Chemotherapy-related cognitive impairment (CRCI) is a common adverse effect of treatment and is characterized by deficits involving multiple cognitive domains including memory. Despite the significant morbidity of CRCI and the expected increase in cancer survivors over the coming decades, the pathophysiology of CRCI remains incompletely understood, highlighting the need for new model systems to study CRCI. Given the powerful array of genetic approaches and facile high throughput screening ability in Drosophila, our goal was to validate a Drosophila model relevant to CRCI. We administered the chemotherapeutic agents cisplatin, cyclophosphamide, and doxorubicin to adult Drosophila. Neurologic deficits were observed with all …

The Association Betweem Total And Types Of Dietary Protein Intake And Prevelence Of Impaired Fasting Glucose And Type 2 Diabetes, Afra Salah Alsammach
Thesis/ Dissertation Defenses
Introduction: People with impaired fasting glues (IFG) and type type 2 diabetes (T2DM) need to modify their dietary composition. This could assist in the management and prevention of the disease. However, the role played by the composition of specific dietary macronutrients on the risk of IFG &T2D remains uncertain. This study sought to examine the association between total and type of dietary protein consumption with prevalence of type 2 diabetes mellitus (T2D) and impaired fasting glucose (IFG). Examining the dietary assessment in nutritional epidemiology provided the data needed to assess the relationship between the variables. Aims: To investigate the association …

MicroRNAs ( miRNAs ) are important regulators of genes expression. Their levels are precisely controlled through modulating the activity of the microprocesser complex ( MC ) . Here, we report that JANUS, a homology of the conserved U2 snRNP assembly factor in yeast and human, is required for miRNA accumulation. JANUS associates with MC components Dicer-like 1 ( DCL1 ) and SERRATE ( SE ) and directly binds the stem- loop of pri-miRNAs. In a hypomorphic janus mutant, the activity of DCL1, the numbers of MC, and the interaction of primary miRNA transcript ( pri-miRNAs ) with MC are reduced. …
Dynamic Regulation Of Store-Operated Calcium Entry By Protein S-Acylation, Goutham Kodakandla
Dynamic Regulation Of Store-Operated Calcium Entry By Protein S-Acylation, Goutham Kodakandla
Theses and Dissertations
Calcium plays a pivotal role in many physiological functions in cells. Cytosolic calcium levels are finely tuned by calcium ion channels, pumps, and intracellular organelles. Store-operated calcium entry (SOCE) is when depletion of endoplasmic reticulum (ER) calcium stores activates a calcium sensor known as stromal interaction molecule 1 (STIM1). Activation of STIM1 leads to a conformational change from a compact state to an extended state. This extended state of STIM1 allows it to bind to a calcium channel in the plasma membrane (PM) known as Orai1. The binding of Orai1 and STIM1 leads to opening of Orai1 channels and calcium …

Novel physically-crosslinked PVA membranes blended with Aloe vera extract were fabricated by solution-casting method. Physically-crosslinking process is depending on the rearrangement of PVA chains forming intermolecular hydrogen bonding with removal of water molecules using propanol as a stabilizing agent. The structure of crosslinked membranes was characterized by FT-IR, SEM, TGA, and XRD analyses and confirmed via gel faction and swelling ratio studies. Caffeine and vitamin C loaded-PVA/Aloe vera membranes were bio-assessed in terms of their impact on the wound healing using Wistar albino rats as an animal model. In vitro evaluation includes protein adsorption showed that the fabricated membranes improved …

Affinity Based Writing Groups: Building Communities Across 9–12 Grade Levels, Melissa Pilakowski
Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–
Writing instruction in secondary classrooms has generally focused on cognitive approaches with feedback and assessment from a limited audience, often only the teacher. However, the burgeoning digital world has made writing an increasingly social activity where individuals can influence a wider audience and engage in dialogue with their readers faster and more efficiently than ever before. This action research aims to study the impacts of online affinity based writing groups that span the 9–12 grade levels, especially in the areas of effort, self-efficacy, and feedback. Data was collected through interviews with participating students and teachers, samples of writing and feedback, …

