Artificial Light At Night Reverses Monthly Foraging Pattern Under Simulated Moonlight, Svenja Tidau, Jack Whittle, Stuart R. Jenkins, Thomas W. Davies
Artificial Light At Night Reverses Monthly Foraging Pattern Under Simulated Moonlight, Svenja Tidau, Jack Whittle, Stuart R. Jenkins, Thomas W. Davies
School of Biological and Marine Sciences
Mounting evidence shows that artificial light at night (ALAN) alters biological processes across levels of organization, from cells to communities. Yet, the combined impacts of ALAN and natural sources of night-time illumination remain little explored. This is in part due the lack of accurate simulations of the complex changes moonlight intensity, timing and spectra throughout a single night and lunar cycles in laboratory experiments. We custom-built a novel system to simulate natural patterns of moonlight to test how different ALAN intensities affect predator–prey relationships over the full lunar cycle. Exposure to high intensity ALAN (10 and 50 lx) reversed the …

Despite reports of a high incidence and various predictors of post-stroke depression (PSD), the underdiagnosis and undertreatment rates of PSD are still high. This study aimed to examine the incidence of depression in stroke patients and identify factors associated with PSD. This was a retrospective cohort study on ischemic stroke patients from the Geisinger Neuroscience Ischemic Stroke (GNSIS) registry. The following statistical analyses were performed to predict PSD in the studied population: a Kaplan-Meier estimator and a Cox proportional hazards model. A total of 5882 patients were included in the study. The median age at the time of an ischemic …

Masturbatory Behaviors Among Older Adult Populations: A Literature Review, Brandon Tischer
Masturbatory Behaviors Among Older Adult Populations: A Literature Review, Brandon Tischer
Journal of Undergraduate Research at Minnesota State University, Mankato
Sexual behaviors, such as masturbation, extend into older adulthood (50+ years in chronological age) and are present at all stages of the aging process. Different living situations and circumstances for older adults can affect masturbatory behavior, such as death of spouse, living in long-term care facilities, and cognitive decline. This literature review evaluates extant research on older adult masturbation, comprising 29 articles included in the final review, all published between 1991 and 2021. They consist of literature reviews, meta-studies, cross-sectional studies, semi-structured interviews, systematic reviews, and case studies. Current evidence suggests that masturbatory behaviors are an important part of older …

The harvesting and hauling operations of bioenergy feedstock is an important area in biofuel production. Production costs can be minimized by maintaining optimal machinery units for these operations. The objective of this study is to design an optimal harvesting unit for bioenergy refinery and estimate harvesting and hauling costs of energy cane. A biorefinery with the annual capacity of processing twenty-five million imp. gallons of ethanol were considered. Given the efficiency of harvesting, a two-row soldier system was considered. Considering the year-round supply of energy cane to the refinery, the optimal machinery unit was designed, and the combined operation costs …

Background: Hair antiretroviral concentrations are an objective and non-invasive measure of adherence to long-term antiretroviral therapy (ART) and can further predict virologic outcomes among people living with HIV/AIDS (PLWH). Zidovudine, one of the mainstream antiretrovirals in China, has been verified to have high reliability in adherence assessment, especially for its hair concentrations. However, data are limited in its predicting virologic outcomes. Therefore, this study aimed to characterize whether hair zidovudine concentrations can predict virologic suppression among Chinese PLWH compared with hair lamivudine concentrations and two self-reported measures, the overall frequency of adherence behaviors and percentage adherence. Methods: This cross-sectional study …

