Background: Indices obtained from lymph node dissection specimens, specifically lymph node yield (LNY) and lymph node ratio (LNR), have prognostic significance in the setting of head and neck squamous cell carcinoma (HNSCCa). However, there are currently no validated tools to estimate adequacy of planned lymph node dissection using preoperative data. The present study sought to evaluate CT-derived estimates of lymphatic tissue volumes as a preoperative tool to guide cervical node dissection. Methods: Fifteen cervical lymph node dissections were performed in 14 subjects with HNSCCa. Preoperative CT-derived estimates of lymphatic tissue volumes were compared with gross pathology tissue volume estimates and …

A Distributed Fmri-Based Signature For The Subjective Experience Of Fear, Feng Zhou, Weihua Zhao, Ziyu Qi, Yayuan Geng, Shuxia Yao, Keith M. Kendrick, Tor D. Wager, Benjamin Becker
Dartmouth Scholarship
The specific neural systems underlying the subjective feeling of fear are debated in affective neuroscience. Here, we combine functional MRI with machine learning to identify and evaluate a sensitive and generalizable neural signature predictive of the momentary self-reported subjective fear experience across discovery (n = 67), validation (n = 20) and generalization (n = 31) cohorts. We systematically demonstrate that accurate fear prediction crucially requires distributed brain systems, with important contributions from cortical (e.g., prefrontal, midcingulate and insular cortices) and subcortical (e.g., thalamus, periaqueductal gray, basal forebrain and amygdala) regions. We further demonstrate that the neural representation of subjective fear …

Color vision is used throughout medicine to interpret the health and status of tissue. Ionizing radiation used in radiation therapy produces broadband white light inside tissue through the Cherenkov effect, and this light is attenuated by tissue features as it leaves the body. In this study, a novel time-gated three-channel camera was developed for the first time and was used to image color Cherenkov emission coming from patients during treatment. The spectral content was interpreted by comparison with imaging calibrated tissue phantoms. Color shades of Cherenkov emission in radiotherapy can be used to interpret tissue blood volume, oxygen saturation and …

Background: The infant intestinal microbiome plays an important role in metabolism and immune development with impacts on lifelong health. The linkage between the taxonomic composition of the microbiome and its metabolic phenotype is undefined and complicated by redundancies in the taxon-function relationship within microbial communities. To inform a more mechanistic understanding of the relationship between the microbiome and health, we performed an integrative statistical and machine learning-based analysis of microbe taxonomic structure and metabolic function in order to characterize the taxa-function relationship in early life. Results: Stool samples collected from infants enrolled in the New Hampshire Birth Cohort Study (NHBCS) …

Background: Depression in children presents a significant health burden to society and often co-exists with chronic illnesses, such as human immunodeficiency virus (HIV). Research has demonstrated that 10–37% of children and adolescents living with HIV also suffer from depression. Low-and-middle income countries (LMICs) shoulder a disproportionate burden of HIV among other health challenges, but reliable estimates of co-morbid depression are lacking in these settings. Prior studies in Rwanda, a LMIC of 12 million people in East Africa, found that 25% of children living with HIV met criteria for depression. Though depression may negatively affect adherence to HIV treatment among children …

Background: Surface plasmon resonance is a label-free biophysical technique that is widely used in investigating biomolecular interactions, including protein-protein, protein-DNA, and protein-small molecule binding. Surface plasmon resonance is a very powerful tool in different stages of small molecule drug development and antibody characterization. Both academic institutions and pharmaceutical industry extensively utilize this method for screening and validation studies involving direct molecular interactions. In most applications of the surface plasmon resonance technology, one of the studied molecules is immobilized on a microchip, while the second molecule is delivered through a microfluidic system over the immobilized molecules. Changes in total mass on …

Recent studies have demonstrated that drought leads to dramatic, highly conserved shifts in the root microbiome. At present, the molecular mechanisms underlying these responses remain largely uncharacterized. Here we employ genome-resolved metagenomics and comparative genomics to demonstrate that carbohydrate and secondary metabolite transport functionalities are overrepresented within drought-enriched taxa. These data also reveal that bacterial iron transport and metabolism functionality is highly correlated with drought enrichment. Using time-series root RNA-Seq data, we demonstrate that iron homeostasis within the root is impacted by drought stress, and that loss of a plant phytosiderophore iron transporter impacts microbial community composition, leading to significant …

