This paper explores research that discusses the impact of a culturally responsive education and lack-of for LGBTQ+ students and recommends a solution to ensure all students are safe and represented in schools. The research cited varies in their definitions and uses of the term LGBTQ+. We report a case of myeloproliferative neoplasm, not otherwise specified (MPN-NOS)-transformed AML with BCR::JAK2 rearrangement. Chromosomal analysis indicated a simple abnormal karyotype 46,XY,t(7;17)(q21;q24),t(9;22)(p24;q11.2). Fluorescence in situ hybridization (FISH) using a BCR/ABL1/ASS1 probe set suggested a possible BCR rearrangement and a reflex JAK2 breakapart probe indicated JAK2 rearrangement, most likely partnered with BCR. Optical genome mapping (OGM) analysis confirmed BCR::JAK2 derived through an inv(9)(p24p13) after a t(9;22)(p13;q11.2) in this case. Due to the complexity of chromosomal aberrations, disruption and/or rearrangement of other genes such as KIF24::BCR, JAK2::KIF24/UBAP1, and CDK6:SOX9 were also identified by OGM. Type II topoisomerases (TOP2) form transient TOP2 cleavage complexes (TOP2ccs) during their catalytic cycle to relieve topological stress. TOP2ccs are covalently linked TOP2-DNA intermediates that are reversible but can be trapped by TOP2 poisons. Trapped TOP2ccs block transactions on DNA and generate genotoxic stress, which are the mechanisms of action of TOP2 poisons. How cells avoid TOP2cc accumulation remains largely unknown. In this study, we uncovered RAD54 like 2 (RAD54L2) as a key factor that mediates a TOP2-specific DNA damage avoidance pathway. RAD54L2 deficiency conferred unique sensitivity to treatment with TOP2 poisons. RAD54L2 interacted with TOP2A/TOP2B and ZATT/ZNF451 and promoted …

Nexavant was reported as an alternative to the TLR3 agonist of Poly(I:C) and its derivatives. The physicochemical properties, signaling pathways, anti-cancer effects, and mechanisms of Nexavant were investigated. The distinctive characteristics of Nexavant compared to that of Poly(I:C) were demonstrated by precise quantification, enhanced thermostability, and increased resistance to RNase A. Unlike Poly(I:C), which activates TLR3, RIG-I, and MDA5, Nexavant stimulates signaling through TLR3 and RIG-I but not through MDA5. Compared to Poly(I:C), an intratumoral Nexavant treatment led to a unique immune response, immune cell infiltration, and suppression of tumor growth in various animal cancer models. Malignant pleural effusions (MPEs) can be utilized as liquid biopsy for phenotyping malignant cells and for precision immunotherapy, yet MPEs are inadequately studied at the single-cell proteomic level. Here we leverage mass cytometry to interrogate immune and epithelial cellular profiles of primary tumors and pleural effusions (PEs) from early and late-stage non-small cell lung cancer (NSCLC) patients, with the goal of assessing epithelial-mesenchymal transition (EMT) and mesenchymal-epithelial transition (MET) states in patient specimens. We evaluated two models to link stressful life events (SLEs) with the psychopathology of schizophrenia spectrum disorders (SSD). We separated SLEs into independent (iSLEs, unlikely influenced by one's behavior) and dependent (dSLEs, likely influenced by one's behavior). Stress-diathesis and stress generation models were evaluated for the relationship between total, i- and d- SLEs and the severity of positive, negative, and depressive symptoms in participants with SSD. Clear cell sarcoma (CCS) is a rare, aggressive malignancy that most frequently arises in the soft tissues of the extremities. It is defined and driven by expression of one member of a family of related translocation-generated fusion oncogenes, the most common of which is EWSR1::ATF1. The EWSR1::ATF1 fusion oncoprotein reprograms transcription. However, the binding distribution of EWSR1::ATF1 across the genome and its target genes remain unclear. Here, we interrogated the genomic distribution of V5-tagged EWSR1::ATF1 in tumors it had induced upon expression in mice that also recapitulated the transcriptome of human CCS. BACKGROUND: Schwannomas are common peripheral nerve sheath tumors that can cause severe morbidity given their stereotypic intracranial and paraspinal locations. Similar to many solid tumors, schwannomas and other nerve sheath tumors are primarily thought to arise due to aberrant hyperactivation of the RAS growth factor signaling pathway. Here, we sought to further define the molecular pathogenesis of schwannomas.
METHODS: We performed comprehensive genomic profiling on a cohort of 96 human schwannomas, as well as DNA methylation profiling on a subset. The gay rights movement has been very active on social media throughout the years. Using a trend analysis this study aimed to answer how social media is being used during a social movement, how a social movement evolves on social media, and how social media is being used by organizations as a public relations strategy to create change in social movements. Overall, the findings revealed 11 different ways social media is being used during a social movement, which shows how important opinion leaders are in helping a social movement gain traction and create the desired impact.

BACKGROUND: The right ventricle (RV) plays a central role in the maintenance of effective cardiac pump function. Despite overwhelming evidence that perioperative RV dysfunction (RVD) and failure (RVF) are associated with poor clinical outcomes, there are very few published recommendations or guidelines for comprehensive, evidence-based RV assessment on the risk of developing either during the perioperative period.

