The Influence Of Post-Visit Emotions On Destination Loyalty, Maksim Godovykh, Asli D. A. Tasci
Faculty Scholarship and Creative Works
Purpose: The influence of different factors including emotional states on loyalty has been previously discussed in the literature. However, the influence of post-visit emotions evoked by emotional stimuli on tourist loyalty lacks empirical attention. The purpose of this study is to investigate the effects of post-visit emotional stimuli on destination loyalty.
Design/methodology/approach: The study applied an online scenario-based experimental design to identify the impact of positive and negative affective pictorial stimuli on destination loyalty. A sample of 500 adult US residents who visited Orlando within the past 12 months was recruited to take part in the experiment. One-way ANOVA was …

Antecedents Of Borderline Personality Disorder And Antisocial Personality Disorder: An Examination Of Gene X Environment Interactions, Amy L. Medina
Dissertations, Theses, and Capstone Projects
Current thinking suggests that genotypes associated with impulse-control disorders and negative emotionality, such as monoamine oxidase-a (MAOA), interact with negative early environmental factors like childhood maltreatment and develop into the disorders know as Borderline Personality Disorder (BPD) and Antisocial Personality Disorder (ASPD). Using existing data from a prospective cohort design study of the consequences of child abuse and neglect, participants (N = 896 represent individuals with documented histories of child abuse and neglect and a matched comparison group that were followed up into adulthood and interviewed. A subsample of 631 participants gave permission for DNA extraction and analyses during …

Mapping Seagrass Habitat And Characterising Dugong Feeding Preferences In Sibu-Tinggi Archipelago, Johor, Heng Wei Khang
Mapping Seagrass Habitat And Characterising Dugong Feeding Preferences In Sibu-Tinggi Archipelago, Johor, Heng Wei Khang
Student Works (2020-2029)
The dugong (Dugong dugon) is a marine mega-herbivore that is a seagrass community specialist. Seagrass meadows are eminently essential to dugongs as their main feeding grounds. The species’ feeding behavioural patterns are often associated with the features of the seagrass habitat. Elucidating such behavioural patterns is vital for understanding how a designated space is used. In Malaysia, the Sibu-Tinggi Archipelago, is regarded as one of the most important habitats for dugongs, hosting a significant local population due to the presence of extensive subtidal seagrass meadows. Field surveys were conducted in 2016 – 2018, across three seasons influenced by the northeast …

Learning To Pre-Train Graph Neural Networks, Yuanfu Lu, Xunqiang Jiang, Yuan Fang, Chuan Shi
Research Collection School Of Computing and Information Systems
Graph neural networks (GNNs) have become the de facto standard for representation learning on graphs, which derive effective node representations by recursively aggregating information from graph neighborhoods. While GNNs can be trained from scratch, pre-training GNNs to learn transferable knowledge for downstream tasks has recently been demonstrated to improve the state of the art. However, conventional GNN pre-training methods follow a two-step paradigm: 1) pre-training on abundant unlabeled data and 2) fine-tuning on downstream labeled data, between which there exists a significant gap due to the divergence of optimization objectives in the two steps. In this paper, we conduct an …

The Meaning Of “Phenomenology”: Qualitative And Philosophical Phenomenological Research Methods, Heath Williams
The Qualitative Report
I show some problems with recent discussions within qualitative research that centre around the “authenticity” of phenomenological research methods. I argue that attempts to restrict the scope of the term “phenomenology” via reference to the phenomenological philosophy of Husserl are misguided, because the meaning of the term “phenomenology” is only broadly restricted by etymology. My argument has two prongs: first, via a discussion of Husserl, I show that the canonical phenomenological tradition gives rise to many traits of contemporary qualitative phenomenological theory that are purportedly insufficiently genuine (such as characterisations of phenomenology as “what-its-likeness” and presuppositionless description). Second, I argue …

Intracellular Ca2+ level is under strict regulation through calcium channels and storage pools including the endoplasmic reticulum (ER). Mutations in certain ion channel subunits, which cause mis-regulated Ca2+ influx, induce the excitotoxic necrosis of neurons. In the nematode Caenorhabditis elegans, dominant mutations in the DEG/ENaC sodium channel subunit MEC-4 induce six mechanosensory (touch) neurons to undergo excitotoxic necrosis. These necrotic neurons are subsequently engulfed and digested by neighboring hypodermal cells. We previously reported that necrotic touch neurons actively expose phosphatidylserine (PS), an "eat-me" signal, to attract engulfing cells. However, the upstream signal that triggers PS externalization remained elusive. Here we …
Cyclic Nucleotide Selectivity Of Protein Kinase G Isozymes, Choel Kim, Rajesh Sharma
Cyclic Nucleotide Selectivity Of Protein Kinase G Isozymes, Choel Kim, Rajesh Sharma
Faculty, Staff and Students Publications
The intrinsic activity of the C-terminal catalytic (C) domain of cyclic guanosine monophosphate (cGMP)-dependent protein kinases (PKG) is inhibited by interactions with the N-terminal regulatory (R) domain. Selective binding of cGMP to cyclic nucleotide binding (CNB) domains within the R-domain disrupts the inhibitory R-C interaction, leading to the release and activation of the C-domain. Affinity measurements of mammalian and plasmodium PKG CNB domains reveal different degrees of cyclic nucleotide affinity and selectivity; the CNB domains adjacent to the C-domain are more cGMP selective and therefore critical for cGMP-dependent activation. Crystal structures of isolated CNB domains in the presence and absence …

