Missense mutations in the DNA binding domain of p53 are characterized as structural or contact mutations based on their effect on the conformation of the protein. These mutations show gain-of-function (GOF) activities, such as promoting increased metastatic incidence compared with p53 loss, often mediated by the interaction of mutant p53 with a set of transcription factors. These interactions are largely context specific. To understand the mechanisms by which p53 DNA binding domain mutations drive osteosarcoma progression, we created mouse models, in which either the p53 structural mutant p53R172H or the contact mutant p53R245W are expressed specifically in osteoblasts, yielding osteosarcoma …

We analyzed findings in a same-gender couple discordant in their human immunodeficiency virus (HIV) status. The HIV+ partner was homozygous for CCR5 while his receptive HIV- partner was a CCR5Δ32 heterozygote with a C20S missense mutation in his CCR5 allele. The cells from the HIV- partner showed significant resistance to R5 fusion/infection and had no chemotactic response to CCL4 (macrophage inflammatory protein 1β). We demonstrated abundant CCR5-specific RNA in the HIV- partner's cells but no detectable CCR5 protein. CCR5 promoter region cloned from each partner's DNA indicated no significant impact on RNA transcription. The compound effect of CCR5Δ32 and C20S …

The quest for a non-hormonal male contraceptive pill for men still exists. Serine protease 37 (PRSS37) is a sperm-specific protein that when ablated in mice renders them sterile. In this study we sought to examine the molecular sequelae of PRSS37 loss to better understand its molecular function, and to determine whether human PRSS37 could rescue the sterility phenotype of knockout (KO) mice, allowing for a more appropriate model for drug molecule testing. To this end, we used CRISPR-EZ to create mice lacking the entire coding region of Prss37, used pronuclear injection to create transgenic mice expressing human PRSS37, intercrossed these …

Five Times More Likely: Haaland V. Brackeen And What It Could Mean For Maine Tribes, Eloise Melcher

Maine Law Review

In the 1970s Native activists realized that states were removing Native children from their families at disproportional rates when compared to non-Native children. The activists pushed for the enactment of the Indian Child Welfare Act, which became law in 1978. The law increases the burden on states before Native children can be taken from their families. As part of a larger movement to attack the Equal Protection Clause in the courts, Haaland v. Brackeen reached the Supreme Court in 2022. The plaintiffs in Brackeen argue that the Indian Child Welfare Act is unconstitutional for a variety of reasons, including that …

Mothers Born Or Produced?: An Analysis Of The Mother-Daughter Relationship In Well-Behaved Indian Women, Shivalika Agarwal, Nagendra Kumar

Journal of International Women's Studies

The word motherhood has been used for centuries without thorough examination of what it encompasses. Literature exhibits the changing reality and needs of mothering irrespective of the outcome: imposed motherhood, and institutionalized mothers. Motherhood has been bifurcated in meaning as “the potential relationship of any woman to her powers of reproduction and to children; and the institution, which aims at ensuring that that potential-- and all women--shall remain under male control” (Rich 13). A woman’s biological capacity to bear and nurture a child has been a significant factor in the existence of human life. Another facet of this is the …

Custodianship And Care: Women And Reading In Anita Desai’S Clear Light Of Day, Aruni Mahapatra

Journal of International Women's Studies

Several scholars have noted how the Indian state has been able to care for women only by placing them in custody of the family or the community, often overseen by male relatives. How do novels by Indian women writers intervene in this difficult social and legal problem? This paper answers this question by integrating feminist scholarship on the place of Indian women in postcolonial India with another scholarly tradition: the ethics of care. Conventionally, these two bodies of writing have not been in direct dialogue. This paper facilitates a conversation by close-reading Anita Desai’s Clear Light of Day, a novel …

