How To Stuff A Duck: Learning Artisan Foie Gras Production In France, Jean Lavigne Oct 2021

How To Stuff A Duck: Learning Artisan Foie Gras Production In France, Jean Lavigne

Environmental Studies Faculty Publications

I have always hated the taste of liver. Calf’s liver, chicken liver, liverwurst, any kind of pâté that contains even a small amount of liver—for me, the taste is repulsive. I was once served a rabbit’s liver at a family meal in Italy, the prize morsel that is offered to guests; I choked it down with an unconvincing “Grazie.” I never imagined I would come to like a specific kind of liver enough to prepare it myself at home, much less to spend six weeks with an artisan foie gras producer in Southwestern France learning how to produce it from …

Developmental Participation Of Vip Interneurons In Cortical Dynamics, Daniel Barson Oct 2021

Developmental Participation Of Vip Interneurons In Cortical Dynamics, Daniel Barson

Yale Graduate School of Arts and Sciences Dissertations

In the mammalian cerebral cortex, local and long-range synaptic connections are believed to relay information related a diverse array of cognitive, emotional, and physiological functions to influence the computations performed by single neurons. While the anatomical connectivity of a single neuron is static on short timescales, its functional involvement in distributed networks may fluctuate rapidly to subserve moment-to-moment changes in behavior. A sparse class of interneurons that express vasoactive intestinal polypeptide (VIP) has been hypothesized to mediate these fluctuations by modulating the gain of cortical micro-circuits in response to neuromodulatory and long-range cortico-cortical inputs. To test this hypothesis, we developed …

Drug Manufacturers, Pricing, And Ethical Obligations, Rebecca E. Wolitz Oct 2021

Drug Manufacturers, Pricing, And Ethical Obligations, Rebecca E. Wolitz

Yale Graduate School of Arts and Sciences Dissertations

High prescription drug prices in the United States and beyond impede patient care, impoverish families, and severely strain institutional budgets. Drug manufacturers routinely launch new products at high prices, increase the prices of existing products, and engage in a range of activities that thwart competition. Yet, while patients grapple with whether today is the day they eat “cat food” or default on their mortgages (let alone have their lives cut short), and health systems are compelled to make difficult choices about program cuts, many drug manufacturers enjoy highly lucrative returns. With this juxtaposition, it is thus no wonder that the …

Molecular Mechanisms Of Aging And Alzheimer’S Disease, Shannon Noble Leslie Oct 2021

Molecular Mechanisms Of Aging And Alzheimer’S Disease, Shannon Noble Leslie

Yale Graduate School of Arts and Sciences Dissertations

Alzheimer’s Disease (AD) is a major public health crisis. AD is a progressive neurodegenerative disease and is the leading cause of dementia worldwide. Currently, there are no disease-modifying drugs available for the treatment of AD. Although autosomal dominant genetic forms of the disease are very rare, they are the foundation for many models of study; in part, because the etiology of the far more common, sporadic AD, remains unknown. While the cause of most AD cases is undetermined, age is a well-known, significant risk factor. The work presented in this thesis relies on studies of non-genetically modified aging rodents and …

Synaptic Density Imaging With Sv2a Using Positron Emission Tomography: Optimization Of Reference Region Analysis And Applications In The Human Spinal Cord And The Developing Nonhuman Primate Brain, Samantha Rossano Oct 2021

Synaptic Density Imaging With Sv2a Using Positron Emission Tomography: Optimization Of Reference Region Analysis And Applications In The Human Spinal Cord And The Developing Nonhuman Primate Brain, Samantha Rossano

Yale Graduate School of Arts and Sciences Dissertations

Positron emission tomography (PET) is a functional imaging modality in which radioactive ligands are used to target physiological functions, including metabolism or receptor densities, in vivo. Once administered into a subject, the PET radioligand distributes throughout the body and undergoes radioactive decay, emitting two gamma rays 180 degree from each other. These gamma rays are detected by the PET camera as coincidence events, and the PET raw data are reconstructed into 4-dimenstional images (x, y, z, t). Dynamic PET images can be quantified by applying various kinetic models to radioactivity concentrations in a volume or region of interest (ROI) over …

Neurodevelopmental Impacts Of Prenatal Exposures To Per- And Poly-Fluoroalkyl Substances; Multiple Outcome Assessments In Childhood Age 5 To 11 Years, Jiajun Luo Oct 2021

