According to American Psychological Association Work and Well-being Survey of 1,501 U.S. adult workers, 79% of employees reported negative impacts of work-related stress, including lack of interest (26%) and effort (19%) at work. Workplace wellness programs offer a means to alleviate stress among employees. The research explored the perceived acceptability and efficacy of a virtual versus hybrid wellness program and some specific health outcomes, total cholesterol, BP, and BMI, perceived well-being and stress among faculty and staff at a northeastern university. Laboratory safety in academic settings has long been a concern for colleges and universities worldwide. Education and training for lab safety is often a one size fits all process. That is, the same training given to faculty and staff is also given to students. However, students are generally younger, less educated and less experienced than faculty or staff. Clostridioides difficile infection (CDI) is classified as an urgent health threat by the Centers for Disease Control and Prevention (CDC), and affects nearly 500,000 Americans annually. Approximately 20–25% of patients with a primary infection experience a recurrence, and the risk of recurrence increases with subsequent episodes to greater than 40%. The leading risk factor for CDI is broad-spectrum antibiotics, which leads to a loss of microbial diversity and impaired colonization resistance. Current FDA-approved CDI treatment strategies target toxin or toxin-producing bacteria, but do not address microbiome disruption, which is key to the pathogenesis of recurrent CDI. Comparative effectiveness evidence from randomized trials may not be directly generalizable to a target population of substantive interest when, as in most cases, trial participants are not randomly sampled from the target population. Motivated by the need to generalize evidence from two trials conducted in the AIDS Clinical Trials Group (ACTG), we consider weighting, regression and doubly robust estimators to estimate the causal effects of HIV interventions in a specified population of people living with HIV in the USA. Presented in this work is an investigation of the application of artificially intelligent algorithms, namely deep learning, to generate segmentations for the application in functional avoidance radiotherapy treatment planning. Specific applications of deep learning for functional avoidance include generating hippocampus segmentations from computed tomography (CT) images and generating synthetic pulmonary perfusion images from four-dimensional CT (4DCT).A single institution dataset of 390 patients treated with Gamma Knife stereotactic radiosurgery was created. From these patients, the hippocampus was manually segmented on the high-resolution MR image and used for the development of the data processing methodology and model testing. Coronary endothelial cell (CECs) damage and rarefaction of coronary microvasculature are implicated in diabetes-induced heart failure with preserved ejection fraction (HFpEF). 4-hydroxy-2-nonenal (4-HNE), a reactive aldehyde that is increased in diabetic hearts, contributes to CEC dysfunction and cardiotoxicity. Aldehyde dehydrogenase (ALDH) 2, a mitochondrial enzyme that detoxifies 4-HNE and confers cardioprotection. However, ALDH2 activity is decreased in diabetic hearts, which results in the augmentation of 4HNE-induced cardiotoxicity. Thus, we hypothesize that activation of ALDH2 in CECs reduces 4-HNE-mediated cell signaling aberrations and preserves coronary angiogenesis. Post-translational modifications (PTMs) are responsible for a variety of cellular processes. One such PTM is protein phosphorylation, which is catalyzed by kinases. Kinase enzymes play important roles in cellular signaling pathways, but dysregulation of kinase-mediated events results in the formation of diseases, which make kinases favorable drug targets. The CXCR4-CXCL12 chemokine signaling axis plays a key role in migration and bone metastasis in prostate cancer (PC). Androgens regulate CXCR4 expression and its receptor activation in lipid-raft micro domains of PC cells, resulting in higher protease expression and invasion. In order to identify some novel CXCR4 co-regulators associated within the lipid-raft, a SILAC (Stable isotope labeling using amino acid in cell culture)-based proteomic analysis was performed with PC3 stable cell-lines over-expressing or knocking-down CXCR4. In this dissertation, we consider a number of matrix-valued random sequences that are modulated by a discrete-time Markov chain having a finite space.Assuming that the state space of the Markov chain is large, our main effort in this work is devoted to reducing the complexity. To achieve this goal, our formulation uses time-scale