Prenatal Exposure To Per- And Polyfluoroalkyl Substances And Child Neurodevelopment, Jiwon Oh
Prenatal Exposure To Per- And Polyfluoroalkyl Substances And Child Neurodevelopment, Jiwon Oh
Earth & Environmental Sciences Dissertations - Archive
Pregnant women are exposed to per- and polyfluoroalkyl substances (PFAS) primarily via ingestion of contaminated food, water, and house dust. PFAS in pregnant women can be transferred to their child through the placenta during pregnancy. Many long-chain PFAS have been frequently detected in blood of pregnant women, amniotic fluid, and umbilical cord blood for the last few decades. Prenatal exposure to PFAS has shown potential to adversely affect offspring’s neurodevelopment in laboratory animals. However, epidemiological evidence on associations between prenatal PFAS exposure and child neurodevelopment remains inconsistent. My dissertation research consists of four sub-studies whose objectives are (1) to examine …
Hplc Analysis Of Potentially Counterfeit Samples Of Amoxicillin, Hunter Noel
Hplc Analysis Of Potentially Counterfeit Samples Of Amoxicillin, Hunter Noel
Undergraduate Honors Theses
The global phenomenon of drug counterfeiting is a threat to both patients and healthcare systems alike. Counterfeit drugs can build bacterial resistance to antibiotics, undermine public trust in prescribed medications, fail to alleviate sickness, or even hurt the patient. Annually, thousands of people die from ingesting fake drugs (Blackstone et al., 2014). In order to combat the global prevalence of drug counterfeiting, it is important to test suspect drugs for legitimacy and then report findings back to relevant authorities that can enforce drug quality standards (Lieberman, 2018). High performance liquid chromatography (HPLC) is an analytical method that can determine the …

Founded in 1993, the Yellowstone to Yukon (Y2Y) vision was one of the earliest large-landscape conservation visions. Despite growing recognition of large-landscape conservation strategies, there have been few tests to date of conservation gains achieved through such approaches. We tested for conservation gains in the Y2Y region of North America following initiation of the Y2Y conservation vision in 1993 using a counterfactual spatiotemporal comparison and tracking change in five different conservation metrics. First, we enumerated the area of land within Y2Y in designated protected areas. We then compared the rate of change of protected area growth before- and after-initiation of …

Development Of Bioaffinity Based Assays For Forensic Applications, Juliana Maria Agudelo Cano
Legacy Theses & Dissertations (2009 - 2024)
In the field of forensic science, body fluids are a major contributor of evidence in investigations. However, body fluids are usually used only for their genetic content, overlooking its unique composition of proteins and low-molecular-weight compounds that often serve as biomarkers in clinical diagnostics. This investigation aimed to develop simple, effective, and economical models that can be used to identify different attributes of the originator. The focus of this research was to develop biocatalytic cascades that could analyze body fluids by using UV-Vis spectroscopy. These models could be implemented in forensic settings, as well as in biometric authentications. The central …

The Assemblage Of The Dead : Speech, Subjectivity, And Being Human, Nazia Manzoor
Legacy Theses & Dissertations (2009 - 2024)
ABSTRACT“The Assemblage of the Dead: Speech, Subjectivity, and Being Human” reimagines the construct of the human as a political subject of the state through a bio(necro)political lens. The project re-envisions the conceptual framework of biopolitics through an engagement with the figure of the living dead, centralized in Giorgio Agamben’s work through his casting of the figure of the Muselmann as the cipher that reveals the limits of humanity and being human. With a bid to counter-narrate the twinning of death and resistance and death and subjectivity as foundational markers of humanity in current critical scholarship of the field, this project …

