BACKGROUND: The identification of differentially expressed tumor-associated proteins and genomic alterations driving neoplasia is critical in the development of clinical assays to detect cancers and forms the foundation for understanding cancer biology. One of the challenges in the analysis of pancreatic ductal adenocarcinoma (PDAC) is the low neoplastic cellularity and heterogeneous composition of bulk tumors. To enrich neoplastic cells from bulk tumor tissue, coring, and laser microdissection (LMD) sampling techniques have been employed. Ecosystem structure—that is the species present, the functions they represent, and how those functions interact—is an important determinant of community stability. This in turn aects how ecosystems respond to natural and anthropogenic crises, and whether species or the ecological functions that they represent are able to persist. Here we use fossil data from museum collections, literature, and the Paleobiology Database to reconstruct trophic networks of Tethyan paleocommunities fromthe Anisian and Carnian (Triassic), Bathonian (Jurassic), and Aptian (Cretaceous) stages, and compare these to a previously reconstructed trophic network from a modern Jamaican reef community. Trained immunity is an innate immune memory response that is induced by a primary inflammatory stimulus that sensitizes monocytes and macrophages to a secondary pathogenic challenge, reprogramming the host response to infection and inflammatory disease. Dietary fatty acids can act as inflammatory stimuli, but it is unknown if they can act as the primary stimuli to induce trained immunity. Here we find mice fed a diet enriched exclusively in saturated fatty acids (ketogenic diet; KD) confer a hyper-inflammatory response to systemic lipopolysaccharide (LPS) and increased mortality, independent of diet-induced microbiome and hyperglycemia. The management of human-shark interactions can benefit from the implementation of effective shark hazard mitigation measures. A Shark-Management-Alert-in-Real-Time (SMART) drumline trial in the Capes region of Western Australia was instigated after several serious incidents involving surfers and white sharks (Carcharodon carcharias). The project aimed to determine whether white sharks (target species), which were relocated after capture, remained offshore using satellite and acoustic tagging. Over a 27-month period, 352 fish were caught, 55% of which comprised tiger sharks (Galeocerdo cuvier). Ninety-one percent of animals were released alive in good condition. The purpose of this qualitative collective case study was to describe parent participation for home-based educators and parents at an Early Head Start home-based program in rural Appalachia. Parental participation in the Early Head Start home-based program was generally defined as the active participation of children with disabilities’ parents in a home-based program guided by home-based educators through the implementation of early intervention services. Maslow’s hierarchy of needs guided this study, as it relates to both identifying barriers that may prevent parents from being involved and illuminates how Early Head Start educators help families meet growth and developmental needs. The purpose of this hermeneutic phenomenological study was to understand the experiences of former college-athletes who persist in medical school. The theories that guided this study were the achievement goal theory by Nicholls and the self-determination theory by Deci and Ryan, as each theory relates to the motivation to succeed in academics, as well as athletic participation. Eleven participants were purposefully selected to address the question: How do former college student-athletes describe their experiences in relation to the attributes and characteristics that allowed them to persist in medical school? Aluminum (Al) is a metallic toxicant at high concentrations following natural or unnatural exposures. Dietary intake is considered as the main source of aluminum exposure in children. We used data from 366 typically developing (TD) children (ages 2−8 years) who participated as controls in an age- and sex-matched case−control study in Jamaica. We investigated additive and interactive associations among environmental factors and children’s genotypes for glutathione S-transferase (GST) genes (GSTT1, GSTM1, GSTP1), in relation to having a detectable blood aluminum concentration (BAlC) of >5.0 μg/L, using multivariable logistic regression models. Exposure to early life adversity has long term consequences on cognitive function. Most research has focused on understanding components of early life adversities that contribute to later risk, including poverty, trauma, maltreatment, and neglect. Whereas these factors, in the aggregate, explain a significant proportion of emotional and cognitive problems, there are serious gaps in our ability to identify potential mechanisms by which early life adversities might promote vulnerability or resilience. Here we discuss early life exposure to unpredictable signals from the caretaker as an understudied type of adversity that is amenable to prevention and intervention. Based on a profound understanding of the great significance of constructing an ecological protection pattern in Northwest China, this paper systematically summarizes the achievements, new problems and new trends of ecological protection in Northwest China, and accordingly, puts forward an overall spatial arrangement for the ecological protection under "two regions (watersheds)-six zones (core ecological function zones)-multiple points (key ecological points)". With the increase in the average life span, the incidence of neurodegenerative diseases also increases, among which Alzheimer's disease (AD) and Parkinson's disease (PD) are the two most common types of neurological disorders. AD and PD share the same pathogenic mechanism that involves a class of proteins called beta-amyloid (Aβ), which, under the induction of some factors, changes from its natural monomeric state to the oligomers containing β-sheet, which is neurotoxic and thus leads to cell damage or apoptosis. There is a growing body of evidence that therapy-related myeloid neoplasms (t-MNs) with driver gene mutations arise in the background of clonal hematopoiesis (CH) under the positive selective pressure of chemo- and radiation therapies. Uncovering the exposure relationships that provide selective advantage to specific CH mutations is critical to understanding the pathogenesis and etiology of t-MNs. In a systematic analysis of 416 patients with t-MN and detailed prior exposure history, we found that TP53 mutations were significantly associated with prior treatment with thalidomide analogs, specifically lenalidomide. We demonstrated experimentally that lenalidomide treatment provides a selective advantage to Trp53-mutant hematopoietic stem …

