This work consists of studies on dwarfism in Miniature horses and a study on breakdowns in Thoroughbreds in relation to gene aggrecan. A correction of the description and designation of D3 dwarf variant in aggrecan (ACAN) from the master’s thesis Chondrodysplasia-like dwarfism in the Miniature horse (2013). Commercial sequencing showed previous sequencing reads presented an artifact and not a single base deletion. Analysis showed a single base missense mutation in exon 8 identified as D3* was the actual cause. Multiple alleles of ACAN associated with chondrodysplastic dwarfism in Miniature horses by Eberth et al. (2018), corrected the …

Accumulating high-speed exercise has been identified as a significant risk factor for catastrophic injuries in racing Thoroughbreds. Injuries, regardless of severity, are a main cause of withdrawal from the racing industry, raising animal welfare concerns and resulting in significant economic losses. While most of the current literature focuses on catastrophic injuries incurred during racing rather than training, the present study aims to help fill this gap as well as discuss the associated risk factors. Though generally regarded as safe, research continues to demonstrate negative side effects of antibiotic administration on the gastrointestinal (GIT) microbiota across species. In horses, antibiotic associated diarrhea (AAD) is a life-threatening side effect linked to the GIT microbiota. This study tested the hypothesis that short term antibiotic administration to healthy horses would negatively impact the fecal microbiota as measured by their ability to digest nutrients and produce short chain fatty acids (SCFAs). Twenty-four horses were assigned to one of four treatments: control (CO); potassium penicillin/gentamicin sulfate (KPG); ceftiofur crystalline free acid (EX); trimethoprim/sulfamethoxazole (SMZ); and treated for 4 days. Musculoskeletal injuries account for the majority of career-ending health concerns in Thoroughbred racehorses. A small but very serious subset of these can be life-threatening and usually involve fractures of the bones in the lower limb that occur during high-speed exercise or racing competitions. Understanding the etiopathogenesis of lower forelimb bone fractures leading to catastrophic musculoskeletal injuries (CMI) is the highest safety and welfare priority in horse racing. This thesis conducted two studies utilizing computed tomography (CT) images of Thoroughbred lower limbs. The first examined bone lesion annotations in 134 horses (268 forelimbs) across 105 anatomical sites to assess the concordance …

The present research incorporated molecular genetic methods to 1) investigate the genetic basis of Juvenile Onset Lordosis or Swayback in the American Saddlebred horses; and 2) conduct a population genetic study to compare the Persian Kurdish, Persian Arabian and American Thoroughbred horse populations.
Juvenile-onset lordosis, or swayback, is a condition in horses where the conformational topline back curvature drops significantly within the first two years of life. The trait has a higher prevalence in Saddlebreds (5%). Influenza D virus (IDV) utilizes bovines as a primary reservoir with periodical spillover to other hosts. We have previously demonstrated by the traditional hemagglutination assay coupled with sialoglycan microarray and functional assays that IDV can recognize and bind both 9-O-acetylated N-acetylneuraminic acid (Neu5,9Ac2) and 9-O-acetylated N-glycolylneuraminic acid (Neu5Gc9Ac). Agricultural animals such as cattle can produce both Neu5,9Ac2 and Neu5Gc9Ac, while humans are genetically unable to synthesize Neu5Gc9Ac. The bifunctional enzyme, glucosamine (UDP-N-acetyl)-2-epimerase/N-acetylmannosamine kinase (GNE) is a master regulator of de-novo sialic acid synthesis while 9-O-Acetylation of sialic acids is catalyzed by …

