Mitochondrial Phosphatidylethanolamine Modulates Ucp1 To Promote Brown Adipose Thermogenesis, Jordan M Johnson, Irfan J Lodhi, Et Al. Feb 2023

Mitochondrial Phosphatidylethanolamine Modulates Ucp1 To Promote Brown Adipose Thermogenesis, Jordan M Johnson, Irfan J Lodhi, Et Al.

2020-Current year OA Pubs

Thermogenesis by uncoupling protein 1 (UCP1) is one of the primary mechanisms by which brown adipose tissue (BAT) increases energy expenditure. UCP1 resides in the inner mitochondrial membrane (IMM), where it dissipates membrane potential independent of adenosine triphosphate (ATP) synthase. Here, we provide evidence that phosphatidylethanolamine (PE) modulates UCP1-dependent proton conductance across the IMM to modulate thermogenesis. Mitochondrial lipidomic analyses revealed PE as a signature molecule whose abundance bidirectionally responds to changes in thermogenic burden. Reduction in mitochondrial PE by deletion of phosphatidylserine decarboxylase (PSD) made mice cold intolerant and insensitive to β3 adrenergic receptor agonist-induced increase in whole-body oxygen …

Field Testing Of A Turnip Growing Protocol On New Zealand Dairy Farms, J. P. J. Eerens, P. M. S. Lane

IGC Proceedings (1977-2023)

Summer droughts are a regular occurrence in central North Island districts of New Zealand, which causes pastures to wilt, lose their nutritive value and stop growing. The resulting summer feed gap depresses farm productivity (Clark et al., 1996). Turnips optimally sown mid-late October (more often sown in November or even December) are grown to fill this feed gap. Recorded average yields of 7.4 t dry matter (DM)/ha are below the economic breakeven point of 8-10 t DM/ha (Clark et al., 1996). A turnip growing protocol was developed from published data (Eerens & Lane 2004) and tested on commercial …

Use Of Alkanes To Estimate Dry Matter Intake Of Beef Steers Grazing High Quality Pastures, G. Scaglia, H. T. Boland, I. Lopez-Guerrero, R. K. Shanklin, J. P. Fontenot

IGC Proceedings (1977-2023)

Pastures remain the most important source of nutrients for ruminant livestock and nutrition is critical to optimize animal production. The daily quantity of dry matter that is consumed by an animal is a critical measurement to make nutritional inferences about feed and subsequent animal response. Researchers are facing the dilemma that, while estimates of individual animal performance are readily obtained, it is still difficult to estimate the herbage intake of individual animals. The objectives of this experiment were to estimate forage intake in beef steers grazing tall fescue (Festuca arundinacea Schreb.) and alfalfa (Medicago sativa)/tall fescue pastures …

Detection And Capture Of Nitrogen Fixing Bacteria From Soil And Roots Using Plant Specific Lectins, Alana Horton Feb 2023

Detection And Capture Of Nitrogen Fixing Bacteria From Soil And Roots Using Plant Specific Lectins, Alana Horton

Natural Sciences and Mathematics | Biological Sciences Master's Theses

Diazotrophs increase plant productivity by providing ammonia directly to plants in the rhizosphere or through association with the roots (Sun et al., 2021). Leveraging diazotrophs can aid in the reduction or application of harmful synthetic nitrogen fertilizers. This thesis aims to use plant root proteins, or lectins, to more rapidly recover viable diazotrophs that are likely to associate with crops. In this study, lectins, are used to recover bacteria from bulk soil by taking advantage of the binding relationship between lectins and bacterial cell surface polysaccharides. When commercially-available lectins were used with a magnetic pulldown to separate the lectin, bacterial …

Objectives And Benefits Of Molecular Breeding In Forage Species, T. Lübberstedt Feb 2023

Objectives And Benefits Of Molecular Breeding In Forage Species, T. Lübberstedt

IGC Proceedings (1977-2023)

  1. The amount of resources and information provided by forage crop genomic programs has dramatically increased during the past few years.
  2. Trait-based forward genetic procedures such as mapping and expression profiling have successfully provided new candidate genes or genome regions affecting forage quality. Respective information can easily be transferred across related forage species.
  3. Since several genes in major biochemical pathways related to forage traits have been isolated, gene-based reverse genetic approaches (transformation, association studies) are promising.
  4. Most genetic experiments are conducted under simplified "artificial" conditions such as on single-spaced plants. Therefore, transferability of respective genetic information to breeding practice needs to …

