While significant efforts have been made in developing pre-clinical treatments for the neuronal ceroid lipofuscinoses (NCLs), many challenges still remain to bring children with NCLs a cure. Devising effective therapeutic strategies for the NCLs will require a better understanding of pathophysiology, but little is known about the mechanisms by which loss of lysosomal proteins causes such devastating neurodegeneration. Research into glial cells including astrocytes, microglia, and oligodendrocytes have revealed many of their critical functions in brain homeostasis and potential contributions to neurodegenerative diseases. BACKGROUND: Improved survival for patients with brain metastases has been accompanied by a rise in tumor recurrence after stereotactic radiotherapy (SRT). Laser interstitial thermal therapy (LITT) has emerged as an effective treatment for SRT failures as an alternative to open resection or repeat SRT. We aimed to evaluate the efficacy of LITT followed by SRT (LITT+SRT) in recurrent brain metastases.
METHODS: A multicenter, retrospective study was performed of patients who underwent treatment for biopsy-proven brain metastasis recurrence after SRT at an academic medical center. BACKGROUND: Glioblastoma (GBM) has a 5-year survival rate of 3%-5%. GBM treatment includes maximal resection followed by radiotherapy with concomitant and adjuvant temozolomide (TMZ). BACKGROUND: Many factors impact survival in patients with glioblastoma, including age, Karnofsky Performance Status, postoperative chemoradiation,
METHODS: We utilized a widely available diagnostic platform (FoundationOne CDx) to perform high-throughput next-generation sequencing on 185 patients with newly diagnosed glioblastoma in our tertiary care center. We performed multivariate analysis to control for clinical parameters with known impact on survival to elucidate the independent prognostic value of prevalent mutant genes and the independent impact of gross total resection.

OBJECTIVE: Whether short-term, low-potency opioid prescriptions for acute pain lead to future at-risk opioid use remains controversial and inadequately characterized. Our objective was to measure the association between emergency department (ED) opioid analgesic exposure after a physical, trauma-related event and subsequent opioid use. We hypothesized ED opioid analgesic exposure is associated with subsequent at-risk opioid use.
METHODS: Participants were enrolled in AURORA, a prospective cohort study of adult patients in 29 U.S., urban EDs receiving care for a traumatic event. Rift Valley fever virus (RVFV) is a veterinary and human pathogen and is an agent of bioterrorism concern. Currently, RVFV treatment is limited to supportive care, so new drugs to control RVFV infection are urgently needed. RVFV is a member of the order Bunyavirales, whose replication depends on the enzymatic activity of the viral L protein. Screening for RVFV inhibitors among compounds with divalent cation-coordinating motifs similar to known viral nuclease inhibitors identified 47 novel RVFV inhibitors with selective indexes from 1.1-103 and 50% effective concentrations of 1.2-56 μM in Vero cells, primarily α-Hydroxytropolones and N-Hydroxypyridinediones. The ENIGMA group on Generalized Anxiety Disorder (ENIGMA-Anxiety/GAD) is part of a broader effort to investigate anxiety disorders using imaging and genetic data across multiple sites worldwide. The group is actively conducting a mega-analysis of a large number of brain structural scans. Workplace exposures to physical, chemical, and psychosocial factors account for a large burden of chronic diseases. Obtaining useful estimates of current and past workplace exposures is challenging, particularly in large general population studies. Job-exposure matrices (JEMs) are a useful tool for exposure assessment, particularly when no individual level exposure data are available. A JEM provides a cross-tabulation of job titles (sometimes combined with industry) and estimated exposures to workers carrying out these jobs during different time periods. Programmed cell death protein-1/ligand-1 (PD-1/PD-L1) blockade, including antibody therapeutics, has transformed cancer treatment. However, a major challenge in the field relates to selecting patients who are likely to respond to immune checkpoint inhibitors. Indeed, biopsy-based diagnostic tests to determine immune checkpoint