Advances in CRISPR/Cas9 gene editing technologies present opportunities to better understand the contribution of individual genes to complex eukaryotic cellular processes. I have applied CRISPR whole genome screen technologies to inform further mechanistic understanding of macrophage fitness and neutral lipid metabolism. This work is important because macrophages play critical roles in immune function and are implicated as causative agents in disease states. The goal of this dissertation was to understand the strategies needed to optimize performance of confinement fed beef cattle in the Northern Plains. This was accomplished through three objectives: 1) evaluate health, growth performance, and antibody titers in previously vaccinated, newly weaned calves administered a respiratory and clostridia vaccine upon arrival compared to no vaccination; 2) determine the influence of manger space restriction on program-fed feedlot heifers during the growing phase; and 3) to evaluate growth performance and carcass traits following transit of feedlot heifers sourced and finished in different regions of the United States. Fish require a variety of resources including prey, thermal refugia, and different physical habitat characteristics to grow and reproduce. All necessary resources are rarely provided by a single habitat, resulting in tradeoffs between occupying different habitats; therefore, the ability to move among heterogenous habitats is critical. These tradeoffs may occur in lakes with connected wetlands where the main lake habitats tend to be more thermally stable, but wetlands often provide more diverse and abundant prey. This study conducted in 2023 aimed to enhance nitrogen use efficiency (NUE) in wheat and corn grown in South Dakota. Based on dynamic weather conditions and other factor interactions, conventional nitrogen (N) recommendations need to be improved. Soil properties information, including electrical conductivity, was used to create management zones. In each zone, three N-rich spots were established as biosensors. Drones and satellites collected imagery data, and an AI-driven approach assessed the crop response to applied N. In this paper, I consider the extent to which caselaw has – either explicitly or implicitly – incorporated the precepts of therapeutic jurisprudence (TJ), a school of legal thought that focuses on the law’s influence on emotional life and psychological well-being, and that asks us to assess the actual impact of the law on people’s lives. Two of the core tenets of TJ in practice are commitments to dignity and to compassion. I conclude ultimately that, with these principles as touchstones, TJ can be an effective tool – perhaps the most effective tool - in rooting out bias, limiting polarization, …

Legal education can help prepare law graduates to do that work. As an added bonus, doing so would in turn help law students understand and retain the subjects they study.

Law professors who teach alternative dispute resolution, lawyering skills, clinics, and sometimes traditional doctrinal courses, have all called for greater inclusion of dispute resolution in the law school curriculum. Some have urged the introduction of specific courses to prepare contemporary law students to work as problem resolvers. This paper explores Mexico’s reactionary migration policies, especially concerning the labeling of African migrants and asylum-seekers as so-called “stateless” and the subsequent unsolicited permanent residence status it offers, and address the numerous international and domestic legal violations contained within. Part I of this article presents the lived experiences of African asylum-seekers traveling through Mexico in 2015 and 2019 to demonstrate the direct impact of Mexican migratory policy changes. Many Black migrants and asylum-seekers, as well as Afro-Mexicans, have raised awareness of anti-Blackness in Mexico and of how racism is pervasive. Astragalus lentiginosus is the most taxomically diverse species in the North American flora with over 40 recognized varieties. The incredible diversity within A. lentiginosus, particularly in respect to its fruit morphology, makes it an exceptional model to study adaptation and evolution. However, it remains unknown whether fruit morphology is an adaptation and, if so, the extent to which selection or neutral evolutionary processes shape the striking diversity in this trait. Dance is a universal human behavior and a crucial component of human musicality. When and how does the motivation and tendency to move to music develop? How does this behavior change as a process of maturation and learning? We characterize infants’ earliest dance behavior, leveraging parents’ extensive at-home observations of their children. Parents of infants aged 0–24 months (N = 278, 82.7% White, 84.5% in the United States, 46.0% of household incomes ≥ $100,000) were surveyed regarding their child’s current and earliest dance behavior (movement by the child, during music, that the parent considered dance), motor development, and their …

Biological diversity peaks in tropical regions followed by a steady decline towards the poles. It is a pattern in γ-diversity that sometimes explains latitudinal patterns in local assemblages (α) and their species turnover (β), which suggests that similar ecological mechanisms might be operating at different latitudes. However, not all taxa have more species towards the equator. Aquatic shredder insect richness peaks in temperate streams relative to tropical streams, where local aquatic shredder insect assemblages have low α-diversity. Purpose: Parental vaccine hesitancy is a known driver of sub-optimal vaccine uptake in the United States.1 Even though a study analyzing parental responses in National Immunization Survey-Teen found that HPV vaccine hesitancy has slightly declined (69% in 2010 v/s 63% in 2019) over the years, only about 52.6% of adolescents in Montana had received all the required doses of the HPV vaccine series in 2021.2,3 Nurses are at the forefront of healthcare provision and possess a unique ability to influence parental vaccine decisions.5 By listening to and addressing parents' concerns about immunizations, nurses can dispel misinformation and help change parents' …

Tonic immobility (TI), a temporary, involuntary state of paralysis marked by motor inhibition and vocal suppression, has been a well-documented occurrence among animals. Humans also experience TI, which has previously been referred to as trauma- or rape-induced paralysis and freezing. TI among survivors of sexual assault has been associated with greater PTSD and increased risk of depression. Existing studies have not adequately explored additional posttraumatic outcomes of TI beyond PTSD diagnostic criteria, nor has prior research sufficiently tested the mechanistic relationship between TI and PTSD. This work develops and applies novel methods for metagenomic and landscape genomic (LSG) investigation of wildlife populations, specifically in Rocky Mountain elk (Cervus canadensis) in the Greater Yellowstone Ecosystem (GYE). Host fecal microbiomes facilitate and reflect interactions between the host and its environment. We conducted metagenomic surveys and developed a machinelearning classifier for microbiomes that provided new insights into elk-microbiome biogeography, and the impact of fecal microbiomes on host health and the ecology of elk-wolf (Canis lupus) predation. Mechanistic understanding of population dynamics requires linking population change and demographic processes. However, for some species, accurate estimation of population parameters can be difficult owing to their life histories, resulting in reduced or biased inference. Mobile species that are not territorial or use dynamic habitats are susceptible to estimation problems arising from variable exposure to sampling, or temporary emigration, and common approaches to account for it like robust design are not feasible. However, restoration is often done without a full understanding of its impact on biological communities. Songbird populations are sensitive to broad habitat changes and therefore can be used to assess and prioritize habitat for biodiversity. Whitebark pine (Pinus albicaulis), recently listed as threatened under the Endangered Species Act, is in steep decline in Glacier National Park, Montana, USA due to the non-native pathogen Cronartium ribicola, causal agent of the fatal disease white pine blister rust. A sample of the park’s population suggests that approximately 70 percent of whitebark pines have died, while 65 percent of the remaining trees are infected. Using landscape and climate variables, we show how geographic location, elevation, aspect, solar radiation, relative humidity, and snowpack interact with tree diameter to affect mortality, disease incidence, cone production, and regeneration.
