Federally inspected slaughter establishments in the United States must adhere to the Humane Methods of Slaughter Act and regulations that enforce it. Failure to comply with this law results in a Humane Handling Enforcement Action (HHEA) issued by the United States Department of Agriculture Food Safety Inspection Service (USDA FSIS). The objective of this study was to systematically analyze and describe HHEAs issued between 2018 and 2020. Enforcement action notification letters were accessed from the USDA FSIS website and date, location, regulatory action, reason for noncompliance, species, and follow up action for each HHEA was recorded. …

Efficient Interface Enabled By Nano-Hydroxyapatite@Porous Carbon For Lithium-Sulfur Batteries, Jia-Yu Wang, Xue-Feng Tong, Qi-Fan Peng, Yue-Peng Guan, Wei-Kun Wang, An-Bang Wang, Nai-Qiang Liu, Ya-Qin Huang
Journal of Electrochemistry
The dissolution and “shuttle effect” of lithium polysulfides (LiPSs) hinder the application of lithium-sulfur (Li-S) batteries. To solve those problems, inspired by natural materials, a nano-hydroxyapatite@porous carbon derived from chicken cartilage (nano-HA@CCPC) was fabricated by employing a simple pre-carbonization and carbonization method, and applied in Li-S batteries. The nano-HA@CCPC would provide a reactive interface that allows efficient LiPSs reduction. With a strong affinity for LiPSs and an excellent electronic conductive path for converting LiPSs, the shuttle effect of LiPSs was confined and the redox kinetics of LiPSs was substantially enhanced. Li-S batteries employing nano-HA@CCPC-modified separators exhibited long cycle life and …
The Shadow Of Fast Fashion: The Quilting Industry, Elizabeth Y. Barnes
The Shadow Of Fast Fashion: The Quilting Industry, Elizabeth Y. Barnes
Honors Theses
The purpose behind this creative research project is to bring the contemporary quilting industry to attention when discussing environmental concerns that are typically associated with the textile industry. Quilting, despite its origins in inadvertent conservatism, has evolved into a consumerist business that can be most adequately compared to that of fast fashion. Through analysis of the different types of fibers, manufacturing processes, and environmental implications, an extensive background of the textile industry was given so that the consequences discussed later in the paper can be fully realized. By comparing examples of need-based quilting, like the quilts of Gee’s Bend, to …

Emotionally Unavailable: A Comparison On The Ability Of Children With Developmental Language Disorders To Interpret Emotions In Humans And Inanimate Objects, Savanna B. Gullett
Honors Theses
Developmental Language Disorder (DLD) is a neuro-developmental condition that emerges in early childhood and can persist into adulthood, manifesting as a variety of symptoms that can be significantly different in each individual. It has an estimated prevalence of 7.58% and is nearly 7 times more common in children than autism spectrum disorder (McGregor, 2020). As a population, individuals with DLD are at risk of facing significant issues in associating areas of language, including reading comprehension, problem-solving abilities, and social skills. Professionals have noticed emotional disconnect amongst individuals with DLD, leading to the proposition that those with DLD may have a …

Reducing Instrumentation Barriers Of Diffuse Correlation Spectroscopy For Low-Cost Deep Tissue Blood Flow Monitoring, Arindam Biswas
USF Tampa Graduate Theses and Dissertations
Cerebral blood flow (CBF) is a good indicator of brain health as blood carries necessary nutrients, oxygen, and metabolic byproducts. Quantitative blood flow information can be used in several clinical and therapeutic applications such as stroke detection, measuring autoregulation, evaluating brain injury, or determining neuronal activity. Over the past few decades, light-based deep tissue hemodynamic detection modalities have become popular for non-invasive CBF measurements. In particular, noninvasive Diffuse Correlation Spectroscopy (DCS), has become a tool of choice for research and clinical applications due to its depth sensitivity (>1 cm), portability, validity against other technologies such as Magnetic Resonance Imaging …

Bioactive Self-Forming Hydrogels For Biomedical Applications, Kirstene Arcuino Gultian Giddings
Bioactive Self-Forming Hydrogels For Biomedical Applications, Kirstene Arcuino Gultian Giddings
Theses and Dissertations
Hydrogels are soft biomaterials that feature a wide range of physical and biochemical properties. Although highly versatile, hydrogels typically require an exogeneous input (e.g., light, pH, temperature) to form, and this limits their use to conditions where a catalyst is present. The goal of this dissertation is to expand the use of catalyst-based hydrogels by developing hydrogels with independently tunable properties which gel in situ without external stimuli. This new class of hydrogels was developed by first modifying macromers with norbornene (Nor) or tetrazine (Tet) moieties. Macromers containing Nor or Tet remain liquid when dissolved, and upon mixing self-form into …

