BACKGROUND AND RATIONALE: Immune checkpoint inhibitor (ICI) therapy is an expanding therapeutic option for hepatocellular carcinoma (HCC). Antibiotics (ATB) taken prior to or early during ICI therapy can impact immunotherapy efficacy across indications; however, the effect of ATB is undefined in HCC.
METHODS: In a large international cohort of 450 ICI recipients from Europe, North America, and Asia, we categorized patients according to timing of ATB focusing on exposure within -30 to +30 days from ICI (early immunotherapy period [EIOP]). EIOP was evaluated in association with overall survival (OS), progression-free survival (PFS), and best radiologic response using RECIST 1.1 criteria. …

Many 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase-producing and oil-degrading rhizosphere actinobacteria were isolated from oil-contaminated soils in the United Arab Emirates. These actinobacterial isolates were further selected based on their ability to solubilize phosphorus, to produce siderophores, and to produce plant growth regulators (PGRs) including auxins and polyamines. The ability of the most-promising isolate #20 (Streptomyces sp.) to promote the growth of the hyper-accumulating plants “Bermuda grass” in soil polluted with Light Arabian Crude Oil (LACO) was evaluated under greenhouse conditions. Emerging evidence suggests a positive association between pesticide exposure and sporadic Parkinson's disease (PD) development. Produced water (PW) is the effluent generated during oil mining and extraction. On average, for every barrel of oil, 4 - 5 barrels of PW are generated worldwide. In UAE, 1.22 billion cubic meters of PW were generated in the year 2017. Proper management of PW is thus very important taking into account the large volumes of it being generated. Human intestinal epithelial organoids (enteroids and colonoids) are tissue cultures used for understanding the physiology of the human intestinal epithelium. Here, we explored the effect on the transcriptome of common variations in culture methods, including extracellular matrix substrate, format, tissue segment, differentiation status, and patient heterogeneity. RNA-sequencing datasets from 276 experiments performed on 37 human enteroid and colonoid lines from 29 patients were aggregated from several groups in the Texas Medical Center. RATIONALE & OBJECTIVE: Pulmonary hypertension (PH) is highly prevalent among patients with chronic kidney disease (CKD) not requiring kidney replacement therapy. We studied the associations of PH with mortality, kidney failure, as well as cardiovascular (CV) and non-CV hospitalization among Medicare beneficiaries with a CKD diagnosis.
STUDY DESIGN: Retrospective, observational study using a matched cohort design.
SETTING & PARTICIPANTS: Patients with PH (based on 2 claims within 2 years) and patients without PH matched on CKD stage from the Medicare 5% CKD sample (1996-2016).
PREDICTOR: Presence of pulmonary hypertension.
OUTCOME: Mortality, kidney failure, and all-cause, CV, and non-CV hospitalization.

Microbial Dark Matter: Culturing The Uncultured In Search Of Novel Chemotaxonomy, Sarah J. Kennedy

Although <1% of environmental isolates can be cultured, insights into the culturable fraction refine future techniques to unlock the cryptic biodiversity. This cryptic biodiversity is omnipresent within culture-independent surveys of environmental bacterial populations. Subsequently, these elusive organisms are termed “microbial dark matter” due to the intrigue surrounding their potential taxonomic and biosynthetic impacts. To this end of investigating bacterial-specific dark matter, we employed high-throughput culturing techniques to examine Gulf of Mexico sediments and sponges for chemotaxonomically important bacteria. Through stepwise implementation of permissive and selective recovery techniques, we succeeded in recovering over 150 unique bacterial isolates. We observed several isolates that pose as chemotaxonomically interesting, including several strains that belong to critical bioremediation taxa. In addition to these biotechnological-amenable isolates, we have also succeeded in culturing taxa historically attributed with antibacterial discoveries. One such isolate was a previously sponge-associated organism belonging to the rarely cultured Verrucosispora genus. Through extensive comparative genomics, we identified it as a novel species and subsequently named it Verrucosispora sioxanthis sp. nov. with the published genome serving as a resource for continued microbial dark matter investigations. Following continued passaging of the organism, we observed improved growth, biomass, and liquid growth dispersal within each increasingly sub-passaged generation of the isolate. As such, the laboratory attenuation, or domestication, of the organism warranted in-depth whole genomic analyses to generate insight into the increased laboratory sustainability. Through phenotypically-guided bioinformatics, we determined that the domestication was mediated through genome-wide selective pseudo- or non-pseudogenization of genes. As the previously sponge-associated organism evolved with the selection pressures of the static laboratory, we demonstrate that isolate effectively underwent genomic atrophy to resolve into a largely stable, sustainable laboratory-domesticated strain. Collectively, our chemotaxonomic discovery pipeline expands on the culturable bacterial biodiversity and ultimately provides a previously uncultured model organism for initial forays into resolving the microbial dark matter knowledge gap.

