The Structural Basis Of Long-Term Potentiation In Hippocampal Synapses, Revealed By Electron Microscopy Imaging Of Lanthanum-Induced Synaptic Vesicle Recycling, John E Heuser Jan 2022

The Structural Basis Of Long-Term Potentiation In Hippocampal Synapses, Revealed By Electron Microscopy Imaging Of Lanthanum-Induced Synaptic Vesicle Recycling, John E Heuser

2020-Current year OA Pubs

Hippocampal neurons in dissociated cell cultures were exposed to the trivalent cation lanthanum for short periods (15-30 min) and prepared for electron microscopy (EM), to evaluate the stimulatory effects of this cation on synaptic ultrastructure. Not only were characteristic ultrastructural changes of exaggerated synaptic vesicle turnover seen within the presynapses of these cultures-including synaptic vesicle depletion and proliferation of vesicle-recycling structures-but the overall architecture of a large proportion of the synapses in the cultures was dramatically altered, due to large postsynaptic "bulges" or herniations into the presynapses. Moreover, in most cases, these postsynaptic herniations or protrusions produced by lanthanum were …

Since first reported in 2019, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is rapidly acquiring mutations, particularly in the spike protein, that can modulate pathogenicity, transmission and antibody evasion leading to successive waves of COVID19 infections despite an unprecedented mass vaccination necessitating continuous adaptation of therapeutics. Small animal models can facilitate understanding host-pathogen interactions, target selection for therapeutic drugs, and vaccine development, but availability and cost of studies in BSL3 facilities hinder progress. To generate a BSL2-compatible

Regulation Of Rod Photoreceptor Function By Farnesylated G-Protein Γ-Subunits, Alexander V. Kolesnikov, Elena Lobysheva, Jaya P. Gnana-Prakasam, Vladimir J. Kefalov, Oleg G. Kisselev Jan 2022

Regulation Of Rod Photoreceptor Function By Farnesylated G-Protein Γ-Subunits, Alexander V. Kolesnikov, Elena Lobysheva, Jaya P. Gnana-Prakasam, Vladimir J. Kefalov, Oleg G. Kisselev

2020-Current year OA Pubs

Heterotrimeric G-protein transducin, Gt, is a key signal transducer and amplifier in retinal rod and cone photoreceptor cells. Despite similar subunit composition, close amino acid identity, and identical posttranslational farnesylation of their Gγ subunits, rods and cones rely on unique Gγ1 (Gngt1) and Gγc (Gngt2) isoforms, respectively. The only other farnesylated G-protein γ-subunit, Gγ11 (Gng11), is expressed in multiple tissues but not retina. To determine whether Gγ1 regulates uniquely rod phototransduction, we generated transgenic rods expressing Gγ1, Gγc, or Gγ11 in Gγ1-deficient mice and analyzed their properties. Immunohistochemistry and Western blotting demonstrated the robust expression of each transgenic Gγ in …

Influences Of Seawater Flows On The Degradation Of Organic Coatings Applied On Offshore Wind Turbines, Amin Vedadi Jan 2022

Influences Of Seawater Flows On The Degradation Of Organic Coatings Applied On Offshore Wind Turbines, Amin Vedadi

Mechanical Engineering and Applied Mechanics

The regular protection methods of offshore wind structures consist of the application of two or three layers of epoxy-based coatings with a polyurethane topcoat. The coating systems of offshore wind turbines are mostly tested on-site, where different coated samples are exposed to the sea water at the specific locations planned for the installations of the turbines. Despite several advantages of laboratory testing, the majority of laboratory-based tests have been limited to the exposure of coated or unprotected metals to stationary electrolytic solutions, while the flow-induced corrosion measurements have not received enough attention until recently. The focus of our work is …

Sexual Dimorphisms In The Adipose Biology Of Vasoactive Intestinal Peptide Deficient Mice, Emma Hawley Jan 2022

Sexual Dimorphisms In The Adipose Biology Of Vasoactive Intestinal Peptide Deficient Mice, Emma Hawley