Cellular signaling generates calcium (Ca2+) ions, which are ubiquitous secondary messengers decoded by calcium-dependent protein kinases, calcineurins, calreticulin, calmodulins (CAMs), and CAM-binding proteins. Previous studies in the model plant Arabidopsis thaliana have shown the critical roles of the CAM-BINDING PROTEIN 60 (CBP60) protein family in plant growth, stress responses, and immunity. Certain CBP60 factors can regulate plant immune responses, like pattern-triggered immunity, effector-triggered immunity, and synthesis of major plant immune-activating metabolites salicylic acid (SA) and N-hydroxypipecolic acid (NHP). Although homologous CBP60 sequences have been identified in the plant kingdom, their function and regulation in most species remain unclear. In …

Exon-Intron Boundary Inhibits M, Zhiyuan Luo, Qilian Ma, Shan Sun, Ningning Li, Hongfeng Wang, Zheng Ying, Shengdong Ke

Faculty Research 2023

Regional bias of N6-methyladenosine (m6A) mRNA modification avoiding splice site region, calls for an open hypothesis whether exon-intron boundary could affect m6A deposition. By deep learning modeling, we find that exon- intron boundary represses a proportion (12% to 34%) of m6A deposition at adjacent exons (~100 nt to splice site). Experiments validate that m6A signal increases once the host gene does not undergo pre-mRNA splicing to produce the same mRNA. Inhibited m6A sites have higher m6A enhancers and lower m6A silencers locally and show high heterogeneity at different exons genome- widely, with only a small proportion (12% to 15%) of …

Recent Advances In Image-Guided Locoregional Therapies For Primary Liver Tumors., Cody R Criss, Mina S Makary

Oncology Articles

Primary liver cancer is the leading cause of cancer-related deaths worldwide. with incidences predicted to rise over the next several decades. Locoregional therapies, such as radiofrequency or microwave ablation, are described as image-guided percutaneous procedures, which offer either a curative intent for early-stage hepatocellular carcinoma or bridging/downstaging for surgical resection or transplantation. Catheter-driven locoregional therapies, such as transarterial chemoembolization and radioembolization, induce tumor hypoxia, can be palliative, and improve survival for early-to-intermediate hepatocellular carcinoma and unresectable intrahepatic cholangiocarcinoma. Herein, we provide a comprehensive overview of the antineoplastic mechanisms underpinning locoregional therapies, different treatment approaches, and the current state of the …

Predicted growth in world population will put unparalleled stress on the need for sustainable energy and global food production, as well as increase the likelihood of future pandemics. In this work, we identify high-resolution environmental zones in the context of a changing climate and predict longitudinal processes relevant to these challenges. We do this using exhaustive vector comparison methods that measure the climatic similarity between all locations on Earth at high geospatial resolution relative to global-scale analyses. The results are captured as networks, in which edges between geolocations are defined if their historical climate similarities exceed a threshold. We apply …

Disease-Associated Mutation A554v Disrupts Normal Autoinhibition Of Dnmt1, Rebecca L. Switzer, Zach J. Hartman, Geoffrey R. Hewett, Clara F. Carroll

Faculty Journal Articles

DNA methyltransferase 1 (DNMT1) is the enzyme primarily responsible for propagation of the methylation pattern in cells. Mutations in DNMT1 have been linked to the development of adult-onset neurodegenerative disorders; these disease-associated mutations occur in the regulatory replication foci-targeting sequence (RFTS) domain of the protein. The RFTS domain is an endogenous inhibitor of DNMT1 activity that binds to the active site and prevents DNA binding. Here, we examine the impact of the disease-associated mutation A554V on normal RFTS-mediated inhibition of DNMT1. Wild-type and mutant proteins were expressed and purified to homogeneity for biochemical characterization. The mutation increased DNA binding affinity …

Trans-Formation: Establishing Interdisciplinary Support For The Transgender Experience Through Historical, Biological, And Religious Lenses, Gaymarie Vaughan

Graduate Liberal Studies Theses and Dissertations

Quite frankly, nothing could be more relevant to a modern discussion about what it means to be human than the emergence of discourse on transgender identity. By looking at the long history of human evolution, the impact that the progressing fields of science and medicine have had on human development, and the human need for bodily autonomy, it should be taken for granted that transgender issues belong in public discourse and that transgender people deserve to enjoy the same rights and freedoms all other humans share. This research advocates for best and most inclusive societal practices for transgender individuals by …