Crohn's disease (CD), is an inflammatory bowel disease that can affect any part of the gastro-intestinal tract (GI) and is associated with an increased risk of gastro-intestinal cancer. In the current study, we determined the role of genetic and small-molecule modulation of STAT3 in a mouse model of CD. STAT3 has 2 isoforms (α, β) which are expressed in most cells in a 4:1 ratio (α: β). STAT3α has pro-inflammatory and anti-apoptotic functions, while STAT3β has contrasting roles. We used an animal model of CD consisting of intrarectal administration of 2,4,6-trinitrobenzene sulfonic acid and examined the severity of CD in …

In endochondral bone development, bone-forming osteoblasts and bone marrow stromal cells have dual origins in the fetal cartilage and its surrounding perichondrium. However, how early perichondrial cells distinctively contribute to developing bones remain unidentified. Here we show using in vivo cell-lineage analyses that Dlx5+ fetal perichondrial cells marked by Dlx5-creER do not generate cartilage but sustainably contribute to cortical bone and marrow stromal compartments in a manner complementary to fetal chondrocyte derivatives under the regulation of Hedgehog signaling. Postnatally, Dlx5+ fetal perichondrial cell derivatives preferentially populate the diaphyseal marrow stroma with a dormant adipocyte-biased state and are refractory to parathyroid …

"The Missus Has Paid For Your Life.": Preconstituted Third-World Women In Chris Cleave's Little Bee, Jeanette P. Barry, Jeanette Parry Barry
Masters Theses
In “Under Western Eyes,” Chandra Mohanty explains how some Western feminist scholars unconsciously other third-world women by imposing on them their Eurocentric universality. Using Mohanty as a primary lens, this thesis argues that in Chris Cleave’s Little Bee (2008), the protagonists, Sarah and Little Bee, are respectively represented as a Western referent and a third-world preconstituted woman, rendering any true collaboration between them impossible. Little Bee is meant to enlighten its readers of the plight of refugees in the United Kingdom, and in some ways it does just that. Little Bee also, however, situates the English protagonist Sarah as a …

Basal-like breast cancers, an aggressive breast cancer subtype that has poor treatment options, are thought to arise from luminal mammary epithelial cells that undergo basal plasticity through poorly understood mechanisms. Using genetic mouse models and ex vivo primary organoid cultures, we show that conditional co-deletion of the LATS1 and LATS2 kinases, key effectors of Hippo pathway signaling, in mature mammary luminal epithelial cells promotes the development of Krt14 and Sox9-expressing basal-like carcinomas that metastasize over time. Genetic co-deletion experiments revealed that phenotypes resulting from the loss of LATS1/2 activity are dependent on the transcriptional regulators YAP/TAZ. Gene expression analyses of …

Biological effects of radioactive particles can be experimentally investigated in vitro as a function of particle concentration, specific activity and exposure time. However, a careful dosimetric analysis is needed to elucidate the role of radiation emitted by radioactive products in inducing cyto- and geno-toxicity: the quantification of radiation dose is essential to eventually inform dose-risk correlations. This is even more fundamental when radioactive particles are short-range emitters and when they have a chemical speciation that might further concur to the heterogeneity of energy deposition at the cellular and sub-cellular level. To this aim, we need to use computational models. In …

Axisymmetric Soft Elastohydrodynamic Squeeze Films: Parametric Studies, Eli Heisey
Axisymmetric Soft Elastohydrodynamic Squeeze Films: Parametric Studies, Eli Heisey
Theses
Theoretical and experimental studies of hydrodynamically lubricated bearings with a soft, compliant surface have shown advantages over conventionally rigid bearings. These bearings operate in the isoviscous, elastohydrodynamic regime of thin film lubrication with low film pressures and relatively large surface deflections. They exhibit less sensitivity to misalignment, particulates in the lubricant, and low levels of lubricant, making them very attractive for bearing designers. However, there is very little published work, either experimental or theoretical, to describe the squeeze-film lubrication behavior of a bearing with a compliant layer. This thesis utilizes a Nodal Inverse Hydrodynamic computation approach to calculate the behavior …

