Blood biomarkers indicative of Alzheimer's disease (AD) pathology are altered in both preclinical and symptomatic stages of the disease. Distinctive biomarkers may be optimal for the identification of AD pathology or monitoring of disease progression. Blood biomarkers that correlate with changes in cognition and atrophy during the course of the disease could be used in clinical trials to identify successful interventions and thereby accelerate the development of efficient therapies. When disease-modifying treatments become approved for use, efficient blood-based biomarkers might also inform on treatment implementation and management in clinical practice. In the BioFINDER-1 cohort, plasma phosphorylated (p)-tau231 and amyloid-β42/40 ratio …

IMPORTANCE: Polygenic risk scores (PRSs), which aggregate the genetic effects of single-nucleotide variants identified in genome-wide association studies (GWASs), can help distinguish individuals at a high genetic risk for Alzheimer disease (AD). However, genetic studies have predominantly focused on populations of European ancestry.
OBJECTIVE: To evaluate the transferability of a PRS for AD in the Korean population using summary statistics from a prior GWAS of European populations.
DESIGN, SETTING, AND PARTICIPANTS: This cohort study developed a PRS based on the summary statistics of a large-scale GWAS of a European population (the International Genomics of Alzheimer Project; 21 982 AD cases …
Segment-Wise Time-Varying Dynamic Bayesian Network With Graph Regularization, Xing Yang, Chen Zhang, Baihua Zheng
Segment-Wise Time-Varying Dynamic Bayesian Network With Graph Regularization, Xing Yang, Chen Zhang, Baihua Zheng
Research Collection School Of Computing and Information Systems
Time-varying dynamic Bayesian network (TVDBN) is essential for describing time-evolving directed conditional dependence structures in complex multivariate systems. In this article, we construct a TVDBN model, together with a score-based method for its structure learning. The model adopts a vector autoregressive (VAR) model to describe inter-slice and intra-slice relations between variables. By allowing VAR parameters to change segment-wisely over time, the time-varying dynamics of the network structure can be described. Furthermore, considering some external information can provide additional similarity information of variables. Graph Laplacian is further imposed to regularize similar nodes to have similar network structures. The regularized maximum a …

Listeria monocytogenes (Lm) is a food-borne pathogen that causes severe bacterial gastroenteritis, with high rates of hospitalization and mortality. Lm is ubiquitous in soil, water and livestock, and can survive and proliferate at low temperatures. Following oral ingestion of contaminated food, Lm crosses the epithelium through intestinal goblet cells in a mechanism mediated by Lm InlA binding host E-cadherin. Importantly, human infections typically occur with Lm growing at or below room temperature, which is flagellated and motile. Even though many important human bacterial pathogens are flagellated, little is known regarding the effect of Lm motility on invasion and immune evasion. …

OBJECTIVE: Metabolomics as an approach to solve biological problems is exponentially increasing in use. Thus, this a pivotal time for the adoption of best practices. It is well known that disrupted tissue oxygen supply rapidly alters cellular energy charge. However, the speed and extent to which delayed mouse tissue freezing after dissection alters the broad metabolome is not well described. Furthermore, how tissue genotype may modulate such metabolomic drift and the degree to which traced
METHODS: By combined liquid chromatography (LC)- and gas chromatography (GC)-mass spectrometry (MS), we measured how levels of 255 mouse liver metabolites changed following 30-second, 1-minute, …

Heterotrimeric G proteins (αβγ subunits) that are activated by G protein-coupled receptors (GPCRs) mediate the biological responses of eukaryotic cells to extracellular signals. The α subunits and the tightly bound βγ subunit complex of G proteins have been extensively studied and shown to control the activity of effector molecules. In contrast, the potential roles of the large family of γ subunits have been less studied. In this review, we focus on present knowledge about these proteins. Induced loss of individual γ subunit types in animal and plant models result in strikingly distinct phenotypes indicating that γ subtypes play important and …

The Role Of Akirin/Nurd Interactions During Heart Development, Mia Jones
Symposium of Student Scholars
Congenital heart defects are often identified during pregnancy or infancy and are the most common birth defect presented in humans, affecting blood circulation resultant from small holes in a blood chamber to deformity-inducing missing structures. Recent advancements in medicine have allowed those affected to live healthier, longer lives. Gene combinations and chromosomal changes play pivotal roles in congenital heart defects, but the specific genes involved remain unknown. The Nowak Lab has discovered several novel regulatory proteins that are critical for embryonic heart development and is studying them for a potential link to congenital heart defects.
During embryogenesis, the heart of …

International Journal of Exercise Science
International Journal of Exercise Science 15(5): 1554-1562, 2022. One’s beliefs about the nature of stress (e.g., stress mindset) play a large role in the extent to which one experiences the detrimental or beneficial outcomes of stress. Stress mindset has been explored in college students, but there is limited research on stress mindsets in student-athletes. Sport can serve as a buffer to the negative impacts of stress for some student-athletes; however, pressures associated with sport participation increase stress in other student-athletes. Therefore, the purpose was to examine potential differences in stress mindset and perceived stress between non-athletes and college student-athletes. We …

