We describe a pipeline for optimized and streamlined multiplexed immunofluorescence-guided laser capture microdissection allowing the harvest of individual cells based on their phenotype and tissue localization for transcriptomic analysis with next-generation RNA sequencing. Here, we analyze transcriptomes of CD3+ T cells, CD14+ monocytes/macrophages, and melanoma cells in non-dissociated metastatic melanoma tissue. While this protocol is described for melanoma tissues, we successfully applied it to human tonsil, skin, and breast cancer tissues as well as mouse lung tissues. The genome of cancer cells contains circular extrachromosomal DNA (ecDNA) elements not found in normal cells. Analysis of clinical samples reveal they are common in most cancers and their presence indicates poor prognosis. They often contain enhancers and driver oncogenes that are highly expressed. The circular ecDNA topology leads to an open chromatin conformation and generates new gene regulatory interactions, including with distal enhancers. The absence of centromeres leads to random distribution of ecDNAs during cell division and genes encoded on them are transmitted in a non-mendelian manner. ecDNA can integrate into and exit from chromosomal DNA. This paper attempts to tackle state revisionism of monumental figures in American history through the case of Martin Luther King Jr., a radical who has been reimagined and whitewashed through conservative efforts for political purposes. I use examples of other historically revised prominent figures by the state to demonstrate the phenomenon. King's case can be connected to Derrick Bell and Critical Race Theory (CRT), an obscure legal study and fellow victim of conservative revisionism. This research project utilizes primary sources from diplomatic communiques, public speeches, political debates, and autobiographies of Israeli political elites in an attempt to better understand early Israeli democracy. Specifically, this project aims to frame this development of Israeli democracy and its institutions in the context of the 1948 Palestine War. The political elites of the Palestinian Jewish community who established the State of Israel came from various political, ethnic, religious, and linguistic backgrounds. This essay explores investigative deceptions, intentional lies used to gather information that is in the public interest, across different areas of law. In some contexts, such falsehoods are viewed as producing great social benefits. For example, civil rights laws embrace intentional lies by testers to smoke out discriminatory conduct. Under federal labor law, union 'salts' are allowed to hide their identity and acquire jobs with employers whose workers they wish to organize. In most cases, law enforcement officers may engage in elaborate subterfuges when operating 'stings' and interrogating witnesses. In these contexts, the law permits a person to lie in …

Anhedonia is a complex symptom of Major Depressive Disorder (MDD) that presents itself through multiple behavioral phenotypes of diminished reward processing. In order to better understand this deficit, we aim to look at a marker of reward that is sensitive to information content and valence, the Reward Positivity (RewP). The source of this signal is, however, up for debate. This study used concurrent EEG and MEG to establish the source of the RewP as a distributed network involving ventromedial prefrontal cortex (vmPFC), anterior midcingulate cortex (aMCC), and insulae.
