Obesity is primarily a consequence of consuming calories beyond energetic requirements, but underpinning drivers have not been fully defined. 5-Hydroxytryptamine (5-HT) neurons in the dorsal Raphe nucleus (5-HTDRN) regulate different types of feeding behavior, such as eating to cope with hunger or for pleasure. Here, we observed that activation of 5-HTDRN to hypothalamic arcuate nucleus (5-HTDRN → ARH) projections inhibits food intake driven by hunger via actions at ARH 5-HT2C and 5-HT1B receptors, whereas activation of 5-HTDRN to ventral tegmental area (5-HTDRN → VTA) projections inhibits non-hunger-driven feeding via actions at 5-HT2C receptors. Background: The tomato psyllid, Bactericera cockerelli Šulc (Hemiptera: Triozidae), is a pest of solanaceous crops such as tomato (Solanum lycopersicum L.) in the U.S. and vectors the disease-causing pathogen ‘Candidatus Liberibacter solanacearum’. Currently, the only effective strategies for controlling the diseases associated with this pathogen involve regular pesticide applications to manage psyllid population density. However, such practices are unsustainable and will eventually lead to widespread pesticide resistance in psyllids. Therefore, new control strategies must be developed to increase host-plant resistance to insect vectors. Pain, the most common symptom reported among patients in the primary care setting, is complex to manage. Opioids are among the most potent analgesics agents for managing pain. Since the mid-1990s, the number of opioid prescriptions for the management of chronic non-cancer pain (CNCP) has increased by more than 400%, and this increased availability has significantly contributed to opioid diversion, overdose, tolerance, dependence, and addiction. Excessive inflammation drives the progression from sepsis to septic shock. Macrophage migration inhibitory factor (MIF) is of interest because MIF promoter polymorphisms predict mortality in different infections, and anti-MIF antibody improves survival in experimental models when administered 8 hours after infectious insult. The recent description of a second MIF superfamily member, D-dopachrome tautomerase (D-DT/MIF-2), prompted closer investigation of MIF-dependent responses. We subjected Mif-/- and Mif-2-/- mice to polymicrobial sepsis and observed a survival benefit with Mif but not Mif-2 deficiency. The mechanisms that underlie the extensive phenotypic diversity in genetic disorders are poorly understood. Here, we develop a large-scale assay to characterize the functional valence (gain or loss-of-function) of missense variants identified in UBE3A, the gene whose loss-of-function causes the neurodevelopmental disorder Angelman syndrome. We identify numerous gain-of-function variants including a hyperactivating Q588E mutation that strikingly increases UBE3A activity above wild-type UBE3A levels. Mice carrying the Q588E mutation exhibit aberrant early-life motor and communication deficits, and individuals possessing hyperactivating UBE3A variants exhibit affected phenotypes that are distinguishable from Angelman syndrome. Global freshwater biodiversity is declining dramatically, and meeting the challenges of this crisis requires bold goals and the mobilisation of substantial resources. While the reasons are varied, investments in both research and conservation of freshwater biodiversity lag far behind those in the terrestrial and marine realms. Inspired by a global consultation, we identify 15 pressing priority needs, grouped into five research areas, in an effort to support informed stewardship of freshwater biodiversity. The proposed agenda aims to advance freshwater biodiversity research globally as a critical step in improving coordinated actions towards its sustainable management and conservation.

Problem-solving skills in the field of biology play an important role in representing problems, identifying relationships, making predictions, and verifying or evaluating solutions. This study aimed to implement an augmented reality-assisted problem-based learning e-module on bioindicator materials to improve students’ problem-solving skills. The research method used was a pre-experimental one-group pretest-posttest design involving 29 biology students at the State University of Malang in the 2020/2021 academic year. The results of the paired t-test showed that the significance value (p) for the pretest and posttest was 0.000 (< 0.05), indicating that the e-module had a significant effect on improving students’ problem-solving skills.

