Frederick Douglass And The Original Originalists, Bradley Rebeiro Jan 2023

Frederick Douglass And The Original Originalists, Bradley Rebeiro

BYU Law Review

Constitutional scholars incessantly grapple over the significance of the Constitution’s original meaning. More specifically, they are preoccupied with, on the one hand, what that meaning is (if such meaning exists) and, on the other hand, the exact nature of that meaning’s authority (if any) over the Constitution and its interpreters. But this debate is hardly novel. In fact, one of the most compelling voices in U.S. history was immersed in similar debates and, out of the constitutional sparring of his time, forged an arresting theory of constitutional interpretation. Frederick Douglass, once a fierce opponent of the U.S. Constitution, evolved into …

Medulloblastoma (MB) is a malignant pediatric brain tumor which shows upregulation of MYC and sonic hedgehog (SHH) signaling. SHH inhibitors face acquired resistance, which is a major cause of relapse. Further, direct MYC oncogene inhibition is challenging, inhibition of MYC upstream insulin-like growth factor/ phosphatidylinositol-4,5-bisphosphate 3-kinase (IGF/PI3K) is a promising alternative. While PI3K inhibition activates resistance mechanisms, simultaneous inhibition of bromodomain-containing protein 4 (BRD4) and PI3K can overcome resistance. We synthesized a new molecule 8-(2,3-dihydrobenzo[b] [1, 4] dioxin-6-yl)-2-morpholino-4H-chromen-4-one (MDP5) that targets both BRD4 and PI3K pathways. We used X-ray crystal structures and a molecular modeling approach to confirm the interactions …

Research outputs 2022 to 2026

Background: Seasonally dry tropical forests (SDTFs) are globally important ecosystems which receive less research attention compared to tropical rainforests but are equally under serious threat. The objectives of this paper are to characterize the vegetation structure, diversity and composition of SDTF of Baluran National Park, East Java, Indonesia, and to assess the impact of burning this SDTF and its post-fire recovery. Methods: In the field, we measured floristic composition and dominance at sites with different fire histories in both SDTF and adjacent savannas of Baluran. Remote sensing image analysis was also employed using the MODIS burn area product and various …

An Entosis-Like Process Induces Mitotic Disruption In Pals1 Microcephaly Pathogenesis, Noelle A Sterling, Jun Young Park, Raehee Park, Seo-Hee Cho, Seonhee Kim Jan 2023

An Entosis-Like Process Induces Mitotic Disruption In Pals1 Microcephaly Pathogenesis, Noelle A Sterling, Jun Young Park, Raehee Park, Seo-Hee Cho, Seonhee Kim

Center for Translational Medicine Faculty Papers

Entosis is cell cannibalism utilized by tumor cells to engulf live neighboring cells for pro- or anti-tumorigenic purposes. It is unknown whether this extraordinary cellular event can be pathogenic in other diseases such as microcephaly, a condition characterized by a smaller than normal brain at birth. We find that mice mutant for the human microcephaly-causing gene Pals1, which exhibit diminished cortices due to massive cell death, also exhibit nuclei enveloped by plasma membranes inside of dividing cells. These cell-in-cell (CIC) structures represent a dynamic process accompanied by lengthened mitosis and cytokinesis abnormalities. As shown in tumor cells, ROCK inhibition completely …

Establishing An In Vitro Model To Study Avian Carotenoid Ketolation, Chidimma Okegbe Jan 2023

Establishing An In Vitro Model To Study Avian Carotenoid Ketolation, Chidimma Okegbe

Electronic Theses and Dissertations Archive

Carotenoid coloration is among the most widely recognized condition-dependent signal of quality in animals. This project first explored the possibility of establishing an in-vitro model to study carotenoid ketolation in two avian species, where both species use red keto-carotenoids for plumage pigmentation. Secondly, this project tested whether the carotenoid ketolation pathway would be affected by antioxidant and mitochondrial uncoupler treatments. We observed that most carotenoid ketolation could happen in in-vitro settings using primary hepatocytes for both House Finch and Domestic Canary. However, interestingly, the in-vitro system using House Finch hepatocytes failed to convert β-cryptoxanthin to 3-HE. Then, we tested the …

