Recent wildlife agency efforts aimed at hunter recruitment, retention, and reactivation (R3) are examples of behavior change interventions. Therefore, these R3 programs and policies should be strategically designed to meet hunters’ goals and motivations. We analyzed survey responses (n=808) from North Carolina resident migratory bird hunters regarding potential Sunday hunting legalization to serve as an illustrative case demonstrating how the capabilities-opportunity-motivation-behavior (COM-B) system can be used to conceptualize and more effectively test potential hunter behavior changes prompted by debated R3 strategies. A growing body of research reflects the ways in which trans students, who do not adhere to society’s rigid gender binary in practices, policies, and/or norms (Kosciw et. al., 2020; Luecke, 2011; Marine & Nicolazzo, 2014), are ill-served by the current social climates in the majority of our nation’s public schools. Trans students are at a disproportionate risk for harassment, low academic achievement, and school dropout (Payne & Smith, 2018), and transgender individuals experience disparate rates of serious mental health concerns including depression, anxiety, and suicidality (Austin, 2022). Background and ObjectivesFrontotemporal dementia (FTD) is a highly heritable disorder. The majority of genetic cases are caused by autosomal dominant pathogenic variants in the chromosome 9 open reading frame 72 (c9orf72), progranulin (GRN), and microtubule-associated protein tau (MAPT) gene. The proximal tubule is a key regulator of kidney function and glucose metabolism. Diabetic kidney disease leads to proximal tubule injury and changes in chromatin accessibility that modify the activity of transcription factors involved in glucose metabolism and inflammation. Here we use single nucleus RNA and ATAC sequencing to show that diabetic kidney disease leads to reduced accessibility of glucocorticoid receptor binding sites and an injury-associated expression signature in the proximal tubule. We hypothesize that chromatin accessibility is regulated by genetic background and closely-intertwined with metabolic memory, which pre-programs the proximal tubule to respond differently to external stimuli. Herbivory by wild and domestic ungulates can influence tree recruitment and understory forest communities throughout the world. Herbivore-driven declines in tree recruitment have been observed for quaking aspen (Populus tremuloides), a foundation species whose health and management is recognized as a critical priority throughout much of its range. Livestock fencing is commonly used to promote aspen regeneration, but its effectiveness is rarely assessed, especially across large spatial scales. Using a livestock-reduction experiment, we evaluated the effects of ungulate herbivory on aspen in the Great Basin and southern Cascades, an expansive and environmentally heterogeneous region where aspen faces the …

A protein’s structure is determined by its amino acid sequence and post-translational modifications, and provides the basis for its physiological functions. Roughly a third of the proteome in mammalian species comprises proteins that contain highly unstructured or intrinsically disordered regions. Proteins with extensive unstructured regions are referred to as intrinsically disordered proteins (IDPs). Recent advances in experimental and computational analyses predicted multiple interacting partners for the disordered regions of proteins, implying critical roles in signal transduction and regulation of biological processes. Protein kinase-mediated phosphorylation plays a critical role in many biological processes. However, the identification of key regulatory kinases is still a great challenge. Here, we develop a trans-omics-based method, central kinase inference, to predict potentially key kinases by integrating quantitative transcriptomic and phosphoproteomic data. Using known kinases associated with anti-cancer drug resistance, the accuracy of our method denoted by the area under the curve is 5.2% to 29.5% higher than Kinase-Substrate Enrichment Analysis. We further use this method to analyze trans-omic data in hepatocyte maturation and hepatic reprogramming of human dermal fibroblasts, uncovering 5 kinases as regulators in the two …

Understanding the B cell response to SARS-CoV-2 vaccines is a high priority. High-throughput sequencing of the B cell receptor (BCR) repertoire allows for dynamic characterization of B cell response. Here, we sequenced the BCR repertoire of individuals vaccinated by the Pfizer SARS-CoV-2 mRNA vaccine. We compared BCR repertoires of individuals with previous COVID-19 infection (seropositive) to individuals without previous infection (seronegative). We discovered that vaccine-induced expanded IgG clonotypes had shorter heavy-chain complementarity determining region 3 (HCDR3), and for seropositive individuals, these expanded clonotypes had higher somatic hypermutation (SHM) than seronegative individuals. The continuation of utilizing fossil fuels as cooking energy sources in rural communities in the Philippines causes more citizens to be at risk of developing numerous health illnesses. Accumulation of food waste and the burning of coal emit harmful chemicals which contribute to environmental problems such as climate change and global warming. Follicular lymphoma (FL) is a B-cell malignancy with a complex tumor microenvironment that is rich in nonmalignant immune cells. We applied single-cell RNA sequencing to characterize the diverse tumor and immune cell populations of FL and identified major phenotypic subsets of FL T cells, including a cytotoxic CD4 T-cell population. We characterized four major FL subtypes with differential representation or relative depletion of distinct T-cell subsets. By integrating exome sequencing, we observed that somatic mutations are associated with, but not definitive for, reduced MHC expression on FL cells.
