Quantitative Super-Resolution Microscopy, Andrew Barentine

The diffraction limit fundamentally restricts the achievable resolution of conventional microscopes, however it can be circumvented if only a subset of fluorophores within a given diffraction-limited volume are simultaneously emitting. Such super-resolution methods routinely achieve tens of nanometer resolution in whole (if not living) cells. However, being relatively young, these techniques still struggle with providing robust, quantitative, and statistically powerful data in a routine manner. Accurate quantification of subcellular features requires an estimate of the achieved spatial resolution, but traditional methods of measuring it do not cope well with these techniques. Understanding the constraints, including biological ones, that may influence mathematical development is of great importance because math ability is a key predictor of career success, income and even psychological well-being. While research in developmental cognitive neuroscience of mathematics has extensively studied the key functional regions for processing numbers, particularly the horizontal segment of intraparietal sulcus (HIPS), few studies have investigated the effects of early cerebral constraints on later mathematical abilities. Cell migration is an essential physiological process, and aberrant migration of epithelial cells underlies many pathological conditions. However, the molecular mechanisms governing cell migration are not fully understood. We report here that growth factor–induced epithelial cell migration is critically dependent on the crosstalk of two molecular switches, namely phosphorylation switch (P-switch) and transcriptional switch (T-switch). As each year passes, a fatal cardiovascular event continues to be the leading cause of death among on-duty firefighters. Risk factors for a fatal cardiovascular event and cardiovascular health disease (CVD) are obesity, sleep deprivation, weight gain, lack of exercise, unhealthy diet, etc. The research in this thesis lays out the poor association between cardiovascular health and firefighters. Interventional programs that aim to improve the cardiovascular health of firefighters must be implemented universally to all fire departments by the International Association of Firefighters. A lack of protocol for screening of social determinants of health (SDH), specifically for pediatric patients, despite national endorsement, is setting a basis for increased rates of negative health outcomes that can follow individuals into adulthood. The PRAPARE tool, or the Protocol for Responding to and Assessing Patients’ Assets, Risks, and Experiences, is a guided national effort to assist healthcare centers in the collection of the necessary social data required to care for patients and act upon their social determinants of health (The National Association of Community Health Centers [NACHC], 2019). Uncovering The Model Minority Narrative: A Case Study On Asian American Students In High-Achieving Schools, Joanne Choi

Asian American students in high-achieving secondary schools often have unique cultural experiences, perspectives, and challenges due to the perpetuation of the “Model Minority” conceptualization coined by sociologist William Peterson. Guided by the theoretical frameworks of Critical Race Theory and Culturally Responsive Teaching from the existing literature by theorists such as Ladson-Billings, Gay, and Paris, my research attempts to devise potential methods for secondary educators to make their classrooms more culturally responsive, address Asian American students' deficits or educational challenges, and eliminate harmful biases.

The presence of a nearby boundary is likely to be important in the life cycle and evolution of motile flagellate bacteria. This has led many authors to employ numerical simulations to model near-surface bacterial motion and compute hydrodynamic boundary effects. A common choice has been the method of images for regularized Stokeslets (MIRS); however, the method requires discretization sizes and regularization parameters that are not specified by any theory. To determine appropriate regularization parameters for given discretization choices in MIRS, we conducted dynamically similar macroscopic experiments and fit the simulations to the data. BACKGROUND: The human tumor microenvironment (TME) is a complex and dynamic milieu of diverse acellular and cellular components, creating an immunosuppressive environment, which contributes to tumor progression. We have previously shown that phosphatidylserine (PS) expressed on the surface of exosomes isolated from human TMEs is causally linked to T-cell immunosuppression, representing a potential immunotherapeutic target. In this study, we investigated the effect of ExoBlock, a novel PS-binding molecule, on T-cell responses in the TME.

METHODS: We designed and synthesized a new compound, (ZnDPA)

Testing The Role Of Wfs1 In Calcium Regulation In Drosophila Cells, Briana Scott, Alyandra Lee

The purpose of the project is to knock out the Wfs1 gene in Drosophila (fruit fly) cells and test the hypothesis that this gene is involved in regulating intracellular calcium levels. To test this hypothesis, the gene will first be knocked out using CRISPR-Cas9 molecular tools that are readily available in Dr. Shanle's lab. This will effectively inactivate the Wsf1 gene in cells. To do this, we will transfect Drosophila cells with CRISPR-Cas9 targeting the gene. Next, we will confirm that the gene is mutated using genomic DNA purification, polymerase chain reaction, and sequencing. Finally, we will treat cells with …

We will investigate the physical impacts both short and long term effects of of substance use. In doing so we will frame an argument for the substance's addictive properties as it relates to both its physically addictive and sociological addictive properties. In doing, so a stronger understanding of how an individual can move through the stages of substance use, to abuse, and finally addiction will be reviewed. We will then address current treatment strategies for the substance and how they address or fail to address both the physical and sociological impacts of its use.
