This perspective on Penn State Extension Services and plain people is based on my personal experiences as a plain person, in which I interacted with Extension Services first as a farmer, and then (while working on a doctorate) as a part of the Extension system. Penn State Extension started over a century ago and was deliberate in reaching out to plain (conservative Anabaptist) farmers since the beginning, which led to a history of trust-based cooperation. For all these successes there remain challenges to effective cooperation with certain plain individuals and subgroups. I suggest these challenges are broadly similar to those …

Certain aspects of Amish agriculture have been studied extensively but much is still unknown about Amish environmental attitudes and beliefs. This is due, in part, to the difficulty of directly soliciting responses from adherents. This article—part of a larger study of the Kishacoquillas Valley Amish settlement in Mifflin County, Pennsylvania—reflects on methodological challenges that arose during a study on Amish environmental attitudes and behaviors. Farmers from two Amish groups, as well as two non-Amish groups used for comparison, were interviewed about their environmental attitudes. Recruiting Amish participants for interviews was difficult due to the limited use of modern telecommunications technology, …

Caring For The Land And The Livestock: Anabaptist Agricultural Practices In Europe And Colonial Pennsylvania, Rebecca Shenton

Anabaptists have a strong history of agricultural innovation and care for the land. Their innovative spirit was forged out of persecution, migration, and the need to survive in challenging circumstances. This article examines the agricultural practices of seventeenth- and eighteenth-century Swiss and South German Anabaptist farmers and those of eighteenth-century Anabaptist immigrants to Pennsylvania. European Anabaptist tenant farmers distinguished themselves by their family-centered mixed agriculture and their investment in both the land (using manure, lime, gypsum, and crop rotation to improve the soil) and livestock (improving natural meadows and planting pastures for fodder, maintaining clean barns, practicing confinement feeding, and …

Purpose: The purpose of this qualitative multi-case study was to describe and analyze how schools that implement a STEAM program include the arts in their integrated program. The study focused on 3 K-12 schools in Southern California that are located in the Antelope Valley high desert region of the state. This multi-case study analyzed an elementary school, a middle school, and a high school that are in different districts in the area.

Cytokine production is a critical component of cell-extrinsic responses to DNA damage and cellular senescence. Here, we demonstrated that expression of the gene encoding interleukin-19 (IL-19) was enhanced by DNA damage through pathways mediated by c-Jun amino-terminal kinase (JNK) and cGAS-STING and that IL19 expression was required for the subsequent production of the cytokines IL-1, IL-6, and IL-8. IL19 expression was stimulated by diverse cellular stresses, including inhibition of the DNA replication checkpoint kinase ATR (ataxia telangiectasia and Rad3-related protein), oncogene expression, replicative exhaustion, oxidative stress, and DNA double-strand breaks. The heart is a major organ whose function is to transport nutrients and waste throughout the body. This organ can become infected by pathogens, such as viruses, bacteria, or parasites. Infection of the middle heart layer, or myocardium, is often caused by a viral agent. This disease has three stages: viral infiltration, adaptive immune system activation, and finally either viral clearance or cardiac cell remodeling. During this process the immune system will begin to secrete cytokines, which are signaling molecules that alert other members of the immune pathways, and also participate in cardiac remodeling. Background: African Americans (AAs) suffer a higher stroke burden due to hypertension. Identifying genetic contributors to stroke among AAs with hypertension is critical to understanding the genetic basis of the disease, as well as detecting at-risk individuals.
Methods: In a population comprising over 10,700 AAs treated for hypertension from the Genetics of Hypertension Associated Treatments (GenHAT) and Reasons for Geographic and Racial Differences in Stroke (REGARDS) studies, we performed an inverse variance-weighted meta-analysis of incident stroke. In this paper, we outline the need for a coordinated international effort toward the building of an open-access Global Ocean Oxygen Database and ATlas (GO2DAT) complying with the FAIR principles (Findable, Accessible, Interoperable, and Reusable). GO2DAT will combine data from the coastal and open ocean, as measured by the chemical Winkler titration method or by sensors (e.g., optodes, electrodes) from Eulerian and Lagrangian platforms (e.g., ships, moorings, profiling floats, gliders, ships of opportunities, marine mammals, cabled observatories). GO2DAT will further adopt a community-agreed, fully documented metadata format and a consistent quality control (QC) procedure and quality flagging (QF) system. GO2DAT …

