Hepatocyte-Hepatic Stellate Cell Axis In Potentiation Of Alcohol And Hiv-Induced Liver Injury, Moses O. New-Aaron

Theses & Dissertations

Human Immunodeficiency Virus (HIV) remains a global threat, with approximately 38.4 million active infections and 33 million HIV-related deaths since the first case. While many may be tempted to think of HIV as a relic of the past, emerging data suggests otherwise. By the end of 2021, approximately 1.5 million HIV incidences and 650 000 mortalities were reported globally. The etiologies of HIV-related mortality are numerous. Liver disease is one of the leading etiologies of HIV-related mortality, especially in recent times of antiretroviral therapy (ART)-induced longevity among people living with HIV (PLWH). While co-infections with hepatotropic viruses notoriously contribute to …

Role Of E3 Ubiquitin Ligase Ubr5 In B Cell Development And Mantle Cell Lymphoma, Tyler J. Gilbreath Dec 2022

Role Of E3 Ubiquitin Ligase Ubr5 In B Cell Development And Mantle Cell Lymphoma, Tyler J. Gilbreath

Theses & Dissertations

UBR5 is part of the ubiquitin proteasome system (UPS), most well-known as a degradation and recycling pathway in which proteins are tagged with a chain of ubiquitin protein and degraded by the proteasome. However, the UPS is multifunctional and other important roles include tagging protein with ubiquitin for cell signaling purposes. UBR5 is a homologous to the E6AP C-terminus (HECT) E3 ubiquitin ligase, meaning ubiquitin is transferred to UBR5 on a cysteine residue within the HECT domain and then onto a target substrate. UBR5 has mutations in many types of cancers: breast, ovarian, liver, and lymphomas. Recently published data shows …

The Role Of Ubiquitin E3 Ligase Fbxo21 In Steady-State, Stressed, And Malignant Hematopoiesis, Karli J. Wittorf Dec 2022

The Role Of Ubiquitin E3 Ligase Fbxo21 In Steady-State, Stressed, And Malignant Hematopoiesis, Karli J. Wittorf

Theses & Dissertations

Hematopoietic stem cells (HSCs) allow for the formation of all cell types in the blood and maintain these populations throughout a person’s life. The process of hematopoiesis is regulated through a variety of molecular mechanisms that are either signaled from the cell’s environment (extrinsic) or from within the cell itself (intrinsic). One intrinsic mechanism that regulates hematopoietic cell fate decisions is the ubiquitin proteasome system (UPS). The UPS controls protein levels by tagging them with polyubiquitin chains and promoting their degradation through the proteasome. The key component of the UPS is the ubiquitin E3 ligase as this protein is the …

The Relationship Between P66shc Expression And Proliferation Of Human Prostate Cancer Cells With Mntnbuoe-2-Pyp Treatment, Thao Nhi Radik Dec 2022

The Relationship Between P66shc Expression And Proliferation Of Human Prostate Cancer Cells With Mntnbuoe-2-Pyp Treatment, Thao Nhi Radik

Theses & Dissertations

Prostate cancer (PCa) is the second leading cause of cancer-related deaths in the U.S. in men, mainly due to the development of castration-resistant PCa (CRPC). MnTnBuOE-2-PyP5+ (BuOE), a superoxide dismutase (SOD) mimic, inhibits human PCa cell proliferation through hydrogen peroxide (H2O2). p66Shc protein level is upregulated in CRPC and is associated with cell proliferation in PCa cells. We propose that the different sensitivity of those PCa cells to BuOE could be due to the expression levels of p66Shc protein.

Our data showed that PCa cells that expressed high levels of p66Shc are more resistant to BuOE …

Frailty Determination And Responsiveness In Left Ventricular Assist Devices, Sarah Schroeder Dec 2022

Frailty Determination And Responsiveness In Left Ventricular Assist Devices, Sarah Schroeder

Theses & Dissertations

Frailty, the state of being vulnerable with susceptibility to decreased age-related physical reserve, is found in 56-79% of adults with end-stage HF receiving durable left ventricular assist device (dLVAD) therapy. Frailty increases risk of adverse events, including increased mortality in the dLVAD population, but less is known about depression, quality of life (QOL), cognition and frailty responsiveness. Therefore, we aimed to describe and compare frailty (5-Meter Walk Test, 5MWT), depression (Patient Health Questionnaire-9, PHQ-9), quality of life (EQ5D Visual Analog Scale, EQ-5D VAS) and cognition (Trailmaking B) from pre- to 12-months post-dLVAD We also compared demographics, clinical characteristics, and post-dLVAD …