Microbiological Sciences

Both VIP and PACAP deficient mice display impaired fat storage, but only the molecular adipose characteristics of PACAP KO mice are known. While mice deficient in these peptides are lean, supplementation could promote weight gain in livestock after establishing tissue distribution. We investigated both sides of this 'metabolic coin' by measuring serum metabolites, adipocyte area, and pscWAT gene expression in VIP deficient mice and screening 15 ruminant tissues for VIP, PACAP, and their receptors using RT-qPCR. We found male VIP deficient mice displayed hypertrophic adipocytes, low serum FFAs, and suppressed relative mRNA expression of TMEM26. In ruminants, VIP and PACAP …

In The Pursuit Of Poultry: Β-Phenylethylamine And Ethyl Acetoacetate As Antimicrobials On Chicken, Erika Shay Hedrick Bauer

Microbiological Sciences

This research examines the effect of β-phenylethylamine (PEA), a natural trace amine commonly found in food, and ethyl acetoacetate (EAA), an FDA approved flavoring agent and food additive, as novel antimicrobials on store-bought chicken thighs in a 5-minute immersion. In the first part of this experiment, 5%, 7.5%, and 10% treatments of β-phenylethylamine and ethyl acetoacetate were compared to control H2O treatments utilized on chicken thighs. 10% treatments of PEA and EAA had significant reductions in counts of total aerobic bacteria and Pseudomonas spp. grown at 20°C by >1 log10 CFU/g of chicken meat. In the next experiments regarded 10% …

The Human Myocardium Harbors A Population Of Naive B-Cells With A Distinctive Gene Expression Signature Conserved Across Species, Kevin C Bermea, Nicolas Kostelecky, Sylvie T Rousseau, Chieh-Yu Lin, Luigi Adamo

2020-Current year OA Pubs

INTRODUCTION: Cardiac immunology studies in murine models have identified a sizeable population of myocardial B-cells and have shown that its modulation represents a promising strategy to develop novel therapies for heart failure. However, scarce data on B-cells in the human heart leaves unclear whether findings in rodents are relevant to human biology.

METHODS: We performed immunohistochemical stains to characterize the amount and distribution of B-cells in human hearts, analyzing both fresh and post-mortem tissue. To gain insight into the biology of human myocardial B-cells we analyzed publicly-available spatial transcriptomics and single-cell sequencing datasets of myocardial and peripheral blood mononuclear cells …

Russian Logics And The Culture Of Impossible: Part Ii: Reinterpreting Algorithmic Rationality, Ksenia Tatarchenko, Anya Yermakova, Liesbeth De Mol Jan 2022

Russian Logics And The Culture Of Impossible: Part Ii: Reinterpreting Algorithmic Rationality, Ksenia Tatarchenko, Anya Yermakova, Liesbeth De Mol

Research Collection School of Social Sciences

This article reinterprets algorithmic rationality by looking at the interaction between mathematical logic, mechanized reasoning, and, later, computing in the Russian Imperial and Soviet contexts to offer a history of the algorithm as a mathematical object bridging the inner and outer worlds, a humanistic vision that we, following logician Vladimir Uspensky, call the “culture of the impossible.” We unfold the deep roots of this vision as embodied in scientific intelligentsia. In Part I, we examine continuities between the turn-of-the-twentieth-century discussions of poznaniye—an epistemic orientation towards the process of knowledge acquisition—and the postwar rise of the Soviet school of mathematical logic. …

Research Collection School of Social Sciences

Political polarization impeded public support for policies to reduce the spread of COVID-19, much as polarization hinders responses to other contemporary challenges. Unlike previous theory and research that focused on the United States, the present research examined the effects of political elite cues and affective polarization on support for policies to manage the COVID-19 pandemic in seven countries (n = 12,955): Brazil, Israel, Italy, South Korea, Sweden, the United Kingdom, and the United States. Across countries, cues from political elites polarized public attitudes toward COVID-19 policies. Liberal and conservative respondents supported policies proposed by ingroup politicians and parties more than …

Old Minds, New Marketplaces: How Evolved Psychological Mechanisms Trigger Mismatched Food Preferences, Michal Folwarczny, Tobias Otterbring, Valdimar Sigurdsson, Lynn K. L. Tan, Norman P. Li Jan 2022

