A Dnak Chaperone System Connects Type Iv Pilus Activity To Polysaccharide Secretion In The Cyanobacterium Nostoc Punctiforme, Heather J. Mcdonald Jan 2023

A Dnak Chaperone System Connects Type Iv Pilus Activity To Polysaccharide Secretion In The Cyanobacterium Nostoc Punctiforme, Heather J. Mcdonald

University of the Pacific Theses and Dissertations

Type IV pili (T4P) systems are widely utilized among bacteria to power and direct surface motility. The production and secretion of a viscous polysaccharide to provide friction and resistance to the extended pilus structure is seen in several species of cyanobacteria including Nostoc punctiforme. The complex coregulation of polysaccharide secretion and T4P motor activity is not fully understood, although studies indicate a consistent relationship between functional motility and intact pathways of polysaccharide secretion and pilus extension in cyanobacteria. Using a combination of protein-protein interaction analysis, cytological studies, and comparative genomics this study proposes a theoretical mechanism for T4P motor influenced …

Chemically Mediated Algal-Bacterial Interactions: Promotion & Inhibition, Kara Talbott Jan 2023

Chemically Mediated Algal-Bacterial Interactions: Promotion & Inhibition, Kara Talbott

University of the Pacific Theses and Dissertations

Algae-bacteria interactions range from mutualistic, Chlamydomonas reinhardtii (green algae) provides carbonate complexes for surface associated bacteria (SAB) and the bacteria in turn provide B-12 for the algae, to pathogenic, the presence of certain bacteria increases the incidence of bleaching in the algae. Algal natural products have been found to display the following activities: anti-fouling, anti-bacterial, anti-cancer, and anti-feedant. Separately, it has been well documented in the literature that algae mediate their SAB, also known as their microbiome, using natural products to attract and inhibit different bacterial strains. My graduate work builds on these findings; our hypothesis is that the algal-bacterial …

Influence Of Female Sex Hormones On Ghb Toxicokinetics And Regulation Of Mcts And Smcts, Hao Wei Jan 2023

Influence Of Female Sex Hormones On Ghb Toxicokinetics And Regulation Of Mcts And Smcts, Hao Wei

University of the Pacific Theses and Dissertations

Gamma-hydroxybutyric acid (GHB) is an endogenous shorty chain fatty acid that is used clinically as Xyrem to treat narcolepsy. GHB is best known for its illicit use and abuse due to its sedative/hypnotic and euphoric effects. It is used in body building, for recreational use and sexual assault. Nonlinear toxicokinetics of GHB has been described in humans and rats with decreased total clearance at higher doses due to capacity-limited metabolism resulting in a higher plasma exposure. Renal clearance increases with increasing dose and becomes the major route of GHB elimination in overdose cases due to saturation of GHB metabolism. Proton- …

Quantifying genetic clusters (=populations) from genotypic data is a fundamental, but non-trivial task for population geneticists that is compounded by: hierarchical population structure, diverse analytical methods, and complex software dependencies. AdmixPipe v3 ameliorates many of these issues in a single bioinformatic pipeline that facilitates all facets of population structure analysis by integrating outputs generated by several popular packages (i.e. CLUMPAK, EvalAdmix). The pipeline interfaces disparate software packages to parse Admixture outputs and conduct EvalAdmix analyses in the context of multimodal population structure results identified by CLUMPAK. We further streamline these tasks by packaging AdmixPipe v3 within a Docker container to …