separation of the Markov chain. The state-space of the Markov chain is split into subspaces. Defects in insulin secretion from pancreatic islet beta- cells leads to long term complications such as type 2 diabetes mellitus. The mechanism and regulation of glucose stimulated insulin secretion (GSIS) as well as factors contributing to pancreatic islet beta-cell demise are yet poorly understood. Previous work from our laboratory demonstrated that Ras-related C3 botulinum toxin substrate 1 (Rac1), a small G- protein is involved in GSIS as well as in culminating dysfunction under metabolic stress. Familial hypercholesterolemia (FH) is an autosomal genetic disease that causes elevated blood levels of low-density lipoprotein (LDL). One of the leading causes of FH is gain-of-function mutations in the gene coding for proprotein convertase subtilisin/kexin type 9 (PCSK9). The PCSK9 protein binds to LDL receptors (LDLR) on the surface of hepatocytes and promotes their degradation, preventing the recycling of LDLRs and thus increasing LDL blood levels. Monoclonal antibody therapies that bind to PCSK9 inhibiting LDLR binding are currently only available as an injection. We piloted a community-based intervention to improve outcomes among adolescents living with HIV who were transitioning to adult-oriented care in Lima, Peru. We assessed feasibility and potential effectiveness, including within-person changes in self-reported adherence, psychosocial metrics (NIH Toolbox), and transition readiness (“Am I on TRAC” questionnaire, “Got Transition” checklist). From October 2019 to January 2020, we enrolled 30 adolescents (15–21 years). The nine-month intervention consisted of logistical, adherence and social support delivered by entry-level health workers and group sessions to improve health-related knowledge and skills and social support. Antimicrobial resistance (AMR) is one of the serious global health challenges of our time. There is an urgent need to develop novel therapeutic agents to overcome AMR, preferably through alternative mechanistic pathways from conventional treatments. Interdisciplinary research in inorganic medicinal chemistry with biology is advancing the knowledge and implementation of transition metal complexes for therapy and is offering a realistic alternative to traditional antibiotics. Metal complexes with 1,10-phenanthroline (phen) ligands have demonstrated promising therapeutic capabilities with diverse biological activity. Due to the widespread effects of COVID-19, mental health professionals need to understand the lived experiences of survivors. Data were collected from YouTube videos (N = 52), which contained firsthand perspectives from COVID-19 survivors. Survivors understood the high stakes of COVID-19, reported the dual threat to physical and mental health, entered “survival mode,” and had both positive and negative reflections on the experience. Implications for mental health professionals and future research directions are included.

Soil compaction, a result of urbanization, may be detrimental to the soil moisture dynamics and health of urban trees which subsequently limits their ecosystem services. This study considers two hypotheses: higher compacted soils will negatively impact soil moisture dynamics of red maple, Norway maple, and honey locust tree species on campus; and there will be a difference in the soil moisture dynamics between tree species due to varying tree traits. Post-traumatic stress disorder (PTSD), a common condition with potentially devastating individual, family, and societal consequences, is highly associated with substance use disorders (SUDs). The association between PTSD and SUD is complex and may involve adverse childhood experiences (ACEs), historical and multi-generational traumas, and social determinants of health as well as cultural and spiritual contexts. Current psychosocial and pharmacological treatments for PTSD are only modestly effective, and there is a need for more research on therapeutic interventions for co-occurring PTSD and SUD, including whether to provide integrated or sequential treatments. Neuropathic pain is a chronic pain condition resulting from damaged peripheral nerves that induces allodynia (pathological touch sensitivity). Our previous studies suggest that prenatal alcohol exposure (PAE) is a risk factor for allodynia. In contrast to control rats, adult PAE rats with minimal nerve injury develop allodynia. Nerve-injured PAE rats display heightened peripheral and central nervous system (CNS) immune cell-derived proinflammatory cytokine production that consequently results in allodynia. This suggests that PAE dysregulates peripheral immune and CNS glial cell function to over-respond to subtle immune challenges leading to allodynia.