FtsZ is an essential bacterial protein abundantly studied as a novel and promising target for antimicrobials. FtsZ is highly conserved among bacteria and mycobacteria, and it is crucial for the correct outcome of the cell division process, as it is responsible for the division of the parent bacterial cell into two daughter cells. Disruption of epithelial barriers is a common disease manifestation in chronic degenerative diseases of the airways, lung, and intestine. Hypertension is a risk factor for major cardiovascular events and it is usually detected and managed by general medical practitioners (GPs) in primary care. However, it is estimated that 4.8 million adults are living with untreated high blood pressure (BP) in the UK. Health authorities are encouraging more collaborative work across health professions to find and refer individuals with undiagnosed hypertension. Students of Color study abroad at rates lower than their white peers, thus missing out on the academic, personal, and intercultural benefits one can gain from studying abroad. The purpose of this thesis was to conduct a qualitative case study to examine how a study abroad program supports its Students of Color during the pre-departure phase of study abroad. This study sought insight into the religious and cultural identity of emerging adults belonging to the Church of Jesus Christ of latter-day Saints(LDS) and the impact such identity had in relation to volunteering motivation. Participants included 24 emerging adult college students belonging to the Church. Thematic analysis was used to determine influencing factors that led to Church membership and how Church membership shaped self-perception. Participants shared volunteer experiences and motivation to volunteer in relation to Deci and Ryan’s (2000) self-determination theory (SDT) was examined. Amidst the amphibian extinction crisis, in situ and ex situ amphibian species conservation initiatives utilize assisted reproductive technologies for optimal genetic management of captive and wild populations. Development of effective, simple, low-cost methods for obtaining gametes for artificial fertilization, sperm biobanking, or natural breeding alleviates obstacles for institutions or programs to apply reproductive technologies. Objectives herein include investigating the efficacy of alternative hormones for inducing spermiation, an alternative hormone administration route for inducing ovulation, exploring the physiological effects of hormone therapy, and utilizing sperm from deceased animals for biobanking.

Teachers want to be in schools where their identities are authentically affirmed. However, some educators in predominantly Black schools perceive that they are not always afforded the same level of value and respect as their colleagues in mostly White or mixed schools. Instead, teachers in predominantly Black schools seem to have experiences associated with frustrations that often go unheard of or dismissed for various reasons. Schools with predominantly Black student populations are often associated with challenges in retaining teachers, commonly referred to as teacher retention or teacher turnover. The digenetic trematode, Bolbophorus damnificus, poses a substantial threat to catfish aquaculture, causing significant economic losses. Infestations lead to suppressed feed consumption, secondary bacterial infections and poor production performance. Survey data reveals widespread infestation in the in the southeastern United States. Current control strategies involve the application of a concentrated copper sulfate solution to reduce snail populations which serve as the first intermediate host of the trematode life cycle. This study aimed to improve treatment efficacy by developing a granular copper sulfate application system. This study examines the impact of a bus driver attendance incentive program implemented in a rural Mississippi school district with the aim of improving bus driver attendance rates. There is a shortage of bus drivers across Mississippi which has made bus driver attendance paramount. Bus driver absenteeism causes disruptions in student transportation services and affects overall school operations. To address this problem, a rural Mississippi school district introduced a bus driver attendance incentive program that provided financial incentives for bus drivers who maintained perfect attendance rates each month.

High-amplitude freestream turbulence and surface roughness elements can excite a laminar boundary-layer flow sufficiently enough to cause streamwise-oriented vortices to develop. These vortices resemble elongated streaks having alternate spanwise variations of the streamwise velocity. Following the transient growth phase, the fully developed vortex structures downstream undergo an inviscid secondary instability mechanism and, ultimately, transition to turbulence. This mechanism becomes much more complicated in high-speed boundary layer flows due to compressibility and thermal effects, which become more significant for higher Mach numbers. The availability of a scalable and explainable rule extraction technique via motif discovery is crucial for identifying the health states of a system. This study evaluates the spatial and temporal impacts of implemented Best Management Practices (BMP) on stream water quality under baseflow conditions in a stream segment affected by livestock production. The BMPs includes 10-m fenced riparian zone and crossing paths along the reach segment. Grab water samples and water quality monitoring were collected biweekly from July 2019 to March 2022. Water quality parameters included temperature, dissolved oxygen, electric conductance, total dissolved solids, turbidity, and pH. Water samples were lab analyzed for sediments and nutrients. Results evidenced poor water quality before BMPs implementation, with nutrient concentrations exceeding the nation's and state's criteria. …

Fibrous air filters are commonly used to capture airborne particles due to their potential for a relatively high capture efficiency and low airflow resistance. Salamanders are one of the most at-risk taxa in the world due to habitat destruction, pollution, climate change, invasive diseases, and more. This has led to a need for conservation breeding programs that are often associated with zoos and aquaria. Salamanders can be difficult to breed in captivity, though, due to their dependence on specific environmental cues and other unknown factors that stimulate sperm and egg production. By the early 20th century, in 1920, Black farmers owned 14% of US farmland. Today, in the 21st century, Black farmers own less than 2% of US farmland. The demise of Black farmers and Black farmland in US Agriculture is a direct result of social, political, and racial weaponization against their foodways, culture, and livelihoods. The history concerning the plight of Black farmers goes beyond USDA's historical discrimination but enters a position where racism is embedded and perpetuated within the structure of US agriculture. “I Tell It Like It Is, Then How It Could Be”: Public Discourse And Public Administration In Popular American Music Since The 1990s, Kristen Dechert

This study is an analysis of lyrics from popular American music albums since the 1990s to understand public discourse about and experience with public administration. Albums included in the study came from two prominent sources: Billboard and Pitchfork decade-end lists. The study is framed by critical, postmodern, and responsiveness theories, methodologically grounded in critical discourse analysis, and technically approached using standard qualitative methods, including especially qualitative content analysis and iterative, inductive coding techniques. Findings from the qualitative study demonstrate public discourse on the themes of institutional and systemic racism and violence within and perpetuated by public administration and public institutions. …
