Current chemoradiation therapy suffers from normal tissue toxicity. Thus, we are proposing incorporating gold nanoparticles (GNPs) and docetaxel (DTX), as they have shown very promising synergetic radiosensitization effects. Here, we explored the effect of a DTX prodrug encapsulated in lipid nanoparticles (LNPDTX-P) on GNP uptake in pancreatic cancer models in vitro and in vivo. For the in vitro experiment, a pancreatic cancer cell line, MIA PaCa-2, was cultured and dosed with 1 nM GNPs and 45 nM free DTX or an equivalent dose of LNPDTX-P. Bees obtain nutrients from flowers. Pollen provides proteins and fats; nectar provides carbohydrates. Pollen is important for providing essential amino acids which honey bees must obtain from their diet for proper development. Commercial honey bee colonies, however, pollinate monocultures, which contain one crop type thus, one unbalanced nutritional resource. We examined how a lack of protein diversity affects honey bee pollen foraging behavior. Bees were raised on three diet treatments: no manipulation, all 10 essential amino acids (EAAs, i.e., protein building-blocks), or only 6/10 EAAs. Bee-collected pollen was trapped upon return to the hive and nutritional content was analyzed. We …

Earth’s increased global temperature as a response to climate change has caused less snowfall and earlier snowmelt in spring. These changes can alter the phenology of organisms such as through plant leafing and flower blooming. This can be harmful to species reliant on plants that are more sensitive to temperature changes. The frosted elfin (Callophrys irus) is a species of concern in 11 states—including RI, MA, and CT. The fetal-to-adult hemoglobin switching at about the time of birth involves a shift in expression from γ-globin to β-globin in erythroid cells. Effective re-expression of fetal γ-globin can ameliorate sickle cell anemia and β-thalassemia. Despite the physiological and clinical relevance of this switch, its posttranscriptional regulation is poorly understood. Here, we identify Pumilo 1 (PUM1), an RNA-binding protein with no previously reported functions in erythropoiesis, as a direct posttranscriptional regulator of β-globin switching. PUM1, whose expression is regulated by the erythroid master transcription factor erythroid Krüppel-like factor (EKLF/KLF1), peaks during erythroid differentiation, binds γ-globin messenger RNA (mRNA), and reduces γ-globin …