Trisomy 21, the genetic cause of Down syndrome (DS), is the most common congenital chromosomal anomaly. It is associated with a 20-fold increased risk of acute lymphoblastic leukemia (ALL) during childhood and results in distinctive leukemia biology. To comprehensively define the genomic landscape of DS-ALL, we performed whole-genome sequencing and whole-transcriptome sequencing (RNA-Seq) on 295 cases. Our integrated genomic analyses identified 15 molecular subtypes of DS-ALL, with marked enrichment of CRLF2-r, IGH::IGF2BP1, and C/EBP altered (C/EBPalt) subtypes compared with 2257 non-DS-ALL cases. We observed abnormal activation of the CEBPD, CEBPA, and CEBPE genes in 10.5% of DS-ALL cases via a …

1. Researchers can investigate many aspects of animal ecology through noninvasive photo–identification. Photo–identification is becoming more efficient as matching individuals between photos is increasingly automated. However, the convolutional neural network models that have facilitated this change need many training images to generalize well. As a result, they have often been developed for individual species that meet this threshold. These single-species methods might underperform, as they ignore potential similarities in identifying characteristics and the photo–identification process among species. 2. In this paper, we introduce a multi-species photo–identification model based on a state-of-the-art method in human facial recognition, the ArcFace classification head. …

The Need For Embodied Dramaturgy: The Laramie Project And Generation Z, Carin Silkaitis, Zoe Rose Kriegler-Wenk

Faculty, Staff, and Students

20 years after the murder of Matthew Shepard, I introduced The Laramie Project to a group of undergraduate students. Observing cast members' first experience of the characters, a troubling lack of empathy to the complexity of this tragic story became apparent. Of primary concern was that decades of progress made towards LGBTQIA equality stripped this story of its relevance. Geographic and generational bias is natural and expected but ultimately requires a new dramaturgical approach. We offer reflections on the methodology developed to address this gap: an embodied dramaturgical approach to our pre-production work culminating in a full cast and crew …

Investigating The Longitudinal Relationship Between Social Motivation And Depression In Autistic Adults, Robyn H. Himelstein

Theses and Dissertations

Autism affects individuals across the lifespan, yet there tends to be limited research and services for autistic adults. This is especially concerning given that autistic adults have high mental health needs, with depression being one of the most common and clinically significant co-occurring conditions. We explored the longitudinal relationships between social motivation, social access (i.e., having opportunities for meaningful social interactions), loneliness, and depression in N=303 autistic adults ages 18-65. Participants completed online surveys about social behavior and wellbeing three times over 3–4 months. We hypothesized that an interaction between higher social motivation and lower social access at Time 1 …

BACKGROUND: Trauma and a subsequent hemorrhagic shock (T/HS) result in insufficient oxygen delivery to tissues and multiple organ failure. Extracellular adenosine, which is a product of the extracellular degradation of adenosine 5' triphosphate (ATP) by the membrane-embedded enzymes CD39 and CD73, is organ protective, as it participates in signaling pathways, which promote cell survival and suppress inflammation through adenosine receptors including the A

METHODS: T/HS shock was induced by blood withdrawal from the femoral artery in wild-type, global knockout (CD39, CD73, A

RESULTS: T/HS upregulated the expression of CD39, CD73, and the A

CONCLUSION: In conclusion, the CD39-CD73-A

The Qualitative Report

This study explores lecturers' perceptions and suggestions on integrating gamified lessons in Malaysian higher institutions. The COVID-19 pandemic has drastically changed the norm of traditional classroom teaching by accelerating digital integration amongst educators and necessitating the need to address classroom gamification. To further understand educators' perceptions, we conducted in-depth interviews with 25 lecturers. We thematically analyzed the interviews by following the steps undertaken by Braun and Clarke (2006) to identify the emerging themes. The findings determine that educators found gamification suitable in teaching and learning activities during the set induction, reinforcement and assessment of specific skills. Several misconceptions also evident …
