Creation And Optimization Of An Optical Trapping Microfluidic Device For The Separation Of Mock Forensic Sexual Assault Samples, Mackenzie Lally
Creation And Optimization Of An Optical Trapping Microfluidic Device For The Separation Of Mock Forensic Sexual Assault Samples, Mackenzie Lally
Master of Science in Forensic Science Directed Research Projects
Cell mixtures are often seen in forensic samples and commonly involve sexual assault cases where mixtures of sperm cells and vaginal epithelial cells are frequently encountered. This produces challenges in downstream analysis in the form of STR mixture profiles. The only method currently in use in crime laboratories for front-end sperm and epithelial cell separation is differential extraction. This method often results in STR mixture profiles due to carryover into both the male and female fractions and suffers from a wide range of efficiency depending on the laboratory or individual processing the sample. Optical trapping offers an alternative method for …

Publications
During the ongoing COVID-19 crisis, subreddits on Reddit, such as r/Coronavirus saw a rapid growth in user's requests for help (support seekers - SSs) including individuals with varying professions and experiences with diverse perspectives on care (support providers - SPs). Currently, knowledgeable human moderators match an SS with a user with relevant experience, i.e, an SP on these subreddits. This unscalable process defers timely care. We present a medical knowledge-infused approach to efficient matching of SS and SPs validated by experts for the users affected by anxiety and depression, in the context of with COVID-19. After matching, each SP to …
Targeting Galectin-1 As A Potential Therapeutic For Glioblastoma, Wayne Luther Glore
Targeting Galectin-1 As A Potential Therapeutic For Glioblastoma, Wayne Luther Glore
MUSC Theses and Dissertations
Glioblastoma (GB) is classified by the World Health Organization (WHO) as a Grade IV astrocytoma characterized by a poor prognosis with a median survival time ranging from 15-16 months. The standard of care for GB is surgery followed by radiation and chemotherapy treatment with Temozolomide, but even with the aggressive treatment, GB recurrence occurs in approximately 90 % of the patient population. New treatment options have been FDA approved which include Novocure’s Optune Device and Genentech’s Avastin, but neither of these options drastically change survival time or quality of life. Galectin-1 (Gal-1), a protein with a high affinity to bind …
Statistical Methods For Integrative Analysis, Subgroup Identification, And Variable Selection Using Cancer Genomic Data, Zequn Sun
MUSC Theses and Dissertations
In recent years, comprehensive cancer genomics platform, such as The Cancer Genome Atlas (TCGA), provides access to an enormous amount of high throughput genomic datasets for each patient, including gene expression, DNA copy number alteration, DNA methylation, and somatic mutation. Currently most existing analysis approaches focused only on gene-level analysis and suffered from limited interpretability and low reproducibility of findings. Additionally, with increasing availability of the modern compositional data including immune cellular fraction data and high-dimensional zero-inflated microbiome data, variable selection techniques for compositional data became of great interest because they allow inference of key immune cell types (immunology data) …

Performance Comparison Between A Distributed Particle Swarm Algorithm And A Centralised Algorithm, Ciarán O’Loughlin
Performance Comparison Between A Distributed Particle Swarm Algorithm And A Centralised Algorithm, Ciarán O’Loughlin
Dissertations
Particle Swarm optimisation (PSO) is a particular form of swarm intelligence, which itself is an innovative intelligent paradigm for solving optimization problems. PSO is generally used to find a global optimum in a single optimisation function. This typically occurs on one node(machine) but there has been a significant body of research into creating distributed implementations of the PSO algorithm. Such research has often focused on the creation and performance of the distributed implementation in an isolated manner or compared to different distributed algorithms.
This research piece aims to bridge a gap in the existing literature, by testing a distributed implementation …