Codling moth (CM) (Cydia pomonella L.), a devastating pest, creates a serious issue for apple production and marketing in apple-producing countries. Therefore, effective nondestructive early detection of external and internal defects in CM-infested apples could remarkably prevent postharvest losses and improve the quality of the final product. In this study, near-infrared (NIR) hyperspectral reflectance imaging in the wavelength range of 900–1700 nm was applied to detect CM infestation at the pixel level for three organic apple cultivars, namely Gala, Fuji and Granny Smith. ACEs are adverse childhood experiences experienced in the first 18 years of life. They are present in over a third of the population and lead to adverse health outcomes (Karatekin, 2017). The goal of this study was to examine whether ACEs could be used to identify stress levels of college students during the COVID-19 pandemic. Data on ACEs and stress levels were collected from students actively enrolled at a college or university (N= 183). Findings indicated that high ACEs were predictive of high stress, specifically considering stressful factors experienced during the COVID-19 pandemic. The results of this study suggest the …

Introduction: Mutations in the KCNN2 gene have been linked to phenotypes of movement disorders and autism. The goal of this experiment was to use the CIUSPR-Cas9 system to create a deletion within the KCNN2 gene and to observe the effect on the expression of movement in zebra fish embryos.
Methods: Linearized Cas9 DNA and guide DNA underwent in vitm transcription (IVT) reactions to produce RNA to be injected into zebrafish embryos at the one cell stage. There are functional benefits to exercise in muscle, even when performed late in life, but the contributions of epigenetic factors to late-life exercise adaptation are poorly defined. Using reduced representation bisulfite sequencing (RRBS), ribosomal DNA (rDNA) and mitochondrial-specific examination of methylation, targeted high-resolution methylation analysis, and DNAge™ epigenetic aging clock analysis with a translatable model of voluntary murine endurance/resistance exercise training (progressive weighted wheel running, PoWeR), we provide evidence that exercise may mitigate epigenetic aging in skeletal muscle. Late-life PoWeR from 22–24 months of age modestly but significantly attenuates an age-associated shift toward promoter hypermethylation. The epigenetic age of muscle …

Hevea brasiliensis (rubber tree) is a large tree species of the Euphorbiaceae family with inestimable economic importance. Rubber tree breeding programs currently aim to improve growth and production, and the use of early genotype selection technologies can accelerate such processes, mainly with the incorporation of genomic tools, such as marker-assisted selection (MAS). However, few quantitative trait loci (QTLs) have been used successfully in MAS for complex characteristics. Recent research shows the efficiency of genome-wide association studies (GWAS) for locating QTL regions in different populations. Objectives
To compare phagocytic activities of monocytes in peripheral blood mononuclear cells (PBMCs) from acute gout patients and normal subjects, examine monosodium urate monohydrate (MSU) crystal-induced IL-1β secretion ± recombinant human proteoglycan 4 (rhPRG4) or interleukin-1 receptor antagonist (IL-1RA), and study the anti-inflammatory mechanism of rhPRG4 in MSU stimulated monocytes.
Methods
Acute gout PBMCs were collected from patients in the Emergency Department and normal PBMCs were obtained from a commercial source. Monocytes in PBMCs were identified by flow cytometry. Cognitive load refers to the amount of used working memory resources, which is limited in both capacity and duration. Predicting cognitive load from raw electroencephalogram (EEG) recordings remains a challenge because of the high degree of noise due to technical variations in the recording process and the multi-factorial nature of the mapping between the EEG data and cognitive load. We present parameter-optimized deep convolutional neural network (CNN) models to predict four levels of cognitive load. In 2021, approximately 248,530 new prostate cancer (PCa) cases are estimated in the United States. Hispanic/Latinos (H/L) are the second largest racial/ethnic group in the US. The objective of this study was to assess DNA methylation patterns between aggressive and indolent PCa along with ancestry proportions in 49 H/L men from Puerto Rico (PR). Prostate tumors were classified as aggressive (n = 17) and indolent (n = 32) based on the Gleason score. Genomic DNA samples were extracted by macro-dissection. DNA methylation patterns were assessed using the Illumina EPIC DNA methylation platform. We used ADMIXTURE to estimate global ancestry proportions. …

