Effect Of Histone H4 Tail On Nucleosome Stability And Internucleosomal Interactions, Tommy Stormberg, Sridhar Vemulapalli, Shaun Filliaux, Yuri L. Lyubchenko

Chromatin structure is dictated by nucleosome assembly and internucleosomal interactions. The tight wrapping of nucleosomes inhibits gene expression, but modifications to histone tails modulate chromatin structure, allowing for proper genetic function. The histone H4 tail is thought to play a large role in regulating chromatin structure. Here we investigated the structure of nucleosomes assembled with a tail-truncated H4 histone using Atomic Force Microscopy. We assembled tail-truncated H4 nucleosomes on DNA templates allowing for the assembly of mononucleosomes or dinucleosomes. Mononucleosomes assembled on nonspecific DNA led to decreased DNA wrapping efficiency. This effect is less pronounced for nucleosomes assembled on positioning …

Batten disease or neuronal ceroid lipofuscinosis (NCL) is a group of rare, fatal, inherited neurodegenerative lysosomal storage disorders. Numerous genes (CLN1-CLN8, CLN10-CLN14) were identified in which mutations can lead to NCL; however, the underlying pathophysiology remains elusive. Despite this, the NCLs share some of the same features and symptoms but vary in respect to severity and onset of symptoms by age. Some common symptoms include the progressive loss of vision, mental and motor deterioration, epileptic seizures, premature death, and in the rare adult-onset, dementia. The ability of healthcare providers to carefully consider all presentations of illness and injury, draw upon research and knowledge to develop a list of potential diagnoses, and further investigate systems expected to be impacted by these diagnoses, is crucial for accurate identification of a condition and subsequent treatment plans to improve the welfare of patients. When an African American male defendant tries to plea bargain an equitable justice outcome, he finds that the deep-rooted racial bias that casts African American men as dangerous, criminal and animalistic, compromises his justice rights. Plea bargaining has become the preferred process used to secure convictions for upwards of 97 percent of cases because of its efficiency. This efficiency, however, comes at a cost. This study investigated the effects blue light from self-luminous devices has on adult sleeping patterns. We investigated sleep quality and duration qualitatively with a randomized, double-blind, cross-over, placebo-controlled design with a wash-out period. Participants wore blue light blocking lenses or placebo lenses whenever they were using a self-luminous device throughout each day for one week. Each morning, participants filled out a survey to report their number of hours slept, time to fall asleep, and amount of time spent on self-luminous devices. Participants took the Pittsburgh Insomnia Rating Scale (PIRS) at the end of the one week trial (See Appendix A). …

Introduction: Lyme disease is an emerging public health threat in Ontario, Canada due to the range expansion of the tick vector, Ixodes scapularis. Tick abundance and density are important predictors of human Lyme disease risk and are typically measured using active tick surveillance (dragging). Since 1998, the United States Department of Agriculture (USDA) has allowed meat companies to increase slaughter line speeds in order to increase production of meat and poultry. The USDA grants approvals known as “line speed waivers” that bypass the requirements of current speed and inspection regulations. During the Covid-19 pandemic, slaughterhouses became epicenters of Covid-19 outbreaks, which called into question safety regulations established at these facilities. The USDA continued to grant line speed waivers despite evidence that faster line speeds result in increased worker injuries, animal welfare violations, environmental protection violations, food safety violations, and higher risk of Covid-19.

The issue of prescription drug costs has dominated the U.S. political discussion, both at the federal and local levels, as the amount individuals must pay to receive the medicines they need continues to increase. In this context, the debate over how best to determine a reasonable drug price and which existing drugs should be subject to such prices has spurred multiple legislation proposals. However, the implementation of a reasonable price mandate to control the cost of prescription drugs is not a novel idea. The treatment of transgender people while incarcerated is an issue that was catapulted into the national spotlight in 2019 when the Ninth Circuit Court of Appeals ruled in favor of providing a transgender inmate gender confirmation surgery. This decision created a circuit split within the United States regarding the medically necessary treatment a state is legally required to provide an inmate with gender dysphoria. The enzyme, 2′-3′-cyclic nucleotide 3′-phosphodiesterase (CNPase) has been known for over fifty years. Nevertheless, the roles this membrane-bound enzyme play have yet to be described completely. Recently, there has been renewed interest in the study of this enzyme due to studies that suggest that CNPase plays a role in the mediation of cellular inflammatory responses in renal and nervous system tissues. Also, this enzyme, found in oligodendrocytes of the nervous system, has been reported to participate in significant regulatory changes associated with age which may be involved in age-related CNS degeneration. When Features in a High Dimension Dataset Are Organized Hierarchically, There is an Inherent Opportunity to Reduce Dimensionality. Since More Specific Concepts Are Subsumed by More General Concepts, Subsumption Can Be Applied Successively to Reduce Dimensionality. We Tested Whether Sub-Sumption Could Reduce the Dimensionality of a Disease Dataset Without Impairing Classification Accuracy. We Started with a Dataset that Had 168 Neurological Patients, 14 Diagnoses, and 293 Unique Features. We Applied Subsumption Repeatedly to Create Eight Successively Smaller Datasets, Ranging from 293 Dimensions in the Largest Dataset to 11 Dimensions in the Smallest Dataset. The Indian Child Welfare Act (ICWA) is a federal statute that protects Indian children by keeping them connected to their families and culture. The Act’s provisions include support for family reunification, kinship care preferences, cultural competency considerations and community involvement. These provisions parallel national child welfare policies. Nevertheless, the Act is relentlessly attacked as a law that singles out Indian children for unique and harmful treatment. This is untrue but, ironically, it will be if challenges to the ICWA are successful. To prevent this from occurring, the defense of the Act needs to change. For too long, this defense has …