Old Minds, New Marketplaces: How Evolved Psychological Mechanisms Trigger Mismatched Food Preferences, Michal Folwarczny, Tobias Otterbring, Valdimar Sigurdsson, Lynn K. L. Tan, Norman P. Li

Research Collection School of Social Sciences

Principally due to unhealthy food choices, almost half of adults worldwide are overweight or obese. Current food retail practices bear some responsibility for such public health issues. We argue that numerous attempts to promote healthy eating have been unsuccessful due to the failure to account for our outdated evolved food selection mechanisms. Building on the evolutionary mismatch hypothesis and contrasting ancestral versus present-day foraging environments, we discuss how marketing activities exploit evolutionarily old food preferences and elicit unhealthy food choices for profit maximization at the expense of public health in terms of food consumption. We conclude by explaining how to …

A Machine Learning Perspective On Fnirs Signal Quality Control Approaches, Andrea Bizzego, Michelle Neoh, Giulio Gabrieli, Gianluca Esposito Jan 2022

A Machine Learning Perspective On Fnirs Signal Quality Control Approaches, Andrea Bizzego, Michelle Neoh, Giulio Gabrieli, Gianluca Esposito

Research Outputs: 2025-Present

Despite a rise in the use of functional Near Infra-Red Spectroscopy (fNIRS) to study neural systems, fNIRS signal processing is not standardized and is highly affected by empirical and manual procedures. At the beginning of any signal processing procedure, Signal Quality Control (SQC) is critical to prevent errors and unreliable results. In fNIRS analysis, SQC currently relies on applying empirical thresholds to handcrafted Signal Quality Indicators (SQIs). In this study, we use a dataset of fNIRS signals (N = 1,340) recorded from 67 subjects, and manually label the signal quality of a subset of segments (N = 548) to investigate …

To this end, we briefly introduce the RRI paradigm (Responsible Research and Innovation) and then highlight seven EU research projects in the context of a transnational COST Action project. This COST Action will investigate how placemaking activities, like public art, civil urban design, and local knowledge production, reshape and reinvent public space, and improve citizens’ involvement in urban planning and urban design, especially in the context of heritage sites. The chapter introduces heritage case studies that either contrast, differentiate, and add to existing knowledge and practices in …

Thermal Death Kinetics Of Three Representative Salmonella Enterica Strains In Toasted Oats Cereal, Matthew Chick, Antonio Lourenco, Alice Maserati, Ryan C. Fink, Francisco Diez-Gonzalez Jan 2022

Thermal Death Kinetics Of Three Representative Salmonella Enterica Strains In Toasted Oats Cereal, Matthew Chick, Antonio Lourenco, Alice Maserati, Ryan C. Fink, Francisco Diez-Gonzalez

Research Outputs: 2025-Present

Several reports have indicated that the thermal tolerance of Salmonella at low-water activity increases significantly, but information on the impact of diverse food matrices is still scarce. The goal of this research was to determine the kinetic parameters (decimal reduction time, D; time required for the first decimal reduction, δ) of thermal resistance of Salmonella in a previously cooked low water activity food. Commercial toasted oats cereal (TOC) was used as the food model, with or without sucrose (25%) addition. TOC samples were inoculated with 108 CFU/mL of a single strain of one of three Salmonella serovars (Agona, Tennessee, …

Unlocking The Functional Potential Of Polyploid Yeasts, Simone Mozzachiodi, Kristoffer Krogerus, Brian Gibson, Alain Nicolas, Gianni Liti Jan 2022

Unlocking The Functional Potential Of Polyploid Yeasts, Simone Mozzachiodi, Kristoffer Krogerus, Brian Gibson, Alain Nicolas, Gianni Liti

Research Outputs: 2025-Present

Breeding and domestication have generated widely exploited crops, animals and microbes. However, many Saccharomyces cerevisiae industrial strains have complex polyploid genomes and are sterile, preventing genetic improvement strategies based on breeding. Here, we present a strain improvement approach based on the budding yeasts’ property to promote genetic recombination when meiosis is interrupted and cells return-to-mitotic-growth (RTG). We demonstrate that two unrelated sterile industrial strains with complex triploid and tetraploid genomes are RTG-competent and develop a visual screening for easy and high-throughput identification of recombined RTG clones based on colony phenotypes. Sequencing of the evolved clones reveal unprecedented levels of RTG-induced …

