Extracellular Vesicle-Associated Biomolecules As Potential Biomarkers For Alzheimer's Disease Diagnosis, Lina Bedoya Martinez
Extracellular Vesicle-Associated Biomolecules As Potential Biomarkers For Alzheimer's Disease Diagnosis, Lina Bedoya Martinez
Electronic Theses and Dissertations, 2020-2023
Alzheimer's disease (AD) involves progressive neurodegeneration leading to the loss of normal neuronal function. Extracellular accumulation of amyloid-beta (Aß), through the abnormal cleavage of the amyloid precursor protein (APP) by ß- and γ-secretases, is one of the hallmarks of AD. Current research focuses on finding potential candidates for biomarkers and techniques with improved sensitivity for early disease detection. Extracellular vesicles (EVs) found in body fluids are a source of biomarkers for AD diagnosis. EVs transport pathologically significant biomolecules, like nucleic acids and proteins, across the blood-brain barrier, mediating local and distant cell-to-cell communication. Therefore, this study evaluated EV-associated DNA and …

Making Emotions Trustworthy, Xiaoyu Ke
Making Emotions Trustworthy, Xiaoyu Ke
Arts & Sciences Graduate Student Theses and Dissertations
I investigate ways by which agents can make their emotions more trustworthy from a perspective that’s informed by recent psychological research on the nature of emotion regulation. This dissertation is a collection of three articles, each addressing one type of cases of untrustworthy emotions in a particular context. Chapter one (Developing Appropriate Emotions) concerns emotions that are untrustworthy because they are irrelevant to the moral or epistemic judgements at hand. By situating the problem in the context of a situationist challenge to neo-Aristotelian virtue ethics, I argue that we can address this problem by developing ameliorative abilities such as emotion …
A Synthesis-Based Approach To The Study Of Fe(Ii) Oxidation In Smectites, Robert Joe Kupper Ii
A Synthesis-Based Approach To The Study Of Fe(Ii) Oxidation In Smectites, Robert Joe Kupper Ii
Arts & Sciences Graduate Student Theses and Dissertations
Smectite clay minerals are present in diverse environments on Earth as a major product of silicate weathering. Although smectites are compositionally varied, they almost always incorporate iron. Iron-bearing smectites are an important component of the biogeochemical iron cycle, serving as a major iron fraction in many soils and altered rocks as well as participating in redox reactions within the surrounding environment. Iron incorporates into smectites in the ferric [Fe(III)] form at the modern, oxygenated surface of Earth but clays forming in anoxic settings instead incorporate ferrous iron [Fe(II)]. Owing to their ubiquity at the surface of the modern Earth, the …

Dealing With Dimensionality: Problems And Techniques In High-Dimensional Statistics, Cezareo Rodriguez
Arts & Sciences Graduate Student Theses and Dissertations
In modern data analysis, problems involving high dimensional data with more variables than subjects is increasingly common. Two such cases are mediation analysis and distributed optimization. In Chapter 2 we start with an overview of high dimensional statistics and mediation analysis. In Chapter 3 we motivate and prove properties for a new marginal screening procedure for performing high dimensional mediation analysis. This screening procedure is shown via simulation to perform better than benchmark approaches and is applied to a DNA methylation study. In Chapter 4 we construct a cryptosystem that accurately performs distributed penalized quantile regression in the high-dimensional setting …

Structure And Morphology Of Photoreactive Monomer–Nanoparticle Mixtures Under Patterned Irradiation, Shreyas Pathreeker
Structure And Morphology Of Photoreactive Monomer–Nanoparticle Mixtures Under Patterned Irradiation, Shreyas Pathreeker
Dissertations - ALL
Establishing processing–structure relationships is central to materials science. In this work, we seek answers to whether morphology can be controlled during Light–Induced Self–Writing (LISW) in polymer composite materials, and if yes, how? LISW is based on the ability of light to undergo divergence–free propagation in photoreactive media due to photopolymerization–induced rise in refractive index of the medium. Notably, LISW in polymeric materials has found use predominantly for the fabrication of optical waveguides but has not been explored in polymer–inorganic mixtures. Using through–mask projection which creates a spatially periodic array of optical beams, this work extends LISW to nanoparticle–monomer mixtures to …

