Aims/hypothesis: In vitro and rodent studies suggest that pioglitazone, a thiazolidinedione, can promote adipogenesis in adipose tissue (AT); however, there is a lack of in vivo studies in humans to support these findings. The objectives of this randomised, placebo-controlled, parallel-arm trial were to test if pioglitazone stimulates in vivo adipogenesis in the subcutaneous adipose tissue depots and if these measures were related to metabolic health outcomes in women with obesity. Methods: Forty-one healthy women with obesity (20 black; 21 white; 29 ± 6 years; BMI 32.0 ± 1.7 kg/m2; 44.0 ± 3.6% body fat) were randomised to consume 30 mg/day …

Office-based activity reduces sedentariness, yet no randomized controlled trials (RCTs) have assessed how such activity influences visceral adipose tissue (VAT). This study examined the effect of an office-based, multicomponent activity intervention on VAT. The WorkACTIVE-P RCT enrolled sedentary office workers (body mass index: 31.4 (standard deviation (SD) 4.4) kg/m2 ) to an intervention (n = 20) or control (n = 20) group. For 3 months, the intervention group received an office-based pedal desk, further to an intervention promoting its use and increased walking. The control group maintained habitual activity. At baseline and follow-up, VAT, cardiometabolic disease risk markers, physical activity, …

This study investigated the amendment of sugarcane bagasse biochar (SCBB) on soil fertility, crop yield, and nutrient loss in two different-textured soils under sugarcane (Saccharum spp.) production. Eleven megagrams per hectare of freshly incorporated biochar increased yield of plant cane by 22% in light-texture soil (LS) and 12% in heavy-textured soil (HS). Although the overall yield of the ratoon crop was lower, the biochar treatment produced 20 and 14% higher yields than the control at the LS and HS sites, respectively. Biochar increased soil carbon (Csoil) across LS and HS sites by 15% and decreased the soil C/N ratio by …

Obesity research progresses in understanding neuronal circuits and adipocyte biology to regulate metabolism. However, the interface of neuro-adipocyte interaction is less studied. We summarize the current knowledge of adipose tissue innervation and interaction with adipocytes and emphasize adipocyte transitions from white to brown adipocytes and vice versa. We further highlight emerging concepts for the differential neuronal regulation of brown/beige versus white adipocyte and the interdependence of both for metabolic regulation.

Glucocorticoids are clinically essential drugs used routinely to control inflammation. However, a host of metabolic side effects manifests upon usage beyond a few days. In the present study, we tested the hypothesis that seven-in-absentia mammalian homolog-2 (SIAH2), a ubiquitin ligase that regulates adipogenesis, is important for controlling adipocyte size, inflammation, and the ability of adipose tissue to expand in response to a glucocorticoid challenge. Using mice with global deletion of SIAH2 exposed or not to corticosterone, we found that adipocytes are larger in response to glucocorticoids in the absence of SIAH2. In addition, SIAH2 regulates glucocorticoid receptor (GR) transcriptional activity …

The inflammatory response is an obstacle to success in both allogeneic and autologous islet transplantation. In autologous islet transplantation (AIT), however, the recipient is also the donor, permitting pretreatment of donor/recipient for a controlled duration prior to transplantation. We sought to exploit this feature of (AIT) by pretreating donor/recipients with chronic pancreatitis undergoing total pancreatectomy and autologous islet transplantation (TPAIT) to test the hypothesis that peri-transplant treatment with the FDA-approved anti-inflammatory hydroxychloroquine (HCQ) improves graft function. In this randomized placebo-controlled pilot clinical study, patients (n = 6) were treated with oral HCQ for 30 days prior to and 90 days …

Dictionary learning and sparse coding techniques overcome limitations of traditional voxel-level analyses of task-based functional magnetic resonance imaging (fMRI) data by identifying broader temporal and spatial patterns of brain activity. However, prior applications of these methods to task-related fMRI data are not simultaneously optimized to find temporal patterns of activity that change in concert with changes in task conditions and spatial patterns that leverage existing neuroscience knowledge. In this study we present a new sparse dictionary learning method that uses prior knowledge of the temporal pattern of task conditions and the locations of brain regions hypothesized to be involved in …

Declining estrogen levels before, during, and after menopause can affect memory and risk for Alzheimer's disease. Undesirable side effects of hormone variations emphasize a role for hormone therapy (HT) where possible benefits include a delay in the onset of dementia—yet findings are inconsistent. Effects of HT may be mediated by estrogen receptors found throughout the brain. Effects may also depend on lifestyle factors, timing of use, and genetic risk. We studied the impact of self-reported HT use on brain volume in 562 elderly women (71–94 years) with mixed cognitive status while adjusting for aforementioned factors. Covariate-adjusted voxelwise linear regression analyses …

Woodrats (genus Neotoma) comprise 24 species found primarily in the United States and México. The Neotoma mexicana species group reaches its southernmost distribution in the highlands of southern México and Central America. Previous research suggested that N. mexi-cana has a discontinuous distribution, whereas N. ferruginea and N. picta have allopatric distributions around the lowlands of the Isthmus of Tehuantepec. However, these hypotheses were suggested with incomplete subspecific sampling near the isthmus. We used samples of N. m. parvidens from the Sierra Sur de Oaxaca and N. m. tropicalis from the Sierra Norte de Oaxaca to assess their taxonomic affinity. Our …