Polyelectrolyte Multilayer Composite Coating On 316 L Stainless Steel For Controlled Release Of Dual Growth Factors Accelerating Restoration Of Bone Defects, Wai Ching Liu
Department of Food and Health Sciences
A composite coating of polyelectrolyte multilayers (PEMs) consisting of collagen, a chitosan barrier, and poly- γ-glutamic acid was fabricated using a spin coating technique to investigate and overcome the limited osseointegration capacity of 316 L stainless steel (316 L SS). To further enhance the biocompatibility, bone morphogenetic protein 2 (BMP-2) and basic fibroblast growth factor-2 (FGF-2) were loaded separately as dual growth factors, allowing for progressive drug release following the natural process of bone regeneration. The first burst release of FGF-2 triggered the proliferation of surrounding cells, and the subsequent release of BMP-2 stimulated their differentiation. The microstructure, surface potential, …

Assessment Of The Potential Environmental And Ecological Risks Associated With Traffic Induced Heavy Metal Contamination In Country Parks Of Hong Kong, King Wah Ngai, Ming-Wai Mak, Kwok Leung Pun
Department of Construction, Environment and Engineering
Hong Kong is a densely populated area with a limited area, so most country parks are adjacent to busy traffic roads and town activities. The soil in country parks may be contaminated by heavy metals emitted by traffic activities. During the holidays, most people go to country parks to play. It is very important to assess the potential risks of residents, especially children. So far, data on pollution in Hong Kong’s country parks is very limited. The heavy metals arsenic (As), cadmium (Cd), chromium (Cr), copper (Cu), lead (Pb), mercury (Hg), nickel (Ni), silver (Ag) and zinc (Zn) were collected …
Statistics Used In The Nebraska Beef Cattle Report And Their Purpose
Statistics Used In The Nebraska Beef Cattle Report And Their Purpose
Nebraska Beef Cattle Reports
The purpose of beef cattle and beef product research at UNL is to provide reference information that represents the various populations (cows, calves, heifers, feeders, carcasses, retail products, etc) of beef production. Obviously, the researcher cannot apply treatments to every member of a population; therefore he/she must sample the population. The use of statistics allows the researcher and readers of the Nebraska Beef Cattle Report the opportunity to evaluate separation of random (chance) occurrences and real biological effects of a treatment. Following is a brief description of the major statistics used in the beef report. For a more detailed description …

Dietary Impact On Antibiotic Resistance In Feedlot Manure, Maranatha Zelt, Amy Schmidt, Noelle Mware, Xu Li, Galen E. Erickson
Nebraska Beef Cattle Reports
There is a growing public concern regarding antibiotic resistance and the use of antibiotics, including in livestock management. Understanding the ecology of antibiotic resistance among microbes, identifying resistance gene reservoirs, and implementing antibiotic resistance mitigation practices in livestock production are critical to protecting animal and human health while meeting increasing food demands. This research is one of several studies seeking to assess risk for livestock- to- human transfer of antibiotic resistance and to identify mechanisms for reducing that risk where possible. This study evaluated the impact of forage concentration and supplemental essential oil in beef cattle finishing diets on antibiotic …

Nebraska Beef Cattle Reports
The purpose of dry aging is to develop novel favors and other sensory characteristics different from wet aged meat. However, leaving meat exposed to air for an extended period of time can have negative effects on meat quality. As the meat is exposed to oxygen for an extended period of time, lipids are oxidized resulting in compounds that negatively affect favor. In this study, oxygen concentration was regulated along with time, temperature, humidity, and air flow. The purpose of oxygen regulation was to determine the effect of oxidation on the quality, specifically flavor preference, of dry aged meats. Sensory analysis …
Does Biochar Improve All Soil Ecosystem Services?, Humberto Blanco-Canqui
Does Biochar Improve All Soil Ecosystem Services?, Humberto Blanco-Canqui
Department of Agronomy and Horticulture: Faculty Publications
Biochar is considered to sequester C and deliver other soil ecosystem services, but an overview that synthesizes the current knowledge of biochar implications on all essential soil ecosystem services is difficult to find in the ample biochar literature. Most previous research and review articles on this topic focused on a single ecosystem service and did not integrate all essential soil ecosystem services. This overview paper (1) synthesizes the impacts of biochar on water and wind erosion, C sequestration, soil water, nutrient leaching, soil fertility, crop yields, and other soil ecosystem services based on published literature and (2) highlights remaining research …

