We describe a pipeline for optimized and streamlined multiplexed immunofluorescence-guided laser capture microdissection allowing the harvest of individual cells based on their phenotype and tissue localization for transcriptomic analysis with next-generation RNA sequencing. Here, we analyze transcriptomes of CD3+ T cells, CD14+ monocytes/macrophages, and melanoma cells in non-dissociated metastatic melanoma tissue. While this protocol is described for melanoma tissues, we successfully applied it to human tonsil, skin, and breast cancer tissues as well as mouse lung tissues. Extrachromosomal Dna (Ecdna): An Origin Of Tumor Heterogeneity, Genomic Remodeling, And Drug Resistance., Lauren T Pecorino, Roel G W Verhaak, Anton Henssen, Paul S Mischel

The genome of cancer cells contains circular extrachromosomal DNA (ecDNA) elements not found in normal cells. Analysis of clinical samples reveal they are common in most cancers and their presence indicates poor prognosis. They often contain enhancers and driver oncogenes that are highly expressed. The circular ecDNA topology leads to an open chromatin conformation and generates new gene regulatory interactions, including with distal enhancers. The absence of centromeres leads to random distribution of ecDNAs during cell division and genes encoded on them are transmitted in a non-mendelian manner. ecDNA can integrate into and exit from chromosomal DNA. An exponential rise in the use of transcranial direct current stimulation (tDCS) to modify the human brain and behavior has been observed over the last 20 years, with studies often yielding mixed outcomes. Such heterogeneity is partially attributed to the lack of a concise understanding of the underlying mechanisms. The purpose of this study was to examine the work of school food service professionals as this has implications for all school food service staff. The imbalance of learning resources between urban and rural China, however, has caused disadvantages for learners in rural China to gain access to quality educational opportunities necessary for becoming well-rounded learners. The Buffalo Creek watershed has experienced water quality impairments for various reasons, including but not limited to, agricultural influence, abandoned mine drainage (AMD), on-site wastewater malfunction, and stormwater. The source and cause of the most prevalent impaired areas remains unknown. The goal of this study was to analyze water chemistry using state water quality standards and mass ratio analysis, then to conduct fish community analysis. We found toxic metal pollution across the watershed, with iron, cadmium, copper, and selenium frequently exceeding criteria. Objective: Serious mental illness (SMI) causes an increased risk for comorbidities like heart disease, obesity, metabolic syndrome, and diabetes. Many times, these comorbidities are due to side effects of antipsychotics and poor self-management from low insight and other symptoms of mental illness. This project aims to enhance client care enrolled in an Assertive Community Treatment (ACT) program by providing evidence-based resources tailored to clients with SMI, providing ACT team members with support to promote healthy behaviors in clients.

This study analyzed the possibility of saving on textbook costs without sacrificing student performance by using Open Educational Resources (OER) at no cost to students as a replacement for high-priced commercially printed books. Approximately two hundred students enrolled in business and economics courses at Kingsborough Community College of the City University of New York from Fall 2016 to Fall 2018 were assessed in this study. On 4 August 2020, a strong explosion occurred near the Beirut seaport, Lebanon and killed more than 200 people and damaged numerous buildings in the vicinity. The interactions between migrants and Mexican local communities have positive and negative outcomes. A report by Human Rights First found that more than 630 violent crimes against asylum seekers were reported in the first few months of the “Remain in Mexico” policy. Still, some migrants have been able to assimilate and stay in Mexico, particularly in large cities such as Tijuana, Baja California and Ciudad Juarez, Chihuahua. This research project combines qualitative data collected through interviews with local NGOs between September 2020 to February 2021 and secondary research data. Mitigation Impact Of Statewide Non-Pharmaceutical Policies On Covid-19: An Application Of Infectious Disease Transmission Model And Partially Observed Markov Process To New Mexico, Xingya Ma

This thesis is an application of epidemiological models for infectious disease transmission and the use of partially observed Markov process (POMP) for model fitting. It focuses on COVID-19 pandemic in the state of New Mexico. The analysis covered March 2020 to June 2021. Daily data of COVID19 cases and deaths and a daily index of eleven statewide government non-pharmaceutical intervention (NPI) policies were collected from six public sources and were validated. These data were integrated through the Susceptible-Exposed-Infected-Removed (SEIR) model. Estimated daily transmission rates between the model compartments quantify the impact of the mitigation policies, and show that transmission rates …

Delineating Differences In How Us High Schools Are Racialized, Dara Shifrer, C. J. Appleton

Schools’ overt or explicit practices are a dominant lens through which education researchers and policymakers attempt to understand how schools are racially inequitable. Yet, Lewis and Diamond argue that contemporary racial inequalities are largely sustained through implicit factors, like institutional practices and structural inequalities. Ray’s framework on racialized organizations similarly outlines how our racialized sociopolitical structure becomes embedded in organizations, legitimating and perpetuating the racialized hierarchy. We apply illustrative cluster analysis techniques to rich data on schools, teachers, and students from the nationally representative High School Longitudinal Study of 2009 to find that structural inequities (e.g., student body, sector, average …

Background: South Asians from Bangladesh, Pakistan, and India have a higher prevalence of diabetes than the general global population and are also more susceptible to developing diabetes due to biological and lifestyle factors. The South Asian diet is high in refined grains and monounsaturated fatty acids, increasing the risk of diabetes.

Local Problem: Congregants of a Sikh gurdwara (place of worship) lacked knowledge of diet and lifestyle in relation to preventing and managing diabetes. Many senior congregants have been diagnosed with diabetes or are at high risk of developing the disease.

Providing necessary health care alone in unpredictable and remote environments are associated with poor outcomes from physical violence. Background: Pepper spray is known for its active ingredient, oleoresin capsicum (OC), as a self-defense tool. OC gel is used worldwide as a non-lethal incapacitating agent. In 2019, the Philadelphia Energy Solutions oil refinery closed after a dangerous explosion. But for years before the accident, members of the local environmental justice organization Philly Thrive had been advocating for its closure. Neighborhood residents who live near the refinery site had been complaining for years of health problems, ranging from asthma to cancer. “Enough is enough,” says Sylvia Bennett, 79, who lives in the Grays Ferry neighborhood near where the refinery is located. Her two daughters have both been diagnosed with cancer.
