"A Sort Of Pain, Which Is New": Unresolved Grief In British Romantic Literature, Eta Farmacelia Nurulhady

This dissertation examines the cultural phenomenon of mourning in relation to British Romantic Literature. In chapters on the work of William Wordsworth, Jane Austen, Felicia Hemans, and Charles Lamb, it argues that the Romantic period, as a time of increased mobility due to three revolutions, wars, and the expansion of empire, was a moment when unresolved grief became a common experience. Using the psychologist Pauline Boss’s concept of “ambiguous loss” as a lens for a new reading of British Romantic writing, and distinguishing this concept from the modern concept of “nostalgia,” this dissertation analyzes poetry, novels, and essays written by …

The alarming rise of multidrug-resistant Gram-positive bacteria has precipitated a healthcare crisis, necessitating the development of new antimicrobial therapies. Here we describe a new class of antibiotics based on a ring-fused 2-pyridone backbone, which are active against vancomycin-resistant enterococci (VRE), a serious threat as classified by the Centers for Disease Control and Prevention, and other multidrug-resistant Gram-positive bacteria. Ring-fused 2-pyridone antibiotics have bacteriostatic activity against actively dividing exponential phase enterococcal cells and bactericidal activity against nondividing stationary phase enterococcal cells. The molecular mechanism of drug-induced killing of stationary phase cells mimics aspects of fratricide observed in enterococcal biofilms, where both …