On 15 July 2022, the Delhi High Court, in Ms X v The Principal Secretary of Health and Family Welfare Department Government of NCT of Delhi (‘Ms X’), denied an unmarried woman permission to terminate her unwanted pregnancy at 23 weeks under Section 3 of the Medical Termination of Pregnancy Act, 1971. A week later, through an ad-interim order, the Supreme Court allowed the woman’s appeal against the Delhi High Court’s order, permitting termination as long as a Medical Board confirms that abortion is medically safe for the pregnant woman. In this post, I discuss both these orders, which differ …
Sweet Taste Signaling: The Core Pathways And Regulatory Mechanisms, Sunil Kumar Sukumaran, Salin Raj Palayyan
Sweet Taste Signaling: The Core Pathways And Regulatory Mechanisms, Sunil Kumar Sukumaran, Salin Raj Palayyan
Department of Nutrition and Health Sciences: Faculty Publications
Sweet taste, a proxy for sugar-derived calories, is an important driver of food intake, and animals have evolved robust molecular and cellular machinery for sweet taste signaling. The overconsumption of sugar-derived calories is a major driver of obesity and other metabolic diseases. A fine-grained appreciation of the dynamic regulation of sweet taste signaling mechanisms will be required for designing novel noncaloric sweeteners with better hedonic and metabolic profiles and improved consumer acceptance. Sweet taste receptor cells express at least two signaling pathways, one mediated by a heterodimeric G-protein coupled receptor encoded by taste 1 receptor members 2 and 3 (TAS1R2 …

The formation of carbon-carbon bonds from prebiotic precursors such as carbon dioxide represents the foundation of all primordial life processes. In extant organisms, this reaction is carried out by the carbon monoxide dehydrogenase (CODH)/acetyl coenzyme A synthase (ACS) enzyme, which performs the cornerstone reaction in the ancient Wood-Ljungdahl metabolic pathway to synthesize the key biological metabolite, acetylCoA. Despite its significance, a fundamental understanding of this transformation is lacking, hampering efforts to harness analogous chemistry. To address these knowledge gaps, we have designed an artificial metalloenzyme within the azurin protein scaffold as a structural, functional, and mechanistic model of ACS. We …

Mutant KRAS (KM), the most common oncogene in lung cancer (LC), regulates fatty acid (FA) metabolism. However, the role of FA in LC tumorigenesis is still not sufficiently characterized. Here, we show that KMLC has a specific lipid profile, with high triacylglycerides and phosphatidylcholines (PC). We demonstrate that FASN, the rate-limiting enzyme in FA synthesis, while being dispensable in EGFR-mutant or wild-type KRAS LC, is required for the viability of KMLC cells. Integrating lipidomic, transcriptomic and functional analyses, we demonstrate that FASN provides saturated and monounsaturated FA to the Lands cycle, the process remodeling oxidized phospholipids, such as PC. Accordingly, …

It is well known that the properties of hematopoietic stem/progenitor cells (HSCs), such as their self-renewal ability and multipotency, are maintained through interactions with mesenchymal stem/stromal cells (MSCs). MSCs are rare cells that are present in the bone marrow and are useful for clinical applications due to their functional ability. To obtain the necessary number of cells, MSCs must be cultured to expand, but this causes a remarkable decrease in stem cell properties, such as multipotency and proliferation ability. In this study, we show that the c-Mpl signal, which is related to the maintenance of hematopoietic stem cells, has an …

Fluid-Structure Interaction With Kelvin-Voigt Damping And Quantized State Integration Methods, Rasika Lasantha Mahawattege
Electronic Theses and Dissertations Archive
The class of fluid-structure interaction models in the physical dimension d = 2,3 has been the object of intense mathematical studies in the past several years. In the case of static interface, justified to be appropriate for small, rapid oscillations of the structure, the typical configuration considers a structure – modeled by a d-dimensional wave-type equation –surrounded by a fluid –modeled by the d-dimensional dynamic Stokes equation involving pressure. Coupling takes place at the common interface of the two media where the individual equations evolve and is given by differential operators. Thus, the overall system provides a physical illustration of …

Development Of Soil Microbial Community Structure In Created And Natural Wetlands, Wendy A. Owens
Development Of Soil Microbial Community Structure In Created And Natural Wetlands, Wendy A. Owens
Theses
Wetlands are ecologically and economically important, providing ecosystem services such as biogeochemical cycling, carbon sequestration, flood mitigation, and wildlife habitat. Many of these services are the result of the unique microbial communities found in wetlands. Degradation and destruction of wetlands, from direct human development or indirect stressors caused by climate change, pollution, or invasive species, disrupts community structure and provision of services. Although restoration has been adopted as a mechanism to counteract net loss of function, the success rate in achieving functional equivalence with natural wetlands is low. To improve wetland conservation and outcomes for wetland creation, a better understanding …