The monitoring and control of mammalian cell culture processes are critical for the safe and cost-effective development of biotherapeutics in drug development. Current control using off line sampling is infrequent and can miss important metabolic shifts which can impact product quality and consistency. Raman spectroscopy can provide in-line monitoring of multiple metabolites and critical process parameters to allow automation and process improvements. In this research a generic model was developed for monitoring multiple cell lines with a platform Chinese hamster ovary (CHO) cell process. Michael Barresi is Professor of Biological Sciences at Smith College, Northampton, MA, USA, where he uses the zebrafish to understand central nervous system development. Michael is also Program Director of the ‘Student Scientists’ outreach project and has made significant contributions to teaching developmental biology, including being co-author and illustrator of the textbook Developmental Biology, producing developmental documentaries and starting the Online Developmental Teaching Forums. He was awarded the 2021 Viktor Hamburger Outstanding Educator Prize from the Society of Developmental Biology (SDB). We caught up with Michael over Teams to hear more about his career and love of learning.

Maltreatment, such as physical or emotional abuse, can alter one’s later emotional regularity and responses to stimuli. To be able to study the effect of maltreatment on later stimuli response, appropriate laboratory paradigms need to be available, which is not currently true. The purpose of the study was to investigate the effect of vocal hostility stimuli on mood change as preliminary steps toward the creation of a laboratory paradigm. The Federal Extension Service (FES) of the United States Department of Agriculture (USDA) was segregated during the Jim Crow era. FES farm agents provided agricultural education and outreach; they answered questions in office, hosted meetings, and made farm visits. Agents also ran 4-H, which educated youth about agriculture through camps, and demonstrated farming activities carried out by participants. This study investigated whether segregation of services led to disparities between white and non-white (mostly African American) farm operators and families among four South Carolina regions. Background/aim: The aim of this descriptive article is to share the experience in Ege University, Turkey with favipiravir in the treatment of severe SARS-CoV-2 pneumonia. Materials and methods: This retrospective descriptive study included patients diagnosed with COVID-19 who presented with or developed severe pneumonia. Results: Forty patients who completed a full course (at least 5 days) of favipiravir were included in the study. At baseline, 30 (75%) patients required treatment for respiratory distress. Background /aim: In this study, we aimed to compare the efficacy of greater occipital nerve (GON) block alone and GON combined with supraorbital nerve (SON) block in the treatment of medication overuse headache (MOH). Material and methods: 82 patients were divided into two groups: 41 patients were administered bilateral GON block while the other 41 patients GON + SON block. Background/aim: Isolated methylmalonic acidemia (MMA) is caused by complete or partial deficiency of the enzyme methylmalonylCoA mutase (mut0 or mut- enzymatic subtype), a defect of its cofactor adenosyl-cobalamin (cblA, cblB, or cblD-MMA), or deficiency of the enzyme methylmalonyl-CoA epimerase. While onset of the disease ranges from the neonatal period to adulthood, most cases present with lethargy, vomiting and ketoacidosis in the early infancy. Major secondary complications are; growth failure, developmental delay, interstitial nephritis with progressive renal failure, basal ganglia injury and cardiomyopathy. Background/aim: The aim of this study was to compare renal and pancreatic apparent diffusion-coefficient (ADC) values of diabetic patients and control subjects and to examine their potential association with several diabetes-related clinical parameters. Materials and methods: A total of 80 sex- and age-matched patients were included in the study. Of them, 40 were patients with type 2 diabetes and 40 were nondiabetic participants. Background/aim: Operative bleeding is one of the major determinants of outcome in liver surgery. This study aimed to describe the impact of intraoperative blood loss on the postoperative course of liver resection (LR). Materials and methods: The data of 257 patients who were treated with LR between January 2007 and October 2018 were retrospectively analyzed. LRs were performed via intermittent portal triad clamping (PTC) under low central venous pressure.
