The purpose of this transcendental phenomenological study was to understand lived experiences in young adults with Asperger’s syndrome (AS) as they transitioned out of high school after completion of quality-of-life training. The theories guiding this study were Ivan Pavlov’s classical conditioning and Adam Morton’s theory of mind in predictive behaviors. Pavlov’s theory relates to understanding whether individuals could be classically conditioned to reinforce a natural reflex or some involuntary behavior that would occur as a response to classical conditioning. Morton’s theory helped focus this phenomenological study by describing the training in predictive behaviors by conceptual structure. Participants were selected from …

Neutralization assays like the plaque reduction neutralization test (PRNT) and focus reduction neutralization test (FRNT) are the most specific ways to measure neutralizing antibodies, which provide a good estimate of protection for RNA viruses. However, these assays are time-consuming, labor-intensive, and low-throughput. Alternative methods that are faster and convenient are necessary to quantify antibody responses in large-scale studies. In this dissertation, I develop a mix of computational and experimental methods to quantify antibody responses to dengue viruses (DENV) and SARS-CoV-2. Background: Enterococcus faecalis is the most commonly isolated enterococcal species in clinical infection. This bacterium is notorious for its ability to share genetic content within and outside of its species. With this increased proficiency for horizontal gene transfer, tremendous genomic diversity within this species has been identified. Many researchers have hypothesized E. faecalis exhibits niche adaptation to establish infections or colonize various parts of the human body. Here, we hypothesize that E. faecalis strains isolated from the human bladder will carry unique genomic content compared to clinical strains isolated from other sources. OBJECTIVES: Primary and post-primary tuberculosis (TB) are distinct entities. The aim of this study was to study the histopathology of primary and post-primary TB by using the unique human autopsy material from the pre-antibiotic era, 1931-1947.

MATERIAL AND METHODS: Autopsy data were collected from the autopsy journals, and the human tissue was collected from the pathology archives at the Department of Pathology, the Gades Institute.

RESULTS: Histological presentations of TB lesions showed great diversity within a single lung. Post-primary TB starts as a pneumonia forming early lesions, characterized by the infiltration of foamy macrophages containing mycobacterial antigens within alveoli, and …

This research was designed to investigate the metabolite profiling, phenolics, and flavonoids content as well as the potential nematicidal properties of decoction (ZpDe), orange-yellow resin (ZpRe) and essential oil (ZpEO) from Argentinean medicinal plant Zuccagnia punctata Cav. Additionally, the antioxidant and antibacterial properties of ZpDe and ZpEO were determined. Metabolite profiling was obtained by an ultrahigh-resolution liquid chromatography MS analysis (UHPLC-ESI-QTOF/OT-MS-MS) and GCMS. The nematicidal activity was assayed by a standardized method against Meloidogyne incognita. This study examined the normative messages that inform youth postsecondary decision making in a predominantly rural state in the northeastern U.S., focusing on the institutionalization and circulation of identity master narratives. Using a multilevel, ecological approach to sampling, the study interviewed 33 key informants in positions of influence in educational, workforce, and quality of life domains. Narrative analysis yielded evidence of a predominant master narrative – College for All – that participants described as a prescriptive expectation that youth and families orient their postsecondary planning toward four-year, residential baccalaureate degree programs. The common marmoset (Callithrix jacchus) is one of the most widely used nonhuman primate models of human disease. Owing to limitations in sequencing technology, early genome assemblies of this species using short-read sequencing suffered from gaps. In addition, the genetic diversity of the species has not yet been adequately explored. Using long-read genome sequencing and expert annotation, we generated a high-quality genome resource creating a 2.898 Gb marmoset genome in which most of the euchromatin portion is assembled contiguously (contig N50 = 25.23 Mbp, scaffold N50 = 98.2 Mbp). Hereditary spastic parapareses (HSPs) are clinically heterogeneous motor neuron diseases with variable age of onset and severity. Although variants in dozens of genes are implicated in HSPs, much of the genetic basis for pediatric-onset HSP remains unexplained. Here, we re-analyzed clinical exome-sequencing data from siblings with HSP of unknown genetic etiology and identified an inherited nonsense mutation (c.523C>T [p.Arg175Ter]) in the highly conserved RAB1A. The mutation is predicted to produce a truncated protein with an intact RAB GTPase domain but without two C-terminal cysteine residues required for proper subcellular protein localization. Several canonical translocations produce oncofusion genes that can initiate acute myeloid leukemia (AML). Although each translocation is associated with unique features, the mechanisms responsible remain unclear. While proteins interacting with each oncofusion are known to be relevant for how they act, these interactions have not yet been systematically defined. To address this issue in an unbiased fashion, we fused a promiscuous biotin ligase (TurboID) in-frame with 3 favorable-risk AML oncofusion cDNAs (PML::RARA, RUNX1::RUNX1T1, and CBFB::MYH11) and identified their interacting proteins in primary murine hematopoietic cells. Severe problems like climate change, pollution, and biodiversity loss demand urgent action. Unfortunately, people can tend to view such problems as distant, failing to feel that they are relevant to their lives. Psychological distance may play a role in such perceptions. In order to understand how environmental documentaries, which are often used to educate the public about the environment, may influence perceptions of psychological distance, environmental film festival attendees and organizers were interviewed. This research aims to evaluate how organizations manage their reputations with publics through compounding crises. The research applies concepts of public relations and crisis management, including situational crisis communication theory and organization-public relationship management. The research investigates a case of compounding health crises at a prominent American University, and assesses how communication managers prioritized publics and issue response, coordinated their communication strategies, and revised their communication practices in the wake of the compounding crises. In this essay, I interrogate Texas Attorney General Ken Paxton’s KP-0401, a 2022 legal opinion letter that frames gender affirming healthcare as dangerous, sterilizing, and therefore, child abuse. Within this framing, trans* youth are presented as both delusional and deviant, written off as victims of abuse and social contagion. In analyzing KP-0401 through a critical perspective of affect and performativity, I demonstrate how particular affects have gained power through their circulation to reify feelings and performances of cisheteronormativity, often through establishing the trans* body as a source of negative affect, like fear or repulsion, and trans* youth as infantile and …