Qualifying Factors That Predict Weight Loss Success Within Diet Interventions, Jordan Losavio
LSU Master's Theses
Evidence-based diet strategies lead to weight loss for some, while for others, the same diet approach could result in trivial weight loss or even in weight gain. This prompts the underlying research question: what factors predict that a given diet strategy will be efficacious for some, but not for others? This research was a secondary analysis of clinical weight loss studies to determine which factors influence an individual’s weight loss success for three diet intervention strategies: low-carbohydrate, low-fat, and low-calorie. A combination of physiological, psychological, behavioral, and dietary factors was evaluated for each diet study. Multiple linear regression and logistic …

Infants with B-cell acute lymphoblastic leukemia (B-ALL) have poor outcomes because of chemotherapy resistance leading to high relapse rates. Tisagenlecleucel, a CD19-directed chimeric antigen receptor T-cell (CART) therapy, is US Food and Drug Administration approved for relapsed or refractory B-ALL in patients ≤25 years; however, the safety and efficacy of this therapy in young patients is largely unknown because children(n = 14). Sixty-four percent of patients (n = 9) achieved minimal residual disease-negative remission after CART and 50% of patients remain in remission at last follow-up. All patients with high disease burden at time of CART infusion (>M1 marrow) …

Coronavirus disease 2019 (COVID-19) is primarily an infection of the respiratory tract, but it can have multisystem manifestations. Cardiac complications of COVID-19 can range from acute myocardial injury, cardiac arrhythmias, or heart failure, amongst others. Heart failure (HF) in COVID-19 can be a de novo process or due to worsening of pre-existing cardiovascular ailment. HF in a patient with COVID-19 not only poses challenges in clinical presentation and management of COVID-19 but also affect prognosis of the patient. This article aims to succinctly revisit the implications of this pandemic regarding pre-existing HF or new-onset HF based on prevailing data. It …

As a well-known glycolysis inhibitor for anticancer treatment, 2-Deoxy-D-glucose (2DG) inhibits the growth and survival of cancer cells by interfering with the ATP produced by the metabolism of D-glucose. In addition, 2DG inhibits protein glycosylation in vivo by competing with D-mannose, leading to endoplasmic reticulum (ER) stress and unfolded protein responses in cancer cells. However, the molecular details underlying the impact of 2DG on protein glycosylation remain largely elusive. With an integrated approach to glycoproteomics and proteomics, we characterized the 2DG-induced alterations in N-glycosylation, as well as the cascading impacts on the whole proteome using the HT29 colorectal cancer …

Alphaviruses can cause severe human arthritis and encephalitis. During virus infection, structural changes of viral glycoproteins in the acidified endosome trigger virus-host membrane fusion for delivery of the capsid core and RNA genome into the cytosol to initiate virus translation and replication. However, mechanisms by which E1 and E2 glycoproteins rearrange in this process remain unknown. Here, we investigate prefusion cryoelectron microscopy (cryo-EM) structures of eastern equine encephalitis virus (EEEV) under acidic conditions. With models fitted into the low-pH cryo-EM maps, we suggest that E2 dissociates from E1, accompanied by a rotation (∼60°) of the E2-B domain (E2-B) to expose …

The capacity to efficiently deliver the gene-editing enzyme complex to target cells is favored over other forms of gene delivery as it offers one-time hit-and-run gene editing, thus improving precision and safety and reducing potential immunogenicity against edited cells in clinical applications. Here we performed a proof-of-mechanism study and demonstrated that a simian adenoviral vector for DNA delivery can be repurposed as a robust intracellular delivery platform for a functional Cas9/guide RNA (gRNA) complex to recipient cells. In this system, the clinically relevant adenovirus was genetically engineered with a plug-and-display technology based on SpyTag003/SpyCatcher003 coupling chemistry. Under physiological conditions, an …