Primary cilia from the brain microvascular endothelial cells (ECs) are specialized cell-surface organelles involved in mediating sensory perception, cell signaling, and vascular stability. Immunofluorescence (IF) analysis of human primary brain microvascular ECs reveals two cilia per cell. To confirm the in vitro observation of the two-cilia phenotype in human primary brain ECs, ECs isolated from mouse brain were cultured and stained for cilium. Indeed, brain ECs from a ciliopathic mouse (polycystic kidney disease or Pkd2−/−) also possess more than one cilium. Primary cilium emerges from the mother centriole. Centriole analysis by IF suggests that in brain ECs, markers …

Avoidance stress coping, defined as persistent internal and/or external avoidance of stress-related stimuli, is a key feature of anxiety- and stress-related disorders, and contributes to increases in alcohol misuse after stress exposure. Previous work using a rat model of predator odor stress avoidance identified corticotropin-releasing factor (CRF) signaling via CRF type-1 receptors (CRF1) in the central amygdala (CeA), as well as CeA projections to the lateral hypothalamus (LH) as key mediators of conditioned avoidance of stress-paired contexts and/or increased alcohol drinking after stress. Here, we report that CRF1-expressing CeA cells that project to the LH are preferentially activated in male …
A Phenomenological Study Of Successful Superintendent Retention In A North-Central New Mexico School District, Adán G. Delgado
A Phenomenological Study Of Successful Superintendent Retention In A North-Central New Mexico School District, Adán G. Delgado
Teacher Education, Educational Leadership & Policy ETDs
Superintendent of Schools is uniquely critical in a school district due to its tremendous effect on the operational and cultural bearing of the organization (Myers, 2011). Superintendent turnover can hinder district reform and improvement (Grissom & Andersen, 2012; Marzano, 2006). The United States and New Mexico struggle with superintendent retention and northern New Mexico is especially plagued with short superintendent tenure (Grissom & Andersen, 2012; NMPED Stat Books, 2021; Sawchuck, 2022). In this study, I investigated the initial selection and subsequent interactions and relationships between school board members and superintendents. Using a phenomenological approach, I interviewed two superintendents and four …

The dependency of cancer cells on iron increases their susceptibility to ferroptosis, thus providing new opportunities for patients with treatment-resistant tumors. However, we show that lipid peroxidation, a hallmark of ferroptosis, was found in various areas of patient samples, indicating the potential resistance of ferroptosis. Using whole deubiquitinases (DUBs) sgRNA screening, we found that loss of ZRANB1 confers cancer cell resistance to ferroptosis. Intriguingly, functional studies revealed that ZRANB1 ubiquitinates and represses SLC7A11 expression as an E3 ubiquitin ligase and that ZRANB1 inhibits glutathione (GSH) synthesis through SLC7A11 degradation, leading to elevated lipid peroxidation and ferroptosis. Deletion of the region …

Understanding the links between development and evolution is one of the major challenges of biology. ‘Heterochronies’, evolutionary alterations in the timings of development are posited as a key mechanism of evolutionary change, but their quantification requires gross simplification of organismal development. Consequently, how changes in event timings influence development more broadly is poorly understood. Here, we measure organismal development as spectra of energy in pixel values of video, creating high-dimensional landscapes integrating development of all visible form and function. This approach we termed ‘Energy proxy traits’ (EPTs) is applied alongside previously identified heterochronies in three freshwater pulmonate molluscs (Lymnaea stagnalis, …

Ovarian cancer (OC) is the most common lethal gynecologic cause of death in women worldwide, with a high mortality rate and increasing incidence. Despite advancements in the treatment, most OC patients still die from their disease due to late-stage diagnosis, the lack of effective diagnostic methods, and relapses. Aptamers, synthetic, short single-stranded oligonucleotides, have emerged as promising anticancer therapeutics. Their ability to selectively bind to target molecules, including cancer-related proteins and receptors, has revolutionized drug discovery and biomarker identification. Aptamers offer unique insights into the molecular pathways involved in cancer development and progression. Moreover, they show immense potential as drug …
