississippi Canyon Block 555, And Sedimentation Rates In The Gulf Of Mexico, Bethany L. Cobb Faulk, Murlene W. Clark

Gulf and Caribbean Research

The Miocene sediments of 2 deep—water boreholes from the northern Gulf of Mexico, from Alaminos Canyon (AC) Block 627 and Mississippi Canyon (MC) Block 555, have been biostratigraphically analyzed using calcareous nannofossils, revealing changes in sedimentation rates and depositional environments between these 2 areas. High nannofossil abundance values and low sedimentation rates generally recorded in the Alaminos Canyon region suggest a condensed section during much of the Miocene, associated with a basinal environment. Mississippi Canyon exhibits lower nannofossil abundance and higher sedimentation rates compared to Alaminos Canyon during the majority of the Miocene. Increased sediment volumes are largely attributed to …

Pancreatic Ductal Adenocarcinoma And New Treatments In Development, Melissa J. Moisan Jan 2023

Pancreatic Ductal Adenocarcinoma And New Treatments In Development, Melissa J. Moisan

Biology and Microbiology Graduate Students Plan B Research Projects

Pancreatic ductal adenocarcinoma (PDAC) is the most frequently mutated (95%) 5exocrine pancreatic cancers. Patients with PDAC are also the least likely to survive more than 5 years with only 8% overall survivability. The median survival for patients with distant metastasis is less than one year. Most pancreatic ductal adenocarcinoma occurs in the pancreatic head, and the remainder can be found within the body (5% to 15%) or tail (10% to 15%) (14). This extremely aggressive cancer has an increasingly large metastatic dissemination rate to surrounding organs such as the liver, lung, and peritoneum, or can be seen as distant metastases …

Role Of Histone Deacetylases In Regulation Of Macrophage Phagocytosis Of Mycobacterium Tuberculosis, Benjamin Esiesimua Jan 2023

Role Of Histone Deacetylases In Regulation Of Macrophage Phagocytosis Of Mycobacterium Tuberculosis, Benjamin Esiesimua

Biotechnology Theses

Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb), remains a global health emergency. Macrophages are critical in protection against TB by phagocytosis of Mtb. Although histone deacetylases (HDACs), are critical in macrophage differentiations, their roles in macrophage phagocytosis of Mtb remain unexplored. To explore this, I first established human monocyte derived macrophages (MDMs) phagocytosis of red zymosan beads. MDM phagocytosed zymosan beads at 37°C in a dose dependent manner in contrast to suppressed phagocytosis by the MDMs incubated with beads at 4°C or in the presence of cytochalasin D, actin polymerization inhibitor. Incubation of MDMs with 11 zinc-dependent HDAC inhibitors did …

Arginine-178 Is An Essential Residue For Itpa Function, Nicholas E. Burgis, Caitlin April, Kandise Vanwormer Jan 2023

Arginine-178 Is An Essential Residue For Itpa Function, Nicholas E. Burgis, Caitlin April, Kandise Vanwormer

Chemistry and Biochemistry Faculty Publications

The inosine triphosphate pyrophosphatase (ITPA) enzyme plays a critical cellular role by removing noncanonical nucleoside triphosphates from nucleotide pools. One of the first pathological ITPA mutants identified is R178C (rs746930990), which causes a fatal infantile encephalopathy, termed developmental and epileptic encephalopathy 35 (DEE 35). The accumulation of noncanonical nucleotides such as inosine triphosphate (ITP), is suspected to affect RNA and/or interfere with normal nucleotide function, leading to development of DEE 35. Molecular dynamics simulations have shown that the very rare R178C mutation does not significantly perturb the overall structure of the protein, but results in a high level of structural …

The incidence of empyema is increasing and associated with a mortality rate of 20% in patients older than 65 years. Since 30% of patients with advanced empyema have contraindications to surgical treatment, novel, low-dose, pharmacological treatments are needed. A Streptococcus pneumoniae-induced rabbit model of chronic empyema recapitulates the progression, loculation, fibrotic repair, and pleural thickening of human disease. Treatment with single chain (sc) urokinase (scuPA) or tissue type (sctPA) plasminogen activators in doses 1.0–4.0 mg/kg were only partially effective in this model. Docking Site Peptide (DSP; 8.0 mg/kg), which decreased the dose of sctPA for successful fibrinolytic therapy in acute …