Characterization Of Metastatic Cutaneous Squamous Cell Carcinoma In The Immunosuppressed Patient, Megan E. Wackel

Theses & Dissertations

Patients who are immunosuppressed, such as solid organ transplant recipients (SOTRs), are known to have a higher risk of developing cutaneous squamous cell carcinoma (cSCC). This population is at a higher risk of metastasis and worse disease-specific survival, with over 1 million cases and an estimated 3,932 to 8,791 deaths each year in the United States. Large cancer genomic databases have reported on genomic drivers for many cancer subtypes, yet cSCC has not been included. The immunosuppressed population is generally ineligible to receive immunotherapy which poses a challenge for treatment options in cases that develop metastasis. After we performed a …

Optimized Production Of Second-Generation Bioethanol From A Spent C4 Grass: Vetiver (Chrysopogon Zizanioides), Sameer Neve, Dibyendu Sarkar, Zhiming Zhang, Rupali Datta

Michigan Tech Publications, Part 1

Vetiver grass (Chrysopogon zizanioides) is well-known for its contaminant phytoextraction potential and its capacity to reduce soil erosion, owing to its massive, dense root system. However, the shoots are not major contributors to either of these processes, and are either not utilized at all or they become part of the waste stream. It is well-recognized that lignocellulosic biomass can serve as a source of raw material to produce second-generation bioethanol. This study investigated the simultaneous saccharification and fermentation (SSF) of acid–alkali pretreated vetiver (VG) shoots by Saccharomyces cerevisiae. Vetiver shoots were obtained from three sources: (1) shoots from VG grown …

Motives For (In)Effective Giving: Comparing Rural And Urban Groups In South Africa., Luvuyo Bulelani Magwaza

Master's Theses

Different cultures have their own set of norms and values that not only shape people’s motives but also influences their decision making. What may be viewed as logical and ethical in one culture, may be seen as illogical or unethical in another. One area that is consistently affected by cross-cultural differences in motives is charitable giving. Recently, there has been an increase in interest around effective altruism— a social movement and philosophy that argues, people should give to charities that do the most good. Prior research that has found that people do not give based on efficiency; instead, people give …

Understanding The Perceived Risk And Biopsychosocial Impact Of The Covid-19 Pandemic On People Living With Hiv And Pre-Exposure Prophylaxis Users In Nebraska: A Mixed Methods Approach, Titilola O. Labisi

Theses & Dissertations

The COVID-19 pandemic disrupted HIV care and prevention services. COVID-19 guidelines required physical and social distancing, resulting in movement restrictions and limited healthcare services. People living with HIV (PLWH) may be more vulnerable to the pandemic’s impact due to immune suppression and socioeconomic status than people without HIV. Consistent use of pre-exposure prophylaxis (PrEP), condoms, and regular sexually transmitted infection tests are critical strategies for HIV prevention. The pandemic’s impact on PLWH have not been thoroughly studied, and whether PLWH may be more impacted than PrEP users is inconclusive.

This research aimed to use an explanatory sequential mixed methods design …

Gender Differences In Breast Cancer Treatment Decisions And Related Survival: A Study Using The National Cancer Database, Shamim Shahi Dec 2022

Gender Differences In Breast Cancer Treatment Decisions And Related Survival: A Study Using The National Cancer Database, Shamim Shahi

Theses & Dissertations

Despite increasing incidence, male breast cancer (MBC) remains understudied in almost all aspects of disease management. MBC patients are more likely to be diagnosed with larger tumors, at an advanced stage, and to experience poorer survival than female patients. There is a need to identify modifiable factors that can improve MBC patients’ survival.

This dissertation aimed to investigate whether there was a difference in the refusal of recommended treatments and related survival, by gender, in breast cancer patients enrolled in the National Cancer Database. Specifically, we conducted separate analyses for four treatments: hormone therapy, chemotherapy, radiation therapy, and surgery. Then, …

Polymeric Chloroquine In Metastatic Pancreatic Cancer Therapy, Rubayat Islam Khan

Theses & Dissertations

Hydroxychloroquine (HCQ) is an established drug that has been used for the treatment of multiple diseases, including malaria, rheumatoid arthritis, and lupus erythematosus. Several recently published studies have highlighted its beneficial use in various types of cancers, and a plethora of clinical trials are currently evaluating its use in cancer therapies, wherein some of the results look promising. The preclinical studies to date suggest HCQ can work effectively in conjunction with traditional cancer treatments, as it has been shown to sensitize tumor cells to a wide range of chemotherapeutic agents. Polymeric HCQ (PCQ), a macromolecular multivalent version of HCQ, has …

An Analysis Of The Genetic And Environmental Mechanisms Behind Aortic Aneurysm Development And Inhibition, Kathryn E. Jespersen Dec 2022