Department of Agronomy and Horticulture: Faculty Publications
Large datasets of greenhouse gas and energy surface-atmosphere fluxes measured with the eddy-covariance technique (e.g., FLUXNET2015, AmeriFlux BASE) are widely used to benchmark models and remote-sensing products. This study addresses one of the major challenges facing model-data integration: To what spatial extent do flux measurements taken at individual eddy-covariance sites reflect model- or satellite-based grid cells? We evaluate flux footprints—the temporally dynamic source areas that contribute to measured fluxes—and the representativeness of these footprints for target areas (e.g., within 250–3000 m radii around flux towers) that are often used in flux-data synthesis and modeling studies. We examine the land-cover composition …

Guidelines For The Use And Interpretation Of Assays For Monitoring Autophagy (4th Edition), Daniel Klionsky, Rodrigo Franco, Guangchao Sun, Richard Wilson, More Than 2000 Coauthors
Department of Agronomy and Horticulture: Faculty Publications
In 2008, we published the first set of guidelines for standardizing research in autophagy. Since then, this topic has received increasing attention, and many scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Thus, it is important to formulate on a regular basis updated guidelines for monitoring autophagy in different organisms. Despite numerous reviews, there continues to be confusion regarding acceptable methods to evaluate autophagy, especially in multicellular eukaryotes. Here, we present a set of guidelines for investigators to select and interpret methods to examine autophagy and related processes, and for reviewers to …
Post-Translational Modification And Degradation Mechanisms Of The Aryl Hydrocarbon Receptor, Yujie Yang
Post-Translational Modification And Degradation Mechanisms Of The Aryl Hydrocarbon Receptor, Yujie Yang
University of the Pacific Theses and Dissertations
The aryl hydrocarbon receptor (AHR) is a transcription factor first discovered to be activated by exogenous ligands, such as dioxins, and helps promote downstream gene (e.g. CYP1A1) transcription to metabolize the toxicants. With the reports of various AHR targets genes, the expression levels and activities of AHR have been implicated in many physiological and pathological situations. Understanding how AHR protein level is regulated would provide more information to target AHR. AHR stays in the cytosol in the absence of ligand in a complex with HSP90, p23 and XAP2. After ligand activation, AHR translocates into the nucleus, fulfilling its transactivation function …

Gliomas are primary brain tumors that originate from glial cells. Classification and grading of these tumors is critical to prognosis and treatment planning. The current criteria for glioma classification in central nervous system (CNS) was introduced by World Health Organization (WHO) in 2016. This criteria for glioma classification requires the integration of histology with genomics. In 2017, the Consortium to Inform Molecular and Practical Approaches to CNS Tumor Taxonomy (cIMPACT-NOW) was established to provide up-to-date recommendations for CNS tumor classification, which in turn the WHO is expected to adopt in its upcoming edition. In this work, we propose a novel …

Biofilm infections in wounds seriously delay the healing process, and methicillin-resistant Staphylococcus aureus is a major cause of wound infections. In addition to inactivating micro-organisms, low-temperature gas plasma can restore the sensitivity of pathogenic microbes to antibiotics. However, the combined treatment has not been applied to infectious diseases. In this study, low-temperature gas plasma treatment promoted the effects of different antibiotics on the reduction of S. aureus biofilms in vitro. Low-temperature gas plasma combined with rifampicin also effectively reduced the S. aureus cells in biofilms in the murine wound infection model. The blood and histochemical analysis demonstrated the biosafety of …