Particulate hexavalent chromium [Cr(VI)] is a well-established human lung carcinogen. RAD51, a key protein in homologous recombination repair pathway, is inhibited after prolonged exposure to Cr(VI), leading to an increase in chromosome instability after prolonged exposures in human lung fibroblasts. chromosome instability is the proposed driver of Cr(VI) carcinogenesis. Since tumors from chromate workers develop from epithelial cells, we sought to translate these findings from human bronchial fibroblasts to human bronchial epithelial cells. We hypothesized Cr(VI) inhibits RAD51 after prolonged exposure leading to an increase in chromosome instability in human bronchial epithelial cells (BEP2D). Part-time will center around a 25-year-old medical school dropout starting a new job at a neighborhood golf clubhouse and gym. This coming-of-age narrative follows Danny as he struggles with his sexual identity, student debt, and job outlook after returning to the privileged community he grew up in. This study investigates how African Americans perceive the racialization process in the criminal justice system following the death of Breonna Taylor. Focusing primarily on police-community relations in Louisville, KY, I find the Breonna Taylor case amplifies preconceived perceptions of race between civilian and non-civilian groups. To test the hypothesis that racialization in the criminal justice system has caused damaging effects on relations between African Americans and police, two virtual focus groups were designed to discuss participants’ experiences with law enforcement. This phenomenological study examined the importance of otherparenting performed by Black student affairs professionals at a predominantly White research-intensive institution in the mid-South. Otherparenting is a U.S. tradition that has West African roots in chattel slavery. Otherparenting is defined as those that assist blood mothers and fathers by sharing parenting responsibilities. Semi-structured interviews were conducted with seven participants across various functional units at the institution. This macro-level study examines the effects of social disorganization predictors on violent crime registered in nonmetropolitan areas of Kentucky from 2012 to 2016. The “human footprint” can be used as a general proxy to estimate human activities across the landscape. The human footprint in the Zambezi Region of Namibia is critically important for regional management of conservation efforts and land use planning. The land covers in the Zambezi Region are characteristically difficult to separate spectrally, due to a highly heterogeneous savanna landscape. Voice is a concept that is both highly sought after and elusive in education. While schools aim to foster students’ voices, many academic structures inadvertently conceal their voices and in turn their identities. Definitions of voice have been assumed, vague, or looked at as a writing trait, with little consideration of voices’ dynamic and mediated structures. Drawing on scholarship grounded in sociocultural theories and dialogism (e.g., Bakhtin, 1986; Engeström; 1987, Leont’ev, 1981; Rosenblatt, 1978; Vygotsky, 1978), I contribute a new, tangible definition of voice, in which voice is a dynamic happening, continually negotiated and constructed. Water is an integral and necessary component of life. It is then, exceedingly remarkable that some species are capable of surviving virtually complete water loss for extended periods of time. Several decades of intense research into anhydrobiosis, or life without water, have given significant insights into the molecular mechanisms governing this phenomenon. Griffithsin (GRFT) is a carbohydrate binding agent (lectin) that was originally identified in the red alga Griffithsia sp. Q-Griffithsin (Q-GRFT) is an oxidation stable analog of GRFT. GRFT has demonstrated inhibitory activity against HIV-1, Coronaviruses, Hepatitis C, influenza and Ebola viruses. The broad-spectrum activity suggests the potential utility of this lectin in a wide range of viral infections. However, the lectin’s activity in mucosal infections has not been extensively studied. This dissertation explores the development and benefits of familial relationships as the primary feature of Afrocentricity at Black Scholars Academy (BSA), a Pre-Kindergarten (Pre-K) through 12th-grade Black homeschool collective in the United States. This qualitative study draws upon Afrocentricity as a theoretical framework to analyze and interpret data collected from classroom observations, individual interviews with current and former teachers and students, and textual artifacts between July and November 2019. Familial relationships—an African time orientation, a personalized learning plan, OurStory, and Rising Meeting—are present in BSA. The vitamin B12 derivates, otherwise known as cobalamin (Cbl), are ubiquitous organometallic cofactors. The biologically active forms of Cbl, such as methylcobalamin (MeCbl) and adenosylcobalamin (AdoCbl), act as cofactors in different physiological reactions for both prokaryotes and eukaryotes. A crucial aspect of the Cbl-mediated systems is the activation of the organometallic Co-C bond that plays a critical role in its catalytic activity. This dissertation is a discursive analysis of the decision in the Dred Scott v Sandford, 1857 case written by Chief Justice Roger Taney. It begins with an overview of the literature on performative speech acts, focusing on the aspects of performatives that relate to Louis Miron and Jonathan Xavier Inda’s thesis that race is a performative speech act. Breaking from their use of race as the analytic, this analysis is situated within a black/nonblack paradigm. Latinx caregivers underutilize traditional youth mental health services (MHS) relative to their White counterparts. Disparities may be explained by barriers preventing engagement in traditional MHS. A potential mismatch between available services and preferred culturally appropriate healing practices may also contribute to treatment disparities. The current study examined Latinx caregivers’ perceived need for and utilization of support services in relation to family characteristics, youth problem-type, and barriers to care. Findings were contextualized by assessing caregiver preferences for addressing youth psychopathology. The study consisted of 598 Latinx caregivers from across the United States. Turtles are perhaps best known for the bony shells that encase them, a unique morphological trait that provides protection against predators. Many taxa have even evolved the ability to enclose themselves using hinges that can be used to create a seal between the carapace and plastron. I measured the hinge closing force of Ornate Box Turtles (Terrapene ornata) to assess the performance of this unusual yet ecologically important trait. Research has shown that bisexual individuals experience poorer mental health outcomes, resulting in depression, anxiety, substance use, and suicidal ideation, than their gay or straight counterparts (Calzo, Antonucci, Mays, & Cochran, 2011; Ross, Dobinson, & Eady, 2010; Taylor, Power, & Smith, 2020). Object: The present study examines the clinical utility of combining a cognitive screener, Montreal Cognitive Assessment (MoCA), with a measure of adaptive functioning, Texas Functional Living Scale (TFLS), to diagnose memory disorders when differentiating between cognition within normal limits (WNL), mild cognitive impairment (MCI), and Alzheimer’s disease (AD). Method: A total of 207 Health First Memory Disorder Clinic patients, ages 64-94, were included in the study. Participants were screened using the MoCA and then they completed a brief neuropsychological evaluation, which included the TFLS. Reactive chlorine species (RCS) - hypochlorous acid (HOCl), hypothiocyanous acid (HOSCN), and chloramines - are powerful antimicrobial oxidants generated by the innate immune system. Bacteria developed defense mechanisms against RCS, which are highly toxic for invading pathogens. One such defense system involves RclA, an enzyme from the flavin disulfide reductase family, which is massively upregulated upon exposure of Escherichia coli to RCS. Recent research suggests that RclA functions as a Cu(II) reductase to provide resistance against RCS. However, the exact mechanism used by RclA was subject to debate.