Ecological Complexity And Avoiding Pest Resurgence: Intuitions From Mathematical Ecology, Theresa Ong, John H. Vandermeer Jan 2023

Ecological Complexity And Avoiding Pest Resurgence: Intuitions From Mathematical Ecology, Theresa Ong, John H. Vandermeer

Dartmouth Scholarship

Pest resurgence is defined by a predictable increase in pests following declines resulting from the implementation of pest management practices. This phenomenon is notorious; familiar in biblical references to cyclical plagues of locust and discussions of pesticide treadmills, where pests resurge following the evolution of pesticide resistance. Targeted “silver-bullet” approaches, whether chemical, mechanical, biological, or genetic in nature, apply a reductionist lens toward the removal of a specific target pest, often ignoring ecological context, by application of said silver-bullet. In this review article, we cover how past and recent developments in theoretical ecology and complex systems dynamics provide a general …

Endothelial activation and sickle red blood cell (RBC) adhesion are central to the pathogenesis of sickle cell disease (SCD). Quantitatively, RBC-derived extracellular vesicles (REVs) are more abundant from SS RBCs compared with healthy RBCs (AA RBCs). Sickle RBC-derived REVs (SS REVs) are known to promote endothelial cell (EC) activation through cell signalling and transcriptional regulation at longer terms. However, the SS REV-mediated short-term non-transcriptional response of EC is unclear. Here, we examined the impact of SS REVs on acute microvascular EC activation and RBC adhesion at 2 h. Compared with AA REVs, SS REVs promoted human pulmonary microvascular ECs (HPMEC) …

Although murine models of coronary atherosclerotic disease have been used extensively to determine mechanisms, limited new therapeutic options have emerged. Pigs with familial hypercholesterolemia (FH pigs) develop complex coronary atheromas that are almost identical to human lesions. We reported previously that insulin-like growth factor 1 (IGF-1) reduced aortic atherosclerosis and promoted features of stable plaque in a murine model. We administered human recombinant IGF-1 or saline (control) in atherosclerotic FH pigs for 6 months. IGF-1 decreased relative coronary atheroma in vivo (intravascular ultrasound) and reduced lesion cross-sectional area (postmortem histology). IGF-1 increased plaque’s fibrous cap thickness, and reduced necrotic core, …

SCARB1 belongs to class B of Scavenger receptors (SRs) that are known to be involved in binding and endocytosis of various pathogens. SRs have emerging role in regulating innate immunity and host–pathogen interactions by acting in co-ordination with Toll-like receptors.Query Little is known about the function of SCARB1 in milk-derived mammary epithelial cells (MECs). This study reports the role of SCARB1 in infection and its potential association in TLR4 signaling on bacterial challenge in Goat mammary epithelial cells (GMECs). The novelty in the establishment of MEC culture lies in the method that aims to enhance the viability of the cells …

Eukaryotic initiation factor-4A2 (EIF4A2) is an ATP-dependent RNA helicase and a member of the DEAD-box protein family that recognizes the 5' cap structure of mRNAs, allows mRNA to bind to the ribosome, and plays an important role in microRNA-regulated gene repression. Here, we report on 15 individuals from 14 families presenting with global developmental delay, intellectual disability, hypotonia, epilepsy, and structural brain anomalies, all of whom have extremely rare de novo mono-allelic or inherited bi-allelic variants in EIF4A2. Neurodegeneration was predominantly reported in individuals with bi-allelic variants. Molecular modeling predicts these variants would perturb structural interactions in key protein domains. …

BACKGROUND: Diffuse midline gliomas (DMG) are highly invasive brain tumors with rare survival beyond two years past diagnosis and limited understanding of the mechanism behind tumor invasion. Previous reports demonstrate upregulation of the protein ID1 with H3K27M and ACVR1 mutations in DMG, but this has not been confirmed in human tumors or therapeutically targeted.