Culture Of Wellness Toward Resiliency, Shelby Stemig Feb 2023

Culture Of Wellness Toward Resiliency, Shelby Stemig

Master of Arts in Criminal Justice Leadership

Stress and burnout are increasingly prevalent amongst law enforcement officers and civilian staff due to job demands and job-related traumas. A culture of wellness planning is how administrations can build resiliency against stress and burnout. A wellness plan should emphasize organizational responsibility, officer responsibility, formal and informal leadership, and external collaborations. Agencies can also utilize the same cognitive behavioral therapies that supervised release agents find beneficial for clients. There are great similarities between the utilization of cognitive behavioral interventions, evidence-based models, and thought behavioral links between supervised release clients and law enforcement staff members. Often, cognitive behavioral therapies are used …

Exploring The Existence Of Transformational Education In The Nys Social Studies Curriculum And Civic Readiness Initiative, Gregory Vincent Massara

Educational Policy & Leadership Other Graduate Student Scholarship

The enormity and perniciousness of economic, social, and environmental sustainability challenges are evident through several avenues. The Intergovernmental Panel on Climate Change is issuing increasingly dire warnings for the future of life on Earth. Threats to social cohesion, evidenced particularly in the institutionalization of injustices and the current fragility of democracy in the United States, highlight the need for increased social cohesion. Furthermore, rising economic inequality, inequities based on gender and race, and the increasing cost of damages caused by climate events highlight economic challenges. In 2015, the United Nations put forth an agenda to address economic, social, and environmental …

Holistic Framework To Contextualize Dietary Quality Assessment: A Critical Review, Jessica M. Phelan, Richard R. Rosenkranz, Connor J. Phelan, Sara K. Rosenkranz

Kinesiology and Nutrition Sciences Faculty Research

Numerous dietary quality indices exist to help quantify overall dietary intake and behaviors associated with positive health outcomes. Most indices focus solely on biomedical factors and nutrient or food intake, and exclude the influence of important social and environmental factors associated with dietary intake. Using the Diet Quality Index- International as one sample index to illustrate our proposed holistic conceptual framework, this critical review seeks to elucidate potential adaptations to dietary quality assessment by considering—in parallel—biomedical, environmental, and social factors. Considering these factors would add context to dietary quality assessment, influencing post-assessment recommendations for use across various populations and circumstances. …

Analysis And Technical Optimization Of Processing Condition For Better Quality Of Robusta Coffee Production, Usman Ahmad, Khalimatus Sa’Diyah, Sathivel Subramaniam

Faculty Publications

The wet processing of robusta coffee starts with harvesting coffee cherries, followed by sorting the cherries, pulping, fermentation, washing, drying, hard skin hulling, bean sorting, and packaging. Soaking fruit before pulping is an alternative for coffee farmers to delay the pulping process if time does not al-low it. The fermentation process aims to remove mucus that is firmly attached to the hard skin. The addition of L. casei bacteria can accelerate the process but affect the final taste. This study aimed to optimize the duration of soaking and fermentation processes for obtaining coffee beans with a minimum number of unpeeled …

Alfalfa In Rotation With Annual Crops Reduced Nitrate Leaching Potential, Arshdeep Singh, Tahseen Afzal, Bryan Woodbury, Charles Wortmann, Javed Iqbal

Department of Agronomy and Horticulture: Faculty Publications

Rotation of perennial alfalfa (Medicago sativa L.) with annual crops has the potential to reduce nitrate-nitrogen (NO3-N) in the vadose zone and increase soil organic carbon (SOC) sequestration. The objective of this study was to determine the long-term effects on SOC, NO3-N, ammonium-N (NH4-N), and soil water in the 7.2 m depth with an alfalfa rotation compared with continuous corn (Zea mays L.). Soils from six pairs of alfalfa rotation versus continuous corn observation points were sampled to 7.2 m depth in 0.3 m increments. The uppermost 0.3 m was divided into …

Introduction: Allergen-specific immunotherapy (IT) is emerging as a viable option for treatment of peanut allergy. Yet, prophylactic IT remains unexplored despite early introduction of peanut in infancy was shown to prevent allergy. There is a need to understand how allergens interact with the immune system depending on the route of administration, and how different dosages of allergen may protect from sensitisation and a clinical active allergy. Here we compared peanut allergen delivery via the oral, sublingual (SL), intragastric (IG) and subcutaneous (SC) routes for the prevention of peanut allergy in Brown Norway (BN) rats.