Despite attending the same universities and working toward the same degrees, trans students and cisgender students do not always have the same perceptions of acceptance at their university. The transmale, gender nonconforming, and gender nonbinary students who participated in this study continue to experience a layer of rejection due to their gender identities and expressions. Congress first developed compassionate release in 1984, granting federal courts the authority to reduce sentences for “extraordinary and compelling” reasons. Compassionate release allows the Federal Bureau of Prisons (BOP) and inmates to apply for immediate early release on grounds of “particularly extraordinary or compelling circumstances which could not reasonably have been foreseen by the court at the time of sentencing.” Questions remain about how the BOP and the courts grant compassionate release and whether the courts apply the compassionate release guidelines consistently. In this brief, we examine the racial and socioeconomic disparities in ninth-grade advanced course placement. We utilized a statistical analysis of course placement patterns and interviewed Arkansas counselors. We suggest districts implement local norm-based placement systems that automatically enroll students in ninth-grade advanced courses to enhance equity in course placement.

Conservation agriculture and the use of cover crops are promising strategies for soil improvement in agricultural systems. These strategies are being promoted in Cambodia to address rapidly declining soil fertility; however, there is a lack of insight into the perceptions of Cambodian smallholders towards cover cropping within a conservation agriculture approach. N95 respirators are the most utilized respirator in the healthcare sector, and when properly fitted, protect workers from biological and chemical exposures. A respirator fit test is a protocol used to assess the seal between the user’s face and the respirator. Quantitative and Qualitative methods are both approved and used for respirator fit testing. Quantitative Condensation Nuclei Counting (CNC) uses instrumentation to measure the particles on the inside and outside of the respirator and calculates a fit factor, indicating how well the respirator fits. The World Heritage Program and nomination process have undergone considerable criticism regarding their impact on host nations. These critics have often described violence as an unintended side-effect of heritage sites, but this research has lacked a systematic analysis of types of violence associated with the inscription of sites. The background to this study explores the application of Galtung’s Triangle of Violence, which considers the symbiotic dynamics of cultural violence, structural violence, and direct violence. In so doing, I constructed the Measures of Structural Violence matrix by which to examine direct and structural violence in states that host heritage sites.