protein levels do not accurately capture the inherent spatial and temporal heterogeneity of PD-L1 tumor expression. As a result, not all PD-L1-positive tumors respond to immunotherapies, and some patients with PD-L1-negative tumors have shown clinical benefits. This qualitative research study aims to examine theThai-Myanmar maritimedispute,applying the dispute settlement mechanism of the 1982 United Nations Convention on theLaw of the Sea (UNCLOS). The research methodology includes documentary research and in-depth interview. After analyzing data collected,itis found that the dispute settlement mechanismof the UNCLOS comprises voluntary and compulsory procedures. As both Thailand and Myanmarare parties to the UNCLOS and their dispute involves an area in the Andaman Sea, thoseprocedures may be applied. Plastics can reach the coastal environment and could impact the marine environment. Mussels are marine organisms which are prone to be exposed to microplastics pollution. Therefore, in this preliminary study, the commercially important green mussel (Perna viridis) collected at the Koh Phee Canal in Phuket, Phuket Province, southern peninsular Thailand, was investigated. The collected mussels from aquaculture farm fell into three age groups, namely 1 year-1 month, 1 year-7 months, and 1 year-9 months. Pollution of water resources by uncontrolled pesticide use is a serious health and environmental issue. In this study, concentrations of three organophosphorus pesticides (diazinon, malathion, and azinphos-methyl) in water and sediment samples from five estuaries (Sefidrud, Chalus, Babolrud, Tajan, and Gorganrud) along the Caspian Sea were investigated. Samples were collected from surface water and sediment during summer to autumn, and pesticides were analysed by gas chromatography-mass spectrometry (GC-MS). Results indicated that salinity and turbidity in Gorganrud were higher (salinity: range 2–8%; turbidity: range 1–9%) compared to other stations. Physicochemical characteristics of porous substrates (longan biochar, corn biochar, and pumice) including specific surface area (SSA), pore volume (Vp), and cation exchange capacity (CEC) were determined. Longan biochar had the highest SSA, Vp, and CEC, followed by corn biochar and pumice. Then, columns filled with each of these 3 substrates together with gravel as a control treatment, were evaluated to compare wastewater treatment efficiency under different hydraulic retention times (HRTs). Heavy metals in coastal sediments can adversely affect human health and the environment. The distribution and metal bioavailability of Pb and Zn in 21 sediment samples collected from Phuket Bay, Phuket, Thailand using the first-two steps of sequential extraction proposed by the Standards, Measurements and Testing programme (known as BCR) was determined. The results showed that Pb formed weak complexes contributing up to 11.2% to 33% of its total concentration (1.7 to 7.5 mg kg-1) in the first fraction (BCR1), while Zn in the BCR1 fraction ranged from 4.9% to 9.9%. Identifying genetic variation underlying human diseases establishes targets for therapeutic development and helps tailor treatments to individual patients. Large-scale transcriptomic profiling has extended the study of such molecular heterogeneity between patients to somatic tissues. However, the lower resolution of bulk RNA profiling, especially in a complex, composite tissue such as the skin, has limited its success. Here we demonstrate approaches to interrogate patient-level molecular variance in a chronic skin inflammatory disease, psoriasis vulgaris, leveraging single-cell RNA-sequencing of CD45+ cells isolated from active lesions. Background: Brain imaging and genetics are fields acquiring data at increasing speed, but more information does not always result in a better understanding of the underlying biology. We developed the ProcessGeneLists (PGL) approach to use genetics and mRNA gene expression data to generate regions of interest for imaging studies.

Background: Brain ventricles have been reported to be enlarged in several neuropsychiatric disorders and in aging. Whether human cerebral ventricular volume can decrease over time with psychiatric treatment is not well-studied. The aim of this study was to examine whether inpatients taking serotonin reuptake inhibitors (SRI) exhibited reductions in cerebral ventricular volume.