Neurodevelopmental Impacts Of Prenatal Exposures To Per- And Poly-Fluoroalkyl Substances; Multiple Outcome Assessments In Childhood Age 5 To 11 Years, Jiajun Luo

Yale Graduate School of Arts and Sciences Dissertations

Per- and poly-fluoroalkyl substances (PFAS) are synthetic fluorine-containing chemicals that are widely used in a variety of industrial and commercial applications. Several PFAS compounds were reported to be ubiquitously detected in blood samples collected from the general population since the 1990s. Concerns are raised that PFAS exposure among pregnant women could affect offspring neurodevelopment, mainly because of two reasons: a) PFAS can transfer across the placenta during early gestation making them a direct threat to fetal brain development, and b) PFAS can interfere with maternal endocrine homeostasis during pregnancy which has been suggested to play a crucial role on offspring …

Cross-Kingdom Expression Of Synthetic Genetic Elements Promotes The Discovery Of A New Class Of Nucleotide Analogs From The Human Microbiome, Jaymin Patel Oct 2021

Cross-Kingdom Expression Of Synthetic Genetic Elements Promotes The Discovery Of A New Class Of Nucleotide Analogs From The Human Microbiome, Jaymin Patel

Yale Graduate School of Arts and Sciences Dissertations

The ability to mobilize functional genetic cargo into a versatile array of hosts is a defining challenge, as scientists increasingly reprogram diverse organisms as living therapeutics, for bioremediation, and as foundries for producing materials and molecules. However porting genetic material to diverse recipients is challenged by barriers to robust expression and mobilization. To address these challenges, we have developed an approach that integrates computer aided redesign of genetic sequences that are subsequently mobilized, expressed, and characterized cross-kingdom in diverse microbial hosts, including Gram negative and positive bacteria and yeast. Using the technology developed in this thesis, we were able to …

Axonal Spheroids And Conduction Defects In Alzheimer’S Disease: Mechanisms And Therapeutic Implications, Mengyang Zhang Oct 2021

Axonal Spheroids And Conduction Defects In Alzheimer’S Disease: Mechanisms And Therapeutic Implications, Mengyang Zhang

Yale Graduate School of Arts and Sciences Dissertations

Alzheimer’s disease (AD) is a neurodegenerative condition characterized by widespread disruption in neural connectivity. One of the major pathological hallmarks of AD is the extracellular deposition of the β-amyloid (Aβ) peptide, which is thought to trigger a cascade of events, eventually leading to cognitive decline. However, the precise mechanisms linking Aβ deposition and neural network disruption are not well understood. Extensive previous work has focused on mechanisms such as cell death and synapse loss as potential substrates for neural dysfunction. However, a significant but understudied pathology in AD is the markedly enlarged neuronal processes found around Aβ deposits, here termed …

Polymer Nanoparticles For Optimized Gene Delivery To Mucosal Tissues, Molly Grun Oct 2021

Polymer Nanoparticles For Optimized Gene Delivery To Mucosal Tissues, Molly Grun

Yale Graduate School of Arts and Sciences Dissertations

After decades of pre-clinical and clinical research, gene therapies are finally achieving clinical success. There is rapidly growing interest in using the potential of nucleic acids to develop new treatment options in diverse fields of medicine, but effective gene therapies are still limited by effective delivery systems. Localized gene delivery to mucosal tissues—which include the eyes, respiratory system, digestive system, and female reproductive tract—are being developed to treat disorders and diseases ranging from cystic fibrosis to HIV. However, agent penetration through the mucus barrier that is common to all of these tissues poses a significant challenge to delivery vector design. …

The Embryology And Evolutionary Biology Of The Tetrapod Limb Musculature, Daniel Smith Paredes Oct 2021

The Embryology And Evolutionary Biology Of The Tetrapod Limb Musculature, Daniel Smith Paredes

Yale Graduate School of Arts and Sciences Dissertations

Tetrapods came out of water in great part thanks to the evolution of fins into limbs. This transition greatly modified the skeletal structure of fins but also its muscular anatomy. Land vertebrates exhibit a wide range of morphologies and lifestyles, enabled in many cases by their different limb morphologies. Different as they might be, a wing of a bird and a flipper of a whale are composed of the same bones, and the same muscles, but while we have a more achieved understanding of the homologies of the skeletal elements, the evolutionary identities and relationships between muscles of the limb …