In the eastern U.S., fire-dependent tree species have historically dominated upland forest communities, but are now experiencing widespread regeneration challenges as a result of 20th century fire suppression policies, and are being replaced by mesophytic species. Wildfires that contain areas of high burn severity may provide an important means of mitigating these challenges and facilitating fire-dependent species regeneration and recruitment into larger size classes. One mechanism by which high-severity fire can accomplish this is by modifying understory microclimate characteristics to be more conducive to these species’ growth. The Forestry Reclamation Approach (FRA) is a recently developed coal mining reclamation method that emphasizes best management practices in forestry, such as the planting of native trees and shrubs. Although the FRA is expected to benefit wildlife, no studies have empirically examined the effects of the FRA on avian species. My study aimed to identify which reclamation approaches and/or landscape features promote breeding songbirds, particularly mature forest avian guilds and species of conservation need. Lyme disease has been of national concern for the past few decades. As our understanding of the role of landscape structure in epidemiology expands, it is essential to apply the principles of landscape ecology to the research of vector-borne and zoonotic disease. This study examined the strength of the relationship between land-cover class, degree of habitat fragmentation and county-level Lyme disease cases. Safe and effective chemical immobilization is critical to minimize stress and risk of injury when capturing free-ranging, wild ungulates. Many traditionally favored high potency opioids have been phased out or become unavailable because of increased regulations, leading to the development of two pre-mixed combination drugs, butorphanol-azaperone-medetomidine (BAM) and nalbuphine-medetomidine-azaperone (NalMed-A). Both drugs have been used to chemically immobilize ungulates, but their efficacy has not been documented in elk captured and transported via helicopter. Nanotechnology has several applications in the agricultural industry, as the small size of nanoparticles and high reactivity enables targeted delivery of pesticides and fertilizers to the intended crop. Copper-based nanopesticides such as Kocide, are proposed as a more efficient method for targeted pest control. Despite the potential benefits of nanopesticides, little is known regarding the transport and implications of nanopesticides under long-term inundation conditions often found in downstream wetlands. The production of cannabidiol (CBD) from the floral material of Cannabis sativa L.is of great interest to the hemp industry. The low bulk density oh hemp floral material presents challenges in terms of storage and handling. To improve handling and storage characteristics, both intact (pre-extraction) and supercritical CO2 extracted hemp floral materials were pelletized. Intact floral material (BaOx) was passed through a hammer mill with a 5 mm screen; while, extracted floral material (Hawaiian Haze) was ball milled through a 60 mesh (250 microns) prior to extraction. Bourbon, or whiskey, production in Kentucky has been estimated to double within the next five years and an increase in the main by-product from bourbon distillation, stillage. Stillage is composed mostly of water along with the fermented grains after distillation. Stillage is expensive to dispose of and difficult to store due to the high biodegradability, posing a risk to the environment given the low pH and high chemical oxygen demand (COD). Anaerobic digestion has been identified as a potential solution for stillage valorization, but little research has been performed. Per- and polyfluoroalkyl substances (PFAS) and neonicotinoid insecticides are two classes of emerging contaminants that have been recently detected in many environmental compartments. Both have been associated with negative impacts on humans and the environment. Wastewater treatment plants (WWTPs) have been identified as likely hotspots for introduction of these contaminants into the environment. Thus, the aim of this study was to investigate the occurrence of PFAS and neonicotinoids in two WWTPs in Central Kentucky. Demand for sustainable food sources has increased because of the rapid growth of the world's population. In this study, microalgae cells of Haematococcus pluvialis were immobilized using the edible fungal strain Aspergillus awamori for potential food applications. The study investigated the impact of fungal loading, pellet geometry, and initial microalgae cell concentration on the immobilization performance and product characteristics. It was found that higher fungal loading and larger fungal pellets contributed to increased immobilization performance while increased initial microalgae concentration inhibited the process. Larger fungal pellets had decreased biomass density, which led to decreased surface concentration of immobilized microalgae but …

Equine indoor arenas are a unique infrastructure investment found at equine farms and facilities. They are semi-indoor structures for exercising horses, exhibiting skills during competitive events, and other equine related activities. These spaces do not always include mechanical ventilation or stirring fans and occupancy by horses and humans can be sporadic and inconsistent, which creates a challenging space for understanding and predicting airflow. Typically, indoor arenas have a sand-based footing over which the horse travels. The impact of the hooves can cause dust to become a concern within the facilities. Death of individuals from the same species represents potential risks from disease, predation or competition for animals. Diverse responses associated with death have evolved ranging from simply avoiding or being attracted to the corpses in solitary animals to complicated undertaking behavioral repertoire in eusocial insects. A systematic review in chapter 1 suggested cannibalism is an ancestral and widespread death-related behavior in all non-human animals. Termites are suggested to switch their undertaking behavioral responses from cannibalism to burial based on interactions between chemicals associated with death to balance risks and benefits associated with decomposition. Tick-borne diseases are an emerging threat to human and animal health. In Kentucky, tick-borne disease surveillance has identified rising incidences of spotted fever rickettsiosis, ehrlichiosis, and Lyme disease. Since these diseases occur through the bites of infected ticks, effective prevention efforts are reliant upon knowing where the risk of exposure to tick bites exists. Historical data on tick distribution in Kentucky is variable, with very little reported on a statewide scale, leaving vector control workers, public health personnel, physicians, veterinarians, and others to rely on outdated, intermittent, or out-of-state information. Genetic strategies of insect pest control are receiving increased interest due to recent advances in genetic engineering. By introducing alleles that make males functionally sterile, genetically modified (GM) insects can be used in area-wide pest management programs similar to the Sterile Insect Technique (SIT). Genetic control strategies carry several potential benefits, including improved efficacy and specificity, lower operation costs, and reduced dependence on chemical insecticides, but their use is still controversial despite extensive technical documentation indicating their innocuity to humans and the environment. This hesitation is likely due to the novel nature of the approach.