Theta-Gamma Coupling As A Cortical Biomarker Of Brain-Computer Interface-Mediated Motor Recovery In Chronic Stroke, Nabi Rustamov, Joseph Humphries, Alexandre Carter, Eric C Leuthardt Jan 2022

Theta-Gamma Coupling As A Cortical Biomarker Of Brain-Computer Interface-Mediated Motor Recovery In Chronic Stroke, Nabi Rustamov, Joseph Humphries, Alexandre Carter, Eric C Leuthardt

2020-Current year OA Pubs

Chronic stroke patients with upper-limb motor disabilities are now beginning to see treatment options that were not previously available. To date, the two options recently approved by the United States Food and Drug Administration include vagus nerve stimulation and brain-computer interface therapy. While the mechanisms for vagus nerve stimulation have been well defined, the mechanisms underlying brain-computer interface-driven motor rehabilitation are largely unknown. Given that cross-frequency coupling has been associated with a wide variety of higher-order functions involved in learning and memory, we hypothesized this rhythm-specific mechanism would correlate with the functional improvements effected by a brain-computer interface. This study …

Increased Virus Dissemination Leads To Enhanced Lung Injury But Not Inflammation During Influenza-Associated Secondary Bacterial Infection, Amanda P Smith, Lindey C Lane, Ivan Ramirez Zuniga, David M Moquin, Peter Vogel, Amber M Smith Jan 2022

Increased Virus Dissemination Leads To Enhanced Lung Injury But Not Inflammation During Influenza-Associated Secondary Bacterial Infection, Amanda P Smith, Lindey C Lane, Ivan Ramirez Zuniga, David M Moquin, Peter Vogel, Amber M Smith

2020-Current year OA Pubs

Secondary bacterial infections increase influenza-related morbidity and mortality, particularly if acquired after 5-7 d from the viral onset. Synergistic host responses and direct pathogen-pathogen interactions are thought to lead to a state of hyperinflammation, but the kinetics of the lung pathology have not yet been detailed, and identifying the contribution of different mechanisms to disease is difficult because these may change over time. To address this gap, we examined host-pathogen and lung pathology dynamics following a secondary bacterial infection initiated at different time points after influenza within a murine model. We then used a mathematical approach to quantify the increased …

Evaluation Of Virtual Screening Strategies For The Identification Of Γ-Secretase Inhibitors And Modulators, Alicia Ioppolo, Melissa Eccles, David Groth, Giuseppe Verdile, Mark Agostino Jan 2022

Evaluation Of Virtual Screening Strategies For The Identification Of Γ-Secretase Inhibitors And Modulators, Alicia Ioppolo, Melissa Eccles, David Groth, Giuseppe Verdile, Mark Agostino

Research outputs 2022 to 2026

γ-Secretase is an intramembrane aspartyl protease that is important in regulating normal cell physiology via cleavage of over 100 transmembrane proteins, including Amyloid Precursor Protein (APP) and Notch family receptors. However, aberrant proteolysis of substrates has implications in the progression of disease pathologies, including Alzheimer’s disease (AD), cancers, and skin disorders. While several γ-secretase inhibitors have been identified, there has been toxicity observed in clinical trials associated with non-selective enzyme inhibition. To address this, γ-secretase modulators have been identified and pursued as more selective agents. Recent structural evidence has provided an insight into how γ-secretase inhibitors and modulators are recognized …

Research outputs 2022 to 2026

Addressing overconsumption of protein-rich foods from high ecological footprint sources can have positive impacts on health such as reduction of non-communicable disease risk and protecting the natural environment. With the increased attention towards development of ecologically sustainable diets, this systematic review aimed to critically review literature on effectiveness of those interventions aiming to promote protein-rich foods from lower ecological footprint sources. Five electronic databases (Medline, Web of Science, Scopus, Embase and Global Health) were searched for articles published up to January 2021. Quantitative studies were eligible for inclusion if they reported on actual or intended consumption of protein-rich animal-derived and/or …