Methods: Psychiatric inpatients, diagnosed mainly with depression, substance use, anxiety, and personality disorders, underwent two imaging sessions (Time 1 and Time 2, approximately 4 weeks apart). With the prospect of semi-permanent and permanent habitable fixtures on the moon and Mars, the complications associated with long-term exposure to microgravity should be investigated exhaustively. Spaceflight Associated Neuro-Ocular Syndrome (SANS), a group of neurological and ocular effects resulting from prolonged exposure to microgravity, is characterized by significant fluid shifts into the cranium, namely cerebrospinal fluid and blood, and an upward shift of the brain relative to the skull. Ottawa 50/70 sand specimens and natural beach sand samples were treated using bio-augmented geomicrobies via a surface percolation technique. Testing was conducted on these specimens to determine how resultant calcium carbonate precipitation changed as a function of temperature, depth from the surface, and in the presence of magnesium. Specifically, x-ray Diffraction (XRD), a Scanning Electron Microscope (SEM), and Energy Dispersive X-ray Spectroscopy (EDS) were used to determine and quantify the presence of calcium carbonate and its associated phase. Acrylonitrile, an industrial chemical, is a multisite carcinogen in rats and mice, producing tumors in four tissues with barrier function, that is, brain, forestomach, Zymbal’s gland, and Harderian gland. To assess mechanism(s) of action (MoA) for induction of neoplasia and to evaluate whether the findings in rodents are indicative of human hazard, data on the potential key effects produced by acrylonitrile in the four rodent target tissues of carcinogenicity were evaluated. Attention is a safety-critical operator ability that needs to be sustained over the course of specific tasks. However, many internal factors (e.g.: cognitive underload or overload, fatigue, etc.) and external factors (e.g.: HMI quality, environmental stressors, noise, etc.), can cause attention to drift away from the task. The development of drug resistance in lung cancer is a major clinical challenge, leading to a 5-year survival rate of only 18%. Therefore, unravelling the mechanisms of drug resistance and developing novel therapeutic strategies is of crucial importance. This study systematically explores the novel biomarkers of drug resistance using a lung cancer model (DLKP) with a series of drug-resistant variants. In-depth label-free quantitative mass spectrometry-based proteomics and gene ontology analysis shows that parental DLKP cells significantly differ from drug-resistant variants, and the cellular proteome changes even among the drug-resistant subpopulations. A group of clearwing moths Sesiidae (Syn.: Aegeriidae) is analyzed here with special emphasis on genetics of currant clearwing moth (Synanthedon tipuliformis, CLERCK 1759). DNA sequences of the longest (1544 nt) available sequence of Sesiidae genes Co1 (cytochrome oxidase subunit-1 of mitochondrial DNA, mtDNA) (mtDNA; NCBI# AJ864359.1) were analyzed after In silico data mining, and dendrogram was edited to reveal molecular diversity and genetic distances among Sesiidae species. Batesian mimicry, sexual dimorphism, and genetics and genomics of Sesiidae are indicated.

Breeding and domestication have generated widely exploited crops, animals and microbes. However, many Saccharomyces cerevisiae industrial strains have complex polyploid genomes and are sterile, preventing genetic improvement strategies based on breeding. Here, we present a strain improvement approach based on the budding yeasts’ property to promote genetic recombination when meiosis is interrupted and cells return-to-mitotic-growth (RTG). We demonstrate that two unrelated sterile industrial strains with complex triploid and tetraploid genomes are RTG-competent and develop a visual screening for easy and high-throughput identification of recombined RTG clones based on colony phenotypes. Background: Trauma and chronic stress are believed to induce and exacerbate psychopathology by disrupting glutamate synaptic strength. However, in vivo in human methods to estimate synaptic strength are limited. In this study, we established a novel putative biomarker of glutamatergic synaptic strength, termed energy-per-cycle (EPC). Then, we used EPC to investigate the role of prefrontal neurotransmission in trauma-related psychopathology.
Methods: Healthy controls (n = 18) and patients with posttraumatic stress (PTSD; n = 16) completed 13C-acetate magnetic resonance spectroscopy (MRS) scans to estimate prefrontal EPC, which is the ratio of neuronal energetic needs per glutamate neurotransmission cycle (VTCA/VCycle).