Mitochondria, the 'powerhouse' of eukaryotic cells, play a key role in cellular homeostasis. However, defective mitochondria increase mitochondrial ROS (mtROS) production and cell-free mitochondrial DNA (mtDNA) release, leading to increased inflammation. Mitophagy is a vital pathway, which selectively removes defective mitochondria through the process of autophagy. Thus, an impairment in the mitophagy pathway might trigger the gradual accumulation of defective mitochondria. Accumulating evidence suggest that inflammation and mitochondrial dysfunction are linked to the pathogenesis of depression. In this study, fresh Lentinula edodes was dehydrated using freeze-drying (FD), hot-air drying (HAD), and natural drying (ND), and the volatile and non-volatile flavor compounds were analyzed. The drying process changed the contents of eight-carbon compounds and resulted in a weaker “mushroom flavor” for dried L. edodes. HAD mushrooms had higher levels of cyclic sulfur compounds (56.55 µg/g) and showed a stronger typical shiitake mushroom aroma than those of fresh (7.24 µg/g), ND (0.04 µg/g), and FD mushrooms (3.90 µg/g). The levels of 5′-nucleotide increased, whereas the levels of organic acids and free amino acids decreased after the drying process. …

This review describes the development, special features and applications of silica hydridebased stationary phases for HPLC. The unique surface of this material is in contrast to ordinary, standard silica, which is the material most frequently used in modern HPLC stationary phases. The standard silica surface contains mainly silanol (Si-OH) groups, while the silica hydride surface is instead composed of silicon-hydrogen groups, which is much more stable, less reactive and delivers different chromatographic and chemical characteristics. This study measured the heart rate (HR) responses to a formation run (group run completed along a set route) performed by higher fitness (HF), moderate fitness (MF), or lower fitness (LF) custody assistant (CA) recruits. Retrospective data from 26 recruits (12 males, 14 females) were analyzed. Prior to academy training, a YMCA step test was administered. Quackgrass (Elytrigia repens (L.) Nevski) is a competitive perennial invader of pastures and hay meadows which is frequently harvested as forage in mixtures with desired forage species. Field experiments were conducted to compare quackgrass with cool-season perennial grasses grown under the same soil and climatic conditions, in terms of forage quality, productivity, and palatability. The forage quality of the hays was influenced by the grass species. The objective of the present study was to assess the influence of supplementing increasing levels of ramón (Brosimum alicastrum) foliage to Pelibuey sheep fed guinea grass (Panicum maximum) hay. Rate and extent of rumen degradation of organic matter (OM) and crude protein (CP) of ramón foliage were high. Dry matter (DM) intake of the diet was increased by the inclusion of ramón foliage. Purpose: Androgen deprivation therapy (ADT) for prostate cancer has multiple adverse effects on musculoskeletal health. This 12-month randomized controlled trial aimed to assess the effects of multicomponent exercise training combined with whey protein, calcium and vitamin D supplementation on bone mineral density (BMD), structure and strength, body composition, muscle strength, and physical function in ADT-treated men.
Methods: Seventy ADT-treated men were randomized to exercise plus supplementation (Ex + Suppl; n = 34) or usual care (control; n = 36). The efficacy of aqueous formulations of 1H-cyclopropabenzene (BC) containing different adjuvants to retard ethylene production and maintain fruit quality of Japanese plums (Prunus salicina Lindl. cvs. ‘Angeleno’, ‘Fortune’ and ‘Tegan Blue’) following 25 d and 40 d cold storage (1 °C) was evaluated.