Introduction/Objective: The efficacy of biologic therapy in the treatment of rheumatoid arthritis (RA) has been well-established but, in practice, a quarter of patients will either not respond to the first biologic agent or will suffer an adverse event requiring a switch to a different drug. While clinical guidelines exist to help guide therapy and previous studies have examined sequential use of anti-TNF agents, there is little data to inform a multiple switch strategy. This project continues to fit the experimental data on the stress-strain behavior of newborn mice’s ascending aorta. The stress-strain behavior of newborn mouse aorta is highly nonlinear because of its unique composition of elastin and collagen in the arterial wall. To mathematically describe this behavior, more strain energy functions of different biological materials were used in fitting. Multiple error functions were used to improve accuracy. K-fold cross-validation was applied in regression to avoid overfitting. The nuclear receptor PPARα is associated with reducing adiposity, especially in the liver, where it transactivates genes for β-oxidation. Contrarily, the function of PPARα in extrahepatic tissues is less known. Therefore, we established the first adipose-specific PPARα knockout (PparaFatKO) mice to determine the signaling position of PPARα in adipose tissue expansion that occurs during the development of obesity. To assess the function of PPARα in adiposity, female and male mice were placed on a high-fat diet (HFD) or normal chow for 30 weeks. Secrecy and the use of “secret information” as capital in the hands of the state is mobilised by affective racialised machineries, cultivated on “security” grounds. Securitised secrecy is an assemblage of concealed operations juxtaposing various forms of invasions and dispossessions. It is a central strategy in the politico-economic life of the state to increase its scope of domination. Secrecy is used and abused to entrap and penetrate political subjects and entities. Chlamydia trachomatis is an obligate intracellular bacterium that has developed sophisticated mechanisms to survive inside its infectious compartment, the inclusion. Notably, Chlamydia weaves an extensive network of microtubules (MTs) and actin filaments to enable interactions with host organelles and enhance its stability. Despite the global health and economic burden caused by this sexually transmitted pathogen, little is known about how actin and MT scaffolds are integrated into an increasingly complex virulence system. Previously, we established that the chlamydial effector InaC interacts with ARF1 to stabilize MTs. We now demonstrate that InaC regulates RhoA to control actin scaffolds. InaC relies on …

Attention-deficit Hyperactivity Disorder is one of the most common childhood mental health disorders, affecting about 5.6% of the population worldwide. Several studies have specifically shown a high prevalence of comorbid mood disorders, such as depression and bipolar disorder (BD), in those diagnosed with ADHD. Several common symptoms of ADHD are also found in BD, which are characterized by alternating periods of euthymia and mood disturbances. The inattention and impulsivity of ADHD can be seen in manic and hypomanic episodes of BD. Ongoing Genocides And The Need For Healing: The Cases Of Native And African Americans, Benjamin P. Bowser, Carl O. Word, Kate Shaw
Genocide Studies and Prevention: An International Journal
The elimination of Native peoples and the enslavement of Africans in the U.S. more than qualify as acts of historical state sponsored genocide. A feature of both genocides is that they ended as institutional practices but have continued culturally and psychologically. The primary contemporary legacy of these genocides is racism which reinforces historical trauma and grief. Suggestions are made for how healing for Native and African Americans can begin despite ongoing racism. This includes psychological counseling for White Americans with beliefs in White supremacy. Suggestions are also made for how reconciliation can begin at the county-level between descendants of slave …