METHODS: Whole exome, RNA, and ChIP-sequencing was performed on the ID1 locus in DMG tissue. Scratch-assay migration and transwell invasion assays of cultured cells were performed following shRNA-mediated ID1-knockdown. In vitro and in vivo genetic and pharmacologic [cannabidiol (CBD)] inhibition of ID1 on DMG tumor growth …

Vulnerability And Resilience In A Tourism Destination, Sergio Alvarez, Alan Fyall

Rosen Research Review

In an industry that is critically vulnerable to hazards, some destinations are hit harder than others by the same or similar events. UCF Rosen College of Hospitality Management researchers, Dr. Sergio Alvarez and Dr. Alan Fyall, have developed a conceptual model that sees the destination's vulnerability as the sum of its physical, social-cultural, economic, ecological/environmental, and institutional vulnerabilities, nestled within its level of exposure to different hazards. Their framework offers to better equip destination management organizations (DMOs) by identifying vulnerabilities and facilitating more effective planning and decision making.

Innovate Ag Tech, Not Ag Tort: Why Legislative And Judicial Policies Favoring Tech Innovation And Big Ag's Ip Rights In Gmos Will Benefit Agricultural Production And Food Security, Gage Patton

Journal of Intellectual Property Law

Genetically modified organisms ("GMOs") play a critical role in technological advances in agriculture. GMOs contribute to generating higher crop yields, reduce the need for pesticide application, and even reduce food waste. Virtually all GMO technologies are developed by a small number of very large agricultural corporations. These corporations are responsible for the vast majority of research and development efforts for agricultural technologies. Agricultural research and development yields technological innovation, which produces better methods of food production. Therefore, the research and development efforts that big ag corporations engage in benefit everyone by providing better methods of food production. However, some critics …

Inventing The Right Drug: Artificial Intelligence May Just Be The Cure For An Antiquated Patent System, Matthew Hashemi

Journal of Intellectual Property Law

The transformation of the inventive process through the application of Artificial Intelligence, or “AI”, technology has radically enhanced the way pharmaceutical companies research and develop new drugs. While AI technology has led to improved accessibility to essential drugs and significant reductions in the time and cost required for drug research and development, it has also left the pharmaceutical industry with the puzzling question regarding how to encourage innovation with AI technology if AI machines cannot obtain patent protection under the current United States patent system. Granting patent protection ensures inventors can profit off their inventions, since patented inventions cannot be …

Whether convicted of a crime or booked into jail for a misunderstanding, many individuals rightfully seek to shed the criminal label from their reputations and start anew. However, photo evidence of the individual’s association with the criminal justice system by way of a published mugshot photograph is often only a click away for someone, such as a potential employer or new neighbor. Mugshots, originally created to aid in the detection of criminals, now serve primarily as a device of public shaming by law enforcement agencies who publish mugshots on their website and groups of private citizens who run social media …

Rapid Evolution Of The Fine-Scale Recombination Landscape In Wild House Mouse (Mus Musculus) Populations., Lydia K Wooldridge, Beth L Dumont

Faculty Research 2023

Meiotic recombination is an important evolutionary force and an essential meiotic process. In many species, recombination events concentrate into hotspots defined by the site-specific binding of PRMD9. Rapid evolution of Prdm9's zinc finger DNA-binding array leads to remarkably abrupt shifts in the genomic distribution of hotspots between species, but the question of how Prdm9 allelic variation shapes the landscape of recombination between populations remains less well understood. Wild house mice (Mus musculus) harbor exceptional Prdm9 diversity, with >150 alleles identified to date, and pose a particularly powerful system for addressing this open question. We employed a coalescent-based approach to construct …
