Mechanistic Insights Into An Iron-Responsive Repressor, Fep1, From Schizosaccharomyces Pombe, Allison Nicole Kimsey
Mechanistic Insights Into An Iron-Responsive Repressor, Fep1, From Schizosaccharomyces Pombe, Allison Nicole Kimsey
Theses and Dissertations
Iron is an essential metal cofactor required for a variety of biochemical pathways in most living organisms. In recent years, the nonpathogenic fungi Schizosaccharomyces pombe has been used to study iron regulation and uncover the molecular mechanisms for controlling intracellular iron levels. Some of these regulation pathways are conserved with those of pathogenic fungi, making these studies applicable for the future development of antifungals that target iron homeostasis. In S. pombe, the GATA-type transcriptional repressor Fep1 is responsible for repressing iron uptake genes during times of iron abundance. Previous reports have proposed that the binding of an iron-sulfur (Fe-S) …

Development Of Peptidomimetic Inhibitors Targeting The Polo Box Domain Of Plk1 As Potential Cancer Therapeutics, Jessy Mariam Stafford
Development Of Peptidomimetic Inhibitors Targeting The Polo Box Domain Of Plk1 As Potential Cancer Therapeutics, Jessy Mariam Stafford
Theses and Dissertations
PLK1 overexpression has shown to be associated with tumor progression and an unfavorable prognosis in a wide variety of cancer types. Interfering with PLK1 genetically and chemically induces apoptotic cell death in cancer cells thus confirming PLK1 as a validated anticancer target. From a drug discovery perspective, PLK1 offers two functionally distinct, essential, and interconnected target sites in one molecule. These are the ATP binding kinase domain (KD) and the unique Polo Box Domain (PBD), where the interplay between these results in dynamic structural regulation of catalytic activity and sub cellular localization. Since the KD is conserved among kinases and …

Characterization Of The Interaction Between Grx4-Bol2 And The Iron-Responsive Transcription Factor Aft1 In Candida Glabrata, Evan A. Talib
Theses and Dissertations
Iron is an indispensable protein cofactor that plays a critical role in biological functions such as cellular respiration. However, excess iron causes cellular damage via the generation of reactive oxygen species. To obtain optimal intracellular iron levels, eukaryotic cells have developed mechanisms to tightly regulate iron levels. The molecular mechanism for iron regulation in the pathogenic yeast Candida glabrata has not yet been explored, although this yeast bears iron regulation pathways that are very similar to the model yeast Saccharomyces cerevisiae. Understanding how C. glabrata regulates iron levels is critical for the treatment of infections caused by this opportunistic …
Identifying Racial Differences In Colorectal Polyp Profile At Screening Colonoscopy Using Traditional Regression And Machine Learning Approaches, Yuqi Wu
Theses and Dissertations
Background
Colorectal cancer (CRC), the second leading cause of cancer and cancer deaths in the United States, can largely be prevented by screening the age-appropriate adult population with colonoscopy to remove precancerous lesions. However, despite the encouraging annual, population-wide decline in CRC incidence and mortality in recent decades, and widespread adoption of screening colonoscopy since 2000 (~60% population coverage rate), the Black population has consistently suffered higher CRC incidence and mortality than Whites, currently 20% and 40% respectively, little changed from the 2000 rats of 26% and 52%.
The reason for this is thought to be that the Black population …

Natural Variation Of Fructose-1,6-Bisphosphatase In Colias Butterflies, Andrea Blair Stokes
Natural Variation Of Fructose-1,6-Bisphosphatase In Colias Butterflies, Andrea Blair Stokes
Theses and Dissertations
In a process known as the evolutionary recursion, it is possible to follow how adaptation affects natural selection at successive stages of a species’ life cycle. Through studying allele and genotype frequencies of enzymes in glycolysis, a central metabolic pathway, it is possible to document how polymorphisms can shift the evolving functionality of enzymes within or among species. Fructose-1,6-bisphosphatase (FBPase) is an enzyme in gluconeogenesis that converts fructose-1,6-bisphosphate to fructose 6- phosphate. It is also known to convert the unused amino acid carbon skeletons of glycolysis into glycogen storage. Appearing to play a vital role in the acquisition, storage, and …