Staphylococcus aureus is a pathogen responsible for a wide variety of life-threatening diseases such as bacteremia, endocarditis, and pneumoniae. S. aureus has been a major concern in recent years due to the rampant spread of antibiotic resistance. The ability of S. aureus to form biofilms aids in the spread of antibiotic resistance as biofilms are a known hotspot for horizontal gene transfer. Biofilms also protect cells from host immune responses and antibiotics, making these infections very difficult to treat. How to deliver best care in various clinical settings remains a vexing problem. All pertinent healthcare-related questions have not, cannot, and will not be addressable with costly time- and resource-consuming controlled clinical trials. At present, evidence-based guidelines can address only a small fraction of the types of care that clinicians deliver. Furthermore, underserved areas rarely can access state-of-the-art evidence-based guidelines in real-time, and often lack the wherewithal to implement advanced guidelines. The incidence of oropharyngeal squamous cell carcinoma (OPSCC) has increased rapidly in the United States, driven by rising human papillomavirus (HPV) infections in the U.S. population. HPV-positive OPSCC patients have a better prognosis than HPV-negative patients. To gain insights into the unique biology of HPV(+) OPSCC that may contribute to its clinical behaviors, we performed a multi-stage epigenome-wide methylation profiling of leukocyte and tumor DNA in OPSCC patients and compared the methylation levels of CpG sites between HPV(+) and HPV(-) OPSCC patients. Hearing loss and the related otologic manifestations are receiving increased scrutiny as significant causes of morbidity in Fabry disease. However, the relative risks of auditory deficits among patients with Fabry disease relative to the general population without a diagnosis of Fabry disease have not been studied. This study aims to explore the associations between Fabry disease and hearing-related manifestations using a nationwide population-based dataset. We identified study patients for this cross-sectional study from the 2015−2017 claims databases of the Taiwan Longitudinal Health Insurance Database 2005. GATA-binding factor 1 (GATA1) is a transcription factor that governs the development and function of multiple hematopoietic cell lineages. GATA1 is expressed in hematopoietic stem and progenitor cells (HSPCs) and is essential for erythroid lineage commitment; however, whether it plays a role in hematopoietic stem cell (HSC) biology and the development of myeloid cells, and what that role might be, remains unclear. Bovine mastitis is a common disease among dairy cattle characterized by the inflammation of the udders and loss of milk production. Mastitis-associated Escherichia coli (MAEC) are frequent causes of the disease, but the features that distinguish them from other E. coli strains remain enigmatic. MAEC infections can range from sub-clinical to severe, acute cases that can be fatal. Historically, the severity of mastitis has been attributed to host factors but more recently, a few bacterial genes have been shown to contribute to virulence in mastitis infections. Large amounts of sediment from upstream erosion and mining flowed to the bay resulting in the downsizing of the bay’s square miles (Environmental Protection Agency, 2022). From September 2021 through December 2022 I worked with birth mothers and adoptees who are making efforts to complicate the narrative around adoption online. This resulted in a multi-platform project, creating a highly successful TikTok channel, an Instagram page, a private Facebook group and a Substack newsletter. The platforms decided on as well as the content on these platforms were decided upon in collaboration with the community, using deep listening and collecting both qualitative and quantitative data. A significant body of literature has been generated related to the detection of measurable residual disease (MRD) at the time of achieving complete remission (CR) in patients with hairy cell leukemia (HCL). Advanced prostate malignancies are a leading cause of cancer-related deaths in men, in large part due to our incomplete understanding of cellular drivers of disease progression. We investigate prostate cancer cell dynamics at single-cell resolution from disease onset to the development of androgen independence in an in vivo murine model. We observe an expansion of a castration-resistant intermediate luminal cell type that correlates with treatment resistance and poor prognosis in human patients. Moreover, transformed epithelial cells and associated fibroblasts create a microenvironment conducive to pro-tumorigenic immune infiltration, which is partially androgen responsive. Preeclampsia (PE) is a pregnancy-specific disorder attributed to deficient extravillous trophoblast (EVT) invasion into the uterus, but the mechanism of EVT invasion remains unclear. In this study, we found significantly elevated expression of microRNA 21 (miR21), which negatively regulates trophoblast invasion and migration, in preeclamptic placentae. Whole-genome RNA sequencing revealed that PPP2R2B, which encodes PP2A Bβ, and the Hippo pathway are downstream targets of miR21. The effects of miR21 on trophoblast mobility were abolished in LATS1T1079A/S909A and YAP-5SA mutants. The cancer metastasis process involves dysregulated oncogenic kinase signaling, but how this orchestrates metabolic networks and signal cascades to promote metastasis is largely unclear. Here we report that inhibition of glutamate dehydrogenase 1 (GDH1) and ribosomal S6 kinase 2 (RSK2) synergistically attenuates cell invasion, anoikis resistance, and immune escape in lung cancer and more evidently in tumors harboring epidermal growth factor receptor (EGFR)-activating or EGFR inhibitor-resistant mutations. Mechanistically, GDH1 is activated by EGFR through phosphorylation at tyrosine 135 and, together with RSK2, enhances the cAMP response element-binding protein (CREB) activity via CaMKIV signaling, thereby promoting metastasis. Pregnancy is a challenging time for anyone. For LGBTQ people there are unique obstacles that make it especially difficult to navigate a medical system that was not built for them. Finn Schubert is trans and six months pregnant. He’s loving the experience but stressed by insurance claim denials, concerns for his safety, and medical incompetence.

This capstone project includes a 20-minute documentary, text, and a photo essay.

This thesis examines how the practice of sciencing, tech-romancing, simulacra, and simulation manifests in both the text and symbols used on audiences of the Church of Scientology’s outreach materials. It specifically examines its official FAQ page. Self-framing was observed that satisfied the definition of sciencing and tech-romancing. Under an Estranged approach, Morrill examined selections of the Church’s FAQ pages, coding for instances of sciencing, tech-romancing, simulacra, and simulation. Biomolecular condensates form via coupled associative and segregative phase transitions of multivalent associative macromolecules. Phase separation coupled to percolation is one example of such transitions. Here, we characterize molecular and mesoscale structural descriptions of condensates formed by intrinsically disordered prion-like low complexity domains (PLCDs). These systems conform to sticker-and-spacers architectures. Stickers are cohesive motifs that drive associative interactions through reversible crosslinking and spacers affect the cooperativity of crosslinking and overall macromolecular solubility. Our computations reproduce experimentally measured sequence-specific phase behaviors of PLCDs. Within simulated condensates, networks of reversible inter-sticker crosslinks organize PLCDs into small-world topologies. The overall dimensions of PLCDs …

Seventeen international consortia are collaborating on a human reference atlas (HRA), a comprehensive, high-resolution, three-dimensional atlas of all the cells in the healthy human body. Laboratories around the world are collecting tissue specimens from donors varying in sex, age, ethnicity, and body mass index. However, harmonizing tissue data across 25 organs and more than 15 bulk and spatial single-cell assay types poses challenges. Here, we present software tools and user interfaces developed to spatially and semantically annotate ("register") and explore the tissue data and the evolving HRA.