Nutrient stoichiometry and trade-offs focusing on the mass balance of multiple nutrient elements play important roles in the response mechanism of plants to environmental stresses in wetlands. Nonetheless, little is known about the response dynamics and trade-offs of nutrient elements of wetland plants, especially potassium (K), under hydrological fluctuation conditions. Greenhouse experiments have been undertaken to assess the effects of initial water level (DW), water-level amplitude (AW), lasted time and their interactions on the response processes of nutrient stoichiometry of Carex schmidtii. Plant nutrient trade-offs and limitation were investigated under multiple hydrological fluctuation conditions. Results showed that plant nitrogen (N), …

Modern whole-genome prediction (WGP) frameworks that focus on multi-environment trials (MET) integrate large-scale genomics, phenomics, and envirotyping data. However, the more complex the statistical model, the longer the computational processing times, which do not always result in accuracy gains. We investigated the use of new kernel methods and modeling structures involving genomics and nongenomic sources of variation in two MET maize data sets. Five WGP models were considered, advancing in complexity from a main-effect additive model (A) to more complex structures, including dominance deviations (D), genotype × environment interaction (AE and DE), and the reaction-norm model using environmental covariables (W) …

Study On The Human Gut Bacteria Under Different Ecological Conditions: Antibiotic Perturbation And Dietary Quercetin, Shruti Shashidharan Menon
Electronic Theses and Dissertations
The human intestine encompasses a vast community of microorganisms known as the gut microbiota that play a crucial role in maintaining health. Common perturbations such as changes in the normal diet, antibiotic treatment, and changes in environmental conditions can alter the gut microbiome. This can create dysbiosis in the gut leading to disease conditions. Therefore, it becomes important to determine the forces that influence the gut microbial ecology. In the first study, we focus on antibiotic perturbations on microbial succession and resilience in a synthetic consortium consisting of the most prevalent gut bacteria in humans. In addition, we investigated the …

Analysis Of Metabolites And Therapeutics For Toxic Inhaled Agent Exposure, Abigail Bemah Donkor
Electronic Theses and Dissertations
The inhalation of toxic inhaled agents (TIAs) such as methyl isocyanate and cyanide can compromise an individual’s general health due to their extremely toxic nature. Nonetheless, these gases are used extensively in the electronics, chemical, welding, electroplating industries, etc., increasing the risk of exposure to both civilians and industrial workers. It is therefore very crucial to develop analytical methods for TIA metabolites and therapeutic agents. Methyl isocyanate (MIC), an intermediate in the synthesis of carbamate pesticides is a toxic industrial chemical that causes irritation and damage to the eyes, respiratory tracts, and skin. Due to its reactivity with proteins, protein …

Quantifying The Benefits Of The Expanded Food And Nutrition Education Program (Efnep) Using Biomarkers For Chronic Disease Risk, Richard Acquah-Sarpong
Electronic Theses and Dissertations
The Expanded Food and Nutrition Education Program (EFNEP) is among the major nutrition education programs funded by the United States Department of Agriculture (USDA) with the aim of reducing food insecurity among low-income families. The program reaches about 70,000 adults and youth of low-income families in the US, District of Columbia, and six U.S. territories. Prior studies have used self-reported data, which possesses measurement errors, to estimate the benefits of the program. This can lead to underestimation or overestimation of results. To address this limitation, I use clinically measured objective biomarkers, such as body mass index (BMI), blood sugar level …
Comparison Of Software Packages For Detecting Differentially Expressed Genes From Single-Sample Rna-Seq Data, Rong Zhou
Electronic Theses and Dissertations
RNA-sequencing (RNA-seq) has rapidly become the tool in many genome-wide transcriptomic studies. It provides a way to understand the RNA environment of cells in different physiological or pathological states to determine how cells respond to these changes. RNA-seq provides quantitative information about the abundance of different RNA species present in a given sample. If the difference or change observed in the read counts or expression level between two experimental conditions is statistically significant, the gene is declared as differentially expressed. A large number of methods for detecting differentially expressed genes (DEGs) with RNA-seq have been developed, such as the methods …

An autonomous household robot passed a self-awareness test in 2015, proving that the cognitive capabilities of robots are heading towards those of humans. While this is a milestone in AI, it raises questions about legal implications. If robots are progressively developing cognition, it is important to discuss whether they are entitled to justice pursuant to conventional notions of human rights. This paper offers a comprehensive discussion of this complex question through cross-disciplinary scholarly sources from computer science, ethics, and law. The computer science perspective dissects hardware and software of robots to unveil whether human behavior can be efficiently replicated. The …

Interdisciplinary Stem Undergraduate Programs And The Effectiveness Of Computing Competencies Within The Curriculum, Katherine G. Herbert, Thomas J. Marlowe, Kees Leune, Robert M. Siegfried, Jeanette Wilmanski
Department of Computer Science Faculty Scholarship and Creative Works
Undergraduate interdisciplinary, multidisciplinary and transdisciplinary computing-related STEM programs are proliferating extensively. Each of these programs requires a robust computing component to be integrated into the curriculum. However, including an introductory, programmingoriented sequence designed for CS majors is not always the best fit for these multifaceted programs. In this work in progress paper, we set out to investigate possibilities and issues for the computing component, focusing on three fields: bioinformatics, data science, and cybersecurity. We explore commonalities and differences, and discuss initial plans to test our hypotheses. In doing so, we consider data organization, data acquisition, and a preliminary survey design.
