Small interfering RNAs (siRNAs) maintain transgenerational gene silencing in C. elegans. siRNAs are loaded into Argonaute proteins to form an RNA-induced silencing complex (RISC), which functions in the nucleus or cytoplasm. In the nucleus, RISC directs the establishment of H3K9 trimethylation (H3K9me3) to silence loci. One nuclear Argonaute protein is HRDE-1. Here, we show that HRDE-1 is required for development and maturation in C. elegans. Brood size assays of hrde-1 mutants and wild-type (N2) hermaphrodites grown at permissive temperature (20℃) and under heat stress conditions (25℃) were performed to examine the number of eggs laid, eggs hatched, and matured progeny. …

The federal government has made education a competition among ourselves. Learning is not a rivalry to be imposed on our physical and cognitive development as humans. Escalating beliefs recognize education is more encompassing than CCSS, assessments and the grade level time frames in which standards are to be taught. On the precipice of anti-intellect, low expectations, and what has been termed “The Dumbing of America” an investigation into what it would take to close the achievement gap and increase student learning is overdue.

“Americans, as a whole, were at least two generations removed from the farm and did not understand even the most rudimentary of processes, challenges, and risks that farmers and the agricultural industry worked with and met head-on every day.” This quote perfectly describes the mindset of agriculture stakeholders in 1981 as they began to realize the drastic steps our education system had taken away from using principles of agriculture in K-12 education. The contradictory nature of U.S. laws, including the laws concerning infant feeding, though supposedly aligned with policies to promote wellness in Americans, can exacerbate gender and race inequality and work against the National Strategy. The overarching goal of U.S. laws concerning infant feeding is to ensure that infants are fed, nourished, and receive proper nutrition. However, the laws often appear to be directly contradictory to one another in the priorities they are promoting.

Since the eighteenth century, animal well-being remains a concern for American citizens. Yet, underlying this concern is the thought that while humans should not be cruel to animals, animals are still private property subject to human ownership. Therefore, multi-faceted questions of what constitutes “animal welfare” find a place in modern American debate. One such question becomes: should the producer or the consumer define welfare practice standards of animals raised for human consumption?7 This note provides an answer to this question by first analyzing the robust history of animal welfare in the United States, along with the domestic and international impact …

Alcohol-associated liver disease (ALD) is a broad spectrum of diseases ranging from steatosis to severe hepatic cirrhosis, none of which have any FDA approved therapies. Downregulation of hepatic Sirtuin 1 (Sirt1) activity by ethanol has been shown to lead to an upregulation of the pro-lipogenic gene targets under its jurisdiction, resulting in increased lipogenesis and triglyceride accumulation. It was hypothesized that ethanol acts on α4*-nicotinic acetylcholine receptors (α4*-nAChRs) in hepatocytes to decrease Sirt1 expression, which ultimately dysregulates lipid metabolic homeostasis. In recent years, research into air pollution has shown that exposure to certain components in air pollution, primarily PM2.5 can accelerate biological aging and thereby lead to increased susceptibility to multiple diseases. We hypothesize that prolonged exposure to air pollutants can result in premature aging leading to extensive tissue dysfunction and susceptibility to diseases. To examine this, we exposed mice to PM2.5 for 9, 15, and 21 days, then measured the telomere lengths, cellular senescence, and histone methylation patterns of multiple cell types. We reported pulmonary artery hypertension (PAH) patients had elevated levels of Cd in both blood and urine, therefore, this study tested whether Cd directly induces PAH or facilitates PAH pathogenesis in mouse models. C57/6J mice were initially exposed to drinking water with or without 5ppm Cd for 8 weeks. Then, half the mice in both control and Cd groups were given SU5416 and hypoxia (SuHx) for 4 weeks to induce PAH, resulting in 4 subgroups: Control, Cd, PAH, and Cd+PAH. Diastolic and systolic functions of the left and right ventricles (LV, RV) were examined with echocardiography before and after PAH. …

Porphyromonas gingivalis is a keystone oral pathogen in the progression of periodontal disease and has been implicated in the etiology of many other systemic human diseases. P. gingivalis produces primary virulence factors termed the gingipains, a group of cysteine proteases. There is a high demand for high purity gingipains for downstream in vitro studies where P.g. is implicated, both oral and systemic. Gingipain production candidate mutant P.g. strains were generated to isolate each gingipain for large scale purification and the purified gingipains characterized. Lung cancer is one of the most common forms of cancer diagnosed, and has the highest lethality rate across all other forms of cancer in the U.S. While current therapeutic options include chemo-, immuno-, and radio-therapy, the benefits of caloric or nutrient restriction on cancer cells has also been investigated. Zinc deficiency is associated with metabolic dysfunction-associated steatotic liver disease (MASLD). Previous studies show zinc supplementation improves steatosis, but its therapeutic effects in established MASLD remain unclear. We developed a model to characterize the effects of zinc supplementation on high-fat diet (HFD) induced MASLD and hypothesized established MASLD would be attenuated. Mice were fed control diet or HFD for 12 weeks and then grouped into normal or zinc-supplemented diets for 8 additional weeks. At euthanasia, plasma and liver tissues were collected for phenotypic analysis. Twelve weeks of HFD altered glucose clearance and body composition. Eight weeks of subsequent zinc supplementation …