Strength Training In Swimming, Klaus Wirth, Michael Keiner, Stefan Fuhrmann, Alfred Nimmerichter, Gregory G. Haff Jan 2022

Strength Training In Swimming, Klaus Wirth, Michael Keiner, Stefan Fuhrmann, Alfred Nimmerichter, Gregory G. Haff

Research outputs 2022 to 2026

This narrative review deals with the topic of strength training in swimming, which has been a controversial issue for decades. It is not only about the importance for the performance at start, turn and swim speed, but also about the question of how to design a strength training program. Different approaches are discussed in the literature, with two aspects in the foreground. On the one hand is the discussion about the optimal intensity in strength training and, on the other hand, is the question of how specific strength training should be designed. In addition to a summary of the current …

Bioturbation By Echidna (Tachyglossus Aculeatus) In A Forest Habitat, South-Western Australia, Shannon J. Dundas, Lara Osborne, Anna J. M. Hopkins, Katinka X. Ruthrof, Patricia A. Fleming

Research outputs 2022 to 2026

Bioturbation by digging animals is important for key forest ecosystem processes such as soil turnover, decomposition, nutrient cycling, water infiltration, seedling recruitment, and fungal dispersal. Despite their widespread geographic range, little is known about the role of the short-beaked echidna (Tachyglossus aculeatus) in forest ecosystems. We measured the density and size of echidna diggings in the Northern Jarrah Forest, south-western Australia, to quantify the contribution echidna make to soil turnover. We recorded an overall density of 298 echidna diggings per hectare, 21 % of which were estimated to be less than 1 month old. The average size of digs was …

Comparison Of Auditory Steady-State Responses With Conventional Audiometry In Older Adults, Hadeel Y. Tarawneh, Hamid R. Sohrabi, Wilhelmina H. A. M. Mulders, Ralph N. Martins, Dona M. P. Jayakody

Research outputs 2022 to 2026

Behavioral measures, such as pure-tone audiometry (PTA), are commonly used to determine hearing thresholds, however, PTA does not always provide reliable hearing information in difficult to test individuals. Therefore, objective measures of hearing sensitivity that require little-to-no active participation from an individual are needed to facilitate the detection and treatment of hearing loss in difficult to test people. Investigation of the reliability of the auditory steady-state response (ASSR) for measuring hearing thresholds in older adults is limited. This study aimed to investigate if ASSR can be a reliable, objective measure of frequency specific hearing thresholds in older adults. Hearing thresholds …

Editorial: Fire Regimes In Desert Ecosystems: Drivers, Impacts And Changes, Eddie J. B. Van Etten, Matthew L. Brooks, Aaron C. Greenville, Glenda M. Wardle

Research outputs 2022 to 2026

Although not commonly associated with fire, many desert ecosystems across the globe do occasionally burn, and there is evidence that fire incidences are increasing, leading to altered fire regimes in this biome. The increased prevalence of megafires (wildfires > 10,000 ha in size and typically damaging) in most global biomes is linked to climate change, although those occurring in deserts have received far less attention, from both a research and policy perspective, than that of forested ecosystems (Linley et al., 2022). Understanding the drivers of desert fires, from climate to landscape patterns of hydrology and soil, and how these may be …

Stem Cell Therapy For Insulin-Dependent Diabetes: Are We Still On The Road?, Lu Yang, Zhu-Meng Hu, Fang-Xu Jiang, Wei Wang

Research outputs 2022 to 2026

In insulin-dependent diabetes, the islet β cells do not produce enough insulin and the patients must receive exogenous insulin to control blood sugar. However, there are still many deficiencies in exogenous insulin supplementation. Therefore, the replacement of destroyed functional β cells with insulin-secreting cells derived from functional stem cells is a good idea as a new therapeutic idea. This review introduces the development schedule of mouse and human embryonic islets. The differences between mouse and human pancreas embryo development were also listed. Accordingly to the different sources of stem cells, the important research achievements on the differentiation of insulin-secreting β …