Abstract Ecosystems in the North American Arctic-Boreal Zone (ABZ) experience a diverse set of disturbances associated with wildfire, permafrost dynamics, geomorphic processes, insect outbreaks and pathogens, extreme weather events, and human activity. Climate warming in the ABZ is occurring at over twice the rate of the global average, and as a result the extent, frequency, and severity of these disturbances are increasing rapidly. Disturbances in the ABZ span a wide gradient of spatiotemporal scales and have varying impacts on ecosystem properties and function. Ribosomal RNAs (rRNAs) are the most abundant cellular RNAs, and their synthesis from rDNA repeats by RNA polymerase I accounts for the bulk of all transcription. Despite substantial variation in rRNA transcription rates across cell types, little is known about cell-type-specific factors that bind rDNA and regulate rRNA transcription to meet tissue-specific needs. Using hematopoiesis as a model system, we mapped about 2,200 ChIP-seq datasets for 250 transcription factors (TFs) and chromatin proteins to human and mouse rDNA and identified robust binding of multiple TF families to canonical TF motifs on rDNA. The study presents a novel approach to objectively assessing the upper-extremity motor symptoms in spinocerebellar ataxia (SCA) using data collected via a wearable sensor worn on the patient’s wrist during upper-extremity tasks associated with the Assessment and Rating of Ataxia (SARA). First, we developed an algorithm for detecting/extracting the cycles of the finger-to-nose test (FNT). We extracted multiple features from the detected cycles and identified features and parameters correlated with the SARA scores. We carried out integrated host and pathogen metagenomic RNA and DNA next generation sequencing (mNGS) of whole blood (n = 221) and plasma (n = 138) from critically ill patients following hospital admission. We assigned patients into sepsis groups on the basis of clinical and microbiological criteria. From whole-blood gene expression data, we distinguished patients with sepsis from patients with non-infectious systemic inflammatory conditions using a trained bagged support vector machine (bSVM) classifier (area under the receiver operating characteristic curve (AUC) = 0.81 in the training set; AUC = 0.82 in a held-out validation set). The guidelines for management of children with 22q11.2 deletion syndrome (22q11DS) highlight the risk for developing hypocalcemia after surgery and recommend monitoring calcium perioperatively. Despite this guidance, little has been published on postoperative hypocalcemia and 22q11DS. Our goals were to evaluate the frequency of perioperative calcium monitoring and examine how often postoperative hypocalcemia was identified. This is a retrospective chart review of patients in our 22q Center's repository. Inclusion criteria were a diagnosis of 22q11DS and a history of a non-cardiac surgical procedure. Data collected included all non-cardiac surgeries and perioperative calcium labs. In total, 68 patients were included and …

In the post-Roe world, can a state rationally claim that the value of human life justifies the imposition of abortion bans but does not demand that a state protect the vulnerable young who are “born human beings”—commonly called “minors” or “children”—and are entitled to protection under a state’s laws? This essay advances the claim that it cannot. Although C.S. Lewis’s Chronicles of Narnia (1950-1956) are still hugely popular today, some critics have accused the books of representing masculinity and femininity in an outmoded way. The three Walden Media films, The Lion, the Witch and the Wardrobe (2005), Prince Caspian (2008) and The Voyage of the Dawn Treader (2010), appear to adopt a more contemporary perspective, especially as far as the representation of fighting girls is concerned. Despite David Abram’s fear that reading disrupts people’s “attunement to environing nature,” fantasy literature can vibrantly convey how to hear our environments as it describes characters attuning their ears to particular places. Garth Nix’s Old Kingdom series (1995-2021) and Patrick Ness’s Chaos Walking trilogy (2008-10) develop an echoing ecopoetics of place through both world-building and style. Their fantasy worlds emphasize that characters must relearn to listen in unfamiliar environments: adjusting their expectations and interpretations of background sounds, recognising significant silences, adapting to new ways of communicating, and seeking meaning in nonhuman sounds rather than dismissing them as noise. Robin Hobb’s Assassin’s Apprentice demonstrates a significant effect of epic fantasy’s conventions for creating the history of a fictional world. By prefacing each chapter with an epigraph from an official in-world historical text before giving a first-person personal narrative, the novel blurs the boundaries between text and paratext, public and private, official history and personal myth-making. This structure raises questions about what is central and marginal in history, suggesting the extent to which historical narrative is constructed in the imagination by taking the facts surrounding a central event from which the historian is absent—a process much like negative space drawing …