Mice with a functional human immune system serve as an invaluable tool to study the development and function of the human immune system in vivo. A major technological limitation of all current humanized mouse models is the lack of mature and functional human neutrophils in circulation and tissues. To overcome this, we generated a humanized mouse model named MISTRGGR, in which the mouse granulocyte colony-stimulating factor (G-CSF) was replaced with human G-CSF and the mouse G-CSF receptor gene was deleted in existing MISTRG mice. We address the problem of de novo design and synthesis of nucleic acid nanostructures, a challenge that has been considered in the area of DNA nanotechnology since the 1980s and more recently in the area of RNA nanotechnology. Toward this goal, we introduce a general algorithmic design process and software pipeline for rendering 3D wireframe polyhedral nanostructures in single-stranded RNA. To initiate the pipeline, the user creates a model of the desired polyhedron using standard 3D graphic design software. Reactive oxygen species (ROS), a by-product of aerobic life, are highly reactive molecules with unpaired electrons. The excess of ROS leads to oxidative stress, instigating the peroxidation of polyunsaturated fatty acids (PUFA) in the lipid membrane through a free radical chain reaction and the formation of the most bioactive aldehyde, known as 4-hydroxynonenal (4-HNE). 4-HNE functions as a signaling molecule and toxic product and acts mainly by forming covalent adducts with nucleophilic functional groups in proteins, nucleic acids, and lipids. Genes with moderate to low expression heritability may explain a large proportion of complex trait etiology, but such genes cannot be sufficiently captured in conventional transcriptome-wide association studies (TWASs), partly due to the relatively small available reference datasets for developing expression genetic prediction models to capture the moderate to low genetically regulated components of gene expression. Drawing upon participant observations and semi-structure interviews with 10 Puerto Rican grassroots organizers from the Tampa Bay area of Florida, this project examines the processes by which Puerto Rican diaspora members build, maintain, and utilize social and symbolic ties as resources for organizing and executing grassroot projects and campaigns with a dual focus on the Puerto Rican community in the Tampa Bay area and in Puerto Rico. Cancer cachexia is a lethal metabolic syndrome featuring muscle wasting with preferential loss of fast-twitching muscle mass through an undefined mechanism. Here, we show that cancer induces muscle wasting by selectively degrading myosin heavy chain (MHC) subtypes IIb and IIx through E3 ligase UBR2-mediated ubiquitylation. Induction of MHC loss and atrophy in C2C12 myotubes and mouse tibialis anterior (TA) by murine cancer cells required UBR2 up-regulation by cancer. Genetic gain or loss of UBR2 function inversely altered MHC level and muscle mass in TA of tumor-free mice. Androgen receptor (AR) signaling is crucial for driving prostate cancer (PCa), the most diagnosed and the second leading cause of death in male patients with cancer in the United States. Androgen deprivation therapy is initially effective in most instances of AR-positive advanced or metastatic PCa. However, patients inevitably develop lethal castration-resistant PCa (CRPC), which is also resistant to the next-generation AR signaling inhibitors. Most CRPCs maintain AR expression, and blocking AR signaling remains a main therapeutic approach. About one-fourth of recurrent estrogen receptor-positive (ER+) breast cancers lose ER expression, leading to endocrine therapy failure. However, the mechanisms underlying ER loss remain to be fully explored. We now show that 14-3-3τ, up-regulated in ∼60% of breast cancer, drives the conversion of ER+ to ER- and epithelial-to-mesenchymal transition (EMT). We identify ERα36, an isoform of ERα66, as a downstream effector of 14-3-3τ. Overexpression of 14-3-3τ induces ERα36 in xenografts and tumor spheroids. The regulation is further supported by a positive correlation between ERα36 and 14-3-3τ expression in human breast cancers. ERα36 can antagonize ERα66 and inhibit ERα66 expression. This study presents the Relocatable Operational Ocean Model (ReOMo), which can be used as a Crisis Ocean Modelling System in any region of the global ocean that is free from ice. ReOMo can be quickly nested into an existing coarser resolution (parent) model. The core components of ReOMo are the NEMO hydrodynamic model and Rose-Cylc workflow management software. Endometriosis is an estrogen-dependent inflammatory disease that develops in reproductive-aged women who experience pelvic pain and infertility. Even though endometriosis is not a new disease, its molecular etiology has not been clearly elucidated. Defects in the immune system might be one of the factors that promote endometriosis progression. For example, elevated levels of proinflammatory cytokines are associated with endometriosis. Interferon is one of the cytokines that is elevated in endometriotic tissues compared with normal endometrium. Therefore, high interferon levels play a crucial role in endometriosis progression. Neural crest cells (NCCs) are multipotent stem cells that can differentiate into multiple cell types, including the osteoblasts and chondrocytes, and constitute the majority of the craniofacial skeleton. Here, we show through in vitro and in vivo studies that the transcriptional regulators Yap and Taz have redundant functions as key determinants of the specification and differentiation of NCCs into osteoblasts or chondrocytes. Primary and cultured NCCs deficient in Yap and Taz switched from osteogenesis to chondrogenesis, and NCC-specific deficiency for Yap and Taz resulted in bone loss and ectopic cartilage in mice. Youth sport offers physical, mental, and social health benefits with numerous sport and recreational enterprises and venues offering respective programming of multiple sports. Participation rates of youth in sport and recreational activities, however, are declining. In addition, the age most noted to drop out of youth sport is approximately age 13. Numerous influencers impact participation and retention in youth sport. The current report examines factors impacting youth sport participation, motivation, and retention. Claytor Lake is a 4,500 acre impoundment of the New River, located in southwestern Virginia and is ideal for pleasure boating, fishing, swimming, and exploration. In fall 2020 a visitor survey was administered to capture boater and adjacent landowner perceptions of the recreation experience, identify safety concerns, and help managers to informed decisions. Almost 200 surveys were collected over a month of data collection.

Results indicate that respondents tend to be a dichotomous group of “locals” who live on or within a mile of the lake, and “tourists” who travel greater than 50 miles to visit the lake. Additionally, these …