Cell Biology Of Axon Degeneration And Regeneration In Vivo, Chen Ding Oct 2021

Cell Biology Of Axon Degeneration And Regeneration In Vivo, Chen Ding

Yale Graduate School of Arts and Sciences Dissertations

Axons are fundamental connective units that mediate information flow in nervous systems. Axon degeneration breaks neural circuits and is a hallmark of neurodegenerative diseases. Axon regeneration has the potential to repair damaged circuits. Here I investigate the molecular and cellular mechanisms underlying both axon degeneration and regeneration in C. elegans in vivo. In the first part of the dissertation, I examine a specific type of axon degeneration that is caused by mitochondria defects. I describe a novel calsyntenin-CaMKII-Sarm1-MAPK pathway involved in this type of axon degeneration. I further demonstrate that by modulating this signaling pathway, degeneration can be dramatically delayed. …

Binary Logic: Race, Expertise, And The Persistence Of Uncertainty In American Sex Research, Beans Velocci Oct 2021

Binary Logic: Race, Expertise, And The Persistence Of Uncertainty In American Sex Research, Beans Velocci

Yale Graduate School of Arts and Sciences Dissertations

Between the mid-nineteenth and mid-twentieth centuries, American researchers tried to sort the living world by sex. Their efforts created more taxonomic problems than they solved, but as they encountered vast quantities of evidence that did not at all show an obvious or stable division between male and female bodies, they used sexual uncertainty to their professional and ideological advantage. This dissertation traces a network of scientists who built their own claims to expertise, as well as theories of racial hierarchy rooted in sexual difference, out of sexually unruly bodies. While much scholarship on the history of sex science describes the …

Toward A Media-Technological Approach To Films: Meaning-Making, Objects, And Procedures, Ido Lewit Oct 2021

Toward A Media-Technological Approach To Films: Meaning-Making, Objects, And Procedures, Ido Lewit

Yale Graduate School of Arts and Sciences Dissertations

Film is a technological medium, and as one which is also mimetic, it often includes other technologies and media among its representations. The dissertation focuses on the intermedial encounter that occurs when film hosts other media and argues that the hosted media have the potential to inflect modes of expression and determine discourse and meaning in films that incorporate them. For the purpose of studying the intermedial encounter as a generator of discourse, the dissertation concentrates on the operation of technologies and media as they are remediated through the medium of the moving image. Within the intermedial encounter the specific operations …

Piwi-Mediated Control Of Tissue-Specific Transposons Is Essential For Somatic Cell Differentiation, Danyan Li Oct 2021

Piwi-Mediated Control Of Tissue-Specific Transposons Is Essential For Somatic Cell Differentiation, Danyan Li

Yale Graduate School of Arts and Sciences Dissertations

All eukaryote genomes lie under the constant threat of transposon invasion. About 45% of the human genome is derived from transposon elements. Although the vast majority of these transposons are inactive, they still lead to a mutagenesis rate of around 1 new insertion per 20 births. Multiple mechanisms have been evolved to silence transposons, among them is the PIWI-interacting RNA (piRNA) pathway. piRNAs rely strictly on PIWI proteins for their biogenesis and function, which includes target RNA cleavage and chromatin state alterations. PIWI and piRNAs are most well known for their transposon repression function in the germline, where PIWI depletion …

Making And Debating Empires In South Asia And The Global British Empire, 1756-1799, Tiraana Bains Oct 2021

Making And Debating Empires In South Asia And The Global British Empire, 1756-1799, Tiraana Bains

Yale Graduate School of Arts and Sciences Dissertations

Why did a strong centralizing British imperial state emerge in South Asia in the eighteenth century? The partial loss of the British Empire in North America, the ferment of partisan politics in Britain and the rise of powerful regional South Asian states during this period amply demonstrate that such a transformation was not inevitable. This dissertation argues that continuous and multi-sited contestation drove British imperial state-building in South Asia. The formation of a centralizing British imperial state in South Asia was not only violent and coercive but, akin to the concurrent emergence of the United States of America and revolutionary …

The Politics Of Play: Digital Youth, New Media, And Social Movement In Contemporary Vietnam, Tri Phuong Oct 2021

The Politics Of Play: Digital Youth, New Media, And Social Movement In Contemporary Vietnam, Tri Phuong