Peromyscus Maniculatus Bairdii And Peromyscus Maniculatus Sonoriensis Polymorphisms, Matthew Dixon Lucius
Peromyscus Maniculatus Bairdii And Peromyscus Maniculatus Sonoriensis Polymorphisms, Matthew Dixon Lucius
Theses and Dissertations
Sequencing technology has advanced greatly over the past couple of decades. This has led to many studies developing hypotheses based on genomic data. This requires many genomic studies to have up-to-date information on the species’ or subject’s genome being investigated. If a study does not have accurate or up-to-date background information, then they cannot fully develop their theories that are being established. In this study, I utilize outbred Peromyscus maniculatus, deer mice that are genetically diverse, to record the polymorphisms that are found throughout the Peromyscus maniculatus genome. I first explore the differences between the polymorphisms found in Peromyscus …
Too Feminine For Execution?: Gender Stereotypes And The Media’S Portrayal Of Women Sentenced To Death, Kelsey M. Collins
Too Feminine For Execution?: Gender Stereotypes And The Media’S Portrayal Of Women Sentenced To Death, Kelsey M. Collins
Theses and Dissertations
Traditional gender norms prescribing women as more nurturing and less aggressive than men have led to both the reluctance to view women as capable of violence, as well as a greater willingness to execute men than women in the United States. To make sense of the instances where women are sentenced to death, the media often pathologizes and/or demonizes them. Scholars have found that demonizing and dehumanizing those executed is a necessity to the implementation of capital punishment, both in cases of male and female defendants. To better understand how the news media have framed the gender and racial narratives …
Decellularization Strategies Of Naturally Derived Biomaterials For Tissue Engineering Applications, Julia Elizabeth Hohn
Decellularization Strategies Of Naturally Derived Biomaterials For Tissue Engineering Applications, Julia Elizabeth Hohn
Theses and Dissertations
In 2017, over 3.5 million peripheral vascular surgeries were performed worldwide with over 400,000 vascular repair or replacement surgeries being performed in the United States each year alone. As the number of vascular repair surgeries, including both coronary and peripheral bypass grafting procedures, continues to increase each year, these statistics indicate an urgent need for more effective and readily available replacement materials. Regenerative medicine and tissue engineering (TE) approaches, including the design, fabrication, and validation of suitable biomaterials in vitro that direct the repair and regeneration of damaged tissues, have been proposed to alleviate this problem. While advanced biomaterials have …

Arctic Ocean And Subarctic Seas Dynamics In A Changing Climate, Sarah B. Hall
Arctic Ocean And Subarctic Seas Dynamics In A Changing Climate, Sarah B. Hall
Theses and Dissertations
Salinity is the primary determinant of the Arctic Ocean’s vertical density stratification in the upper ocean, which has major implications on the ocean’s physical dynamics alongside a period of rapidly declining sea ice. In recent decades, the Arctic’s freshwater content (FWC) has increased as a result of the accumulation of freshwater source inputs. Additional freshwater exported to the North Atlantic may hinder overturning processes that are vital to the regulation of global climate. This dissertation employs in situ measurements, satellite observations, and ocean model simulations to better understand salinity and freshwater changes in the Arctic Ocean during this changing climate. …

Perceived Sexual Intent: Power, Relationship Status, And Gender, Joseph Eric Padgett
Perceived Sexual Intent: Power, Relationship Status, And Gender, Joseph Eric Padgett
Theses and Dissertations
Does lacking power cause people to think potential partners are less interested in engaging with them sexually? Do men and women perceive the interests of potential sex partners differently? Does the amount of sexual intent perceived by people who are in a romantic relationship differ from that of singles? Power has been shown to impact perceptions in other contexts, and the way people rate the attractiveness of potential mates is shown to differ depending upon their own relationship status. Similarly, gender differences are a central theme in discussions of sex related perceptions, preferences, and behaviors. In this study, I utilize …

We are facing two converging waves of racial retrenchment. The first, which arose following the Civil Rights Movement, is nearing a legal milestone. This term or the next, the Supreme Court will prohibit affirmative action in higher education. When it does, the Court will cement decades of conservative jurisprudence that has systematically eroded the right to remedy racial inequality.
The second wave is more recent but no less significant. Following 2020’s global uprising for racial justice, rightwing forces launched a coordinated assault on antiracism itself. The campaign has enjoyed early success. As one measure, GOP officials have passed, proposed or …

Sticky Me: Self-Relevance Slows Reinforcement Learning, M Golubickis, Cn Macrae
School of Psychology
A prominent facet of social-cognitive functioning is that self-relevant information is prioritized in perception, attention, and memory. What is not yet understood, however, is whether similar effects arise during learning. In particular, compared to other people (e.g., best friend) is information about the self acquired more rapidly? To explore this matter, here we used a probabilistic selection task in combination with computational modeling (i.e., Reinforcement Learning Drift Diffusion Model analysis) to establish how self-relevance influences learning under conditions of uncertainty (i.e., choices are based on the perceived likelihood of positive and negative outcomes). Across two experiments, a consistent pattern of …