Methods: BN rats were …

Selenium Nanoparticles Attenuate Bleomycin-Induced Pulmonary Oxidative Stress And Fibrosis In Rats, Azza M A. Abouelella, Bahig A. El Deeb, Eman Mohammed Ali

Bulletin of Faculty of Pharmacy Cairo University

Pulmonary fibrosis is a fatal progressive lung disorder. Bleomycin (BLM) has been used for induction of experimental pulmonary fibrosis. Selenium nanoparticles (SeNPs) have been synthesized and investigated for their ability to ameliorate BLM-induced pulmonary fibrosis. SeNPs (0.5 mg⁄kg. ip) were administered for four weeks; two weeks pre and two weeks post BLM (2.5 U⁄kg. ip). BLM induced biochemical and histological deterioration of lungs. SeNPs induced significant histopathological and biochemical improvement of oxidative stress (MDA, SOD and GSH), inflammatory (IL6 and IL10) and fibrotic markers (hydroxyproline). In conclusion; SeNPs has potential therapeutic rather than protective effect on management of pulmonary damage.

Soybean Seed Protein Concentration Is Limited By Nitrogen Supply In Tropical And Subtropical Environments In Brazil, E. H. Figueiredo Moura Da Silva, N. Cafaro La Menza, G. G. Munareto, A. J. Zanon, Santos Carvalho, F. R. Marin

Department of Agronomy and Horticulture: Faculty Publications

Soybean production contributes to ca. 60% of global plant-based protein used for food and feed. Brazil is the largest soybean producer and exporter, with 60% from tropical and 40% from subtropical environments. Nitrogen (N) can play an essential role in the storage of proteins in seeds; thus, it could be a key factor in increasing the quantity and quality of seeds in high-yielding soybean crops. Unlike in temperate environments, there is a gap of knowledge on whether soybean grown under tropical and subtropical climates are limited by N-fertilization to sustain the seed yield increase without detriments in seed protein concentration. …

Pakistan’s economic situation requires fresh thinking and a clear approach. The blame game where each incoming government passes the buck to its predecessor has reached saturation level. People expect the incumbents to show them the path of relief to resolve their present difficulties, particularly inflation. All governments are scared of taking tough, but necessary decisions, for fear of alienating the awam (public), attacks by the opposition parties and media, and erosion of their political capital. Let’s see how this fear is misplaced.

For a continuous healthcare or environmental monitoring system, it is essential to reliably sense the analyte concentration reported by electrochemical sensors. However, environmental perturbation, sensor drift, and power-constraint make reliable sensing with wearable and implantable sensors difficult. While most studies focus on improving sensor stability and precision by increasing the system’s complexity and cost, we aim to address this challenge using low-cost sensors. To obtain the desired accuracy from low-cost sensors, we borrow two fundamental concepts from communication theory and computer science. First, inspired by reliable data transmission over a noisy communication channel by incorporating redundancy, we propose to measure …

Obesity is a major risk factor for end-stage kidney disease. We previously found that lysosomal dysfunction and impaired autophagic flux contribute to lipotoxicity in obesity-related kidney disease, in both humans and experimental animal models. However, the regulatory factors involved in countering renal lipotoxicity are largely unknown. Here, we found that palmitic acid strongly promoted dephosphorylation and nuclear translocation of transcription factor EB (TFEB) by inhibiting the mechanistic target of rapamycin kinase complex 1 pathway in a Rag GTPase-dependent manner, though these effects gradually diminished after extended treatment. We then investigated the role of TFEB in the pathogenesis of obesity-related kidney …

Adenosine 50 triphosphate (ATP) is the energy currency of life, which is produced in mitochondria (~90%) and cytosol (less than 10%). Real-time effects of metabolic changes on cellular ATP dynamics remain indeterminate. Here we report the design and validation of a genetically encoded fluorescent ATP indicator that allows for real-time, simultaneous visualization of cytosolic and mitochondrial ATP in cultured cells. This dual-ATP indicator, called smacATPi (simultaneous mitochondrial and cytosolic ATP indicator), combines previously described individual cytosolic and mitochondrial ATP indicators. The use of smacATPi can help answer biological questions regarding ATP contents and dynamics in living cells. As expected, 2-deoxyglucose …

Asprosin, a recently identified adipokine, activates agouti-related peptide (AgRP) neurons in the arcuate nucleus of the hypothalamus (ARH) via binding to protein tyrosine phosphatase receptor δ (Ptprd) to increase food intake. However, the intracellular mechanisms responsible for asprosin/Ptprd-mediated activation of AgRPARH neurons remain unknown. Here, we demonstrate that the small-conductance calcium-activated potassium (SK) channel is required for the stimulatory effects of asprosin/Ptprd on AgRPARH neurons. Specifically, we found that deficiency or elevation of circulating asprosin increased or decreased the SK current in AgRPARH neurons, respectively. AgRPARH-specific deletion of SK3 (an SK channel subtype highly expressed in AgRPARH neurons) blocked asprosin-induced …