Modulation Of Rho Termination : Expectation Vs Reality, Kavya Sri Sai Chegireddy
Legacy Theses & Dissertations (2009 - 2024)
Transcription termination factor Rho is an essential protein in many bacteria. Rho is an ATP-dependent RNA helicase that accounts for 20-50% of termination events in Escherichia coli and plays an important role in preventing pervasive transcription that otherwise might be lethal to bacteria. Rho termination usually occurs in the 3’ UTRs and non-coding regions of the DNA, and the actively transcribing RNA is believed to be protected by translating ribosomes from Rho termination since both transcription and translation are a simultaneous process in prokaryotes. However, in reality, Rho terminates transcription of some protein-coding genes, and fails to terminate transcription of …

Sex Differences In Context Fear Neural Circuitry And Behavior Across Development, Lorianna Marie Colon
Legacy Theses & Dissertations (2009 - 2024)
Over the span of five decades, staggering progress has been made in elucidating the neural circuits involved in fear learning. Research focused on rodents and healthy individuals has been instrumental in elucidating how abnormalities in this circuitry may lend to increased prevalence of various fear and anxiety disorders. However, many of the key findings concerning the acquisition, storage, and retrieval of fear memories have been established in the adult male rodent, leaving a gap in knowledge as to whether females and developing animals process fear and stress inducing information in a similar fashion. Compared to men, women are twice as …

Empowering Melatonin Therapeutics With Drosophila Models, Cassandra Millet-Boureima, Caroline C. Ennis, Jurnee Jamison, Shana Mcsweeney, Anna Park, Chiara Gamberi
Biology
Melatonin functions as a central regulator of cell and organismal function as well as a neurohormone involved in several processes, e.g., the regulation of the circadian rhythm, sleep, aging, oxidative response, and more. As such, it holds immense pharmacological potential. Receptor-mediated melatonin function mainly occurs through MT1 and MT2, conserved amongst mammals. Other melatonin-binding proteins exist. Non-receptor-mediated activities involve regulating the mitochondrial function and antioxidant cascade, which are frequently affected by normal aging as well as disease. Several pathologies display diseased or dysfunctional mitochondria, suggesting melatonin may be used therapeutically. Drosophila models have extensively been employed to study disease pathogenesis …
Adverse Childhood Experiences And Social Determinants Of Health In Relation To Pediatric Intensive Care Unit Admissions, Kasey Davis
The Eleanor Mann School of Nursing Undergraduate Honors Theses
This literature review was conducted in order to examine the correlation between adverse childhood experiences (ACEs), social determinants of health and pediatric intensive care unit (PICU) admissions. Oftentimes, those admitted as patients in the PICU come from homes in which social determinants of health, such as poverty and lack of access to healthcare, and ACEs, such as abuse and neglect, are prevalent. Systematic studies of existing literature and experience in the hospital setting revealed the ways in which these conditions often cause children to reach such poor health conditions that they must be admitted to the PICU. In order to …
Implementing 2nd Milk’S Use Of The World Health Organization Standardized Growth Charts To Better Track The Growth And Development Of The Malnourished And Orphaned Infants In Their Supplemental Nutrition Program In Malawi, Allie Wycoff

McKelvey School of Engineering Graduate Student Theses & Dissertations
The advent of the industrial era was precipitated by the discovery of fossil fuels, and ushered in unprecedented changes for humanity included but not limited to the development of rapid transit and communications, improvements to food distribution and preservation, the mass production of goods, and a radical rearrangement of communities from relatively small enclaves to metropolises. With all the benefits, however, come considerable costs, especially to the global environment. Greenhouse gas emissions, built up over centuries of unregulated combustion, have precipitated a rate of global temperature change unparalleled in the 4.5 billion-year history of this planet. In order to preserve …

Aim: Biodiversity is a multidimensional property of biological communities that represents different information depending on how it is measured, but how dimensions relate to one another and under what conditions is not well understood. We explore how taxonomic, phylogenetic, and functional diversity can differ in scale-of-effect dependence and habitat-biodiversity relationships, and subsequently how spatial differences among biodiversity dimensions may arise. Location: Nebraska, United States. Taxon: Birds. Methods: Across 2016 and 2017, we conducted 2,641 point counts at 781 sites. We modeled the occupancy of 141 species using Bayesian Bernoulli-Bernoulli hierarchical logistic regressions. We calculated species richness (SR), phylogenetic diversity (PD), …