Non-Standard Viral Genome-Derived Rna Activates Tlr3 And Type I Ifn Signaling To Induce Cdc1-Dependent Cd8+ T-Cell Responses During Vaccination In Mice, Devin G Fisher, Victoria Gnazzo, David J Holthausen, Carolina B López
2020-Current year OA Pubs
There is a critical need to develop vaccine adjuvants that induce robust immune responses able to protect against intracellular pathogens, including viruses. Previously, we described defective viral genome-derived oligonucleotides (DDOs) as novel adjuvants that strongly induce type 1 immune responses, including protective Th1 CD4+ T-cells and effector CD8+ T-cells in mice. Here, we unravel the early innate response required for this type 1 immunity induction. Upon DDO subcutaneous injection, type 1 conventional dendritic cells (cDC1s) accumulate rapidly in the draining lymph node in a Toll-like receptor 3 (TLR3)- and type I interferon (IFN)-dependent manner. cDC1 accumulation in the lymph node …

Despite advances in sequencing, lack of standardization makes comparisons across studies challenging and hampers insights into the structure and function of microbial communities across multiple habitats on a planetary scale. Here we present a multi-omics analysis of a diverse set of 880 microbial community samples collected for the Earth Microbiome Project. We include amplicon (16S, 18S, ITS) and shotgun metagenomic sequence data, and untargeted metabolomics data (liquid chromatography-tandem mass spectrometry and gas chromatography mass spectrometry). We used standardized protocols and analytical methods to characterize microbial communities, focusing on relationships and co-occurrences of microbially related metabolites and microbial taxa across environments, …
A Multiple Input Multiple Output Framework For The Automatic Optical Fractionator-Based Cell Counting In Z-Stacks Using Deep Learning, Palak Dave
USF Tampa Graduate Theses and Dissertations
Quantifying cells in a defined region of biological tissue is critical for many clinical and preclinical studies, especially in pathology, toxicology, cancer, and behavior. Unbiased stereology is the state-of-art method for quantification of the total number and other morphometric parameters of stained objects in a defined region of biological tissue. As part of a program to develop accurate, precise, and more efficient automatic approaches for quantifying morphometric changes in biological tissue, our group has shown that both deep learning-based and hand-crafted algorithms can estimate the total number of histologically stained cells at their maximal profile of focus in extended depth …

Myotonic dystrophy (DM) is a highly variable, multisystemic disorder that clinically affects one in 8000 individuals. While research has predominantly focused on the symptoms and pathological mechanisms affecting striated muscle and brain, DM patient surveys have identified a high prevalence for gastrointestinal (GI) symptoms amongst affected individuals. Clinical studies have identified chronic and progressive dysfunction of the esophagus, stomach, liver and gallbladder, small and large intestine, and rectum and anal sphincters. Despite the high incidence of GI dysmotility in DM, little is known regarding the pathological mechanisms leading to GI dysfunction. In this review, we summarize results from clinical and …

Who Am I When I Am Teaching? Self In Yoga Practice, Krzysztof T. Konecki
The Qualitative Report
The paper shows the situational transitions between the different identities of a teacher in hatha yoga practice: teacher, instructor, trainee, student, and researcher. The analysis is based on self-reports of my feelings about practicing and teaching hatha yoga. The explication is based on the self-observation of my lived experiences and the observation of interactions with hatha yoga session participants. The self emerges from the interactions or lack of interactions. Sometimes I am a participant, sometimes an instructor, and sometimes a student, even in the same yoga session. I analyze my self-reports using the first-person perspective and third-person mode. There are …

Memes From Confinement: Disorientation And Hindsight Projection In The Crisis Of Covid-19, David Divita
Pomona Faculty Publications and Research
Memes have been described as textual forms of “(post)modern folklore” (Shifman, 2014: 5). Photos or short videos, they highlight current cultural phenomena, and they spread exponentially through person-to-person sharing on social media platforms. For this article, I created a corpus of memes that circulated in March 2020, during the first weeks after statewide lockdown orders were issued in the U.S. in response to the COVID-19 pandemic. Drawing on Bakthin’s (1981) concept of the chronotope, I analyze a subset of these memes that specifically addressed the experience of time in confinement, illuminating two interrelated trends: the disruption of temporal order in …

Water is essential for survival, but one in three individuals worldwide (2.2 billion people) lacks access to safe drinking water. Water intake requirements largely reflect water turnover (WT), the water used by the body each day. We investigated the determinants of human WT in 5604 people from the ages of 8 days to 96 years from 23 countries using isotope-tracking (2H) methods. Age, body size, and composition were significantly associated with WT, as were physical activity, athletic status, pregnancy, socioeconomic status, and environmental characteristics (latitude, altitude, air temperature, and humidity). People who lived in countries with a low human development …