In recent years, it has become apparent that the gut microbiome can influence the functioning and pathological states of organs and systems throughout the body. In this study, we tested the hypothesis that the gut microbiome has a major role in the disruption of the blood-brain barrier (BBB) in the spontaneously hypertensive stroke prone rats (SHRSP), an animal model for hypertensive cerebral small vessel disease (CSVD). Loss of BBB is thought to be an early and initiating component to the full expression of CSVD in animal models and humans. To test this hypothesis, newly born SHRSP pups were placed with …

Traumatic brain injury (TBI) continues to be a major healthcare problem and there is much to be explored regarding the secondary pathobiology to identify early predictive markers and new therapeutic targets. While documented changes in thrombosis and inflammation in major trauma have been well described, growing evidence suggests that isolated TBI also results in systemic alterations in these mechanisms. Here, we review recent experimental and clinical findings that demonstrate how blood-brain barrier dysfunction, systemic immune response, inflammation, platelet activation, and thrombosis contribute significantly to the pathogenesis of TBI. Despite advances in the links between thrombosis and inflammation, there is a …

We currently find ourselves in the midst of a global coronavirus disease 2019 (COVID-19) pandemic, caused by the highly infectious novel coronavirus, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Here, we discuss aspects of SARS-CoV-2 biology and pathology and how these might interact with the circadian clock of the host. We further focus on the severe manifestation of the illness, leading to hospitalization in an intensive care unit. The most common severe complications of COVID-19 relate to clock-regulated human physiology. We speculate on how the pandemic might be used to gain insights on the circadian clock but, more importantly, on …

Proprioception provides crucial information necessary for determining limb position and movement, and plausibly also for updating internal models that might underlie the control of movement and posture. Seminal studies of upper-limb movements in individuals living with chronic, large fiber deafferentation have provided evidence for the role of proprioceptive information in the hypothetical formation and maintenance of internal models to produce accurate motor commands. Vision also contributes to sensorimotor functions but cannot fully compensate for proprioceptive deficits. More recent work has shown that posture and movement control processes are lateralized in the brain, and that proprioception plays a fundamental role in …

Modified DNA aptamers incorporated with amino-acid like side chains or drug-like ligands can offer unique advantages and enhance specificity as affinity ligands. Thy-1 membrane glycoprotein (THY1 or CD90) was previously identified as a biomarker candidate of neovasculature in pancreatic ductal adenocarcinoma (PDAC). The current study developed and evaluated modified DNA X-aptamers targeting THY1 in PDAC. The expression and glycosylation of THY1 in PDAC tumor tissues were assessed using immunohistochemistry and quantitative proteomics. Bead-based X-aptamer library that contains 108 different sequences was used to screen for high affinity THY1 X-aptamers. The sequences of the X-aptamers were analyzed with the next-generation sequencing. …

Do Aquatic Ectotherms Perform Better Under Hypoxia After Warm Acclimation?, Michael Collins, Manuela Truebano, Wilco C.E.P. Verberk, John I. Spicer
School of Biological and Marine Sciences
ABSTRACTAquatic animals increasingly encounter environmental hypoxia due to climate-related warming and/or eutrophication. Although acute warming typically reduces performance under hypoxia, the ability of organisms to modulate hypoxic performance via thermal acclimation is less understood. Here, we review the literature and ask whether hypoxic performance of aquatic ectotherms improves following warm acclimation. Interpretation of thermal acclimation effects is limited by reliance on data from experiments that are not designed to directly test for beneficial or detrimental effects on hypoxic performance. Most studies have tested hypoxic responses exclusively at test temperatures matching organisms' acclimation temperatures, precluding the possibility of distinguishing between acclimation …

Immune dysfunction contributes to the higher risk of communicable and non-communicable diseases among diabetics. HLA-DR expression is a robust marker of immune competence in mononuclear cells, including antigen presentation to CD4 lymphocytes. Given the high prevalence of obesity among diabetics, we evaluated the independent association between hyperglycemia and dyslipidemias with respect to HLA-DR expression in blood monocytes from type 2 diabetes patients. The monocytes from individuals with (n=16) or without diabetes (n=25) were phenotyped by flow cytometry to assess the differential expression of HLA-DR on their three subpopulations (classical, intermediate and non-classical monocytes). Diabetes was independently associated with lower HLA-DR …

During drug development, evaluation of drug and its metabolite is an essential process to understand drug activity, stability, toxicity and distribution. Liquid chromatography (LC) coupled with mass spectrometry (MS) has become the standard analytical tool for screening and identifying drug metabolites. Unlike LC/MS approach requiring liquifying the biological samples, we showed that spectral imaging (or spectral microscopy) could provide high-resolution images of doxorubicin (dox) and its metabolite doxorubicinol (dox’ol) in single living cells. Using this new method, we performed measurements without destroying the biological samples. We calculated the rate constant of dox translocating from extracellular moiety into the cell and …

Adoption of native warm-season grasses (NWSGs) in the tall fescue belt is limited despite studies documenting the potential contribution of these forages to profitable beef production. On the basis of two surveys conducted in Tennessee, a survey of beef producers and a survey of agricultural professionals, we evaluated perceptions of NWSG forages and how those perceptions could influence their adoption. Although agricultural professionals were more familiar with NWSGs than producers, both populations had limited knowledge regarding these forages, indicating that additional Extension education is needed. Our results provide useful guidance for developing NWSG forage educational programs for producers and agricultural …

This story is an extension of the paper "Mathematics and Voter Choice" published in this same issue of the Journal of Humanistic Mathematics. It explores what elections and politics might look like from the voter perspective if some of the ideas from the paper were implemented. The story is also an attempt to highlight how mathematics and data science are done, much the way a colleague of mine refers to shows like CSI as recruiting tools that use "dramatic pipetting" in the labs to show day-to-day science in action. The story is, as you will no doubt see, heavily …