An Analysis Of The Genetic And Environmental Mechanisms Behind Aortic Aneurysm Development And Inhibition, Kathryn E. Jespersen

Theses & Dissertations

Aortic aneurysm is a widespread condition, characterized by dilation and weakening of the aortic vessel wall. Dissection and rupture of the aorta due to uncontrolled aneurysm progression carries a high risk of death. Although both thoracic aortic aneurysms (TAA) and abdominal aortic aneurysms (AAA) share similar risks of rupture and mortality, the pathophysiology of each disease is unique. TAA is a hallmark feature of Marfan syndrome (MFS), a connective tissue disorder characterized by malformation of functional elastic fibers in the aortic extracellular matrix (ECM). This malformation of elastic fibers is attributed to mutation of the gene encoding the fibrillin-1 protein, …

Novel Mechanisms Of Protein Kinase C Α Regulation And Function, Xinyue Li Dec 2022

Novel Mechanisms Of Protein Kinase C Α Regulation And Function, Xinyue Li

Theses & Dissertations

Protein kinase Cα (PKCα) is a member of the PKC family of serine/threonine kinases, which have been implicated in regulation of many cellular processes, including cell proliferation, differentiation, survival, and transformation. A large body of evidence from the Black laboratory and others support an anti-proliferative function of PKCα in normal epithelial tissues, including the intestinal mucosa and endometrial epithelium. PKCα is also tumor suppressive in epithelial cancers, such as colorectal cancer (CRC) and endometrial cancer (EC). However, a major obstacle to harnessing the tumor suppressive functions of PKCα to benefit patients is the widespread loss of PKCα expression in tumors. …

The Role Of Parkin In Mitochondrial Dna, Eliezer Lichter Dec 2022

The Role Of Parkin In Mitochondrial Dna, Eliezer Lichter

Theses & Dissertations

Mitochondria are at the center of biological phenomena such as aging and diseases, especially neurodegenerative diseases. While the discovery of mitochondria only came approximately 200 years after the cell was discovered, a lot of progress has been made since. The mitochondrial genome encodes proteins vital for mitochondrial function. These proteins are only a subset of the proteins present in mitochondria; the rest are nuclear encoded. The nucleus also encodes cytosolic proteins vital for mitochondrial maintenance. One of these is Parkin, an E3 ubiquitin ligase that ubiquitinates mitochondrial proteins as mitochondria become depolarized. Its activity has been shown to be involved …

Underlying Mechanism Of Ovarian Reserve Preservation From Gonadotoxicity, Yi Luan Dec 2022

Underlying Mechanism Of Ovarian Reserve Preservation From Gonadotoxicity, Yi Luan

Theses & Dissertations

Cancer therapies often lead to premature ovarian insufficiency by ovarian damage and follicle depletion. However, the oocyte death pathway induced by chemotherapies has been debated. Here we clarified the precise mechanism of ovarian depletion induced by cyclophosphamide and doxorubicin. Cyclophosphamide induced direct apoptosis in oocytes rather than overactivation of primordial follicles. Dormant oocytes instead of activated oocytes with high PI3K activity were more sensitive to cyclophosphamide. Although the ABL inhibitor, GNF2, has been proposed to ameliorate the apoptosis of primordial follicles against cyclophosphamide, GNF2 did not prevent the loss of primordial follicles induced by cyclophosphamide in the systems tested. Oocyte-specific …

Metoprolol Disrupts Sterol Biosynthesis Through Inhibition Of 7-Dehydrocholesterol Reductase (Dhcr7), Luke B. Allen Dec 2022

Metoprolol Disrupts Sterol Biosynthesis Through Inhibition Of 7-Dehydrocholesterol Reductase (Dhcr7), Luke B. Allen

Theses & Dissertations

Cholesterol is essential for life. It is particularly important in the brain as it relies on de novo synthesis of cholesterol following the formation of the blood brain barrier (BBB). As such, disrupting sterol biosynthesis during neurodevelopment can have devastating outcomes. The most common post-lanosterol sterol biosynthesis disorder, Smith-Lemli-Opitz Syndrome, arises from a faulty DHCR7 enzyme. DHCR7 has also been shown to be inhibited by several psychotropic medications. Here we assess six beta-blockers and their effects on sterol biosynthesis in vitro. Two beta-blockers, metoprolol and nebivolol strongly inhibit DHCR7 in four separate in vitro models of both mouse and …

Deciphering The Mechanism And Function Of Hsp100 Unfoldases From Protein Structure, Grace Lee, Rebecca S Kim, Sang Bum Lee, Sukyeong Lee, Francis T F Tsai Dec 2022