The COVID-19 pandemic has greatly affected the tourism industry, resulting in consequences such as travel fear and a decline in travel and tourism. Recognizing the needs of study in pandemic travel, this study explored individuals’ pandemic travel decision-making mechanism by investigating relationships between pandemic travel fear, coping strategies, resilience against travel fear, and pandemic travel intentions. Further, this study investigated the moderating role of risk tolerance in those relationships. Columbian sharp‐tailed grouse (Tympanuchus phasianellus columbianus) are endemic to grassland and shrub‐steppe ecosystems of western North America, yet their distribution has contracted to <10% of their historical range. Primary threats to Columbian sharp‐tailed grouse include loss of native habitat and conversion to agriculture, reductions in habitat once provided by the Conservation Reserve Program (CRP), wildfire, and drought conditions, yet population‐level consequences of these threats and their spatio‐temporal scales of effect are poorly understood. We evaluated multi‐scale effects of land cover, weather, and fire histories on patterns of abundance and productivity for Columbian sharp‐tailed grouse populations during 1995–2020 in Idaho, USA, using mixedeffects generalized regression and remotely sensed data. We demonstrated negative effects of fire, tree encroachment, and bare ground, positive effects of spring and summer precipitation and cover of shrubs and perennial forbs and grasses, and positive effects of CRP on grouse abundance that changed in magnitude with cover of perennials and shrubs near leks (i.e., strongest effects when average cover of shrubs and perennial forbs and grasses were less abundant). We also demonstrated per capita recruitment of Columbian sharp‐tailed grouse is positively associated with late‐summer greenness. Our results show that several suspected threats have measurable, population‐level impacts to Columbian sharp‐tailed grouse within Idaho. Moreover, our results suggest ongoing changes occurring within the core range of Columbian sharp‐tailed grouse, including loss of CRP cover to tilled agriculture and changes to wildfire and precipitation dynamics are likely to have negative effects on populations.

Marine heat waves (MHWs) have been associated with extensive harmful algal blooms (HABs) in the northeast Pacifc Ocean, but the degree to which these large-scale oceanographic events are mirrored in nearshore environments has not been well established. We compared phytoplankton assemblages in the Lower Columbia River Estuary (LCRE) during two Pacifc MHWs that took place in 2015 and 2019, with observations from 2017, a year with no MHW. These data were paired with environmental data from the summers of 2015–2019 to characterize differences in estuarine conditions during MHWs that promote phytoplankton assemblage transitions and identify HAB-conducive conditions. The genetic resources of oysters in Louisiana and the Gulf of Mexico are threatened due to high unexplained oyster mortality. Germplasm repositories are collections of cryopreserved genetic material stored alongside associated information that are used to protect genetics resources and facilitate breeding programs in agricultural industries. Therefore, there is great need for oyster repositories. Development of repositories has been slow despite research on high-Throughput cryopreservation protocols because of logistical complexities. Through the efforts of the Cambodian people and historians, the world is familiar with the tragic human story that unfolded over decades of war and the brutal rule of the Khmer Rouge. This thesis is an attempt to tell the equally tragic but untold story of how decades of conflict shaped the Cambodian landscape and the lives and populations of elephants and other animals.

Otolith organs of the inner ear are innervated by two parallel afferent projections to the brainstem and cerebellum. These innervations were proposed to segregate across the line of polarity reversal (LPR) within each otolith organ, which divides the organ into two regions of hair cells (HC) with opposite stereociliary orientation. The relationship and functional significance of these anatomical features are not known.Here, we show regional expression of Emx2 in otolith organs, which establishes LPR, mediates the neuronal segregation across LPR and constitutes the bidirectional sensitivity function. In the overall history of technological innovations, social media has only existed for a brief time, however its influence is undeniable. Researchers have found that it can be used to influence elections, spread health misinformation, and aid with financial pump-and-dump schemes. Keeping all this in mind, it is clear that more research is needed to predict the spread of information on social media in order to combat its malicious use.

Remdesivir received the first emergency use authorization from the FDA for the treatment of COVID-19. Multiple adverse drug reactions (ADR) have been reported since its approval in October 2020. Bradycardia, defined by a decrease in heart rate has been reported as an adverse event for patients receiving remdesivir for COVID-19 treatment. The purpose of the research is to systematically investigate the frequency of occurrence of bradycardia in adults receiving remdesivir using clinical data derived from the FDA Adverse Event Reporting System (FAERS) database.