Weaving Subjects At The Obraje De Chincheros: Indigenous Contributions To The Colonial Aesthetic, Maria Smith
Weaving Subjects At The Obraje De Chincheros: Indigenous Contributions To The Colonial Aesthetic, Maria Smith
Dissertations - ALL
This dissertation highlights the contributions that colonial textile producers at the colonial textile mill, the Obraje de Chincheros (c. 1570-1820), influenced the colonial Peruvian aesthetic. Peruvian weavers were seen as superior weavers at the time of the Spanish invasion and obrajes were established to salvage this skill while they were simultaneously subjected to colonial control strategies. At the Obraje, rural indigenous and mestizo textile producers were disciplined into the colonial hierarchy via their labor and conversion to Catholicism. At the same time, the Obraje workforce, largely women and children, shaped the colonial aesthetic via their labor. It was their laborial …

Introduction: Limbic predominant age related TDP-43 encephalopathy neuropathological change (LATE-NC) is a recently characterized brain disease that mimics Alzheimer’s disease (AD) clinically. To date, LATE-NC is difficult to diagnose antemortem using clinical information or biomarkers. Recent studies suggest concentrations of extracellular vesicle (EVs) protein cargo derived from neuronal and glial cells may serve as useful diagnostic biomarkers for AD and other neurodegenerative diseases.
Methods: TDP-43 was evaluated in neuronal (NDEVs), astrocyte (ADEVs), and microglial derived extracellular vesicles (MDEVs). EV preparations were isolated from the plasma of research subjects with autopsy-confirmed diagnoses, including many with LATE (n = 22). …

System Shock: Using -Omics Data To Characterize Biological Remodeling In The Microbiome, Resistome, And Transcriptome Of Bacteria, Winston Eugene Anthony
Arts & Sciences Graduate Student Theses and Dissertations
The human experience is indelibly linked to microbial life. So much so, that the holobiont theory has been coined to define the assemblage of host and microbe as a discrete ecological unit. Perturbation of the commensal microbiome can create niche space for pathogens, which through the concomitant rise of antimicrobial resistance represent an ever-evolving danger. In Chapter 2 of this dissertation, I describe the results of an interventional study designed to directly perturb the healthy gut microbiome and observe the dynamics of taxonomic composition and functional recovery. I observe significant decreases after 5 days of antibiotic treatment in the gold …

Insights Into The Non-Osmoregulatory Function Of A Pollen-Specific Mechanosensitive Ion Channel, Kari Miller
Arts & Sciences Graduate Student Theses and Dissertations
Pollen, the male gametophyte of flowering plants, delivers the sperm cells to the ovule to carry out sexual reproduction. During this process, the pollen grain undergoes dramatic physical changes. Survival requires careful control of cell mechanics, particularly the balance between protoplast expansion and cell wall resistance. One control mechanism is the use of a mechanosensitive (MS) ion channel, MscS-Like (MSL)8. This pollen-specific protein was previously shown to be essential for pollen survival during hydration and was proposed to function as a tension-gated osmoregulator. However, direct proof of osmoregulation during initial hydration has not yet been found. In fact, studies of …

Arts & Sciences Graduate Student Theses and Dissertations
The mammalian brain has unparalleled diversity of cell types with distinct molecular, morphological, connectional and functional properties. The specialization of these cells occurs during development through a series of spatially and temporally controlled changes in gene expression that are critical for proper assembly and function. Neurons in particular have complex and elaborate gene regulatory systems which allow specific combinations of genes to be expressed at distinct levels and at discrete developmental stages, giving rise to heterogenous neuronal subtypes. Identifying unique neuronal gene regulatory factors that contribute to the dynamic changes in gene expression are critical to understanding the molecular mechanism …
