As Immune checkpoint inhibitors are being expanded for use in gynecologic malignancies, rare immune-related adverse events are more frequently being reported. Here we describe a 63-year-old with Stage IIIB mismatch repair deficient uterine adenocarcinoma who underwent six cycles of carboplatin and paclitaxel with partial response but persistent disease. She was then started on single agent pembrolizumab. After six cycles of pembrolizumab, she developed bilateral vision changes and was diagnosed with posterior scleritis. Pembrolizumab was held and she was treated with oral prednisone, with rapid resolution of symptoms. One month after completion of prednisone, vision changes were again reported and she …

Tensor factorization has received increasing interest due to its intrinsic ability to capture latent factors in multi-dimensional data with many applications including Electronic Health Records (EHR) mining. PARAFAC2 and its variants have been proposed to address irregular tensors where one of the tensor modes is not aligned, e.g., different patients in EHRs may have different length of records. PARAFAC2 has been successfully applied to EHRs for extracting meaningful medical concepts (phenotypes). Despite recent advancements, current models' predictability and interpretability are not satisfactory, which limits its utility for downstream analysis. In this paper, we propose MULTIPAR: a supervised irregular tensor factorization …

Most uveal melanoma cases harbor activating mutations in either GNAQ or GNA11. Despite activation of the mitogen-activated protein kinase (MAPK) signaling pathway downstream of Gαq/11, there are no effective targeted kinase therapies for metastatic uveal melanoma. The human genome encodes numerous understudied kinases, also called the "dark kinome". Identifying additional kinases regulated by Gαq/11 may uncover novel therapeutic targets for uveal melanoma. In this study, we treated GNAQ-mutant uveal melanoma cell lines with a Gαq/11 inhibitor, YM-254890, and conducted a kinase signaling proteomic screen using multiplexed-kinase inhibitors followed by mass spectrometry. We observed downregulated expression and/or activity of 22 kinases. …

Localization, Tissue Biology, And T Cell State — Implications For Cancer Immunotherapy, Jason M Schenkel, Kristen E Pauken

Faculty, Staff and Student Publications

Tissue localization is a critical determinant of T cell immunity. CD8+ T cells are contact-dependent killers, which requires them to physically be within the tissue of interest to kill peptide-MHC class I-bearing target cells. Following their migration and extravasation into tissues, T cells receive many extrinsic cues from the local microenvironment, and these signals shape T cell differentiation, fate and function. Because major organ systems are variable in their functions and compositions, they apply disparate pressures on T cells to adapt to the local microenvironment. Additional complexity arises in the context of malignant lesions (either primary or metastatic), and this …

Performing Motorized-To-Non-Motorized Crash Analysis Using Multi-Model Lbs Traffic Data Calibrated Through Random Forest Models, Nicholas Aldridge

Department of Civil and Environmental Engineering: Dissertations, Theses, and Student Research

Traditionally, traffic prediction has been an engineering challenge due to insufficient quantities of data. In this thesis, the use of a new type of data called location-based service (LBS) data from StreetLight (StL) and a large database of multi-modal traffic counts are used to infer vehicle, pedestrian, and bicycle volumes on all road segments in Lincoln, Nebraska. Additional land use features are incorporated into the model from the EPA Smart Location Database (EPA-SLD). The use of random forest models provides strong measures of fit for all modes, with R2 scores of 0.98 for both the bicycle and pedestrian modes …

Stopping The Revolving Door: Mdp-Based Decision Support For Community Corrections Placement, Xiaoquan Gao, Pengyi Shi, Nan Kong

Research Collection Lee Kong Chian School Of Business

Community corrections (CC) programs offer an alternative to incarceration that can reduce jail overcrowding and recidivism rates. The aim is to address the root causes behind criminal behavior, ultimately breaking the cycle of reincarceration. However, placing all eligible individuals in CC may strain case managers, resulting in reduced supervision, increased violations, and higher recidivism rates, which undermines the intended benefits for all participants in the programs. We take the first step in building a comprehensive analytical framework based on a queueing system to support the placement decisions and related decisions such as capacity planning. We develop a Markov Decision Process …

Globally, crickets are gaining recognition as a valuable alternative protein source for human consumption due to their lower resource requirement and ecological footprint compared to traditional livestock. In this paper, we examine strategies that may expedite the sustainable domestication of crickets as a food source. Using survey data from 306 households in western Kenya, we find that supplying cricket production starter kits, granting access to credit facilities, encouraging participation in farmer groups, and fostering partnerships can enhance the adoption of cricket farming. Moreover, we provide new evidence that institutional training significantly increases cricket yields while embracing cricket consumption (i.e. entomophagy) …

Objectives

Sudden unexpected death in epilepsy (SUDEP) is a leading cause of death for patients with epilepsy; however, the pathophysiology remains unclear. Focal‐to‐bilateral tonic–clonic seizures (FBTCS) are a major risk factor, and centrally‐mediated respiratory depression may increase the risk further. Here, we determined the volume and microstructure of the amygdala, a key structure that can trigger apnea in people with focal epilepsy, stratified by the presence or absence of FBTCS, ictal central apnea (ICA), and post‐convulsive central apnea (PCCA).

Methods

Seventy‐three patients with focal impaired awareness seizures without FBTC seizures (FBTCneg group) and 30 with FBTCS (FBTCpos group) recorded during …