Panarchy Suggests Why Management Mitigates Rather Than Restores Ecosystems From Anthropogenic Impact, David G. Angeler, Ran Hur
Panarchy Suggests Why Management Mitigates Rather Than Restores Ecosystems From Anthropogenic Impact, David G. Angeler, Ran Hur
School of Natural Resources: Faculty Publications
Panarchy, a model of dynamic systems change at multiple, interconnected spatiotemporal scales, allows assessing whether management influences ecological processes and resilience. We assessed whether liming, a management action to counteract anthropogenic acidification, influenced scale-specific temporal fluctuation frequencies of benthic invertebrates and phytoplankton assemblages in lakes. We also tested whether these fluctuations correlated with proxies of liming (Ca:Mg ratios) to quantify scale-specific management effects. Using an ecosystem experiment and monitoring data, time series analyses (1998–2019) revealed significant multiscale temporal (and thus panarchy) structure for littoral invertebrates across limed and reference lakes. Such patterns were inconsistent for sublittoral invertebrates and phytoplankton. When …

Elements of perceived control are associated with recidivism in offender populations. We investigated the application of locus of control to the frequency of personal involvement with the law and to beliefs surrounding the likelihood of future contact with the legal system. We hypothesized that, as the number of sentencings or legal experiences increased, locus of control would externalize. We also predicted that increased legal involvement would lead to greater belief in the likelihood of future involvement. A statistically significant path model suggests that locus of control appears to be a predictor of increased criminality, as opposed to the other way …

A Common Mechanism Links Epstein‐Barr Virus Infections And Autoimmune Diseases, Luwen Zhang
Nebraska Center for Virology: Faculty Publications
Epstein‐Barr virus (EBV) infection is associated with a variety of the autoimmune diseases. There is apparently no unified model for the role of EBV in autoimmune diseases. In this article, the development of autoimmune diseases is proposed as a simple two‐step process: specific autoimmune initiators may cause irreversible changes to genetic materials that increase autoimmune risks, and autoimmune promoters promote autoimmune disease formation once cells are susceptible to autoimmunity. EBV has several types of latencies including type III latency with higher proliferation potential. EBV could serve as autoimmune initiators for some autoimmune diseases. At the same time, EBV may play …

Background: Brain age is an MRI-derived estimate of brain tissue loss that has a similar pattern to aging-related atrophy. White matter hyperintensities (WMHs) are neuroimaging markers of small vessel disease and may represent subtle signs of brain compromise. We tested the hypothesis that WMHs are independently associated with premature brain age in an original aging cohort.
Methods: Brain age was calculated using machine-learning on whole-brain tissue estimates from T1-weighted images using the BrainAgeR analysis pipeline in 166 healthy adult participants. WMHs were manually delineated on FLAIR images. WMH load was defined as the cumulative volume of WMHs. A positive difference …

Interconnected Precarity: A Contemporary Reframing Of Bodily And Earthly Health In Wendell Berry's The Unsettling Of America: Culture And Agriculture, Abigail Pinegar
Theses and Dissertations
Published in 1977, Wendell Berry's book The Unsettling of America: Culture and Agriculture gained widespread popularity. More than half a century later, many of the notions of the body and the earth presented in its seventh chapter, "The Body and the Earth," remain relevant and important for environmental discourse today. Berry's discussion of the body and the earth examines their mutuality and codependence from an ontological, theological, agricultural, and even biological perspective. The coupling of this text with Judith Butler's, Frames of War: When is Life Grievable? contemporizes his argument through its more socio-political and philosophical claims regarding life and …

Student Scholar Symposium Abstracts and Posters
Human immunodeficiency virus (HIV) attacks the immune system and if left untreated, could cause acquired immunodeficiency syndrome (AIDS). The HIV matrix protein (HIV-MA) is involved in replication and regulation of the HIV virus. Calmodulin (CaM), a calcium-binding protein found in all eukaryotes, has a potential role in the viral replication of HIV-MA which plays a key role in the replication of HIV. In order to investigate the interactions between calmodulin and the HIV-MA, a series of titrations with CaM are performed using circular dichroism. Circular dichroism (CD) uses circularly polarized light to observe the secondary structure of a molecule. The …

On June 24, 2022, the U.S. Supreme Court ruled to overturn Roe v. Wade (1973)—a groundbreaking case that legalized the right to have an abortion—which signified a deep rift in the nation between the opinions of its lawmakers and citizens in the wake of a widening partisan gap. Biopower, according to Foucault, can be defined as the governing of bodies wherein citizens are stripped of bodily autonomy and are closely regulated by the nation-state. Manifested in political consequences, this can be defined as biopolitics, or when the nation-state’s ideas are made into a reality in the political realm. …