Deciphering The Mechanism And Function Of Hsp100 Unfoldases From Protein Structure, Grace Lee, Rebecca S Kim, Sang Bum Lee, Sukyeong Lee, Francis T F Tsai

Faculty, Staff and Students Publications

Hsp100 chaperones, also known as Clp proteins, constitute a family of ring-forming ATPases that differ in 3D structure and cellular function from other stress-inducible molecular chaperones. While the vast majority of ATP-dependent molecular chaperones promote the folding of either the nascent chain or a newly imported polypeptide to reach its native conformation, Hsp100 chaperones harness metabolic energy to perform the reverse and facilitate the unfolding of a misfolded polypeptide or protein aggregate. It is now known that inside cells and organelles, different Hsp100 members are involved in rescuing stress-damaged proteins from a previously aggregated state or in recycling polypeptides marked …

While the role of common genetic variants in multiple sclerosis (MS) has been elucidated in large genome-wide association studies, the contribution of rare variants to the disease remains unclear. Herein, a whole-genome sequencing study in four affected and four healthy relatives of a consanguineous Italian family identified a novel missense c.1801T > C (p.S601P) variant in the

Mitochondrial Autophagy In Ischemic Aged Livers, Jae-Sung Kim, William C Chapman, Yiing Lin Dec 2022

Mitochondrial Autophagy In Ischemic Aged Livers, Jae-Sung Kim, William C Chapman, Yiing Lin

2020-Current year OA Pubs

Mitochondrial autophagy (mitophagy) is a central catabolic event for mitochondrial quality control. Defective or insufficient mitophagy, thus, can result in mitochondrial dysfunction, and ultimately cell death. There is a strong causal relationship between ischemia/reperfusion (I/R) injury and mitochondrial dysfunction following liver resection and transplantation. Compared to young patients, elderly patients poorly tolerate I/R injury. Accumulation of abnormal mitochondria after I/R is more prominent in aged livers than in young counterparts. This review highlights how altered autophagy is mechanistically involved in age-dependent hypersensitivity to reperfusion injury.

Variation In The M-Opioid Receptor (Oprm1) And Offspring Sex Are Associated With Maternal Behavior In Rhesus Macaques (Macaca Mulatta), Zachary Baron Dec 2022

Variation In The M-Opioid Receptor (Oprm1) And Offspring Sex Are Associated With Maternal Behavior In Rhesus Macaques (Macaca Mulatta), Zachary Baron

Theses and Dissertations

A m-opioid receptor (OPRM1) single-nucleotide-polymorphism, found in both humans and rhesus macaques mediates the mother-infant attachment bond. Because mothers treat their sons and daughters differently, it is somewhat surprising that the role of infant sex has not been assessed in the context of a maternal-OPRM1-genotype-by-infant-sex interaction. The present study investigates the effect of maternal-OPRM1-genotype and infant sex on mother-infant behaviors. Over the first six months of offspring life, mother-infant behavioral data assessing attachment quality was collected twice weekly from a large number of rhesus monkey mother-infant pairs (N=161 dyads; n=64 female infants, n=97 male infants). Mothers were genotyped for OPRM1 …

Sustaining Ireland, Body And Soul: A Woman Leader's Story Of The Cooperative Movement, Elizabeth Summerfield Dec 2022

Sustaining Ireland, Body And Soul: A Woman Leader's Story Of The Cooperative Movement, Elizabeth Summerfield

The Journal of Values-Based Leadership

This article tells the story of the Cooperative Movement in Ireland during the late nineteenth and early twentieth centuries from the perspective of one of its woman leaders. It does so in order to distil lessons for the contemporary thought leadership of sustainability from a period before the term was coined. It does so with the warrant of Albert Einstein:

The distinction between the past, the present and the future is only a stubbornly persistent illusion.

Its evidence base is historical literature, but its argument and analysis draw on recent research in leadership studies, neuroscience and theology.

A Changed Understanding Of Miracles In Religious Tourism, Stephen F. Haller Dec 2022

A Changed Understanding Of Miracles In Religious Tourism, Stephen F. Haller

International Journal of Religious Tourism and Pilgrimage

In this modern age, an unsceptical acceptance of supernatural events–those which cannot be explained as part of the natural order of things–is less common than it once was. This trend is reflected in the declining frequency of miracle-cures certified by the Medical Bureau at Lourdes. Yet miracles past, and the promise of possible miracles in the present, still attract multitudes of religious pilgrims and tourists to sacred sites all over the world. While the frequency of miracles goes down, the appeal of miracles goes on, and the number of religious visitors has not declined. What role do miracles now play …