Yale Graduate School of Arts and Sciences Dissertations

Abstract The Politics of Play:Digital Youth, New Media, and Social Movement in Contemporary Vietnam Tri Mac Phuong 2021 This dissertation examines the role of media and the mediascape of contemporary Vietnam some 40 years after national reunification as provocation, palimpsest, and portent to a number of citizen-subject formations, sociopolitical negotiations, and cultural upheavals. In particular, I look at how the Internet and related digital technologies influence urban youth’s civic life, communal ties, and social imaginations, and how these so-called new media intersect and interact with older forms of media at particular conjunctures to produce new values, beliefs, habits, functions, and …

Pictures As Mirrors For Shah Tahmasb: How Manuscript Paintings Shaped Court Culture In Early Modern Iran, Selin Unluonen Oct 2021

Pictures As Mirrors For Shah Tahmasb: How Manuscript Paintings Shaped Court Culture In Early Modern Iran, Selin Unluonen

Yale Graduate School of Arts and Sciences Dissertations

This dissertation investigates how sixteenth-century manuscript paintings shaped courtly identity and behavior at a time of sweeping political change in Iran. During the first half of the sixteenth century, rulers of the new Safavid dynasty in Iran commissioned richly illustrated manuscripts. Turning to a corpus of paintings from a royal copy of the medieval Persian epic Khamsa, this study examines these paintings in light of the numerous extratextual inscriptions incorporated into the pictorial world (an unstudied body of evidence despite the book’s renown), and puts them in dialogue with autobiographical writings, court chronicles, diplomatic letters, and literary anthologies from the …

Organized Dynamics Of Neonatal Visual Structures In The Absence Of Retinal Input, Maria Sol Bernardez Sarria Oct 2021

Organized Dynamics Of Neonatal Visual Structures In The Absence Of Retinal Input, Maria Sol Bernardez Sarria

Yale Graduate School of Arts and Sciences Dissertations

This dissertation features my Ph.D work, under the supervision of Dr. Michael Crair, on spontaneous brain dynamics in the neonatal visual system. Early in mammalian development, spontaneous correlated bursts of activity in the retina, called retinal waves, dominate the visual pathway. Because diverse features of visual circuits are present by eye-opening, a leading hypothesis states that retinal waves are key for the establishment of these circuits. Given the robustness of retinal input, and the immaturity of most cortical and long-range networks, the field has maintained a retina-centric perspective on the development of the visual system. However, whether non-retina-driven activity is …

System View Of The Talent Gap In The Commercial Property And Casualty Insurance Sector: A Pre-Mortem, Bibi R. Khan Oct 2021

System View Of The Talent Gap In The Commercial Property And Casualty Insurance Sector: A Pre-Mortem, Bibi R. Khan

Full-Text Theses & Dissertations

The commercial property and casualty insurance industry play an essential role by providing critical strategic benefits to society and fueling the economy through risk transfer. The literature review concludes that the industry faces a looming talent crisis, and although the industry leaders are aware of the problem, there is no apparent strategy to address it. Therefore, ascertaining strategies to address the problem requires an understanding of the current state of the property and casualty system.

This study applied system thinking methodology and interactive planning method of intervention to understand how and why the system operates the way it currently does, …

Supraspinatus-To-Glenoid Contact Occurs During Standardized Overhead Reaching Motion, Gaura Saini, Rebekah L. Lawrence, Justin L. Staker, Jonathan P. Braman, Paula M. Ludewig Oct 2021

Supraspinatus-To-Glenoid Contact Occurs During Standardized Overhead Reaching Motion, Gaura Saini, Rebekah L. Lawrence, Justin L. Staker, Jonathan P. Braman, Paula M. Ludewig

Orthopedics Articles

Background: Rotator cuff tears may result from repeated mechanical deformation of the cuff tendons, and internal impingement of the supraspinatus tendon against the glenoid is one such proposed mechanism of deformation.

Purpose: To (1) describe the changing proximity of the supraspinatus tendon to the glenoid during a simulated overhead reaching task and (2) determine the relationship between scapular morphology and this proximity. Additionally, the patterns of supraspinatus-to-glenoid proximity were compared with previously described patterns of supraspinatus-to-coracoacromial arch proximity.

Study Design: Descriptive laboratory study.

Methods: Shoulder models were created from magnetic resonance images of 20 participants. Standardized kinematics were imposed on …