Steatotic liver disease is brought upon by differing etiologies and its global prevalence is expected to keep increasing over the next several decades. Particularly, alcohol-associated liver disease (ALD) manifests from excessive alcohol consumption where the liver retains lipids and progresses with metabolic dysfunction. Previous studies have demonstrated that environmental pollutants, polychlorinated biphenyls (PCBs) promote metabolic dysfunction associated steatotic liver disease (MASLD), in diet-induced obesity models. Testosterone deficiency (TD), which has been linked to some of these sexual dysfunctions, is characterized by low production of testosterone by the testes. These male reproductive outcomes may develop due to exposure to various risk factors, including environmental exposures. A protection zone model represents a patchy environment with positive growth over the protection zone and strong Allee effect growth outside the protection zone. Generally, these models are considered through the corresponding eigenvalue problem, but that has certain limitations. In this thesis, a general protection zone model is considered. This model makes no assumption on the direction of the traveling wave solution over the Strong Allee effect patch. Enhancing physical human-robot interaction in modern robotics relies on refining the tactile perception of robot skin sensors. This research focuses on crucial aspects of the development process, including fabrication techniques, miniaturization, and integration for a more efficient collaborative human-robot interface. The fabrication process of robot skin sensors, designed to mimic human skin, is explored both within and outside cleanroom environments. An enhanced technique is presented to increase fabrication yield and create more miniaturized sensor designs with feature sizes in the tens of microns. Both pre-race meet and daily turf surface condition measurements are required by regula- tions adopted as part of the Horseracing Integrity and Safety Act (HISA). The Orono Biomechanical Surface Tester (OBST) is the primary device used for characterizing a racing surface and is used for the pre-meet inspections. Age-associated alterations in bladder control affect millions of older adults, with a heavy burden added to families both economically and in quality of life. Therapeutic options are limited with poor efficacy in older adults, lending to a growing need to address the gaps in our current understanding of urinary tract aging. This review summarizes the current knowledge of age-associated alterations in the structure and function of the brain-bladder axis and identifies important gaps in the field that have yet to be addressed. The aim of my capstone project was to advance the field of animal-assisted therapy (AAT) and promote the incorporation of AAT within occupational therapy (OT) services. Foodborne pathogens, including Escherichia coli, Salmonella enterica, and Listeria monocytogenes, are major causes of gastrointestinal disease globally. The Contamination Sanitization Inspection and Disinfection (CSI-D) device is a new handheld fluorescence-based imaging system designed to disinfect food contact surfaces contaminated with microorganisms using ultraviolet-C (UVC) illumination. This study aimed to determine the optimal parameters for the disinfection of foodborne bacteria using the CSI-D device. Warthin's tumor is the second most common neoplasm of the parotid gland and consists of 2 components, including lymphoid stroma and glandular epithelium. Malignant transformation in this tumor is mostly seen in the lymphoid component; however, the carcinomatous transformation of the epithelial component is extremely rare. Cases of latter reported in the literature include squamous cell carcinoma, adenocarcinoma, mucoepidermoid carcinoma, oncocytic carcinoma, Merkel cell carcinoma, and undifferentiated carcinoma. We describe an extremely rare case of salivary duct carcinoma arising in a Warthin tumor in a 64-year-old male. Colorectal cancer (CRC) is the third most common cancer and is also the third leading cause of cancer-related death in the USA. Understanding the mechanisms of growth and progression of CRC is essential to improve treatment. Macronutrients such as glucose are energy source for a cell. Many tumor cells exhibit increased aerobic glycolysis. Increased tissue micronutrient iron levels in both mice and humans are also associated with increased colon tumorigenesis. However, if iron drives colon carcinogenesis via affecting glucose metabolism is still not clear. Here we found the intracellular glucose levels in tumor colonoids were significantly increased after iron treatment.

This is often attributed to a warming climate but may also result from increased observer activity and usage of eBird. Late dates are probably the most important aspect of this report, most prominent the record late fall dates for Ruby-throated Hummingbird, Sanderling, Bank Swallow, and Cassin’s Sparrow. Although these species are taxonomically diverse, there were species group concentrations of late dates, notably among shorebirds. Yellow-throated Warbler (Setophaga dominica) is a rare to uncommon migrant in eastern and central Nebraska. It is possibly a regular breeder locally at sites containing sycamores in several southeastern Nebraska counties bordering the Missouri River. There are few Nebraska reports after July, with more reports November–January than August–October. This species has three previous documented Nebraska Panhandle records, all from May.

The interactions between the lacewing Ceraeochrysa claveri (Navás) (Neuroptera: Chrysopidae) larva and the coconut mealybug Nipaecoccus nipae (Maskell) (Hemiptera: Pseudococcidae) were recorded. The third-instar lacewing larva constructed a dorsal packet using mealybug wax from both male pupal cases and the bodies of adult females. Surprisingly, live nymphs were also frequently placed into the dorsal packet. Prey mealybugs were discarded and not incorporated into the packet after consumption. When disturbed, adult female mealybugs reflex bled from their dorsal ostioles, contacting the mouthparts of the lacewing. Dysmicoccus brevipes (Cockerell) and Dysmicoccus neobrevipes Beardsley are major pests of pineapples, ornamentals, and vegetable crops in many countries around the world. The potential distribution of these mealybug pests into South Korea remains a prime concern because of their high incidence in interceptions screened during inspection. Hence, these species prompted a modelling effort to assess their potential risk of introduction. Potential risk maps were developed for these pests with the CLIMEX model based on occurrence records under environmental data.
